Analysis of Immunomodulating Activities in Methanol Extracts from Several Kinds of Grapes (포도 품종별 메탄올 추출물로부터 면역활성 분석)

  • Fruits and vegetable extracts are well-known as healthy foods. Such foods have been used as herbal medicines or traditional therapies for centuries. To assess biological activities in grapes, we examined the immunomodulating activities of water extracts from four kinds of grapes (cultivars Kyoho, Delaware, Campbell, and Niagara). We explored possible antioxidant and anticancer activities using antioxidant assays such as the 1,1-diphenyl-2-picrylhydrazyl (DPPH) reduction assay, the ferric iron reducing ability of plasma (FRAP) assay, a cell proliferation assay, an NO inhibition assay, a wound healing assay, and an IL-4/IL-13 elicitation assay. Methanol extracts of grapes were tested. The results showed that each grape extract had potent antioxidant activities. The grape extracts increased cell proliferation and NO production activities in tumor cell lines. IL-4 and IL-13 cytokine levels were decreased in mouse primary spleen cells by treatment with any extract. These results suggest that grape extracts can be used as biomaterials with immunomodulating activities.

Evaluation of Neoadjuvant Chemotherapy Effect in Osteosarcoma (골육종에서 술전 항암화학요법의 효과 판정)

  • Purpose: Various diagnostic imaging modalities have been used to evaluate the effect of neoadjuvant chemotherapy for osteosarcoma early and noninvasively. We evaluated the effectiveness of imaging studies of plain radiographs and positron-emission tomography/computed tomography (PET/CT) in predicting neoadjuvant chemotherapy effect for osteosarcoma and tried to establish a general principle in interpretation of PET/CT parameters. Materials and Methods: Eighteen patients who underwent two cycles of neoadjuvant chemotherapy and surgical excision for osteosarcoma were enrolled. There were 13 males and 5 females, with a median age of 19 (11-63) years. Fifteen patients of 18 had the American Joint Committe on Cancer (AJCC) stage IIB. They had plain radiographs and PET/CT before and after neoadjuvant chemotherapy. The resected tumor specimens were pathologically examined to determine histological response grade using a conventional mapping method. Statistical analysis was performed to evaluate the correlation between histopathological necrosis rate, and radiographic finding category, post-chemotherapy maximum standardized uptake value (SUVmax), average standardized uptake value and metabolic tumor volume (MTV) as well as reduction rates of them. Results: Eight patients were good responders to neoadjuvant chemotherapy based on histological evaluation. Median SUVmax reduction rate was 73 (23-77) % in good responders and 42 (-32-76) % in poor responders. Median MTV reduction rate was 93.5 (62-99) % in good responders and 46 (-81-100) % in poor responders. While radiographic finding category was not different according to histological response (p=1.0), SUVmax reduction rate was significantly different (p=0.041). Difference in MTV reduction rates approached statistical significance as well (p=0.071). Conclusion: While radiographic finding category was not reliable to assess neoadjuvant chemotherapy effect for osteosarcoma, reduction rate of SUVmax was a useful indicator in this study. As parameters of PET/CT can be influenced by various factors of settings, different centers have to make an effort to establish their own standard of judgement with reference of previous studies.

  • 지난 3월 12일부터 나흘간 미국 올란도 오렌지카운티에서 열린 미국 정형외과연구학회 연례학술대회는 25개국에서 1천8백여명이 참석하여 1천1백여편의 논문이 발표되었다. 정형외과의 모든 기초연구 분야의 논문들이 발표되는 이 학술대회에서 필자는 악성골종양에 항암제의 효과를 높이기 위한 방법의 하나로 항암제를 뼈의 약성종양수술에 흔히 사용되는 골시멘트에 혼합하여 약성종양세포 성장이 억제되는 내용의 논문을 발표했다.

