  • To investigate antitumor components of Korean higher fungi, the carpophores of Ramaria formosa $(Fr.)Qu\acute{e}l$. were collected in Gang Won Province and extracted with hot water. The extract was concentrated and precipitated by four volumes of ethanol. The precipitate was centrifugated and purified by dialyzing through Visking tube and a protein-bound polysaccharide fraction was obtained. The fraction was tested for antitumor activity against sarcoma 180 implanted in mice. The tumor inhibition ratio of the fraction was 66% in the dose of 50mg/kg/day for the period of ten days. The tumor in two of the eight mice was completely regressed. The components of the fraction were found to be a polysaccharide and a protein. The chemical analysis of the fraction showed that the polysaccharide moiety consisted of four monosaccharides and that the protein moiety contained fourteen amino acids.

Physiological Effects and Utilization of Corbicula elatior Products -Effect of Cockle Extracts on Carcinogen-induced Cytotoxicity and Immune REsponse RElated to Its Antitumor Activity- (재첩가공품의 생리학적 특성과 이용 -재첩추출물의 항암효과와 면역활성증강 효과-)

  • Inhibitory effects of cockle extracts on carcinogen-induced cytotoxicity in C3H/10T1/2 cells were studied. Soup (62$\mu\textrm{g}$/mL), solubility (28$\mu\textrm{g}$/mL) and liposolubility (9 $\mu\textrm{g}$/mL) of the cockle inhibited 3-methyl-cholanthrene(MCA)-induced cytotoxicity in C3H/10T1/2 cells by 53 and 94%, respectively. These results suggest that the extracts cockle might have anticarcinogen-induced cytotoxicity of C3H/10T1/2 cells. The effects of cockle extracts on the immune response related to its antitumor activity in vitro and in vivo were investigated. The cockle extracts showed a direct cytotoxic effect on sarcoma-180 cells, tumor cells in vitro. Soup (0.49 mg/mL), solubility (0.11 mg/mL) and liposolubiliy (0.05 mg/mL) of the cockle markedly decreased the total numbers of sarcoma-180 cells, but not their viability. The phagocytic acitivity of peritoneal macrophage of mice was significantly augmented by these extracts of the cockle compared with that of control in vivo. These extracts also raised the phagocuytic index, indicating that the number of phagocytize dmicrobes per macrophage increased. Thus, cockle extracts might show a antitumor activity by enhancing the phagocytic cell activities.

Changes of Lectin Activity of Kidney Beans by floating and Fermentation (강낭콩의 열처리 및 발효에 의한 렉틴의 활성변화)

  • Synthesis of new active protein was investigated by heat-treatment and fermentation of kidney beans with Bacillus subtilis ATCC 51189. The amount of water-soluble protein in raw kidney bean (raw protein, RP) was greatly reduced by heating (heated protein, HP) and several new amino acids were synthesized by fermentation. The molecular weights of proteins determined by SDS-PAGE were 118 kDa for RP and a new band of 18.0 kDa For protein (fermented protein, FP) in kidney beans heated and fermented with B. subtilis ATCC 51189. Hemagglutinating activities of RP, HP and FP were 128 HU, 4 HU and 32 HU respectively. Both of RP and FP showed anticancer activity against stomach cancer cell line (SNU-1) at 50 $\mu\textrm{g}$g/mL and lymphocyte stimulating activity at 1 $\mu\textrm{g}$/mL, and stimulated PBMC to secrete IFN-${\gamma}$ and IL-12. However, HP did not show any kinds of activities. Taken together, these results suggested that lectin in kidney beans was destroyed by heating, hut new active lectin-like Protein was derived by fermentation with B. subtilis ATCC 51189.

Immuno-potentiating and Antitumor Effects against Mouse Sarcoma 180 by Crude Polysaccharides Extracted from Fruiting Body of Russula rosacea (장미무당버섯(Russula rosacea)의 자실체에서 추출한 조다당류의 생쥐 Sarcoma 180에 대한 면역증강 및 항암 효과)

  • Russula rosacea, one of edible and medicinal mushroom belonging Agaricales of Basidiomycota, has been known to have good inhibitory effects on sarcoma 180 and Ehrlich carcinoma of mice. Neutral saline soluble (0.9% NaCl), hot water soluble and methanol soluble substances (hereinafter referred to Fr. NaCl, Fr. HW and Fr. MeOH, respectively) were extracted from fruiting body of the mushroom. In vitro cytotoxicity tests, crude polysaccharides were not cytotoxic against cancer cell lines such as Sarcoma 180, HepG2, HT-29 and NIH3T3 at the concentration of $10{\sim}2000\;{\mu}g/ml$. Intraperitoneal injection with crude polysaccharides exhibited life prolongation effect of $21.4{\sim}45%$ in mice previously inoculated with Sarcoma 180. Fr. HW improved the immuno-potentiation activity of B lymphocyte by increasing the alkaline phosphatase activity by 6.8 fold compared with control at the concentration of $500\;{\mu}g/ml$. In case of Fr. NaCl, the numbers of peritoneal exudate cells and circulating leukocytes were increased by 6 and 2.6 folds at the concentration of 50 mg/kg, respectively. Therefore, the antitumor effect against mice Sarcoma 180 by Russula rosacea could be due to immunomodulating activity.

