Studies on the Storage of Fat-Containing Foods (I) -Effect of Storage Factors on the Rancidity of Fried Instant Noodle- (유지함유식품(油脂含有食品)의 보존성(保存性)에 관(關)한 연구(硏究) (제1보) -보존조건(保存條件)이 인스턴트 Noodle의 산패도(酸敗度)에 미치는 영향(影響)-)

  • Fried instant noodles, manufactured on laboratory scale, preserved in sunlight, room temperature, removed package, at $30{\pm}2^{\circ}C$ and corrugated fiber board box to investigate the oxidative stability of fried products under a few factors of storage for 150 days. And obtained results were as follow. 1) Peroxide value P.O.V. and Acid value A.V. of fried instant noodles were increased slightly during the storage in orange color polycello package in the corrugated fiber board box in the dark room as shown in Fig. 1 and 2. They had not rancid odor and showed P.O.V. 18.8 after 150 days. 2) Preserving in room temperature removed package or with polycello package exposed to sunlight, P.O.V. and A.V. increased rapidly up to 100, 5 and they had rancid odor within 30 days. 3) Peroxide value showed increasing tendency on the storage at $30{\pm}2^{\circ}C$ and scattered light and rancid odor appeared from 90 to 120 days. 4) As peroxide value and refractive index increased, iodine value were decreased as Fig. 3. 5) In these test, P.O.V. is correlated with the organoleptic evaluation of rancidity however, they were not coincided exactly.

Quantification of nitric oxide concentration in the hemocytes of Manila clam Ruditapes philippinarum by using 4,5-diaminofluorescein diacetate (DAF-2) detection method (바지락 (Ruditapes philippinarum) 혈구의 일산화질소 (nitric oxide) 정량)

  • Nitric oxide (NO) is an important intra-intercellular signaling molecule that regulates many physiological processes and participates in the development some pathological conditions in animals. In this study, we compared different methods for determining NO concentration in the hemocytes of Manila clam Ruditapes philippinarum. For measuring the intracellular NO levels, we used the specific fluorescent probe 4,5-diaminofluorescein diacetate (DAF-2 DA), and the quantification methods that were compared were based on image analysis, spectrophotometry, and flow cytometry. NO concentration could be determined using all the 3 methods, and the concentration varied significantly depending upon the presence of NO regulators in the hemocytes; NO concentration increased in the presence of L-arginine, while it decreased in the presence of N-nitro-L-arginine methyl ester. In particular, it is found that estimation of NO using a flowcytometry is more economical, reliable and accurate compared to image analysis and spectrophotometry. Accordingly we believe that determining NO concentration by using flowcytometry will be useful in evaluating physiological and pathological conditions in marine bivalves.

The Association between Number of Present Teeth and Oral Function in Korean Adults Aged 55~84 Years (55~84세 한국 성인의 현존치아수와 구강기능의 관계)

  • The purpose of this study is to analyze the factors of oral function associated with number of present teeth (NT) in Korean elderly adults. The research was designed as cross-sectional study using the data of the 4th Korea National Health and Nutritional Examination Survey, and the samples were selected by stratified clusters sampling. The subjects aged 55 to 84 years were 4,780 that joined the oral examination survey and questionnaires. The respective variables reflecting socioeconomic status, subjective health status and oral function factor were analyzed and their distributional differences were confirmed by complex sampling logistic regression analysis. Multivariable liner regression models considering covariates were applied to explain the associations between the number of present teeth and other variables. Gender, educated level, illegal dental treatment, self-rated oral health and oral functional factors, including chewing behavior and speaking behavior, were associated with the number of present teeth (p<0.05). Subjects with better self-rated oral health had more NT than those with worse. Subjects with worse speaking behaviors had less NT than those with better. Interventions aiming to enhance oral health in elderly adults should consider socioeconomic, demographic, and oral functional factors.

