Microbial desulfurization characterlstics of a bituminous coal have been determined by using Thiobacillus ferrooxidans. The effects of process variables (such as coal pulp density, particle size and addition of surfactants) on pyrite removal have been investigated in both shake and airlift-bioreactor culture experiments. In shake experiments, pyrite could be removed over 78% for pulp densifies below 20% (w/v) and removed below 40% for pulp densities over 30% (w/v) in 8 days. Pyrite removal decreased with increasing pulp densities, and it also decreased sharply with increasing particle sizes. In airlift bioreactor experiments, pyrite at 50% (w/v) pulp density could be removed about 50%. Its value is much higher than 15% at the same pulp density in a shake experiment. With addition of surfactants, pyrite removal was enhanced in shake experiments significantly, whereas it was slightly decreased in an airlift bioreactor experiment.
The metabolites released from cultures of rhizosphere bacteria can inhibit plant growth. Bacillus subtilis IJ-31 inhibited plant growth by the production of hydrocinnamic acid (HCA). The production of HCA by plant-growth inhibiting rhizobacterium B. subtilis IJ-31 was optimized. $90.5\;{\mu}g/ml$ of HCA was obtained under the condition of 1% rice bran as carbon source, 0.5% tryptone as nitrogen source, 0.1% $ZnCl_2$ as metal source at $37^{\circ}C$ for 60 h (pH 7.0). The optimal condition for the HCA production by B. subtilis IJ-31 in the jar fermenter was established using response surface methodology (RSM) of statistical analysis system(SAS) program. The production of HCA by B. subtilis IJ-31 in the jar fermenter culture reached $102.99\;{\mu}g/ml$ when 2.24% soil extracts was added and agitation speed was 290 rpm under the same condition. And the experimental value of HCA production is $102.5\;{\mu}g/ml$ in the same culture condition. The production of HCA by B. subtilis IJ-31 is higher as 12% than that from the flask culture.
The quantitative and rapid method for measuring the biodegradation of polymer materials in soil was developed. In this study, cellophane film was used as a model biodegradable polymer and the biodegradation was assayed by measuring the amount of glucose which was produced by a hydrolysis reaction using HCl after collecting the film from soil. Cellophane film was degraded 41.2% in 4 months during winter while it was degraded 76.5% in 2 months during summer. It means that biodegradation in soil is affected by environmental conditions. The biodegradation was also measured in an incubator (30$^{\circ}C$, humidity 50-55%) to exclude the environmental variations. Cellophane film was degraded 94% in that condition in 40 days. The biodegradation showed the first order kinetics and the rate constant was 0.067 (1/day). Acceleration of the biodegradation in soil was also studied. We added cultured soil microorganisms or nutrients such as N, P, and S into the soil. While the addition of microorganisms showed the temporary increase of rate constant, the addition of nutrients not only showed the increase of rate constant from 0.096 (1/day) to 0.21 (1/day) but also maintained the effect continuously.
Altogether twenty isolates were collected from Wakayama, Japan. The optimum mycelial growth of I. obliquus was observed in BMYA and MCM. The optimum temperature and pH for the mycelial growth were $25^{\circ}C\;and\;6.0{\sim}7.0$, respectively. The optimum carbon and nitrogen sources were dextrose and yeast extract, respectively. Similarly, the optimum mineral salt was $K_{2}HPO_{4}$. The optimum number of mycelial discs for the mycelial growth was $6{\sim}7$ per 100 ml. Similarly, the optimum culture period was $21{\sim}22$ days in liquid broth. The optimum brown rice : water ratio was 1 : 1. No difference in mycelial growth was observed in all the four types of tree stumps used.
Encapsulated Aspergillus niger was prepared in order to inspect the effect of oxygen supply on the production of citric acid. A. niger cells which had been immobilized in the calcium alginate capsule grew and mycellia penetrated through the capsule membrane after two days of cultivation and covered over all of the capsule after eight days. The mycellia became loose when the nitrogen source was sufficient of oxygen was deficient. The larger amount of encapsulated cells were put into a given growth medium, the smaller quantity of citric acid was produced. The increase of volumetric oxygen transfer coefficient from 1.8 $hr^-$ to 2.55 $hr^-$ in the flask culture accelerated cell growth rate but did not influence the production of citric acid. The high oxygen supply rate($k_La:\;150\;hr^-$) in the concentric air lift reactor hastened the growth of cells and hindered the production of the citric acid. The reduction of nitrogen source level in the growth medium in the concentric air lift reactor increased citric acid production by 40 percent of that of flask cultivation and the culture period was shortened by 3 days. The variation of the geometry of the concentric air lift reactor did not influence the growth rate of encapsulated cells and production rate of citric acid.
