Continuous Production of Agarooligosaccharides Using Packed-Bed Reactor (Packed-Bed 반응기를 이용한 한천올리고당의 연속생산)

  • Enzymatic hydrolysis of agar was carried out continuously to produce agarooligosaccharides by immobilized agarase in Packed-Bed Reactor. The reactor was constructed using a acryl tube with an internal diameter of 10 mm and a useful height of 140 mm. The Packed-Bed Reactor was 11 mL reactor volume as its length : diameter ratio was 14 : 1. The operation condition of reaction was performed with an 1 g/L agar concentration at 40$^{\circ}C$, 10 mM MOPS buffer(pH 7.0) and with the flow rate 3 mL∼48 mL/h at a dilution rate of 1.09∼5.45 h$\^$-1/. The hydrolysis products was identified DP6, DP4 and DP2 by HPLC. The conversion rate of agar was about 80% and amount of total agarooligosaccharide was 0.88 mg/mL at Packed-Bed Reactor.

Physicochemical Properties of Agarooligosaccharides for Using as Food Stuffs (식품소재로서의 한천올리고당의 이화학적 특성)

  • A marine bacterium Bacillus cereus ASK202 showing a high agar degrading activity, was incubated in the culture medium containing agar. After incubation for 30 hr, the productivity of agarase in the culture broth reached to maximum value (160.8 units/L). As the results of TLC and HPLC analysis, agarooligosaccharides (degrees of polymerization 2, 4 and 6) were produced from the hydrolysis of agar by using the crude agarase. Physical and chemical properties of agarooligosaccharides were compared with the manufactured products of other oligosaccharides (fructooligosaccharide; isomaltooligosaccharide; maltotetraoligosaccharide) and agarooligosaccharides showed higher viscosity, higher contents of oligosaccharides, higher stability at low pH's and higher temperatures, and lower sweetness than other oligosaccharides.

Medium Supplementation and Atmospheric Condition for Growth of Campylobacter pylori isolated from gastric biopsy tissue (위점막 생검 조직에서 분리된 Campylobacter pylori의 성장을 위한 배지 첨가물 및 대기조건)

  • Experiments were conducted to define the optimal constituents of culture medium and atmospheric condition for growth of Campylobacter pylori. Two clinical isolates were streaked onto various media, incubated in two different atmospheric conditions (microaerophilic condition and carbon dioxide incubator), and growth was assessed semiquantitavely according to relative colony size and extent of growth through the streak. The growth obtained on Campy media, composed of GC agar base plus 1% hemoglobin, 0.2% activated charcoal, 1% IsoVitaleX, vancomycin 6mg/L nalidixic acid 20mg/L and amphotercin 2mg/L, was used as reference. Our conclusions were as follows: Tryptic soy agar base was not acceptable for the growth of C. pylori. As a sole supplementation, GC agar containing 1% hemoglobin was relatively adequate for the growth of C. pylori. The organism grew in both atmospheric conditions, but generally showed a scantier growth in the carbon dioxide incubator than under the microaerophilic condition, however GC agar containing 1% hemoglobin and 0.2% activated charcoal supported well the growth of C. pylori in the carbon dioxide incubator. The authors have found that the GC agar base supplemented with 1 % hemoglobin and 0.2% charcoal was the most satisfactory medium and a microaerophilic condition was optimal atmospheric condition for the growth of Campylobacter pylori in this study.

Comparison of Four Different Isolation Media for Staphylococcus aureus (황색포도상구균 분리배지 비교)

  • Performance test was carried out between selective media which are generally used in Staphylococcus aureus isolation from food. Sensitivity, determined according to the appearance of characteristic colonies when 30 different S. aureus strains were tested, resulted as Baird-Parker agar (RPF)> $Petrifilm^{TM}$ Staph Express plate> Baird-Parker agar> Mannitol salt agar. Also, the four different media showed the same selectivity because all tested media did not produce the false positive colonies. Recovery efficiency from the artificially inoculated foodstuff was almost the same for the tested media. Presumptive colonies were collected from the dried fishery product using Mannitol salt agar and collected strains were tested on 4 different selective agar. Almost presumptive strains did not show the false positive colonies except for S. carnosus ssp carnosus. This strain was identified as false positive colonies on Mannitol salt agar, Baird-Parker agar and $Petrifilm^{TM}$ Staph Express plate. But Baird-Parker agar (RPF) did not show the false positive colonies with the same strains. So, it was concluded that the Baird-Parker agar (RPF) has more higher selectivity than other tested media in this experiment.

  • Gracilaria verrucosa and Gelidium amansii collected from Wando, Yeosoo and Namhae districts Gracilaria sp. imported from Manila, and Ecklonia cava from Cheju island were investigated to raise the extraction yield of agar or alginic acid. The results are summarized as follows. In agar extraction from domestic seaweeds, Gelidium sp. showed 8-10% yield increase by gamma-irradiation whereas Gracilaria sp. exhibited no effect. In Manila seaweed, gamma-irradiation (1.5 Mrad) caused 25-30% increase in agar extraction and the properties of agar sample by these methods were acceptable. In alginic acid extraction from Ecklonia sp., gamma-irradiation (0.3 Mrad) showed 6% yield increase.