Relationship between Mastication Problem and Quality of Life among Middle-aged Korean (한국 중·장년의 저작문제와 삶의 질의 관련성)

  • The purpose of this study was to investigate the relationship between mastication problems and quality of life among middle-aged Koreans. The study used data from the 6th Korea National Health and Nutrition Examination Survey(KNHANES VI). Data on 7,125 respondents aged 40 to 64 were analyzed to evaluate quality of life as measured by the Euro Quality of Life-5D(EQ-5D) index and mastication problems. Quality of life was classified as either low or high based on 0.96. The data were analyzed using SPSS 20.0. The prevalence of low quality of life was significantly higher in the group with mastication problems, at 41.9%, than in the group with no mastication problems(24.4%). After adjusting the general characteristics and oral health status, the odds of a lower quality of life were higher in the those with mastication problems, at 1.77 (95% CI: 1.53-2.06), than in those with no mastication problems. Mastication problem was significantly related to the quality of life. The studies are needed to reflect the oral factors in diverse age groups.

Greenhouse Whitefly and Thrips Management Model Using Natural Enemies in Semi-forcing Culture of Tomato (토마토 반촉성 시설 재배에서 천적중심 온실가루이와 총채벌레 관리모델)

  • To investigate the control effect of insect pests by natural enemies, sticky traps were used in commercial tomato greenhouses in Chun-cheon and the experiment fields of Gangwon-do Agricultural Research and Extension Services, Republic of Korea using semi-forcing culture (February to June). We selected low toxicity pesticides, environment-friendly agricultural materials (EFAM), and natural enemies of the study species, combinations of which have been previously used in farms to control insect pests. In this study, Trialeurodes vaporariorum and thrips, which are major agricultural insect pests, were studied in experimental greenhouses. The adult T. vaporariorum population was observed in mid-April and the population of thrips showed occurrences in early April. Regarding seasonal fluctuation, T. vaporariorum peaked in mid-May and thrips peaked after June. one insecticide, spiromesifen suspension concentrate (SC); one fungicide, cyazofamid SC; and two EFAMs containing neem tree extract were shown to be slightly toxic to Encarsia formosa and Orius laevigatus. The results of this study could be used to develop management models using natural enemies of control the insect pests; T. vaporariorum and thrips in semi-forcing culture of tomato.

In Vitro Maturation, Fertilization and Development of Mouse Oocytes Derived from In Vitro Grown Preantyal Follicles (체외성장된 Preantral Follicle에서 유래된 생쥐난자의 체외성숙, 수정 및 발달)

  • The objective of this study was to examine maturation, fertilization and developmental rate of the in vitro-grown mouse oocytes, and to compare these results with those of oocytes grown and matured in vivo. The preantral follicles isolated from 12-day-old mice were cultured on Transwell-COL membrane inserts. After in vitro growth and maturation, 72.5 % of oocytes grown in vitro produced polar body which can be comparable to in vivo growth (70.5 %). However, the mean oocyte diameter of the in vitro group (69.6$\pm$2.1$\mu$m) was smaller than that of the in vivo group (73.3$\pm$3.0$\mu$m). The fertilization rate was significantly lower (p<0.05) in the in vitro group (76.5%) than in the in vivo group (90.2%), however, there was no difference in the percentage of monospermic and polyspermic oocytes between two groups. The capacities of in vitro grown ova to cleave and develop to blastocyst were (57.8 and 14.4%, respectively) significantly lower (p<0.001) than those of the in vivo counterpart (84.4 and 56.6%, respectively). Moreover, the mean number of cells per blastocyst was significantly lower (p<0.05) in the in vitro group (39.0$\pm$10.8) than in the in vivo group (60.5$\pm$12.5). Live young were produced from transferred 2-cell embryos derived from in vitro-grown and matured oocytes. In conclusion, the results show that in vitro-grown oocytes did not achieve the developmental capacity of in vitro-grown oocytes.

Effects of Wasabi (Wasabia japonica Matsum) on the Physicochemical Characteristics of Baechu Kimchi during Fermentation (고추냉이가 배추김치의 발효 중 이화학적 특성에 미치는 영향)

  • In this study, we explored the application of Wasabi addition to baechu kimchi with regard to quality improvement, by reviewing the optimum Wasabi level and its effects on the product physicochemical characteristics during fermentation. The final weight percentages of Wasabi in the baechu kimchi were adjusted to 0, 2, 3, 4, and 5% per weight of baechu, respectively, and the samples were fermented for 35 days at $10^{\circ}C$. Following fermentation, the treatments over 3% Wasabi contained the highest pH and lowest total acidity values. Total vitamin C and reducing sugar content increased initially, up to certain fermentation times depending on the level of Wasabi contents within the treatments over 3% Wasabi, and decreased afterwards. The water soluble pectin content increased as the fermentation period increased; however, it decreased with increasing levels of Wasabi. In conclusion, the optimum levels of added Wasabi in the baechu kimchi were 3% and 5% per weight of baechu, to have a fermentation-retarding effect on the product.

