Cinnamic acid 유도체들의 SOS 반응을 지표로한 항돌연변이 효과에 관한 연구

  • 목적: Cinnamic acid의 유도체들중에서 항돌연변이 효과가 보고된 바 있어, 본 연구에서는 E. coil PQ37균주를 사용한 SOS chromotest를 이용하여 약 170개 cinnamic acid의 유도체들의 항돌연변이 효과를 확인하고자 하였고, 1차로 확인된 수종의 유도체들에 대한 결과를 보고하고자 한다. 방법: SOS chromotest는 sul A:: lacz fusion strain 인 E. coli PQ37 에서의 chemical로 인한 DNA damage에 대한 SOS response를 $\beta$-galactosidase enzyme activity level로서 측정하는 실험이며, m-RNA 또는 protein synthesis에 대한 test chemicals의 cytotoxic저해효과를 알아보기 위하여 alkaline phosphatase를 병행측정하여 보완하였다. 결과 및 고찰: (-)S-9 경우는 4-nitroquinoline 1-oxide(4-NQO)를 유도물질로 하였으며 대략 Induction factor가 9.7 정도였고, (+)S-9의 경우는 aflatoxinB$_1$을 유도물질로 하였고 이때의 Induction factor는 13.8 정도였다. 이결과 1차로 test된 cinnamic acid유도체 6개중에서 RK001, RK002, RK003, RK004, RK005는 거의 항돌연변이 효과를 나타내지 않았으나, RK006은 항돌연변이 효과를 보여주어 이들의 Structure-Activity Relationship (SAR) 및 Dose-response와 RK006의 항돌연변이 효과의 mechanism에 관해 M13 mp2 viral DNA의 유전자 염기서열을 이용하여 항돌연변이 기전연구를 수행중이다.

Effects of Culture Conditions on the Antimutagenic Activity of Lactobacillus plantarum KLAB21 against 4-Nitro-O-Phenylenediamine (NPD) (Lactobacillus plantarum KLAB21의 배양조건에 따른 4-Nitro-O-Phenylenediamine(NPD)에 대한 항돌연변이 활성)

  • Lactobacillus plantarum KLAB21 isolated from kimchi has been shown to produce antimutagenic subtance(s) into the culture medium using Salmonella typhimurium TA100 and S. typhimurium TA98 (Rhee and Park, Korean J. Appl. Microbiol.. Biotechnol., 1999, in press). In this study, the effects of culture conditions were investigated to maximize the production of antimutagenic substance(s) against 4-nitro-O-phenylenediamine(NPD) by the strain KLAB21. Glucose(255) as a carbon source and yeast extract or bactopeptone(1%) as a nitrogen source showed the highest production of the antimutagenic substance(s). Optimal initial pH of the culture medium, culture temperature and shaking speed for the antimutagenic substance(s) production were pH 7.0, 37$^{\circ}C$ and 150rpm, respectively. Under the optimal conditions, the antimutagenic activity of L. plantarum KLAB21 culture supernatant against NPD on Salmonella typhimurium TA100 and S. typhimurium TA98 were 73.95% and 59.74%, respectively.

  • Antimutagenic effects of boiled water extract and tannin from persimmon leaves were studied by using Ames test, spore rec assay and SOS chromotest. Strong antimutagenic activities toward aflatoxin B1(AFB1), dimethyl-aminobiphenyl(DMAB), N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) and 4-nitroquinoline-1-oxide(4-NQO) were observed when boiled water extract and tannin from the persimmon leaves were added in the Salmonella typhimurium TA 100. In spore rec assay using Bacillus subtilis H17($rec^{+}$) and M45($rec^{-}$), boiled water extract and tannin from the persimmon leaves considerably inhibited the mutagenesis induce by MNNG. In SOS chromotest using Escherichia coli PQ37, these samples also exhibited strong antimutagenic activity toward 4-NQO. The tannin was more effective than boiled water extract of persimmon leaves in the antimutagenicity tests.

Antimutagenicity and Anticancer Activity of Soybean Fractions Extracted by Solvents (대두 분획물의 항돌연변이 및 항암활성 효과)

  • Inhibitory effects of several solvent fractions from soybean on mutagenicity using Salmonella typhimurium TA 100 in Ames test and growth of human cancer cells (AGS gastric adenocarcinoma, Hep 3B hepatocellular cancinoma and HT-29 colon cancer cells) were studied. The treatment of dichloromethane and ethylacetate fractions (2.5 mg/assay) extracted from soybean to Ames test system inhibited aflatoxin $B_1\;(AFB_1)$ induced mutagenicity by 83%, respectively, and showed a higher antimutagenic effect than other solvent fractions. In case of N-methyl-N#-nitro-N-nitrosoguamidine (MNNG) induced mutagenicity, the ethylacetate fraction showed the highest inhibitory effect (by 67%) among solvent extracts, although the inhibitory effect was not stronger compared with $AFB_1$ induced mutagenicity. In sulforhodamine B (SRB) assay, the treatment of ethylacetate fraction (2 mg/assay) significantly inhibited the growth of AGS, Hep 3B and HT-29 cancer cells by 66%, 73% and 77%, respectively, followed with the intermediate and dichloromethane fractions. These results indicated that soybean fraction extracted with ethylacetate had higher inhibitory effects on $AFB_1$ and MNNG in Ames test and growth inhibition activity to human cancer cells was appeared, suggesting that soybean fraction extracted with ethylacetate may contain the biologically active compounds.

