Comparison of Antioxidant and Antimicrobial Activities of Bracken (Pteridium aquilinum Kuhn) according to Cooking Methods (조리방법에 따른 고사리의 항산화활성 및 항균활성 비교)

  • This study was carried out to evaluate the yield of extract, antioxidant compounds (total phenolic and total flavonoid), antioxidant (DPPH assay, ABTS assay and reducing power), and antimicrobial activities of bracken (Pteridium aquilinum Kuhn), according to cooking methods (non-blanched, blanched and seasoned). The yield of seasoned bracken extracts showed a high value of (4.59%) followed by non-blanched bracken and blanched bracken with 2.69% and 0.30%, respectively. In the total polyphenol and flavonoid contents, seasoned bracken extracts showed higher antioxidant compounds ($96.11{\pm}0.34mg\;GAE$/100 g RW, $20.90{\pm}0.mg\;CE$/100 g RW) than non-blanched and blanched. The total antioxidant activities (DPPH assay, ABTS assay and reducing power) were shown to be in the order of seasoned bracken > non-blanched bracken > blanched bracken. In the antimicrobial activities, non-blanched bracken extracts showed antimicrobial activity against B. cereus, B. subtilis, E. cloacae, E. coli, S. enterica, and P. aeruginosa except for S. aureus. The non-blanched bracken extracts (5 and 10 mg/disc) especially showed strong antimicrobial activity against P. aeruginosa ($10.00{\pm}0.71$ and $10.25{\pm}0.35mm$). The inhibition zone diameter from the extracts of blanched bracken and seasoned bracken was not detected. Many seasonings added in the process of cooking can increase the antioxidant capacities. The overall results of this study demonstrate that the cooked bracken with seasoning would be the most efficient way of ingesting antioxidant compounds.

Antibacterial Activity of Medicinal Plant Extracts to S. aureus KCCM12256 and V. parahaemolyticus KCCM11965 (한약재 추출액의 S. aureus KCCM12256과 V. parahaemolyticus KCCM11965에 대한 항균 활성)

  • This study was conducted to investigate the antibacterial activities of medicinal plant extracts. The results were as follows. Crude water and methanolic extracts of Sanguisorba officinalis Linne, Coptis chinensis Makino, and Portulaca oleracea Linne, among 31 kinds of medicinal plants, had strong antibacterial activities against S. aureus and V. parahaemolyticus, with the methanolic extract more effective than the water extract. The methnolic and water extracts of S. officinalis Linne had strong antibacterial activities against S. aureus and V. parahaemolyticus. Antibacterial activity against S. aureus and V. parahaemolyticus was observed with extracts of S. officinalis Linne at dilutions of 5 and 10%, respectively. The EtOAc, BuOH, and $H_2O$ fractions of the extract of S. officinalis Linne had antibacterial activities against S. aureus and V. parahaemolyticus. Specifically, the fractions had antibacterial activity against S. aureus at dilutions of 5, 2, and 1%, respectively. Regarding V. parahaemolyticus, the EtOAc, $H_2O$, and BuOH fractions had antibacterial activities at dilutions of 5, 2, and 1%, respectively.

Development of a Recombinant Strain of Pichia pastoris with Antibacterial Activity (항균활성을 보유한 재조합 Pichia pastoris 균주의 개발)

  • To develop a yeast strain of Pichia pastoris producing an antibacterial peptide, we have attempted the expression and secretion of an insect defensin. The nucleotide sequences corresponding to mature defensin were chemically synthesized by 6 oligomers, assembled in vitro and the synthesized gene was identified by nucleotide sequencing. The prepro sequence of yeast mating factor $\alpha$1 and the defensin gene were recombined into a Pichia expression vector, pPIC9K. The resulting plasmid, pPIDE, was transformed into P. pastoris GSl15 and transformants selected on histidine-deficient minimal plates were tested for antibacterial activity against Micrococcus luteus. Four strains with different antibacterial activity were selected for further analysis. Southern hybridization and RT-PCR verified the defensin gene was maintained and transcribed in a host. Four strains were cultivated in YPD broth for 96 hours to compare cell growth and antibacterial activity, They showed no difference in cell growth, however, each strain showed different antibacterial activity pattern with culture time. The maximal activity was about 550 AU/ $m\ell$.

