Effects of Hot-Water Extract of Mulberry Leaf Tea Fermented by Monascus pilosus on Body Weight and Hepatic Antioxidant Enzyme Activities in Mouse Fed a Normal Diet (정상식이 마우스의 체중과 간 조직 항산화계 효소활성에 미치는 Monascus pilosus 발효 뽕잎차 열수추출물의 효과)

  • In this study, to evaluate the anti-obesity effects of mulberry leaf tea and its fermented product by Monascus pilosus, we investigated body and organ weight, blood and liver biomarkers in mice fed 1% tea infusions instead of water for 8 weeks. Mice were divided into three groups such as a normal control (NC), unfermented mulberry leaf tea infusion (UMI) and fermented mulberry leaf tea infusion (FMI). Although it is not significant, tea infusion groups showed reduction of body weight gains compared with NC group. Moreover, contents of LDL-cholesterol and lipid peroxide (LPO), altherogenic index, and xanthin oxidase (XO) activity were significantly decreased, and glutathione S-transferase (GST) activity was significantly elevated. The results from this study suggested that UMI and FMI may have an anti-obesity activity, upregulate antioxidant enzymes and reduce levels of oxidants related to liver damage.

Hypoglycemic and Antioxidant Effect of Dietary Hamcho Powder in Streptozotocin-induced Diabetic Rats (Streptozotocin-유발당뇨쥐에서 함초첨가 식이의 항당뇨 및 항산화 효과)

  • Male Sprague-Bawler rats were blocked into four groups which were normal rats fed control diet (NC) diabetic rats fed control diet (DC), normal rats fed Hamcho powder diet (NH), and diabetic rats fed Hamcho powder diet (DH). Diabetes was induced by single injection of streptozotocin (60 mg/kg B.W. i.p.). The animals were fed ad libitum for 5 weeks. Malondialdehyde (MDA), glucose 6-phosphtase (Gspase), glutathione S-transferase (GST) glutathione Peroxidase (GPx), and glutathione reductase (GR) activities were measured in the homogenates of liver and kidney, and total lipid, total cholesterol, triglyceride, and HDL-cholesterol concentrations in the blood serum. Food and water intakes were markedly higher in diabetic groups than those of normal groups and were not significantly decreased by Hamcho powder supplementation, But, FER (Feed efficiency ratio) of DH Brood was higher than that of U group. Total cholesterol level of DH group was decreased in the second and third week, and the weekly change of blood sugar was also decreased in the 5th week. Dietary Hamcho intake showed 41.2% of hypoglycemic effect in diabetics rats. Levels of total lipid and triglycerides of DH group were lower than those of DC group. Hepatic GR activity of DH group was higher than those of other groups. However, renal GR activity was lower than those of other groups. Hepatic G6Pase activity was significantly high in DH group and reduced by Hamcho powder supplementation. GST was reduced by Hancho diet in diabetic rats. In conclusion Hamcho supplementation decreased serum lipid and glucose concentration in STZ-induced diabetic rats and this effects of Hamcho might exert antidiabetic effect of Hamcho powder diet.

Effect of Enzymatic Hydrolysate of Hamcho (Salicomia herbacea) on Antioxidative Defense System in Rats Fed High Cholesterol Diet (함초(Salicornia herbacea)의 효소적 가수분해물이 고콜레스테롤 식이 흰쥐의 항산화방어계에 미치는 영향)

  • The purpose of the present study was to investigate the effect of enzymatic hydrolysate (extract) of Salicomia herbacea L. (Korean name: Hamcho) on antioxidative defense system in rats fed high cholesterol diet. Rats were divided into six experimental groups which are composed of normal diet group, normal diet group supplemented with 2% Hamcho extract, high cholesterol diet group, high cholesterol diet groups supplemented with 1%, 2% and 4% Hamcho extracts. The activity of serum glutamate oxaloacetae transaminase in rats was not different among all experimental groups, while the activity of glutamate pyruvate transaminase in groups supplemented with Hamcho extract was significantly lower than that of high cholesterol control group. Supplementation of Hamcho extract (SHE) to the high cholesterol fed rats resulted in increased activities of hepatic superoxide dismutase and glutathione peioxidase. However, there was no significant difference in the activity of hepatic catalase among all experimental groups. SHE also resulted in decreased levels of hepatic thiobarbituric acid reactive substances and mitochondrial carbonyl values. Those effects were higher to some extent in 2% and 4% Hamcho extract groups than those of high cholesterol control group. These results suggest that enzymatic hydrolysate of Hamcho may reduce oxidative damage by activation of antioxidative defense system in rats fed high cholesterol diets.

