  • 대나무잎(신의대)을 70% ethanol로 $80\pm2^{\circ}C에서$ 추출하고 극성이 다른 4가기 용매로 확분하여 생리활성 및 항균성을 검토하였다. 70% ethanol extract는 높은 항산화력을 나타내었고, 70% ethanol extract로부터 얻어진 각 용매별 추출 획분이 지질과 산화물 생성 억제와 malonaldehyde 생성 억제 효과는 ethyl acetate 획분 $\geq$ diethyl ether 획분 $\geqpetroleum$ ether 획분 > n-butanol 획분 > 수용성 획분의 순이었다. 전자공여능, 아질산염 제거능 및 항보체 활성은 n-butanol 획분이 다른 획분에 비하여 가장 높은 효과를 보였고 70% ethanol extract의 경우에도 전자고영능, 아질산염 제거능 및 항보체 활성이 높게 나타났다. 미생물인 Vib-rio parahaemoluticus, Pseudomonas aeruginosa, Salmonella typhimurium, Enterobacter aerogenes, Staphylococcus aureus 등에서 항균력을 나타내었다.

  • Anticomplementary activity of 61 strains of Korean higher fungi was screened for immunostimulation. Extracts from 11 of 61 strains including 5 of Ganoderma lucidum, 3 of Lentinus edodes, 2 of Cordyceps sp. and 1 of Agaricus campestris, showed higher anticomplementary activity than krestin which was immunopotent extract from Japanese Coriolus versicolor. The most potent anticomplementary activity was found with extract from Lentinus edodes IY105 whose complement consumption was 31.7%.

Purification and Chemical Properties of Anti-complementary Polysaccharide from Capsici Fructus (고추의 항보체 다당의 정제와 특성)

  • Water-soluble crude polysaccharide (CAP-0) obtained from the Capsici Fructus(the fruits of Capsium annuum L.) showed a potent anti-complementary activity. The anti-complementary activity did not change by pronase digestion of CAP-0, but decreased by the periodate oxidation. CAP-0 was fractionated into four polysaccharide fractions, CAP-1,2,3 and 4, by the addition of cetyltrimethylammonium bromide. CAP-1, showing the highest anti-complementary activity, was refractioned by anion-exchange chromatography to give three major fractions(CAP-1-III, IV and V). CAP-1-III was shown to have low anti-complementary activity, but CAP-1-IV and V had high activity. CAP-1-III and IV were purified on Sephadex G-100 to give each two fractions(CAP-1-IIIa and IIIb, CAP-1-IVa and IVb), respectively. From the results of gel filtration and electrophoresis, these four fractions and CAP-1-V were found to be homogeneous polysaccharides. High molecular polysaccharies(M.W. CAP-1-IIIa 70,000, IVa 195,000, V 140,000) showed relatively higher anti-complementary activity than low molecular polysaccharides(CAP-1-IIIb and IVb).

Isolation and Characterization of Complement System Activating Polysaccharides from the Hot Water Extract of the Leaves of Capsicum annuum L. (고추잎 열수추출물로부터 보체계 활성화 다당의 분리 및 특성)

  • It was observed that the hot water extract of the leaves of Capsicum annuum L., a Korean edible plant, had a potent anti-complementary activity. Crude polysaccharide fraction(CAL-0) was obtained by methanol reflux, ethanol precipitation, dialysis and lyophilization. CAL-0 contained 51.8% of total sugar, 8.2% of uronic acid and 16.8% of protein, and consisted of mainly arabinose, galactose and glucose as neutral sugars and galacturonic acid as uronic acid. The anti-complementary activity of CAL-0 decreased greatly by periodate oxidation, but was not changed by pronase treatment. Also, the anti-complementary activity of CAL-0 was reduced partially in the absence of the $Ca^{2+}$ ion. The crude polysaccharide CAL-0 was found to activate the C3 component both in the presence and in the absence of $Ca^{2+}$ through the crossed-immunoelectrophoresis suggesting that those involved in both classical and alternative complement pathway CAL-0 was further separated to an unabsorbed fraction(CAL-1) and six absorbed fractions(CAL-2longrightarrowCAL-7) on DEAE Sepharose CL-6B ion exchange column. Among them four major fractions in activity and yield were obtained, and consisted mainly of arabinose, galactose and glucose with various molar ratios. The major fraction, CAL-2, was purified to give a high molecular fraction(CAL-2-I) and a low molecular fraction(CAL-2-II) on Sepharose CL-6B column. The anti-complementary activity of CAL-2-I, a molecular weight of about 61,000, was higher than it of CAL-2-II.-II.