This study was conducted to establish a practical masspropagation system of Phalaenopsis clones from basal shoot segments. The frequency of PLB (protocorm like body) induction was compared with various explants. Basal shoot segments showed the most successful result of 45%, while root tips, stalk node segments, stalk leaves and mature leaves represented low frequency (below 5%). The PLB induction ratio in the culture of basal shoot segments was examined with 11 different Phalaenopsis varieties, and the majority of varieties, including pink flower lines, showed an about 30% rate of PLB formation. Especially, when whole basal shoot parts without cutting were inoculated onto PLB induction medium, giant PLB was induced from explant. This giant PLB was green color and big in size compared with normal PLB. When dissected giant PLB segments inoculated onto PLB multiplication medium, only normal size of PLBs were induced from them. PLBs induced by basal shoot culture were transferred onto proliferation medium and then shooting medium, from which normal plants were formed. Therefore, this culture method is considered as effective and practical protocol for Phalaenopsis mericlone production. In addition, it is suggested that clones of an infinite number can be produced consecutively by this culture system through repeated cycles of PLB induction and proliferation using the basal shoot segment of flask plant.

Tapioca alcoholic distillery waste was utilized as dual purposes to produce citric acid and to reduce the amount of waste to be treated. Primarily an attempt was made to optimize the process conditions by Aspergillus niger in shake bath. The effects of pH, temperature, nitrogen and phosphorus sources on citric acid production were investigated. Maximum concentration of citric acid was made at temperature of $30^{\circ}C$ and pH of 4.3, while maximum cell dry weight was obtained at $35^{\circ}C$. The addition of methanol or ethanol to culture medium promoted citric acid production remarkably, but the addition of $NH_4NO_3,\;KH_2PO_4$ and Manganese as mineral source decreased the acid production.

The main objectives of this research were to study the cultural and nutritional characteristics of Tricholoma matsutake and to establish its liquid culture system. The optimum growth of T. matsutake was observed in HA and TMM agar media. Similarly highest growth was observed in PDB and TMM liquid media. The optimal temperature for the mycelial growth was $25^{\circ}C$. The most suitable carbon source was dextrin among 12 different carbon sources tested. Yeast extract and peptone were best nitrogen sources among 17 different sources tested. The optimum mineral salts were $Fe_{2}(SO_{4})_{3}{\cdot}H_{2}O$ and KCl among 9 different sources tested. Shaking culture gave higher mycelial growth compared to stationary culture. Similarly, optimum medium amount for shaking culture was 100 ml per 250 ml flask. The highest mycelial growth was obtained when $5{\sim}7$ mycelial discs were inoculated in 100 ml of medium and incubated for $8{\sim}9$ weeks, respectively. The highest proportion of mycelial growth was observed at 40 : 1 ratio of medium to inoculum volume in 8 l air-lift fermenter.

Ginsenosides in ginseng berry has been known as functional materials showing physiological effect to the human. Specially, ginseng berry contains plenty of ginsenoside Re, but the study of extraction processes were not enough performed. Accordingly, the purpose of this study was to establish the optimized extraction process for obtaining ginsenoside Re from ginseng berry. The extraction process of ginsenosides was performed in 250 mL extraction flask containing 150 solvent and 10 g of dried ginseng berry. The extracted ginsenoside Re, Rg1 and Rd and total crude ginsenosides from ginseng berry were evaluated by TLC according to the treated conditions (the ratio of alcohol to water, extraction temperature, extraction period, and extraction times). Optimized conditions for extraction was 70% to 30% of the ratio of alcohol to water, $80^{\circ}C$ of extraction temperature, 4 h of extraction period, and 2 times of extraction frequency. The amount of total crude ginsenosides of the extract obtained from the optimized process was 88.6 mg/g based on dried ginseng berry. The composition of ginsenosides from the extracted was 5.5% of Rb1, 5.2% of Rc, 14.3% of Rd, 51.5% of Re, 8.1% of Rf, and 15.7% of Rg1. A protopanaxtriol ginsenosides of whole ginsenosides extracted was about 80%.

