Effects of Bombycis corpus on Amyloid-induced Lipid Peroxidation Antioxidative Enzymes and NO Synthesis in Rat Astrocytes (흰쥐의 뇌 astrocyte에서 $amyloid-{\beta}25-35$로 유발된 지질의 과산화와 항산화 효소계 및 NO 생성에 미치는 백강잠의 효과)

  • 목적 : 본 연구는 식풍해경(熄風解痙), 소산풍열(疏散風熱) 효능이 있는 백강잠이 치매에 미치는 영향을 알아보기 위하여 실험을 행하였다. 방법 : 치매 유발물질인 $amyloid{\beta}(A{\beta})$ 25-35를 흰쥐의 뇌 astrocyte에 처리한 후 대표적인 항산화 효소인 catalase, superoxide dismutase,glutathione peroxidase 및 glutathione-S-transferase의 활성 변화와 NO 생성 변화를 관찰하였다. 결과 $A{\beta}$ 25-35 처리로 catalase와 superoxide dismutase 활성이 현저히 감소하였으나 백강잠을 처리한 경우는 이들 효소 활성이 크게 증가하였다. 그리고, $A{\beta}$ 25-35의 농도에 의존적으로 증가된 NO 생성은 백강잠의 농도에 의존적으로 유의성 있게 억제되는 것으로 나타났다. 결론 : 백강잠은 항산화계 효소의 활성화 및 $A{\beta}$처리와 같은 치매유발 물질의 독성에 대한 적응능력 향상을 통하여 astrocyte를 보호하는 효능을 가지는 것으로 사료되며, 아울러 노인성 치매 등 임상적 응용에 그 효과가 기대된다.

Effect of Styrene on Hepatic Activities of Antioxidant Enzymes in Rats (스티렌이 흰쥐의 간 조직 중 항산화계 효소 활성에 미치는 영향)

  • Styrene is a commercially important chemical used mainly in the production of raw materials and plastics. To determine the effect of styrene on hepatic activities of antioxidant enzymes, styrene was treated to Sprague-Dawley rats at 50 mg/kg, 200 mg/kg and 400 mg/kg (i.p) twice a day for 4 days. There were determined the significantly increased activities of serum AST (aspartate aminotransferase), ALT (alanine aminotransferse), and the increased content of MDA (malondialdehyde) at the dose of 400 mg/kg compared to the control. The hepatic activities of XO (xanthine oxidase) and CYPdAH (cytochrome P450 dependant aniline oxidase) in the dose of 400 mg/kg compared to the dose of 200 mg/kg were more increased, which means the excessive ROS (reactive oxygen species)s were produced during Phase I. In addition, significantly decreased were rates of the hepatic activities of GPx (glutathione peroxidase), CAT (catalase), SOD (superoxide dismutase) and GST (glutathione S-transferase) at the dose of 400 mg/kg compared to the control. And, the group at the dose of 400 mg/kg showed more significantly decreased GSH (glutathione) level than the group at the dose of 200 mg/kg. The decrease in GSH could ascribe to the toxic metabolites of styrene, such as styrene oxide. In conclusion, these results indicate that the excessive ROSs and the toxic metabolites of styrene may result in the hepatotoxicity, and be related to their imbalanced activities for antioxidant enzymes.

The Effects of Panax Ginseng Extract on Antioxidative Enzyme Activity in Ovariectomized Rats (인삼 추출물이 난소를 절제한 백서의 항산화효소 활성에 미치는 영향)

  • In order to observe the bioactivity of ovariectomized rats, nonovariertoized (sham) group, ovariectomized (Ovx) group, ovariectomized ginseng total saponin (GTS)-treated (Ovx+ GTS) group and ovariectomized ginseng water extract (GW)-treated (Ovx+CW) group were made. We measured AST (L-aspartate aminotransferase) and ALT (L-alanin aminotransferase) in sera, and MDA (malondialdehyde:lipid peroxidation), SOD (superoxide dismutase), catalase, total-glutathione (GSH + GSSG) and GPx (glutathione peroxidase) in liver tissue total homogenates of rat. AST activity of serum in Ovx group was 2.11 times increased, but ALT activity was not changed compared to Sham group. In AST activity, they tend to decrease significantly in each substance such as GTS and GW administered group. Lipidperoxides of each fraction in Ovx group were highly increased compared to Sham group. Extracts of ginseng-treated group markedly inhibited lipid peroxidation by 62% ∼72%. And as the result of the measurements of SOD, catalase, total-glutathione and GPx which are antioxidant enzyme, antioxidant enzymes in Ovx group much lower than in Sham group. But they were significantly increased in each substance such as GTS and GW, administered group. Based on the results, it is supposed that more produced free radicals decreased antioxidant enzyme. And it is also thought that extracts of ginseng can inhibit aging by reducing antioxidant enzyme.

