The Dyeability and Antimicrobial Activity of Cotton Fabric Dyed with Sophora Radix Extracts on Skin Microorganisms (고삼 추출액를 이용한 염색 면포의 염색성와 피부 미생물 억제효과)

  • The aim of this study was to elucidate dyeability and antimicrobial activity of cotton fabrics dyed with Sophora Radix extracts according to various mordants. Dyes were extracted from Sophora Radix using ethanol. Then, cotton fabrics were dyed with extracts two times by post-mordanting method in which the extract was 60% (owf, the mordant was 3% (owf), L.R was 1:20, the temperature was 60~7$0^{\circ}C$, the time of dyeing was 60min., and the time of mordanting was 60min.. The dyeability was evaluated by surface color and color fastness. The skin microorganism was evaluated on S. sureus, B. subtilis, S. epidermidis, P. acnes, P. aeruginosa, E. coli, A. niger, C. albicans and T. mentagrophytes. The results are as follows; 1. When mordants were treated, surface color was 3.3Y to 0.1 GY in H (hue) value which indicated greenish yellow to yellow 2. The color fastness to perspiration, dry-cleaning, rubbing, and washing stain fabric showed 4~5 degree. The color fastness to light was improved to 4 degree by treatment of mordants. The color fastness to washing was 2 degree which was somewhat poor. 3. Cotton dyed with ethanol extracts was excellent on S. aureus, B. subtilis, S. epidermidis and p. antis. But that showed poor antibacterial activities on P. aeruginosa and E. coli such as gram negative baterials 4. Antibacterial activity of cotton fabrics dyed didn't be improved by treatment of mordant 5. Antifungal activity of cotton dyed with ethanol extracts was excellent on T. mentagrophytes. Especially, on T. mentagrophytes there was no growth of fungus during 72 hours in cotton dyed mordanting with SnCl$_2$.$_2$$H_2O$.

Complete genome sequence of multidrug-resistant Moraxella osloensis NP7 with multiple plasmids isolated from human skin (사람의 피부에서 분리한 다약제 내성이며 다수의 플라스미드를 갖는 Moraxella osloensis NP7 균주의 유전체 서열 분석)

  • Moraxella osloensis NP7 was isolated from human skin of a collage male and showed resistance to ${\beta}-lactam$ and aminoglycoside antibiotics. Herein, we report the complete whole-genome sequence and gene annotations of M. osloensis NP7. It possesses single circular chromosome and seven plasmids. Chromosome is 2,389,582 bp in length with the G + C content of 43.9% and encodes 2,065 protein-coding genes. The combined seven plasmids are 654,202 bp in size with the average G + C content of 40.5% and code for a total of 667 protein-coding genes. The chromosome of NP7 strain contains four ribosomal RNA operon copies, one transfer-messenger RNA gene, forty-seven tRNA genes, three riboswitch genes and three CRISPR arrays. Additional CRISPR array is found in the plasmid pNP7-1. The genes conferring resistance to ${\beta}-lactam$ and aminoglycoside antibiotics were predicted to reside in the plasmid pNP7-1.

Synergistic Antimicrobial Effect of Sargassum serratifolium (C. Agardh) C. Agardh Extract against Human Skin Pathogens (피부 병원균에 대한 톱니모자반 추출물의 항균 시너지 효과)

  • The object of this study was to develop an alternative way to treat human skin pathogens using marine algae. During this study, we observed that the ethanolic extract of the edible brown algae [Sargassum serratifolium (C. Agardh) C. Agardh] exhibited potential antimicrobial activity against pathogenic commensal bacteria related with acne vulgaris (Propionibacterium acnes, Staphylococcus epidermidis, Staphylococcus aureus and Pseudomonas aeruginosa), and Candida albicans which causes cutaneous candidiasis. Among the solvent-soluble fractions from the ethanolic extract, a hexane-soluble fraction showed the strongest antimicrobial activity against all tested human skin pathogens with MIC values ranging from 32 to $512{\mu}g/mL$. In addition, the hexane fraction exhibited a synergistic antimicrobial activity with commercial antibiotics used in the treatment of acne vulgaris or cutaneous candidiasis. Thus, this study suggests that S. serratifolium extract could be a potential source of natural antimicrobial agents or a pharmaceutical component against human skin pathogens.