Effect of structural variation of medium chain fatty acids on antibacterial activities against pathogenic bacteria (중쇄지방산의 구조적 차이에 따른 병원성 세균에 대한 항균활성 변화)

  • Broad range of fatty acids were reported to show antimicrobial activities against broad range of microorganisms. However, possible changes of the antibacterial activity of a fatty acid based on structural variations are largely unknown. This study was focused on determination and comparison of the antimicrobial activities of the medium chain fatty acids, based on the position of carboxyl groups on either terminal end, against the representative food-pathogenic bacteria. Over all, mono-carboxyl medium chain fatty acids (MC-MCFA) presented stronger antimicrobial activities against the food-pathogenic bacteria tested including methicillin-resistant Staphylococcus aureus (MRSA) than di-carboxyl medium chain fatty acid (DC-MCFA). In addition, some of MC-MCFA and DC-MCFA showed high possibility to be used as a synergistic adjuvant for both the commercial β-lactam family antibiotics and aminoglycoside family antibiotics against MRSA.

Antimicrobial Activities of 'Formosa' Plum at Different Growth Stages against Pathogenic Bacteria (생육 시기에 따른 자두류 중 후무사의 식중독균에 대한 저해효과)

  • To determine the antimicrobial activity of methanol extracts form 'Formosa' plum against 4 kinds of pathogenic bacteria, the fermosa were got at different growth stages and were extracted using methanol, respectively. The Formosa methanol extracts treated with 5.0 mg/disc showed the highest antimicrobial activity against 4 kinds of pathogenic bacteria and those of Formosa 1-4(immature fruit), which thin out 10∼25 days before foal harvest, showed higher antimicrobial activity against gram positive and gram negative microorganisms than Formosa 5-6(mature fruit). Especially, the methanol extracts of Formosa 1 and 2 were exhibited the strongest growth inhibiting activities to these bacteria. The minimal inhibitory concentrations(MIC) of immature Fomosa methanol extracts was 320 $\mu\textrm{g}$/mL against Escherichia coli 0157:H7 and 160 $\mu\textrm{g}$/mL against Staphylococcus aureus respectively. The MIC of immature Formosa methanol extracts to Salmonella typhimurium and Listeria monocytogenes were 640 $\mu\textrm{g}$/mL. These results suggest that methanol extracs of immature formosa can be used as an effective natural antimicrobial agent in food.

Antimicrobial Activity of Dryopteris rhizoma against Some Food Spoilage Microorganisms (일부 식품부패성 미생물에 대한 관중의 항균활성)

  • This study was carried out to know the active faction of Dryopteris rhizoma on antimicrobial activity against some food spoilage microorganisms. Also, antimicrobial activities were investigated for the aqueous and ethanolic extracts of four herbs such as Teminaliae fructus, Eugeniae flos, Salviae miltior-rhizae radix and Dryopteris rhizoma. Antimicrobial activities of three herbs except for Terminaliae fructus showed higher activities in 75% ethanolic extracts than in aqueous extract. Ethanolic extract of Dryopteris rhizoma showed the highest antimicrobial activity among extracts of four herbs. Antimicrobial activity intensities of solvent fractions of Dryopteris rhizoma extracted by 75% ethanol were order to CHC1$_3$fraction>EtOAc fraction >BuOH fraction>$H_2O$ fraction. The CHC1$_3$, EtOAc and BuOH fractions also inhibited growth of food spoilage microorganisms as the concentration increased, respectively. In case of EtOAc fraction, 1000 ppm of fraction almost inhibited completely the growth of microorganisms tested.

Isolation of Antimicrobial Substance from Schizandra chinensis Baillon and Antimicrobial Effect (오미자로부터 항균활성 물질의 분리 및 항균효과)

  • In order to isolate antimicrobial substances from Schizandra chinensis, the dried fruits were extracted with the methanol and the extract showed a strong antimicrobial activity. Also, the methanol exract was further fractionated with hexane, dichloromethane, ethylacetate and buthanol. The ethyl acetate-soluble fraction showed the strongest antimicrobial activity. These fraction were further separated by using various chromatographic methods including thin layer chromatography, silicagel open column chromatography and prep. HPLC. A major component S-EA-5-T1 and S-EA-5-T3 from the ethyl acetate fraction, which showed a strong antimicrobial activity was identified by Mass and NMR spectrometry. Two compounds were isolated and identified as trimethylcitrate and the essential oil of Schizandra chinensis and was estimated as gomisin C, respectively. The growth of S. typhimurium was also inhibited about 1.65 to 2.86 log cycle in minced pork by the addition 1% of Schizandra chinensis extract for 12 days at $4^{\circ}C$. These results suggested that these compounds have a strong potential as a natual food preservatives.