Activities of Antioxidant Enzymes in Serum of Rats Feeding Soybean Extract (콩 추출물 투여가 흰쥐 혈청의 항산화효소 활성에 미치는 영향)

  • This study was conducted to determine antioxidative enzyme activity in serum of rats as affected by soybean cultivar, extract feeding concentration and fat dietary. In all cultivars, activities of antioxidant enzymes in treatment of various fat compositions and cholesterol followed by feeding soy-extracts were higher than in nonfeeding control, however, no significant differences existed in statistically. The activities of SOD and GSHpx in serum of rats when fed with various fat dietary were higher in Geomjeongkong 1 than in Tawonkong and Hwangkeumkong. Enzyme activities of SOD and GSHpx with each fat compositions in treatment soy-extracts was no significant differences statistically. There was significant differences among cultivars and concentrations of soy-extracts in interaction effect. Also, the activities of GSHpx was significant differences among cultivars and concentrations of soy-extracts and fat composition in interaction effect.

Responses in Hepatic Xenobiotic Metabolizing and Antioxidant Enzymes in Javelin Goby Acanthogobius hasta Collected at Shihwa Lake (시화호에서 채집한 풀망둑 Acanthogobius hasta의 간장 약물대사효소계 및 항산화계의 반응)

  • The aim of this study was to assess the responses of mixed function oxygenase (MFO) and antioxidative systems of feral Javelin goby, Acanthogobius hasta, caught in two sites of different pollution level in Shihwa lake, which has been a highly polluted lake by organic pollutants from nearby industrial complexes and sites. Enzymes analyzed in phase I of MFO system are cytochrome P450 (CYP), NADPH-cytochrome P450 reductase (P450R), NADH-cytochrome b5 reductase (b5R), and ethoxyresorufin deethylase (EROD). Phase II enzyme of glutathione S-transferase (GST) in MFO system was also investigated. Moreover, oxidative-enzyme system including catalase (CAT), glutathione reductase (GR) and total-glutathione peroxidase (GPX) activities and glutathione concentration in both of oxidized (GSSG) and reduced form (GSH) were determined. P450R, b5R, and GST activities of fish are relatively high in the polluted area, whereas hepatic EROD activity levels of fish in polluted area were lower than those of unpolluted area. CYP concentrations are not different between areas. These results indicated that feral Acanthogobius hasta were adaptive to highly polluted environment and exposed to oxidative stress in Shihwa lake.

  • Hermansky-Pudlak Syndrome (HPS) 환자에서 조기에 발생되는 폐섬유화의 원인을 알아보고자, 생쥐 HPS 모델인 ep/ep,pe/pe 돌연변이종의 폐 항산화계의 환성과 방사선에 대한 반응을 측정하였다. HPS 폐에서는 대조군에 비해 glutathione이 더 산화되어 있었고, catalase, glutathione S-transferase(GST) 등의 항산화효소의 활성이 저하되어 있었으며, 10 Gy의 방사선을 조사하였을 때, glutathione 양이 감소하였고, 대조군 폐에서 보여지는 방사선에 의한 ${\gamma}$-glutamylcysteine ligase(GCL), glutathione peroxidase(GPx) 활성의 유의성 있는 증가가 관찰되지 않았다. 이 결과로부터 HPS 환자의 폐는 항산화계 활성이 저하되어 있을 뿐 아니라, 산화적 스트레스가 가해 졌을 때 적응 반응이 매우 취약하여 산화적 환경에 노출된 폐의 병증을 유발할 수 있음을 추측할 수 있다.