Regulation of Quinone Reductase Activity in Mice by Dehydroglyasperin C Isolated from Licorice (감초에서 분리된 데하드로글라이아스페린 C에 의한 마우스 모델계에서 quinone reductase 활성의 조절)

  • Licorice, Glycyrrhizae radix, is one of the oldest and most frequently used botanicals in the oriental medicine. Our previous study showed that dehydrolyasperin C (DGC) isolated from licorice had antioxidant activity and induced phase 2 detoxifying enzymes in mouse hepatoma cells. Therefore, this study was conducted to investigate the effect of exposure time to DGC on quinone reductase (QR), one of the anticarcinogenic biomarkers, and antioxidant potential of plasma using animal model. ICR mice were divided into 7 groups, in which mice in each group were injected with DGC (5 mg/kg b.w.) for 0, 2, 4, 6, 8, 12, 24 hours respectively. Following the treatment the organs including liver, kidney, lung, stomach, large intestine, small and large intestines were collected and subjected to QR activity assay, western blotting, and FRAP assay. Exposure to DGC caused a significant induction of QR activity in stomach and large intestine of mice. Ferric reducing activity of plasma, a typical biomarker for antioxidative potentialshowed that DGC improved antioxidant potential in mice. However, no significant effect of DGC was observed in the other organs.

Effects of Scopoletin Supplementation on Insulin Resistance and Antioxidant Defense System in Chronic Alcohol-Fed Rats (Scopoletin 보충이 만성 알코올을 급여한 흰쥐의 인슐린저항성 및 항산화방어계에 미치는 영향)

  • This study investigated the effects of scopoletin (6-methoxy-7-hydroxycoumarin) supplementation on insulin resistance and the antioxidant defense system in chronic alcohol-fed rats. Rats were fed a Lieber-Decarli liquid diet containing 5% ethanol with or without two doses of scopoletin (0.01 and 0.05 g/L) for 8 weeks. Pair-fed rats received an isocaloric carbohydrate liquid diet. Chronic alcohol did not affect fasting serum glucose levels, although it induced glucose intolerance and hyperinsulinemia compared with the pair-fed group and led to insulin resistance. Both doses of scopoletin similarly improved glucose intolerance, serum insulin level, and insulin resistance. Scopoletin supplementation significantly activated phosphatidyl inositol 3-kinase, which was inhibited by chronic alcohol. Two doses of scopoletin up-regulated hepatic mRNA expression and activity of glucokinase as well as down-regulated mRNA expression and activity of glucose-6-phosphatase compared with the alcohol control group. Both doses of scopoletin significantly reduced cytochrome P450 2E1 activity and elevated aldehyde dehydrogenase 2 activity, resulting in a lower serum acetaldehyde level compared with the alcohol control group. Chronic alcohol suppressed hepatic mRNA expression and activities of antioxidant enzymes such as superoxide dismutase, catalase, and glutathione peroxidase; however, they were reversed by scopoletin supplementation, which reduced hydrogen peroxide and lipid peroxide levels in the liver. These results indicate that dietary scopoletin attenuated chronic alcohol-induced insulin resistance and activated the antioxidant defense system through regulation of hepatic gene expression in glucose and antioxidant metabolism.

  • 사람 기관지상피세포주인 BEAS-2B에 담배연기농축액(CSC)을 처리하여 유도된 1198 세포주는 대조군 세포주인 1799에 비해 현저하게 낮은 glutathione 농도와 낮은 glutamate-cysteine ligase(GCL), glutathione peroxidase(GPx), glucose-6-phosphate dehydrogenase(G6PD), catalase 효소활성을 보였다. 두 세포주를 포도당 존재 하에서 4시간 hypoxia 처리 후 reoxygenation 하면서 시간에 따른 세포의 항산화계 활성을 측정한 결과, 1799 세포주에서는 의미 있는 변화가 관찰되지 않은 반면, 1198 세포주에서는 hypoxia 처리에 의해 glutathione의 농도 및 GSH/GSSG 비와 G6PD 활성이 감소되었고, reoxygenation 기에는 GPx, glutathione reductase(GRd), G6PD, superoxide dismutase 활성이 감소되었다. 그러나 reoxygenation 2시간 이후에는 GRd와 G6PD 활성의 회복이 관찰되었으며, 그 결과 GSH/GSSG 비율이 회복되었다. 이 실험 결과는 CSC가 능력을 현저히 저하시킬 수 있음을 보여준다. Glutathione은 hypoxia-reoxygenation에 의한 산화적 스트레스 하에서 항산화제로서의 역할뿐 아니라, 세포 내 GSH/GSSG 비의 변화를 통해 산화적 스트레스에 대한 항산화계의 적응 반응 여부를 결정하는 중요한 인자로 작용할 것으로 보여진다.

Antioxidant activity of white ginseng extracts prepared by enzyme treatment on V79-4 cells induced by oxidative stress (효소처리에 의한 백삼 저분자 화합물의 V79-4 세포주에 대한 항산화 활성)

  • This study examined the extraction yields, total phenolic compounds content and the antioxidant activities on V79-4 cells of white ginseng extracts prepared by enzyme treatment. Yields of crude extract were 29.5-76%, and total phenolic compounds content showed 0.45-2.2% according to enzyme treatments. Pectinase treatment group showed the highest values of extraction yields and total phenolic compounds content. Pectinase and a-amylase treatment groups protected V79-4 cell viability(above 50%) against $H_2O_2$-induced oxidative damage. In the result of antioxidant enzyme activity evaluation in cells, enzyme treatments did not show the significant difference of SOD activity (p>0.05). However, pectinase treatment group exhibited increased CAT and GPx activities (p>0.05). Also, pectinase and protease treatment group inhibited MDA formation (>50%) in the lipid peroxidation protection experiment.