  • Lactobacillus plantarum KLAB21 isolated from Kimchi has been reported to produce antimutagenic subtance(s) in the culture medium. In this study, antimutagenic effects of the strain KLAB21 were investigated to under various culture conditions maximize the production of antimutagenic substance(s) against N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) on Salmonella typhimurium TA100 and 4-nitroquinoline-1-oxide(NQO) on S. typhimurium TA98. Glucose(2%) as a carbon source and yeast extract(1%) as a nitrogen source resulted in the highest production of the antimutagenic substance(s) against both mutagens in the culture supernatant of L. plantarum KLAB21. The most effective concentrations of bactopeptone as a nitrogen source were 1% against MNNG and 1.5% against NQO. Optimal pH of the medium, culture temperature, and shaking speed for the antimutagenic substance(s) production were pH 7.0, $37^{\circ}C$ and 150 rpm, respectively. Under the optimal condition, the antimutagenic effects of L. plantarum KLAB21 culture supernatant were 98.4% against MNNG on S. typhimurium TA100 and 57.3% against NQO on S. typhimurium TA98.

  • Methanol extract of garlic was fractionated to chloroform and aqueous fractions. The cholorform fraction possessed the highest antimutagenic activity in Salmonella typhimurium TA 100 and TA98, and was further fractionaed into four Allium sativum chromatography fractions (ASC F1, 2, 3 and 4) by column and thin layer chromatographies. The ASC F2 exhibited the higher antimutagenic activity and contained 18 chemical compounds tentatively identifed by GC-MS, NMR and FT-IR. Among the 18 compounds, methyl linoleate was a major compound to exhibit the antimutagenicity.

  • The nitrite-scavenging and antimutagenic effects of various solvent extracts from different parts of the loquat. were investigated to develope as source of functional food. The nitrite-scavenging and antimutagenic effects of fractions obtained from methanol extract were most highest in ethyl acetate fraction. Ethyl acetate fraction showed better results in the nitrite scavenging activity of 80% in leaf, 81% in seedless fruit, 73% in peel and 63% in seed. The antimutagenic effect showed strong ethyl acetate fraction of 53.1%, 48.3% in leaf and seedless fruit. The antimutagenic effect increased with increasing concentration of ethyl acetate fraction against mutagenicity induced by direct mutagen of IQ in Salmonella typhimurium TA 100. Addition of 2.5 mg and 5 mg of ethyl acetate fraction showed inhibitory effect of 65.5%, 71.2% in leaf and 63.4%, 69.2% in seedless fruit, respectively.

  • Antimutagenic and antimicrobial effects of the ethanol extracts from sea-mustard and sea-tangle were investigated. Antimutagenic effects of ethanol extracts from sea-tangle were higher than those of sea-mustard. Seventy and ninety percent ethanol extracts from sea-tangle showed high antimutagenic effects on NDMA-induced mutations in TA100 and TA102. Fifty percent of ethanol extract from sea-mustard showed high antimicrobial effect against S. cerevisiae, while 70 and $90\%$ ethanol extracts from sea-tangle against B. subtilis and E. coli.

  • Leuconostoc mesenteroides subsp. cremoris DLAB19 were investgated under various culture conditions to maximize the production of antimutagenic substance(s) against N-methyl-N\`-nitro-N-nitrosoguandine(MNNG) on Salmonella enterica serovar typhimurium TA100 and 4-nitroquinoline-1-oxide(4-NQO) on S. enterica serovar typhimurium TA98. The MRS medium containing glucose (2%) as a carbon source and yeasty extract (1%) as a nitrogen source resulted in the highest production of the antimutagenic substance(s) against both mutagens in the culture supernatant of Leu. mesenteroides subsp. cremoris DLAB19. Optimal pH of the culture medium, culture temperature and shaking speed for the antimutagenic substance(s) production were pH 7.0, 3$0^{\circ}C$ and 150 rpm, respectively. Under the optimal condition, the antimutagenic effects of Leu. mesenteroides subsp. cremoris DLAB19 culture supernatant were 96.4% against MNNG on S.enterica serovar typhimurium TA100 and 53.8% against 4-NQO on S. enterica serovar typhimurium TA98.

  • Antimutagenic and antimicrobial effects of cucumber extracts were investigated. Antimutagenic effects of cucumber extract against aflatoxin (AFB$_1$) as indirect mutagen and N-methyl-N'-nitro-N-nitrcsoguanidine (MNNG) as direct mutagen using the Ames assay system with Salmonella typhimurium TA100 were studied. 1.25~5.0% of methanol extract exhibited 11 ~ 17% of antimutagenity against AFB$_1$ and 46~85% of antimutagenity against MNNG. Among fractions of methanol extract, hexane fraction exhibited the highest antimutagenic effect against AFB$_1$ (89%) and butanol fraction exhibited the highest antimutagenic effect against MNNG (95%). Antimicrobial effects of cucumber extract were investigated on the eleven microorganisms. Methanol extract showed anitimicrobial effect on eight microorganisms. Among these tested microorganisms, Klebsiella pnemonia KCTC 2208, pseudomonas aeruginosa KCTC 2004 were the most sensitively inhibited with 13 mm clear zone on holo test. Hexane fraction showed anitimicrobial effect only on Vibrio parahaemolyticus KCTC 2471. Chloroform and ethyl acetate fractions showed a weak effect. V. parahaemolyticus showed the lowest minium inhibitory concentration (MIC) (500 ppm) among eleven tested microorganisms by methanol extract. Sterilization effect of 1% methanol extract on P. aeruginosa incubation is 10 times stronger than 0.5% methanol extract. It estimated to need 26 min for the sterilization of 90% P. aeruginosa cell counts by 1% methanol extract but 250 min by 0.5% methanol extract.