Algal Contribution to the Occurrence of Refractory Organic Matter in Lake Paldang, South Korea: Inferred from Dual Stable Isotope (13C and 15N) Tracer Experiment (팔당호 난분해성 유기물에 대한 조류기원 유기물의 기여)

  • While a fairly large amount of organic matter is produced daily via phytoplankton photosynthesis in Lake Paldang, South Korea, knowledge of the role of algal-derived organic matter (OM) as a refractory OM source is not adequate. To understand the contribution of algal-derived OM to the refractory pool, biodegradation experiment and $KMnO_4$ oxidation experiment were conducted for 60 days using $^{13}C$ and $^{15}N$ labeled natural phytoplankton assemblage. The assemblage was collected from Lake Paldang on May 20, 2010. The photosynthetically produced total organic carbon ($TO^{13}C$), particulate organic carbon ($PO^{13}C$), and particulate nitrogen ($P^{15}N$) remained at 26%, 20%, and 17% of the initial concentrations, respectively, in the form of non-biodegradable organic matter. In addition, 12% and 38% of $PO^{13}C$ remained after $KMnO_4$ treatment on Day 0 and 60, respectively. These results indicate that photosynthetic products could be an important source of refractory organic matter after microbial degradation. Moreover, the microbially transformed algal-derived OM could contribute to the oxidation rate of the chemical oxygen demand.

Evaluation of the Availability of Bolting Angelica gigas Nakai (추대 참당귀의 이용가능성 평가)

  • Angelica gigas Nakai (AGN) is a perennial herb belonging to the family Apiaceae. Its root has been utilized as a traditional medicine in Korea. Also, it contains decursin (D) and decursinol angelate (DA) as major active components. This study was performed to compare the change in content of active components and antioxidant activity between bolting and non-bolting AGN. Bolting AGN (B1) and non-bolting AGN (NB1) were harvested in the end of August. Additional non-bolting AGN (NB2) was harvested in the end of October. The total weight of B1 was 728.7 g, which was 98.5% of NB2 weight. Next, the AGN root (B1, NB1 and NB2) was divided into main root and lateral root. And the AGN aerial part (B1, NB1) was divided into flower, leaf and stem. The two active components, D and DA, and antioxidant activity were analyzed. The D content of B1 was 0.35-1.33% according to the plant parts and the DA content was 0.29-1.07%. In addition, B1 flower and leaf showed higher antioxidant activity than NB2 roots. The results show that B1 contained 15-56% of total major components compared with NB2 main roots, suggesting that B1 could be used as a potential material.

Analysis of Mineral and Volatile Flavor Compounds in Pimpinella brachycarpa N. by ICP-AES and SDE, HS-SPME-GC/MS (ICP-AES와 SDE, HS-SPME-GC/MS를 이용한 참나물의 무기성분과 향기성분)

  • Mineral and volatile flavor compounds of Pimpinella brochycarpa N., a perennial Korean medicinal plant of the Umbelliferae family, were analyzed by inductively coupled plasma-atomic emission spectroscopy (ICP-AES) and simultaneous steam distillation extract (SDE)-gas chromatography mass spectrometry (GC/MS), head space solid phase micro-extraction (HS-SPME)-GC/MS. Mineral contents of the stalks and leaves were compared and the flavor patterns of the fresh and the shady air-dried samples were obtained by the electronic nose (EN) with 6 metal oxide sensors. Principal component analysis (PCA) was carried out using the data obtained from EN. The 1st principal values of the fresh samples have + values and the shady air-dried have - values. The essential oil extracted from the fresh and the shady air-dried by SDE method contain 58 and 31 flavor compounds. When HS-SPME method with CAR/PDMS fiber and PDMS fiber were used, 34 and 21 flavor compounds. The principal volatile components of Pimpinella brachycarpa N. were ${\alpha}$-selinene, germacrene D, and myrcene.