신규베타락탐계 항생제 합성과 항균성

  • Cephem ring의 C-7위치에 aminothiazoly lmethoxyimino기를 가진 화합물들이 항균활성을 증가시키고, G(-)균의 외막투과성을 촉진시킬뿐 아니라 광범위항균 spectrum을 갖게 하고 $\beta$-lactamase에 안정하며 PBP(penicillin binding protein)에 대한 결합친화성을 증가시킨다는 보고에 따라 본 저자는 C-7위치에 cefotaxime구조와 같이 aminothiazolylmethoxyimino acetamido moiety를 고정시키고 항균활성을 증가시키기 위하여 약리활성이 기대되는 5-(substituted)-2H-tetrazole유도체들을 합성하여 C-3 위치에 도입시킨 새로운 화합물 7$\beta$-〔(z)-2-(2-aminothiazol-4-yl)-2_(methoxyimino)acetamido〕-3-〔5-(substituted)tetrazol-2-yl〕 methyl-3-cephem-4-carboxylic acid 유도체를 합성하여 B. subtilis ATCC 6633, M. luteus ATCC 1004, E. coli KCTC 1039, E. coli ESS, K. pncumonia KCTC 1560, P. aeruginosa IF0 13130, S. typhimurium KCTC 1925, S. typhimurium SL 1102, 및 C. albicans ATCC 10231 등의 균과 fungus에 대하여 기존의 cefotaxime과 cefazoline을 대조물질로 사용하여 항균력을 비교하였다. 이들 화합물들은 대체적으로 M. luteus ATCC 6633, E. coli ESS, S. typhimurium SL 1102 균에 대해서는 cefotaxime보다 항균력이 우수하였으나 P. aeruginosa IF0 13130에 대해서는 항균력이 저하되었고 cefazolin보다는 대체적으로 항균력이 우수하였다.

Search for Plant-originated Antibacterial Compounds Against Pathogen (Acidovorax avenae subsp. citrulli) of Watermelon Bacterial Fruit Blotch (수박 과실썩음병 병원균(Acidovorax avenae subsp. citrulli)에 대한 식물유래 항균 활성물질 탐색)

  • 133 Species of medicinal plants were used for the development of natural agrichemicals with anti-microbial activity against Acidovorax avenae subsp. citrulli, a pathogen of bacterial fruit blotch in watermelon. The MeOH-extracts of these medicinal plants were examined for anti-microbial activity by bioassay. The MeOH-extract of Citrus unshiu Markovich had the strongest antibacterial activity against Acidovorax avenae subsp. citrulli. To identify anti-microbial compounds from Citrus unshiu Markovich, solvent-fractionation was used. The fraction of hexane, which showing the highest value of anti-microbial activity, was analyzed by GC-MS. Each mass spectra, corresponding to each peak of chromatogram, was compared to mass database of Wiley library. As a result, d-Limonene, ${\gamma}$-terpinene, ${\beta}$-linalool, terpineol, palmitic acid, 9,12-octadecadienoic acid, Linolenic acid, and stigmasterol were identified. Among them, d-Limonene, ${\gamma}$-terpinene, ${\beta}$-linalool, and terpineol confirmed to be shown the anti-microbial activity by bioassay. Especially, d-Limonene and ${\gamma}$-terpinene found to have strong activity. In conclusion, we thought d-limonene and ${\gamma}$-terpinene from Citrus unshiu Markovich. Latin, had anti-microbial activity against Acidovorax avenae subsp. citrulli and could be candidates for the control agents for the control of bacterial fruit blotch in watermelon.

Activity of Nature Plants Extracts against Pseudomonas aeruginosa ATCC 27853 (Pseudomonas aeruginosa ATCC 27853에 대한 약용식물 추출물들의 활성)

  • Anti-Pseudomonas aeruginosa activities for ethanol extract of 34 medicinal plants widely used in the folk medicine were evaluated to screening of anti-Pseudomonas aeruginosa agents. The ethanol extracts of Gardenia jasminoides, Arctium lappa, Citrus unshiu, and Phellodendron amurense showed antimicrobial activities against P. aeruginosa ATCC 27853. The ethanol extracts of Gardenia jasminoides among these medicinal plants showed significant antimicrobial activity against P. aeruginosa. Therefore, we expect that these medicinal plants will be useful for nature antimicrobial agent against P. aeruginosa in future.
