Effects of the Addition of Amino Acids and FBS on the In Vitro Development of Korean Native Cow IVP Embryos (아미노산과 FBS의 첨가가 한우난자의 체외발달에 미치는 영향)

  • These studies were carried out to investigate the effects of the addition of amino acids and FBS as source of exogenous nitrogen fixation added to medium on in vitro production of blastocyst derived from bovine follicular oocytes. The base medium was TCM-199 solution for in vitro maturation(IVM) of bovine follicular oocytes and Fer-TALP solution for in vitro fertilization(IVF) and YS solution for in vitro culture(IVC). IVC used the fertilized oocytes of 24-hr culture (day 1) after IVF. Rmbryos were cultured in drop-culture that contained 25 embryos per 10 ${mu}ell$. The results obtained are as follows: 1 The developmental rates of fertilized oocytes to blastocyst that developed from YS solution with NEAA derived from MEM alone were higher than those of YS solution without NEAA. 2. The developmental rates of fertilized oocytes to blastocyst that developed from YS solution with EAA derived RPMI 1640 alone were significantly higher than those of YS solution without EAA (p<0.05). 3. When added to EAA on day 5 after NEAA supplementation on day 1, the developmental rates of hatched blastocyst and blastocyst to hatched blastocyst were improved. 4. When removed to EAA on day 3, day 4 and day 5 from medium after NEAA and EAA supplementation on day 1. the developmental rates of blastocyst to hatched blastocyst were reduced. 5. When added to FBS as source of exogenous nitrogen fixation, the developmental rates of blastocyst and hatched blastocyst that developed from the later culture higher(day 5) than those of the early culture.

Change of amino acids contents of Gastrodia elata Blume with harvest times and seed tuber (수확시기와 자마의 특성별 천마의 아미노산 함량 변화)

  • Gastrodia elata Blume, an achlorophyllous orchid plant, has been used in traditional medicine and harvests in spring and fall. Here we investigated the quantitative changes of amino acids in G. elata by harvest times and seed tubers. In the results, we found that there was not big difference in contents of total amino acids but the distribution of amino acids differed depending on harvest times. 19% of total amino acids were asparagin and valine, an essential amino acid, occupied 10% within total amino acids in the tubers harvested in October. Among amino acids in the tubers harvested in April, serine occupied 23% and arginine, an essential amino acid, occupied 10% within total amino acids in tuber. Interestingly, the use of sexually propagated seed tubers produced high concentration of total amino acids compared to vegetatively propagated seed tubers. As for sexually propagated seed tubers, essential amino acids contents similar to the tubes harvested in October and nonessential amino acids contents similar to the tubers harvested in April. In this study, we found that amino acids contents in G. elata tubers altered depending on various cultivation practices. Therefore if these results can be applied to food industry, the value of G. elata as a natural food resource will be enhanced to a great extent.

A Bioassay on Amino Acid Availability of Various Domestic Fish Meal (국내산 어분의 종류별 아미노산 이용율에 관한 연구)

  • Proximate, calcium, phosphorus and amino acid composition were determined for sardine fish meal (SM), herring fish meal (HM), anchovy fish meal (AM), alaskapolack by-product meal (ABM) and file fish by-product meal (FBM) produced domestically. These fish protein sources were fed to adult male of meat type (Waren G) broiler chicks to determine the true availability of amino acids. The true availability of essential amino acids were closely related to various fish meals with a few exceptions (e.g. valine, methionine, isoleucine and leucine). Average true availability of essential amino acids (9 amino acids) were 95, 93, 93, 91 and 91 percent for AM, SM, FBM; HM and ABM, respectively. Significant differences in the true availability of each individual amino acid were observed among fish meal tested (P<0.01). Lysine availability was low in HM, ABM and FBM whereas phenylalanine was the lowest in FBM and ABM, respectively. AM showed the highest true amino acid availability among all fish meal tested.

