Natural Dyeing of Silk Fabrics with the Extract of Alnus firma Tree Branches (사방오리나무가지 추출액을 이용한 견직물의 천연염색)

  • The purpose of this study is to investigate the optimal dyeing conditions and human-friendly properties of the extract from the branches of Alnus firma tree which is widely used for forestation and to provide basic information for quantification and commercialization of natural dyeing by discovering such as dye material. The optimal dyeing conditions, as a result, were shown at 100% of dye concentration, $90^{\circ}C$ of dyeing temperature and 40 minutes of dyeing time. In terms of dye uptake depending on the mordanting methods, it showed the best result in order of Fe, Sn, Cu and Al at pre-mordanting while found strong in order of Fe, Cu, Al and Sn at post-mordanting. Color fastness to washing and dry cleaning was found strong at grade 4-5 and fastness to light was rated at 2. The grade of change in color to rubbing and perspiration was good at 4-5. In aspects of functional properties, it showed excellent results of 98% deodorization rate at 120 minutes of dyeing time, 96.1% UV protection rate and 99.9% bacterial reduction against Staphylococcus aureus and Klebsiella pneuminiae. It is considered, therefore, the extract from Alnus firma tree is of great value as an eco-friendly natural dyestuff.

Characteristics and Antimicrobial Effects of Novel Burkholderia cepacia No. 15-2 Isolated from Compost (퇴비로부터 분리된 Burkholderia cepacia No.15-2의 특성과 항균 효과)

  • To develop the functional-compost containing antifungal substance by using antagonistic microorganisms, Spinacia oleracea L and Rhizoctonia solani Kuhn O-28 were used as a model plant and phytopathogen, respectively. Total 80 strains were isolated from the compost of various waste foods mixture processed for a year. Among them, No.15-2 strain was selected due to its highest antifungal activity against R. solani Kuhn O-28 and was identified phyno- and phylogenotypically as Burkholderia cepacia genomovar V. which is rare probability in pathogen, by 16S rDNA sequencing and specific primer pair PCR method. B. cepacia No.15-2 preferentially dominated during the compost and its cell numbers were maintained almost $${\times}$10^{13}$ cuf/g for 15 days. The morbidity caused by R. solani Kuhn O-28 in S. oleracea L cultivation was reduced to 40% by addition of B. cepacia No.15-2. In conclusion, the antifungal compost using B. cepacia No.15-2 could be applied to biocontrol of various crops blights caused by fungal pathogen.

Improvement of Comfortability and Ability on Nonwoven Fabric for Disposable Work Clothing Using Yellow Soil Printing (황토 날염을 이용한 일회용 작업복 소재의 쾌적성 및 기능성 향상에 관한 연구)

  • The purpose of this study were to investigate characteristic changes on nonwoven fabric for disposable work clothes by the yellow soil printing. It separate grind yellow soil as two different size of particles $45\sim52{\mu}m$ and $53\sim65{\mu}m$ for hand screen printing on three kind of nonwoven fabrics. To examine the effect of yellow soil printing on nonwoven fabric were to observe, dyeability by using spectrophotometer, moisture regain by oven method, air permeability, anion property and antibacterial activity. The results were as follows: When yellow soil concentration increased from 5 to 10%, K/S value also increased from 1.05 to 1.88. When yellow soil concentration increased, moisture regain also increased. In same concentration, moisture regain occurred higher as particle of small size. Air permeability decreased when the charcoal printing concentration increased. Anion occurrence appeared $140\sim160ion/cc$ from three different kinds of nonwoven fabrics in 3% and 9% yellow soil concentration. Therefore, occurred anion ineffectively. In concentration of 3%, rate of deodorization measured as 89%, 83% and 87%, and 9% concentration caused 96%, 86% and 93% of high deodorization. Antibacterial activity examination in nonfinished nonwoven fabric resulted range of 60%, however, 3% and 9% concentration finished nonwoven fabric resulted 99.9% of excellent antibacterial activity Surface temperature increased $1.5\sim2^{\circ}C$ by yellow soil finishing.