The Beneficial Effects of Extract of Pinus densiflora Needles on Skin Health (솔잎추출물의 피부건강 개선효과)

  • Pinus densiflora Sieb. et Zucc. (P. densiflora) contains several phenolic compounds that exhibit biological activities, such as antimicrobial, antioxidant, and antihypertensive effects. However, the anti-inflammatory effect of P. densiflora on skin has rarely been reported. Malassezia furfur (M. furfur) is a commensal microbe that induces skin inflammation and is associated with several chronic disorders, such as dandruff, seborrheic dermatitis, papillomatosis, and sepsis. The aim of our study was to identify the anti-inflammatory effects of P. densiflora needle extracts on skin health subjected to M. furfur-induced inflammation. The methanolic extract of the pine needles was partitioned into n-hexane, EtOAc, n-BuOH, and water layers. We measured the anti-inflammatory effects (in macrophages) as well as the antioxidant, antifungal, and tyrosinase inhibitory activity of each of these layers. The antioxidant activity of the individual layers was in the order EtOAc layer > n-BuOH layer > water layer. Only the n-BuOH, EtOAc, and n-hexane layers showed antifungal activity. Additionally, all the layers possessed tyrosinase inhibition activity similar to that of ascorbic acid, which is used as a commercial control. The EtOAc layer was not cytotoxic toward the RAW 264.7 cell line. Interleukin 1 beta and tumor necrosis factor (TNF)-α expression levels in M. furfur-stimulated RAW 264.7 cells treated with the EtOAc layer were decreased markedly compared to those in cells treated with the other layers. Taken together, we believe that the needle extracts of P. densiflora have potential application as alternative anti-inflammatory agents or cosmetic material for skin health improvement.

  • Cutibacterium acnes is a member of normal flora of human skin, conjunctiva, intestinal tract, the external auditory canal as well as oral cavity. It has been identified as an opportunistic pathogen related to acne vulagris, endocarditis infections, sarcoidosis, brain abscess, periodontitis, and osteomyelitis of the humerus. C. acnes KCOM 1315 (= ChDC KB81) was isolated from a human jaw osteomyelitis lesion. Here, we present the complete genome sequence of C. acnes KCOM 1315.

Whitening Effects of Lactobacillus rhamnosus Associated with Its Antioxidative Activities (Lactobacillus rhamnosus 파쇄물의 항산화 및 미백효과)

  • This study was performed to investigate the whitening effects of Lactobacillus rhamnosus in addition to its antioxidative activities. The cytotoxicity of the Lactobacillus rhamnosus was 7.6% at 10.0% (v/v) concentration. Its cytotoxicity was lower than 3.2% of Lactobacillus casei when adding the same concentration. Lactobacillus rhamnosus exhibited high antioxidative activities at 14.9% of DPPH radical scavenging activity, and a lower reducing power was measured. Lactobacillus casei exhibited relatively lower antioxidative activities at 13.4%. The tyrosinase inhibition activity of Lactobacillus rhamnosus was observed at 31.3% when adding 10.0% (v/v), as compared to 17.7% for Lactobacillus casei. Lactobacillus rhamnosus demonstrated strong inhibition activity for melanin synthesis at 58.6% when adding 10.0% (v/v), while Lactobacillus casei increased to 80.6%. It was also observed that the high antioxidative activities of Lactobacillus rhamnosus were strongly correlated to whitening activities, due to the inhibition of both tyrosinase and melanin synthesis. These results support the expanded use of lactic acid bacteria as a functional bioresources in the cosmetics industry.

Screening of Anti-acne Activity of Natural Products against Propionibacterium acnes (피부 여드름 치료제 개발을 위한 천연물의 항균활성 검색)

  • Acne is a chronic inflammatory follicular disorder of the skin, occurring in specialized pilosebaceous units on the face, and Propionibacterium acnes, a strict anaerobic pathogen, plays an important role in the pathogenesis of acne. To develop a reliable and effective anti-acne agent, we have evaluated antibacterial activity of 500 plant extracts, prepared from 335 plants, against P. acnes. Based on the results of disc-paper method, 25 plant extracts, including the extracts of Chloranthus japonicus (aerial part), Sophora flavescens (radix), Evodia officinalis (fructus), Ginko biloba (semem), Morus alba (root bark), Aralia continentalis (whole) and Reynoutria elliptica (radix), were selected as possible sources of anti-acne agent. Among them, the extract of S. flavescens (radix) was finally selected and kuraridin and kurarinone were identified as major active compounds of S. flavescens. These results suggested that medicinal and wild plants could be the potential source of anti-acne agent.