Synthesis of Novel Pyrazolinecarbothioamide and Evaluation of Its Anti-Cancer Activity (새로운 피라졸린카르보티오아미드 화합물의 합성과 항암효과)

  • Novel pyrazolinecarbothioamide (5) was synthesized from chalcone (3) which was prepared from 2'-hydroxy-1'-acetonaphthone (1) and 2-methoxy benzaldehyde (2). Treatment of pyrazolinecarbothioamide (5) on HCT116 cancer cell showed upregulation of p21 and downregulation of cyclin D1 protein. Flowcytometer analysis revealed that pyrazolinecarbothioamide (5) controls the expression of cell cycle regulatory proteins, which blocks cell cycle progression of HCT116 cancer cell at the G1 phase.

Synthesis of Salens and Their Cobalt Complexes from Meso-1,2-Diamine and Their Anti-Cancer Property (메소-1,2-디아민을 이용한 Salen-코발트 착화합물의 합성과 항암효과)

  • New salens (3) and their Cobalt complexes (4) were prepared from meso-1,2-bis(ortho-hydroxyphenyl)-1,2-diaminoethane (1) and substituted salicylic aldehydes (2). In contrast to symmetric structure of salen ligand (3), salen-Co(III) complexes (4) showed dissymmetric molecular structure due to participation of three hydroxyl groups in complex formation. One of the salens (3b) revealed decrease in Cyclin D1 expression, which represents anti-cancer property.

항암제로서 새로운 Anthracycline 유도체의 합성

  • 1960년대 이후 광범위한 항암제로서 그 임상효과가 탁월한 물질로 알려진 Anthracycline계 항생물질이 화학구조상 B환의 Hydroquinone형에 의한 생체내 산화환원 반응으로 이해 심근 독성에 영향을 준다고 알려져 있다. 이에 본 연구실에서는 Anthracycline의 Aqlycone 부분 12번 탄소의 전자친화력을 증가시키는 11번 탄소의 Hydroxy group이 제거되고 9번 탄소의 Hydroxy group을 Amine으로 대체함과 동시에 4번 탄소의 Methoxy group이 제거된 보다 독성이 적을 것이라 생각되는 Anthracycline의 새로운 Aglycone 유도체를 Design 하여 전합성 하였다.

  • Nucleoside계통의 유도체를 합성하여 체내 DNA 또는 RNA 합성대사에 영향을 줌으로써 궁극적으로 항암성 또는 항 virus성 작용을 나타내는 화합물을 얻고자 하였다. 본 연구에서는 천연의 nucleoside 구조중에서 특히 당부분에 변형을 준 일련의 acyclonucleoside 유도체들을 합성하였다. 이는 그간 많이 연구되어온acyclonucleoside 계열인 acyclovir계와는 달리 ribose 당부분을 $C_1$에서 $C_{5}$까지 거리가 유사한 acycloalkyl 결합형태로 결합하고 3'위치에 변형을 가져온 pyrimidine계 유도체를 합성하고자 일련의 반응을 시도하였다. 목적하는 화합물 계열을 얻지 못하고, 몇종류의 관련 유도체들을 분리하여 구조를 규명하고자 하였고, 이들중 일부에 대해 in vitro L1210 cell 증식억제효과를 검색하여 그 결과를 보고하고자 한다.

  • Novel pyrazoline (4) was synthesized from chalcone (3) which was prepared from 2'-hydroxy-l'acetonaphthone (1) and 4-methoxy benzaldehyde (2). Pyrazoline (4) forms resonance assisted hydrogen bond between naphthol hydroxyl group and imine nitrogen in a pyrazoline ring. Pyrazoline (4) shows Poly ADP-ribose Polymerase (PARP) cleavage ability as a proof of apoptosis in cancer cell, which reveals its anti-cancer property.

  • 1. Extracts of the leaves of Eurya emarginata markedly inhibited the growth of leukemia cells such as HL-60, KG-1, U937, K562, and Jurkat. 2. When HL-60 cells were treated with the extract, DNA fragmentation, morphological changes and sub-G1 hypodiploid cells were observed. The expressions of c-myc mRNA were also dramatically decreased. 3. Cornoside and Eutigoside were isolated from the leaves of Eurya emarginata. 4. Eutigoside induced apoptosis through down-regulation of Bcl-2 expression and activation of caspases.