Isolation and Identification of Rice Root Endophytic Antagonistic Serratia marcescens (벼 뿌리 내생 항균성 Serratia marcescens의 분리 및 동정)

  • Twenty-three strains of Serratia sp., isolated from surface-sterilized rice roots collected in Chonbuk and Chungnam province, were identified and characterized. They were Gram-negative, rod shaped and red pigmented typically and their endophytism was confirmed by inoculation and reisolation of the strains in planta. Their antifungal activity against 4 rice pathogenic fungi was compared and ranged from 62.4 to 85.2% against Rhizoctonia solani and 68.0 to 88.5% against Pyricularia grisea. Among the 23 strains tested, strain Rsm220 showed the strongest inhibition activity against 4 pathogenic fungi. The strain was, therefore, selected as a biocontrol candidate for both the pathogens and its bacteriological characteristics and 165 rDNA sequences were analyzed. Phenotypic and biochemical characteristics of the selected Rsm220 were highly related to the type strain of S. marcescens and 165 rDNA sequencing of Rsm220 showed a homology of 98.2% to the type strain of S. marcescens. The strain Rsm220 was identified as S. marcescens and the inhibition result of this endophytic strain indicates that it is a potential biocontrol agent for R. solani and R grisea.

Dyeing Properties and Functionality of Cotton Dyed with Extract from Spirodela polyrhiza and Mixture Extracts from Spirodela polyrhiza and Salvia plebeia R. Br. (개구리밥 추출물 및 개구리밥과 곰보배추 혼합 추출물로 염색한 면직물의 염색성 및 기능성)

  • This study looks into the dyeing properties and functionality of cotton fabrics dyed in both the Spirodela polyrhiza extract and the extract resulting from the mixture of Salvia plebeia R. Br. and Spirodela polyrhiza. Since the UV-Vis Spectrum of the methanol extract of Spirodela polyrhiza shows absorption peaks at 256, 268nm, and 345nm, it can be inferred that the compound that Spirodela polyrhiza contains is a flavonoid. In addition, it can also be presumed that, by analyzing the infrared absorption spectrum of Spirodela polyrhiza, the plant contains flavonoid compounds, just like Salvia plebeia R. Br.. The UV protection factors of the cotton fabrics dyed in both the Spirodela polyrhiza extract and the extract from the mixture of Salvia plebeia R. Br. and Spirodela polyrhiza were 50+, presenting outstanding UV protection factors. The deodorization rate of the cotton dyed in the Spirodela polyrhiza extract was between 30 and 120 minutes, and the rate rose from 92% to 97% as time passed. The deodorization rate of the cotton dyed in the extract from the mixture of Salvia plebeia R. Br. and Spirodela polyrhiza increased from 88% to more than 91%. The result also revealed that overall the fastness of color, including color fastness to washing related to change in color, as well as the color fastness to light of the fabric dyed in the extract from the mixture of the two plants improved, compared to the cloth dyed only in Spirodela polyrhiza extract. Furthermore, the antibacterial activity was also strengthened.

Protective effects of extracts from spent mushroom substrate of Lentinula edodes on gray mold disease of ginseng (표고버섯 수확 후 배지추출물의 인삼잿빛곰팡이 병 방제 효과)

  • This study aims to investigate the antifungal and protective effects of water- and 70% methyl alcohol-extracts from spent mushroom substrate (WESMS and MeOHSMS) of Lentinula edodes, on Botrytis cinerea- the causative agent for gray mold disease in ginseng. MeOHSMS inhibited mycelial growth and spore germination of Botrytis cinerea, by 75% and 95%, respectively. MeOHSMS could suppress gray mold disease of ginseng seedlings by 80% and effectively reduce the disease severity by 60%. Compared to the treatment of ginseng leaves with WESMS and DL-${\beta}$-aminobutyric acid (BABA), the MeOHSMS treatment increased the phenolic compounds in the leaves by 36% and 18%, respectively. These results suggest that the SMS extracts suppress gray mold disease in ginseng via dual functions: antifungal activity and increase in a plant defense factor-phenolic compounds.

  • The aim of this study was to identifyisothiocyanates (ITCs) from wasabi (Wasabi japonica Matsum) using supercritical carbon dioxide ($SCO_2$) and to compare the composition in the extracts between $SCO_2$ and organic solvents extraction. A semi-continuous high pressure apparatus was used to extract wasabi (roots, stems and leaves) at following conditions pressure 80$\sim$120 bar, temperature $40\sim50^{\circ}C$. Ether, ethanol, chloroform and dichloromethane were used as organic solvents. The ITCs extracted by means of both separation technologies were analyzed by a gas chromatography system. As the results of study, AITC and ITCs were highly extracted at 40$^{\circ}C$ and 80 bar. To extract AITC from wasabi, $SCO_2$ extraction is more effective than organic solvents extraction, resulted in thermal degeneration and remaining of organic solvents.