Effect of Giant Embryonic Rice Supplementation on the Lipid Peroxide Levels and Antioxidative Enzyme Activities in the Plasma and Liver of Streptozotocin-induced Diabetic Rats (거대배아미 식이가 streptozotocin 유도 당뇨 흰쥐의 혈장과 간 조직 중 지질과산화물 농도와 항산화 효소 활성에 미치는 효과)

  • Effects on the feeds of streptozotocin-induced diabetic rats with a giant embryonic rice on lipid peroxides level and antioxidative enzyme activites in plasma and liver tissues were investigated. Along with the experimental periods, all animals in diabetic groups had a lower increase rate in body weight than the normal control group. A giant embryonic rice-fed group showed a inhibition in the decrease of body weight, and a increase in feed intake compared to the normal control group. The organ weights of the diabetic control group were heavier than those of the normal control while rice-fed groups including the giant embryonic rice-fed group were found to have lower organ weights, and its blood sugar level was found to be lower than those of the normal group. Lipid peroxides of the giant embryonic rice-fed animals showed a lower lipid peroxidation values compared to that of the diabetic control group. Plasma vitamin A and E concentrations of the diabetic control group were significantly decreased compared to the normal control while those of the giant embryonic rice-fed group were found to be significantly higher than those of the diabetic control. Of the hepatic antioxidative enzymes, SOD activity of the giant embryonic rice-fed group was higher than that of the diabetic control group. Taken these together, low lipid peroxidation values and, in contrast, high antioxidative enzyme activities were thought to be a cause for decreasing hepatic oxidative damages.

A Study on the Mechanism of Calcium Binding Inhibition of Cardiac Sarcoplasmic Reticulum by Oxygen Free Radicals (산소대사물에 의한 심장근 Sarcoplasmic reticulum의 칼슘운반 억제 기전에 관한연구)

  • Mechanism of calcium transport inhibition of cardiac sarcoplasmic reticulum (SR) by oxygen free radicals was examined. Effects of oxygen free radicals generated by xanthine/xanthine oxidase (X/XO) system on isolated porcine ventricle SR were studied with respect to its calcium binding, lipid peroxidation, SH-group content and alteration of membrane protein components. The results are as follows. 1) Calcium binding of isolated SR was markedly inhibited by X/XO. 2) During the incubation of sarcoplasmic reticulum with xanthine/xanthine oxidase, there were marked inclose in lipid peroxidation and reduction of SH-group content. 3) An antioxidant, p-phenylenediamine effectively prevented the lipid peroxidation but partially prevented the calcium binding inhibition of X/XO treated SR. 4) The reduction of SH-group content of SR treated with X/XO was partially prevented by p-phenylendiamine. 5) When modifying SH-group of SR by treatment with DTNB, the inhibition of calcium binding activity was partially prevented. 6) On gel-permeation chromatography of X/XO-treated sarcoplasmic reticulum, there was an increase of small molecular weight products, probably protein degradation products. 7) Semicarbazide, which prevents the cross-linking reaction of protein components, did not affect the calcium binding inhibition of X/XO-treated SR. From these results, it is suggested that the inhibition of calcium binding of SR by oxygen free radicals results from the consequence of multiple changes of SR components, which are lipid peroxidation, SH-group oxidation and degradation of protein components.

Effects of Coenzyme Q10 on the Antioxidant System in Blood and Liver of Laying Hens (산란계에서 Coenzyme Q10 급여가 혈액 및 간 조직의 항산화 작용에 미치는 영향)

  • The objective of this study was to investigate the effects of dietary coenzyme Q10 (CoQ10) sources on the antioxidant defense system in the blood and liver of laying hens. Thirty-six 40-wk old Lohmann Brown hens were randomly assigned to three groups based on body weight, with four cages with three layers each. Laying hens were divided into one of the following groups: control (CON), powdered CoQ10 (PCoQ, 100 mg/kg diet), and emulsified CoQ10 (ECoQ, 100 mg/kg diet). All hens were fed a control diet or a control diet supplemented with powdered or emulsified CoQ10 ad libitum for five weeks. There were no differences in body weight, weight gain, and organ weights among the treatment groups, including the liver and spleen. The blood total antioxidant power (TAP) in the ECoQ group increased (P<0.05) by approximately 2-fold compared to that in the CON group. However, there was no significant difference in blood TAP levels between the PCoQ and ECoQ groups, although a decreasing trend (P<0.13) was observed for levels of TAP in the ECoQ group. The mRNA expression and specific activities of superoxide dismutase, glutathione peroxidase, and catalase in the liver were not affected by dietary CoQ10 or type of CoQ10. However, hepatic lipid peroxidation in the ECoQ group was lower (P<0.05) than in the CON group. In conclusion, emulsified CoQ10 increased blood TAP and decreased hepatic lipid peroxidation without affecting antioxidant enzymes, suggesting that emulsified CoQ10 might be more applicable as an active antioxidant supplement than powdered type in laying hens.