식품 소재로서 감국의 화학성분 조성

  • 다양한 식품 소재의 창출을 목적으로 그 기초자료로서 감국에 대한 화학적조성을 조사하였다. 일반성분은 감국의 수분함량이 10.51%, 조단백 9.38%, 지방 3.47%, 회분 4.87%, 환원당 13.12%, 조섬유가 9.03%, 비타민 C의 함량은 0.74%였다. 총 페놀 화합물의 함량 0.43mg%이였으며, 총 카로티노이드 함량은 0.84mg%로 나타났다. 아미노산 조성은 전체 아미노산 중 필수아미노산이 차지하는 비율은 41.42% 이고, 비필수아미노산의 비율은 58.58%였다. 감국의 아미노산 함량은 glutamic acid가 13.54%로 가장 많았고, 그 다음이 alanine(11.25%), aspartic acid(10.35%) 순으로 나타났다.   • Changes in proximate composition, amino acid, free sugar, reducing sugar and vitamin contents of Curcuma longa L. (autumn woolgeom in Korean) and Curcuma atomatica Salib (spring woolgeom) were investigated according to picking time. Moisture, crude ash, crude protein and crude lipid were increased and non nitrogen substances was decreased by extending the picking time. Contents and ratio of total amino acid to essential amino acids was increased by extending the picking time. Free amino acid was increased by extending the picking time. Ratio of essential amino acids to total amino acids was decreased. Fructose was gradually increased, and then glucose, sucrose and total free sugars were decreased by extending the picking time. Amino acid and sugar contents of Curcuma longa. L. were higher than those of Curcuma atomatica Salib. In contents of vitamin C and $B_1$, Curcuma longa L. was decreased and Curcuma atomatica Salib was increased by extending the picking time.

Comparisons of Physicochemical Composition of Korean and Chinese Crataegi Fructrus (한국산 산사와 중국산 산사의 이화학적 성분 비교)

  • This study was conducted to compare the major chemical components of Korea Crataegi fructrus (KCF) and Chinese Crataegi fructrus (CCF). Among the proximate compositions, the curde fat content of KCF was lower than that of CCF, whereas their crude protein, crude ash and carbohydrate contents were similar. CCF had a higher total free sugar content than KCF. The major free sugars of KCF and CCF were identified as fructose and glucose. The value of glutamic acid was greater in the amino acids of KCF and CCF, and KCF had higher total amino acids and essential amino acids contents than CCF. KCF also had a higher level unsaturated fatty acids than CCF. CCF had a higher organic acid content, but both KCF and CCF had high citric acid levels. and Chinese The vitamin C contents of KCF and CCF were 272.69 mg per 100 g and 262.38 mg per100 g, respectively. The mineral content of KCF was higher than that of CCF, in the following order : K > Ca > Mg > Fe. The results showed that KCF had higher total amino acid, essential amino acid, unsaturated fatty acid and mineral contents and CCF had higher free sugar and organic acid contents.

  • This study was conducted to investigate the effects of gender on sensory scores, fatty acid and amino acid of Berkshire pork A total of 40 pigs (60 kg) were divided into 2 group (gilt and barrow) and raised up to 110 kg live weight. Pigs were conventionally slaughtered, and then chilled overnight. Pork loin (longissimus dorsi) muscle was removed from each left side and meat qualities were evaluated. The results obtained were as follows; In sensory evaluation, color, marbling score, aroma and overall acceptability of brrow groups were significantly higher than that of gilt groups (p<0.05). In fatty acid analysis, linoleic acid and arachidonic acid of gilt groups were higher than those of barrow groups, EFA (essential fatty acid) and UFA (unsaturated fatty acid) were lower in barrow groups compared to the gilt groups. In amino acid compositions, alanine, tyrosine and arginine of barrow groups were significantly higher and threonine, proline and histidine lower than those of gilt groups (p<0.05).

  • Three experiments were conducted to show effects of adding amino acids to a broiler starter diet (0-3 weeks of age) low in protein. A diet of 22% protein with adequate levels of all essential amino acids was used as a reference diet. Supplementing an 18% protein diet with adequate amounts or methionine and lysine significantly improved performance of chicks. However, adding excessive amounts of these amino acids was detrimental, indicating that the excess aggravated deficiency or the third amino acid. Supplemental arginine tended to alleviate this aggravating effect.

  • This study investigated the seasonal changes of body composition and elasticity between wild and cultured brown croaker, Miichthys miiuy. The wild fish were analyzed and compared with cultured fish in moisture, crude protein, lipid, ash and many kinds of amino acids. Cultured fish was higher in moisture content and lower in crude lipid and protein content than those of wild one. The wild fish were more abundant in the total amino acid compositions than those of cultured one. As result E/A ratio there was a little significant differences between wild and cultured. In highly unsaturated fatty acid, EPA (Eicosapentaenoic Acid) and DHA (Docosahexaenoic Acid) content of cultured fish were higher than wild one. On the other hand, the gel strength, max weight and hardness of wild fish were higher than cultured one.