  • 최근 이상기후로 인한 집중호우의 증가로 인해 하천의 홍수방어능력을 초과하는 침수피해가 급증하고 있다. 특히, 제주도는 태풍의 길목에 위치하고 있는 강우량이 약 2,061mm에 달하는 우리나라 최다우 지역으로 평시에는 건천인 상태의 하천이 집중호우 시 급격한 유출 발생으로 하류도심지역에 하천 범람으로 인한 많은 피해가 발생하고 있다. 제주도에서는 이를 방어하기 위해 하도 내 첨두 홍수량을 조절하여 하류지역의 피해를 경감시키기 위한 구조물인 저류지를 상류에 설치하여 운영하고 있다. 하지만 저류지의 건설을 통한 실제 홍수조절효과에 대한 정량적인 분석은 미흡한 실정이다. 이에 본 연구에서는 태풍 '나리' 때 가장 큰 피해가 발생한 한천의 한천1, 2 저류지를 대상지역으로 선정하여 2차원 수치모형인 Nays2d Flood 모형을 활용한 한천 저류지의 운영에 따른 홍수조절효과를 분석하였다. 지형자료는 드론을 활용하여 측정된 약 7cm 간격의 정밀한 하상 측량자료를 사용하였고, 태풍 '차바' 발생 시 계측된 저류지내의 수심 측정 센서 자료를 활용하여 모형의 검보정을 수행하였다. 보정된 모의결과를 활용하여 태풍 '차바' 때의 저류지가 활용되는 모습을 재현할 수 있었고, 이를 통해서 현재 설치된 저류지의 구조적인 문제점과 운영방법에 따른 홍수조절효과 개선방안을 제시하고자 하였다. 또한 저류지 설계 시 사용된 100년 빈도의 설계 홍수량을 바탕으로 모의를 수행하여, 기존에 제시된 한천저류지의 홍수 저감량과의 비교를 통해서 실제 저류지의 운영에 따른 홍수조절효과를 정량적으로 분석하였다.

  • In an attempt to find a satisfactory medium which would facilitate abundant conidial production of Piricularia oryzae agar and liquid media containing various nutrients were tested as to the efficiency of conidial production. Among the media tested most number of conidia occurred on V-8 juice broth and tomato juice agar and the least on rice straw medium. Thus in conidial productivity, V-8 juice broth and tomato juice agar far exceeded the rice polish and yeast starch agar which were known to be best by previous workers.

  • 2010년 태풍 '뎬무'가 제주지역에 내습했을 때, 한천 상 하류에 위치한 하천유출량 관측소의 자료와 저류지 유입량 자료를 이용하여 한천저류지 운영이 하류지역 홍수저감에 미치는 영향을 분석하였다. 분석결과, 하류지역에서의 실제 하천수위는 3.44m를 기록하였으나, 수위-유출량 관계식, 유출전파속도, 저류지 유입량를 활용하여 저류지가 운영되지 않았을 경우 하류지역 하천수위는 4.16m로 예측되어, 저류지 운영으로 하천유출수의 일부가 저류지로 유입됨으로써 하류지역의 하천수위를 0.72m 하강시킨 것으로 분석되었다.

  • Hancheon reservoir, which is located upstream of Jeju city, has been built for flood mitigation after Typhoon Nari, 2007. To verify flood mitigating effect of the this reservoir on the downstream area, runoff analysis based on the measured data (two stream discharge monitoring stations and inflow data to the reservoir) is carried out during torrential rain followed by typhoon Dainmu, 2010. The stream water level was recorded as 3.14 m for the peak at the down gradient station. The stream water level under the assumption of absence of Hancheon reservoir is calculated as 4.16 m using the estimated rating curve, stream water propagation velocity, and the bypassed volume of water to the reservoir. This result shows that clear effect of reservoir operation which is capable of mitigating peak discharge in the downstream area.

Isolation and Characterization of a Marine Derived Bacterium Glaciecola sp. SL-12 Producing β-agarase (한천분해효소를 생산하는 해양유래 세균 Glaciecola sp. SL-12의 분리 및 특성)

  • A novel agar-degrading bacterium SL-12 was isolated from seashore of Kijang at Busan, Korea, and cultured in marine broth 2216 media. Isolated bacterium SL-12 was identified as Glaciecola genus by 16S rDNA sequencing with 98% identity. The optimum pH of the enzyme activity was 7.0 and the optimum temperature for the reaction was $30^{\circ}C$. The enzyme hydrolyzed neoagarohexaose to yield neoagarobiose as the main product, indicating that the enzyme is ${\beta}$-agarase. Thus, isolated bacterium and the enzyme would be useful for the industrial production of neoagarobiose.