Effect of Aerobic Exercise with Resistance Exercise Programs on Blood MDA and SOD, GPx Activities in Elderly Women (유산소 운동을 병행한 근 저항성 운동이 노인 여성의 혈중 MDA 및 SOD, GPx 활성에 미치는 영향)

  • The purpose of this study was to investigate the effects of aerobic exercise(folk dance) with resistance exercise(elastic band) for 12 weeks on blood MDA concentration and SOD GPx activities in the elderly women. The subjects consisted 12 elderly women between 65-75 years exercise were folk dance(HRmax 50-60% levels, 60min, two per a week) and elastic band(yellow band, 60min, two per a week) program for 12 weeks. SOD, GPx activities in the before combined exercise were significantly increased than that in after combined exercise. These results show that aerobic exercise with resistance exercise program in considered to contribute enforced of antioxidant enzyme system by increased SOD and GPx activities in elderly women.

타고난 면역반응이 활성화된 육계병아리의 생산성과 혈액 항산화계에 미치는 사료중 미역제품 수준의 영향

  • Effect of dietary brown seaweed (Undaria pinnatifida) levels on the performance, nutrients utilization, and blood anti-oxidant system was studied in broiler chicks activated innate immune response. Brown seaweed 2.0 % diet improved performance of broiler chicks and resulted in enhanced feed efficiency due to the increased NB and decreased UAN excretion significantly (P<0.05). Dietary brown seaweed reduced SOD activity in erythrocyte cytosol and enhanced peroxidase activity in plasma significantly(P<0.05). Activation of innate immune response increased SOD activity and peroxide levels in blood. The results indicated that dietary brown seaweed affected SOD and peroxidase activity and the increased performance in birds fed brown seaweed 2.0 % diet related with decreased decomposition of body protein and the change of anti-oxidant systems in blood of broiler chicks during activation of innate immune response.

  • Effects of dietary killed salmonella and vancomycin on the performance and immune response was investigated in broiler chicks inoculated with Salmonella typhimurium. During 3 week(27 d) of age, experimental diet did not affect daily gain, feed intake and feed efficiency. Dietary killed salmonella and vancomycin decreased MnSOD activity. At 7day after Salmonella typhimurium inoculation, dietary killed salmonella and vancomycin increased, daily gain and feed efficiency of broiler chicks. At 7 and 15 day after salmonella inoculation, dietary killed salmonella and vancomycin decreased erythrocyte peroxidase activity, but elevated proliferation of PBMC stimulated with LPS and supernatant IL-1 level secreted by the PBMC. The results suggested that dietary killed salmonella and vancomycin improved the performance of broiler chicks due to modulate antioxidant system and innate immune response of broiler chicks innoculated with Salmonella typhimurium.

The Effect of Kimchi on Production of Free Radicals and Anti-oxidative Enzyme Activities in the Brain of SAM (김치가 노화촉진쥐 뇌의 유리기 생성 및 항산화효소 활성에 미치는 영향)

  • This study was Performed to investigate the effect of kimchi intake on antiaging characteristics in the brain of senescence-accelerated mouse (SAM) in terms of free radical production and anti-oxidative enzymes. Two hundreds twenty SAM (20 mice) were divided into four groups and fed kimchi diet for 12 months. Experimental groups were kimchi free AIN-76 diet (control) group, Korean cabbage kimchi diet (KCK)group, 30% mustard leaf added Korean cabbage kimchi diet (MKCK) group, and mustard leaf kimchi diet (MLK) group. Concentrations of total free radical, OH radical and $H_2O_2$ of control group increased up to 123%, 262% and 174% of initial value (p<0.05) after one year. Increase in free radical production in kimchi groups due to aging was decreased by kimchi feeding. Among kimchi groups. MKCK and MLK groups showed greater inhibiting effect against free radical production than KCK. The concentration of TBARS in the bruin of control group also significantly increased up to 362% of initial value as aged (p<0.05) and production of TBARS in kimchi groups were decreased. When the activities of Cu, Zn-SOD, Mn-SOD, GSH-px and catalase of kimchi groups were compared to those of control at the same experimental period, anti-oxidative enzyme activities of kimchi groups were lower than those of control (p<0.05). But GSH/GSSG in kimchi groups were higher compared to control. In conclusion, decrease in free radical production and increase in anti-oxidative enzyme activities were observed from kimchi groups suggesting that kimchi might have important role on retarding aging. Among kimchi variety tested in this experiment, MKCK and MLK seem to have greater effect on inhibiting free radical production and increasing anti-oxidative enzyme activities than KCK.