이 연구는 x-선 투과영상을 이용하여 홍삼의 내공을 검출하기 위하여 수행되었다. 이를 위하여 x-선관에 조사되는 x-ray 양의 차이에 따른 위치별 밝기 값 차이를 보정하고 내공 검출에 적합한 영상처리 알고리즘을 개발하였다. 주요연구 결과를 요약하면 다음과 같다. 1) x-선관에 조사되는 x-ray양은 중심에서 가장자리로 갈수록 감소하며, 이것은 위치에 따른 gray 값의 분포가 달라지게 되는 원인이 되었다. 2) 홍삼을 2치화 하기 위해서는 동일한 x-선 조사강도에서 입력된 원 영상에서 빈 영상을 뺀 감산 영상을 만들어 줌으로써 가능하였다. 3) 조사강도 별 감산영상처리 결과 36kV/4.15mA와 39kV/4.15mA에서는 조사량이 많아서 홈삼 지근 부분의 영상이 손실되는 경우가 발생하였다. 4) 내공 검출을 위해서는 정상부분과 내공부분의 자기 값 차이를 크게 하는 전처리 영상을 만들 필요가 있었고, multiple 감산영상에서 뚜렷한 골짜기가 나타났으나 주근의 내공부분의 밝기값 보다 지근의 정상부분의 자기 값이 더 낮게 나타나 이를 보정하기 위하여 홍삼 위치에 따라 부분 보정된 알고리즘을 개발하였다. 5) x-선관의 놓인 위치에 따라서 내공판정시험 결과 중심에 있을 때에 비해 가장자리에 있을 때는 영상이 일부 손실되었으며, 중심 위치에서 서로 다른 굵기의 홍삼에 대해서는 모두 양호한 결과가 나타났다. 6) 완전한 홈삼으로 내공판정시험 결과 내공주위의 정상부분가지도 일부 내공으로 잘못 검출되었으나 이것을 재차 line profile에 의해 한 라인씩 문턱 값을 설정하여 내공만을 정확하게 판정하는 알고리즘을 개발하였다.양체의 접종작업은 모든 배양실이 인력에 의존하였으며, 배양체를 배지와 분리하여 불필요한 부분을 제거하고 배양작물에 따라 생육정도를 2~3등급으로 구분하여 배양용기의 배지 위에 치상하는 과정으로 수행되었으며, 작업능률은 호접란의 경우 배양병에 25본을 접종하는데 시간당 6병, 심비디움은 원형 플라스크에 25본을 접종하는데 시간당 10병 정도였다. 바. 식물체의 대량증식에 사용되는 플라스크, 배양병, PE용기 등 배양용기의 세척작업은 농원의 1개배양실에서 간이식 세척기, 이 외의 9개배양실은 모두 물에 담겨 두었다가 세제와 브러쉬 등을 사용하여 인력으로 세척하고 있어 생력화 기술개발이 요구되었다.도가 빠를수록 건조속도가 빨라졌으며, 건조에너지도 1,334kcal/kg.water로 비슷하게 소요되었다. 마. 시험구와 대비구의 건감률은 시험구에서 1.08~1.36w.b./h로 나타나 대비구보다 약 9.9~18.3%가 높게 나타났고, 건조에너지는 10.2~14.6%가 절감되었다. 발아율은 열풍온도가 낮을수록 높게 나타났고 시험구가 대비구보다 발아율이 낮게 나타났으며, 동할률 증가량도 원적외선.열풍 복합건조방법이 높게 나타나 이것은 곡물 표면에 원적외선 방사에의한 복사열이 전달되어 열장해를 받았기 때문으로 판단되며, 금후 더 연구하여 적정 열풍온도 및 방사체 크기를 구명해야 할 것이다.으로 보여진다 따라서 옻나무 유래 F는 포유동물의 생식기능에 중요하게 작용하는 것으로 사료된다.된다.정량 분석한 결과이다. 시편의 조성은 33.6 at% U, 66.4 at% O의 결과를 얻었다. 산화물 핵연료의 표면 관찰 및 정량 분석 시험시 시편 표면을 전도성