Cnestis palala (Lour.) Merr. extract suppresses Propionibacterium acnes-induced inflammation (Propionibacterium acnes에 의해 유도되는 염증반응에서 Cnestis palala (Lour.) Merr. 추출물의 억제효과)

  • Acne is an inflammatory skin disease that occurs in puberty and young people. Propionibacterium acnes (P. acnes) is known to be a major cause of inflammation in acne. P. acnes proliferates within hair follicles blocked by overproduced sebum in the skin, and thereby activates monocytic cells to promote the secretion of pro-inflammatory cytokines. In this study, we investigated the possibility of Cnestis palala (Lour.) Merr. extract to diminish P. acnes-mediated inflammatory responses. We found that C. palala extract significantly attenuated P. acnes-induced pro-inflammatory cytokine expressions, such as $IL-1{\beta}$, IL-6, $TNF-{\alpha}$, iNOS, and COX-2 in mouse macrophage RAW264.7 cells. Moreover, we observed that C. palala extract inhibited $NF-{\kappa}B$ transcriptional activation, which is the major transcription factor of inflammatory cytokine expression. Therefore, it is expected that C. palala extract has a potential as a therapeutic agent or supplement for the treatment P. acnes-induced inflammatory responses.

Dietary effect of Lactobacillus plantarum CJLP55 isolated from kimchi on skin pH and its related biomarker levels in adult subjects (김치유산균 Lactobacillus plantarum CJLP55 섭취가 성인 남녀의 피부 산도 및 관련 구성 인자의 변화에 미치는 영향)

  • Purpose: The skin pH is maintained by epidermal lactate, free fatty acids (FFAs), and free amino acids (FAAs). As a significant determinant of skin health, the skin pH is increased (less acidic) under abnormal and aged skin conditions. In a search for dietary alternatives that would promote an acidic skin pH, this study investigated the dietary effects of Lactobacillus plantarum CJLP55 isolated from Korean kimchi on the skin pH, and epidermal levels of lactate, FFAs, and FAAs in adult subjects. Methods: Seventy eight subjects (mean age $24.9{\pm}0.5years$, range 19 ~ 37 years) were assigned randomly to ingest CJLP55, Lactobacillus strain from kimchi, (n = 39, CJLP group) or placebo supplements (n = 39, placebo group) for 12 weeks in a double-blind, placebo-controlled trial. Skin pH and epidermal levels of lactate, FFAs and FFAs were assessed at 0, 6 and 12 weeks. Results: Although significant decreases in skin pH were observed in both the CJLP and placebo groups at 6 weeks, the skin pH was decreased significantly only in the CJLP group at 12 weeks. In parallel, the epidermal level of lactate in the CJLP group was also increased by 25.6% at 12 weeks. On the other hand, the epidermal level of FAAs were not altered in the CJLP and placebo groups, but the epidermal level of total FFAs, including palmitic acid and stearic acid, was lower in the CJLP group than in the placebo group over 12 weeks. The changes in the other FFAs, such as palmitoleic acid and oleic acid, were similar in the CJLP and placebo groups over 12 weeks. Conclusion: Overall, a dietary supplement of CJLP55 promotes acidic skin pH with a selective increase in epidermal lactate in adult subjects.

A Study on the Distribution of Microorganisms in Department of Radiography (영상의학과 촬영실의 미생물 분포에 관한 연구)

  • To prevent the secondary hospital-acquired infection (cross-infection) from occurring in the general radiographic room in the department of radiology, the microbial measurement was conducted at the points making direct close contact with radiologists and patients. For the case of radiologists, the microbial measurement and incubation were focused on the x-ray tube handle of the radiation generating device, and, for the case of patients, the microbial measurement and incubation were focused on the chin supporting device, chest-contact point, and handle. Once disinfected with Aniosurf, the sterilized media were gathered and identified, and the microorganisms were confirmed. Based on the identification results, it was confirmed that the points making direct close contact with radiologists showed a value of 103 CFU for Proteus mirabilis, Staphylococcus epidermidis, Bacillus spp. and Candida spp., and that the points making direct close contact with patients showed a value of 103~5 CFU for Proteus mirabilis, Enterococcu faecium, Pseudomonas aeruginosa, NTM(Non-Tuberculosis Mycobacteria) and Candida spp.. It was also confirmed that the types and number of microorganisms gathered from the points making direct close contact with patients were greater. Fortunately, most of the involved microorganisms were observed to be on the skin surface and are known to become extinct when disinfected in accordance with the hospital-acquired infection control rules. However, since even minor exposure to such microorganisms may be lethal for patients with reduced immunity, caution must be taken. In particular, since the points making contact with patients showed a high level of microbial measurement, it was thought that it would be necessary for radiologists and personnel having frequent access to strictly disinfect the parts, such as instruments and handles, making frequent contact with patients. The purpose of this study was to announce the importance of safe microbial control in the radiographic inspection room in hospital, and this study is expected to be used as the baseline data for preventing hospital-acquired secondary infection and Nth infectious diseases.
