Qualitative Enzyme-Linked Immunosorbent Assay (ELISA) for the Diagnosis of Edwardsiellosis (Edwardsiellosis의 진단을 위한 정성적 ELISA법)

  • Optimization and standardization of solid phase enzyme immunoassay were done for the diagnosis of edwardsiellosis in fish. The analyzed degree of immobilized antibody on surface of solid phase with peroxidase saturation method showed the optimized result by using partially purified $50{\mu}g/ml$ of rabbit anti-E. tarda Edk-2 antibody in sodium bicarbonate buffer for overnight incubation to cover the surface of polystyrene beads. Optimized immunoreaction was observed in the treatment of $50{\mu}g/ml$ of biotin conjugated antibody followed extravidin-peroxidase diluted 1 : 2,000 in PBS. The detectable concentrations of the this method were $1{\times}10^5$ cells/ml and $1{\times}10^5$ cells/ml expressed as the source of antigen amount for EDTA extraction and heat extraction, respectively. High cross-reaction of solid phase ELISA with the prepared rabbit and-E. tarda Edk-2 was observed against E. tarda strains isolated from flounder suffering from edwardsiellosis in aquatic farms of Korea. It suggested that the potential of this solid phase of ELISA technique is very powerful for the application to different strains of E. tarda isolated in farms of many different areas.

Analysis of Characteristics and Optimization of Photo-degradation condition of Reactive Orange 16 Using a Box-Behnken Method (실험계획법 중 Box-Behnken(박스-벤켄)법을 이용한 반응성 염료의 광촉매 산화조건 특성 해석 및 최적화)

  • The aim of our research was to apply experimental design methodology in the optimization of photocatalytic degradation of azo dye(Reactive orange 16). The reactions were mathematically described as a function of parameters amount of $TiO_2(x_1)$, and dye concentration($x_2$) being modeled by the use of the Box-Behnken method. The results show that the responses of color removal(%)($Y_1$) in photocatalysis of dyes were significantly affected by the synergistic effect of linear term of $TiO_2(x_1)$ and dye concentration($x_2$). Significant factors and synergistic effects for the $COD_{Cr}$, removal(%)($Y_2$) were the linear term of $TiO_2(x_1)$ and dye concentration($x_2$). However, the quadratic term of $TiO_2(x_1^2)$ and dye concentration($x_2^2$) had an antagonistic effect on $Y_1$ and $Y_2$ responses. Canonical analysis indicates that the stationary point was a saddle point for $Y_1$ and $Y_2$, respectively. The estimated ridge of maximum responses and optimal conditions for $Y_1:(X_1,\;X_2)$=(1.11 g/L, 51.2 mg/L) and $Y_2:(X_1,\;X_2)$=(1.42 g/L, 72.83 mg/L) using canonical analysis was 93% and 73%, respectively.

Performance Analysis on Speed Quality of LTE-Maritime Considering Scenario of e-Navigation Service and the Modulation Coding Scheme(MCS) (이내비게이션 서비스 시나리오 및 변조코딩방식(MCS)을 고려한 초고속해상무선통신망(LTE-Maritime)의 전송속도 성능 분석)

  • LTE-maritime, which was built as part of the Korean e-Navigation project, has been providing a service that meets the goal of establishing a network up to 100km from the coast since J anuary 31, 2021. To provide appropriate communication services for LTE-maritime from the perspective of real users, additional performance analysis of LTE-maritime is required considering service implementation, and efficient use of spectrum resources. This paper presents the requirement of transmission speed according to the e-Navigation service scenario, and available transmission speed based on LTE modulation coding scheme (MCS) according to sea-specific ship distribution scenario and frequency resource allocation, and is cross-analyzed with results of the sea test.

Ultrasensitive Enzymeimmunoassay for Testosterone in Human Saliva (사람 타액내 Testosterone의 초감도 효소면역측정법)

  • A few enzymeimmunoassay (EfA) for testosterone (T) have been reported but was not suitable for all biological samples. The present study was designed to develop a rapid, ultrasensitive EIA and to apply this technique for study the physiological changes of T in biological samples. Saliva samples were collected at 06:00~09:00 hour during one menstrual cycle from 18 normally menstruating women and on 09:00~10:00 hour from 20 normal men. The present study shows an established EIA for testosterone, using horseradish peroxidase (HRP), which was covalently bonded to testosterone-3-carboxymethyloxime (T-3-CMO). One batch of T-antisera was also covalently linked to microcrystalline cellulose particles by a mixed anhydride method in order to facilitate separation of bound and free steroids. The established EIA was validated in terms of sensitivity, accuracy, specificity, precisions etc., comparing with conventional radioimmunoassay. The sensitivity of the established EIA was less than 25 pg/tube. The correlation coefficients between the expected T-values and observed T-values measured by EIA or RIA were r=0.985 and r=0.941 respectively. The cross reactivity of antiserum in EIA was a little higher than that of RIA, especially by 5 ${\alpha}$-DHT. The intra- and inter-assay precisions of the present EIA were similar to those of RIA. The present study also demonstrates that the normal T-values in saliva of Korean male & female samples are 265.65${\pm}$15.80 pmol/l and 109.74${\pm}$ 12.01 pmol/l, respectively. The present EIA seems to be established and suitable for use in the endocrinological studies. The advantages of this EIA system also might make the present T-EIA an ideal procedure for use in a routine assay of ordinary laboratory with a conventional spectrophotometer.