Antioxidant Activity of Dietary Fibers from Tubers and Stalks of Sweet Potato and Their Anti-cancer Effect in Human Colon Cancer (고구마 괴근과 잎자루 식이섬유의 항산화 효과 및 인간 대장암세포에 대한 항암작용)

  • Contents of the total polyphenols and flavonoids in the dietary fiber from tubers and stalks of domestic sweet potatoes were investigated. In addition, their antioxidant activity as well as the potent anti-cancer effects through the growth inhibition in human colon cancer cells (HT-29) in vitro were tested. The total flavonoids as naringin equivalents in dietary fiber from tubers and stalks of sweet potatoes were $0.5{\pm}0.001$ naringin/g extract and $2.0{\pm}0.008$ mg naringin/g extract dry basis, respectively. The amounts of the total polyphenols as gallic acid equivalents were $2.8{\pm}0.01$ mg gallic acid/g dry basis and $6.3{\pm}0.03$ mg gallic acid/g dry basis, respectively. 1,2-Diphenyi-1-picrylhydrazyl (DPPH) radical-scavenging activity of the dietary fiber from stalks was 2.4 times higher than that of the dietary fiber from tubers. Interestingly, a strong growth inhibition on HT-29 cells was observed in both dietary fibers originated from stalks and tubers of sweet potato in a dose-dependent manner. In addition, we found that the dietary fiber from tubers and stalks of sweet potato increased the gene expression of tumor suppressor p53. The great potential value in the prevention of various diseases including cancer the potential value could be confirmed through effects of the dietary fiber from tubers and stalks of sweet potato on antioxidant activity and anticancer in human colon cancer.

Differential Sensitivities of Human Multidrug-resistant Cancer Cells to BIIB021 and Modulation of Hsp90 Inhibitors by NSAIDs and Niclosamide (항암제 다제내성(MDR) 암세포의 Hsp90 저해제 BIIB021에 대한 감수성의 차이 및 NSAIDs 및 Niclosamide에 의한 Hsp90 저해제의 활성 변화)

  • The critical role of heat shock protein 90 (Hsp90) in tumorigenesis led to the development of several first- and second-generation Hsp90 inhibitors, which have demonstrated promising responses in cancers. In this study, we found second-generation Hsp90 inhibitor BIIB021-resistant multidrug-resistant (MDR) human cancer cells, although BIIB021 was shown to be active in first-generation Hsp90 inhibitor 17-allylamino-17-demethoxygeldanamycin (17-AAG)-resistant MDR cells. MCF7-MDR and HeyA8- MDR cells were more resistant to BIIB021 than their parental counterparts, indicating that BIIB021 cannot be applicable to all cancer cells expressing MDR proteins. We revealed that dimethyl-celecoxib (DMC), one of the non-steroidal anti-inflammatory drugs (NSAIDs), potentiated cytotoxicity of BIIB021 against both BIIB021-resistant and BIIB021-sensitive MDR cells. The effectiveness of NSAIDs involving celecoxib and DMC in combination with BIIB021 led to the autophagic degradation/down-regulation of mutant p53 (mutp53) that overexpressed MDR cells and the suppression of Hsp70 induction. This resulted in sensitization of MDR cells to BIIB021. Moreover, autophagy induction by sulindac sulfide, another type of NSAID, and niclosamide, an FDA-approved anthelmintic drug, potentiated 17-AAG-mediated autophagic degradation/down-regulation of mutp53 and c-Myc, client proteins of Hsp90. Therefore, our results suggest that NSAIDs and niclosamide positively enhance the anticancer activity of Hsp90 inhibitors through an autophagic pathway. They may also be new candidates for sensitizing MDR cells to Hsp90 inhibitors.

Antiproliferation Effects of Germinated-Korean Rough Rice Extract on Human Cancer Cells (한국산 발아 벼 추출물의 여러 가지 암세포주에 대한 증식 억제 효과 비교)

  • This study was conducted to investigate the effects of 70% ethanol extracts of various germinated-rough rice cultivars: ('Ilpum', 'Goami2', 'Keunnun', 'Sulgaeng', 'Baegjinju', and 'Heugkwang') on proliferation of human cancer cell lines (MKN-45, HCT116 and NCI-H460). Antiproliferation effects of rough rice on different cancer cell lines were higher in after germination than before germination. The viability of HCT-116 colon cancer cells was lowest at 18.89% in after germination of 'Heugkwang' at 1.0 mg/mL. The cell viability of MKN-45 lung cancer cells and MKN-45 stomach cancer cells were in the range of 5~10% in after germination of 'Ilpum', 'Goami2', 'Baegjinju', and 'Sulgaeng', and 'Heugkwang' at 1.0 mg/mL. These results suggest that germinated rough rice might have a potential preventive effect on human cancer cells.