Isolation and Chemical Analysis of Potent Anti-Complementary Polysaccharides from Fruiting Bodies of the Fomes fomentarius (말굽버섯 자실체에서 분리한 항보체 활성 다당체의 화학적 분석)

  • The five anti-complementary polysaccharides (MFKF-NP, MFKF-AP1${\alpha}$, ${\beta}$, and MFKF-AP2${\alpha}$, ${\beta}$) were separated from hot water extracts of fruiting bodies of Fomes fomentarius by two subsequent column chromatography using DEAE-sepharose FF and Concanavalin A-sepharose 4B. The order of anti-complementary activity was MFKF-AP1${\beta}$ > MFKF-AP1${\alpha}$ > MFKF-AP2${\alpha}$ > MFKF-AP2${\beta}$ > MFKF-NP > Polysaccharide Krestine (PSK). Especially, MFKF-AP1${\beta}$ among those showed the most excellent anti-complementary activity (70% of ITCH50 value at $20{\mu}g/ml$). The monosaccharide composition analysis by gas chromatography indicates that MFKF-AP1${\alpha}$ and ${\beta}$ are a kind of homoxylan consisted mainly of xylose above 97%. Molecular weight of MFKF-AP1${\beta}$, major anti-complementary polysaccharide, was estimated to be about 12,000 by high performance liquid chromatography (HPLC). After the incubation of the serum with MFKF-AP1${\beta}$ in the presence or absence of $Mg^{++}$ and $Ca^{++}$ ions, its anti-complementary activity was investigated. This result indicated that MFKF-AP1${\beta}$ seems to be activator both on the classical and the alternative pathway of complement activation.

Screening and characteristics of anti-complementary polysaccharides from Chinese medicinal herbs (한약재로부터 항보체 활성 다당의 검색 및 특성)

  • We conducted screening on Chinese medicinal herbs to examine their anti-complementary activity by hemolytic complementary assay $(TCH_{50})$. Among 55 kinds of herbs, several herbs showed relatively potent anti-complementary activity which decreased $TCH_{50}$, more than 70% in comparison with control. Then, hot water extracts of the following herbs, Curcuma aromatica, Areca catechu, Gleditsiae spina, Euonymus alata, Acanthopanax senticous. Lonicera japonica, Aconitum carmichaeli, Curcuma zedoaria and Cinnamoum cassia, which were shown relatively potent anti-complementary activity were partially purified and analyzed their chemical properties. These activities were resistant to digestion with pronase but decreased by treatment with $NaIO_4$. These results may indicate that the complement activating ability in their herbs is due to polysaccharide. Furthermore, the anti-complementary activity of Areca catechu which was showed the most potent activity, was reduced partially in the absence of the $Ca^{++}\;ion$. After incubation of the normal human serum with partially purified polysaccharide of A. catechu in the absence of $Ca^{++}\;ion$, a cleavage of C3 in the serum was found to have occurred through immunoelectrophoresis using rabbit anti-human C3 serum. These results indicate that the mode of complement activation by polysaccharide of A. catechu is via both the alternative and classical pathway.

Characterization and Action Mode of Anti-Complementary Substance Prepared from Lactobacillus plantarum (Lactobacillus plantarum 균체 중 항보체 활성물질의 특성과 작용양식)

  • Among 12 lactic acid bacteria examined for their abilities to activate the complement system by hemolytic complement assay $(TCH_{50})$, Lactobacillus plantarum previously isolated from Kimchi showed high anti-complementary activity. The anti-complementary activity of the cell wall fraction of L. plantarum was more potent than that of the cytosol fraction, and both activities showed dose dependency. These high activities of the cytosol and the cell wall fractions were relatively resistant to the digestion with pronase, but sharply decreased after the treatment of $NaIO_4$. These results suggested that the complement activation by the cytosol and the cell wall fractions was mainly due to their polysaccharides. By the cross-immunoelectrophoresis using anti-human C3, the C3 activation products from both fractions were identified in $Ca^{++}$-free condition. Anti-complementary activity $(ITCH_{50})$ of the cell wall fraction was retained under the same condition, whereas that of the cytosol fraction was reduced considerably. From these results, it was inferred that the mode of complement activation by the cell wall fraction was mainly via alternative pathway, and that of the cytosol fraction was via both alternative and classical pathways.

  • The major protein (GMP) from the roots of Panax ginseng C. A. Meyer was purified, using gel filtration and ion exchange chromatography followed by reversed-phase and ion exchange FPLC. Staining analysis indicated that the protein has a carbohydrate moiety, which was also shown by band shift experiments using various glycosidases. Electrophoretic and gel permeation studies showed that GMP has an apparent molecular weight of 63 kDa composed of possibly two subunits of 25 kDa containing carbohydrate moiety. GMP showed an anticomplementary activity on the hemolysis of red blood cells, which is a screening tool for inflammation mediator search.

Optimun Conditions for Production of Mycelia and Extracellular Polysaccharide from Inonotus obliquus and Their Immunomodulating Acitivities (차가버섯의 균사체 및 세포외다당체의 생산조건과 면역활성)

  • Optium conditions for the production of mycelia and extracellular polysaccharide (EXPS) from submerged mycelial culture of Inonotus obliquus and their immunomodulating activities were investigated. The optmium production of mycelia and EXPS from I. obliquus was observed in mushroom complete medium (MCM). The optimum pH, temperature, and agitation speed for the production of mycelia and EXPS were 5.5, $25^{\circ}C$, and 150 rpm, respectively. The culture period for maximum production of mycelia (10.89 g/l) and EXPS (1.25 g/l) in shake flask cultivation was 11 days. The anticomplementary activity of intracellular polysaccharide (INPS) and EXPS form I. obliquus increased in a dose-dependent manner. Lysosomal enzyme activity of EXPS and INPS increased by 2.0- and 2.2-fold at $100{\mu}g/ml$ concentration, respectively, compared to the control group.