Effect of embryo and recipient condition on pregnancy rate following bovine embryo transfer (소 수정란이식(受精卵移植)에 있어서 수란우(受卵牛)와 수정란(受精卵)의 조건이 수태율(受胎率)에 미치는 영향)

  • This study was carried out to determine suitable selection factors for recipients and embryos which could improve pregnancy rates following bovine embryo transfer. The experiment included 52 surgical transfers from February, 1985 through June, 1986 performed on Kyungpook Breeding Center in southern Korea. The pregnancy rate was highest when recipients were in estrus within 6 hours before the donor to 12 hours after the donor (78.3% versus 50% for recipients in estrus earlier or later). Pregnancy rates were acceptable following culture under field conditions for up to 17 hours. More recipients over 15 months of age (76.1%) remained pregnant than those under 15 months (66.7%). Embryos transferred during the months from February to July resulted in higher pregnancy rates than those transferred during the remaining 6 months (77.3% versus 57.1%). Transferrable embryos were classified A (best) to C (worst); those graded A or B resulted in significantly higher pregnancy rates than those graded C (81.8% and 73.3% versus 25.0%, p<.05). Pregnancy rates among recipients of the Korean native breed tended to be higher than among Holstein recipients (100% versus 71.1%). Similarly, when the embryo was transferred to the right uterine horn, pregnancy rates tended to be higher than when it was transferred to the left (81.3% versus 65%). Pregnancy rates did not differ according to the stage of development of the embryo; they were for morulae, tight morulae, blastocysts, and advanced blastocysts, respectively: 75.0%, 66.7%, 75.0%, and 77.4%.

Establishment of an Analytical Method of Fluoroquinolones in Milk by HPLC (HPLC를 이용한 우유에서의 fluoroquinolones 시험법 확립)

  • A high-performance liquid chromatography (HPLC) method was established for the determination of fluoroquinolones in milk. Protein was removed by using trichloroacetic acid in order to increase a mean recovery of milk. The extracts were using $Strata^{TM}$-X solid-phase extraction cartridge. The analytes were detected by HPLC on a $C_{18}$ column. HPLC method with fluorescence detection system (Ex: 278 nm, Em: 456 nm) provided a high degree of sensitivity in detecting fluoroquinolones. The limits of quantitation (LOQ) and mean recoveries of fluoroquinolones were 40 ${\mu}g$/kg and 73.6-95.2% (ofloxacin), 10 ${\mu}g$/kg and 77.3-91.9% (norfloxacin), 20 ${\mu}g$/kg and 91.6-94.3% (ciprofloxacin), 10 ${\mu}g$/kg and 81.0-87.8% (enrofloxacin), 10 ${\mu}g$/kg and 71.3-81.0% (sarafloxacin), 10 ${\mu}g$/kg and 89.4-90.8% (orbifloxacin), 2 ${\mu}g$/kg and 69.4-85.5% (danofloxacin).

Quality Changes of Satsuma Mandarin during Storage by Storage Warehouse (저장고 형태에 따른 온주밀감의 저장 중 품질변화)

  • Quality changes of satsuma mandarin (Citrus unshiu Marc. var. miyagawa) during storage by storage warehouse were investigated. Citrus were treated with 2000-folds diluted iminoctadime-triacetate solution and 1.5% chitosan with 0.5% CaCl$_2$ solution, and were at 30$\^{C}$ for 24 hr before storage. The citrus of about 12kg/26L plastic container were stored at room temperature, and at 4$\^{C}$ with 87% relative humidity. Decay ratio of citrus with precise temperature and humidity control were lower than the others during storage. Penicillium italicum Monilia candida Alternara citri, Mucor hiemalis, Phomopsis citri Botrytis cinerea, Phoma citricarpa Clomererella cingulata, Penicillium digitatum were identified as putrefactive microorganisms in citrus storage. Weight loss, moisture content of peel and flesh were decreased slowly during storage. 24% of original acid content were decreased at room temperature on 120 days storage, compared to 15∼18% loss on cold storage. Total sugar of citrus was decreased rapidly after 90 days, and vitamin C content were also decreased rapidly after 60 days during storage.