Inhibitive Effect of Mouse Sarcoma 180 by Crude Polysaccharide Extracted from Fruiting Body of Aramillaria mellea (뽕나무버섯(Armillaria mellea)의 자실체에서 추출한 조다당류가 생쥐의 Sarcoma 180에 미치는 억제효과)

  • Armillaria mellea, one of edible and medicinal mushroom belonging to Tricholomataceae of Basid-iomycota, has been known to have outstanding inhibitive effects on the sarcoma 180 and Erhrlich carcinoma of mice. Neutral salt soluble (0.9% NaCl), hot water soluble and methanol soluble substances (hereinafter referred to Fr. NaCl, Fr, HW and Fr. MeOH, respectively) were extracted from the mushroom. In vitro cytotoxicity tests, crude polysaccharide were not cytotoxic against cancer cell lines such as NIH3T3 and Sarcoma 180 at the concentration of $1000{\mu}g/ml$. Intraperitoneal injection with crude polysaccharides exhibited life prolongation effect of $60{\sim}67.5%$ in mice inoculated with Sarcoma 180, respectively. Fr, NaCl improved the immunopotentiation activity of B lymphocyte by increasing the alkaline phosphatase activity by $1.8{\sim}3.0$ folds, respectively. In case of Fr. NaCl, the numbers of peritoneal exudate cells and circulating leukocytes were increased by 10 and 2 folds, respectively.

The Immuno-Modulatory and Antitumor Effects of Crude Polysaccharides Extracted from Daedaleopsis tricolor (삼색도장버섯(Daedaleopsis tricolor)에서 추출한 조다당류의 면역 활성 및 항암 효과)

  • Neutral salt soluble [0.9% NaCl (Fr. NaCl)], hot water soluble (Fr. HW) and methanol soluble (Fr. MeOH) materials were extracted from Daedaleopsis tricolor. In vitro cytotoxicity tests, Fr. HW was not cytotoxic against cancer cell lines such as Sarcoma 180, HepG2 and HT-29 at the concentration of $0{\sim}2,000\;{\mu}g/ml$, while Fr. NaCl and Fr. MeOH were cytotoxic to the cell lines. Intraperitoneal injection with Fr. NaCl showed antitumor effect with life prolongation of 77.4% in mice inoculated with Sarcoma 180. Fr. NnCl and Fr. HW improved proliferation of spleen cells and the immunopotentiation activity of B lymphocyte by increasing spleen cells and the alkaline phosphatase activity by $1.7{\sim}2.4\;and\;2.2{\sim}8.7$ folds, respectively. Fr, NaCl generated $90\;{\mu}M$ of nitric oxide (NO) when cultured with RAW 264.7 at the concentration of $50\;{\mu}g/ml$, while lipopolysaccharide, a positive control, produced $79{\mu}M$. Intraperitoneal injection with Fr. NaCl (50 mg/kg body weight) increased the numbers of peritoneal exudate cells and circulating leukocytes by 10 folds and two folds, respectively, than in the control group. The antitumor effect of D, tricolor was likely due to immunopotentiation activity.

Inhibitory Effects of S-allylcysteine on Cell Proliferation of Human Cervical Cancer Cell Line, HeLa (S-allylcysteine의 자궁경부암세포주 HeLa에 대한 세포증식 억제효과)

  • S-allylcysteine (SAC) is a water-soluble organosulfur compound abundant in the aged garlic extract and has been drawing attention as a diet-derived alternative agent not only for the effects of anti-oxidation and anti-inflammation but also for the prevention and treatment of various types of cancer. However, there is no report about the anticancer effects of SAC on cervical cancer cells. The aim of this study was to analyze the inhibitory effects of SAC on cell proliferation of cervical cancer cell line, HeLa and to examine its effects on the apoptosis and cell cycle as the cellular mechanisms of anti-proliferation. For this, we examined effects of different concentrations of SAC on cell proliferation according to treatment periods. Treatment with SAC not only induced morphological changes but also resulted in the reduction of cell viability and the inhibition of concentration- and time-dependant cell proliferation of HeLa. Furthermore, SAC also induced fragmentation of DNA in both DNA fragmentation and TUNEL assays, and induced cell cycle arrest at the G2/M phase in cell cycle analysis. These results suggest that SAC inhibits proliferation of HeLa at least in part through the induction of apoptosis and the cell cycle arrest.
