Comparison of Mycelium Cultivation and Fruiting Body Characteristics of Lentinula edodes According to the Sawdust Media Nutrients and Inoculation and Cultivation Conditions (표고 톱밥배지 영양원, 접종 및 배양조건에 따른 균사배양 및 자실체 특성 비교)

  • The cultivation conditions of shiitake mushrooms (Lentinula edodes) influence the production and quality of fruiting bodies. We conducted this study to improve the productivity and quality of shiitake mushrooms by modifying the cultivation conditions. Two types of spawns (sawdust and liquid spawn) were used, and corn flour was used as a nutritional source for the sawdust medium. A blue light-emitting diode (LED; 300 lux) was also used instead of a white LED during the incubation period. Sanbaekhyang was used as the experimental variety. When using corn flour, the mycelial growth rate increased 1.1 to 2.7 times the growth rate of the control up to 21 days of incubation, and the weight loss rate of the media was also higher. Mushroom productivity increased 1.2 times when the liquid spawn was used compared to when the sawdust spawn was used, and the blue LED also increased fruiting body production by 1.1 times compared to the white LED. Mushroom productivity increased when the liquid spawn was used, and the blue LED also increased fruiting body production. Fruiting body weight and the size of the cap were greater when sawdust spawn was used. The fruiting body weight and the stipe diameter were greater when the blue LED was used. Taste analysis showed that the saltiness increased when corn flour was used, and the sourness increased when the blue LED was used.

Comparison of vitamin K contents in different meats commonly consumed in Korea (국내에서 소비되는 육류의 부위별 비타민 K 함량 분석 및 비교)

  • Vitamin K is a group of fat-soluble vitamins that naturally exist in phylloquinone (vitamin K1) and menaquinone (vitamin K2). In this study, the vitamin K content in different meats commonly consumed in Korea was analyzed using HPLC, and the analytical method was validated. Vitamin K1 was not detected in any of the meat samples. Vitamin K2 contents in different cuts of beef ranged from 0.00 to 5.87 ㎍/100 g, whereas the corresponding value in different parts of chicken ranged from 16.59 to 46.64 ㎍/100 g. In the case of pork, vitamin K2 contents varied from 4.33 to 22.90 ㎍/100 g. Among the different types of meat, the highest vitamin K2 content was found in boiled chicken meat and skin (46.64 ㎍/100 g). The analytical method was found to be reliable and had high accuracy. These results provide accurate nutritional information and contribute a food composition database for meat consumption.

  • Sampling inspection methods for quality control have been proposed a lot in the industry. However, the sampling inspection method for reliability, which is a quality over time, has been relatively less presented, and there are not many literatures that are clearly summarized. Therefore, this paper focuses on the reliability conformity test to verify that the reliability evaluation scale value of the target is satisfied during the reliability test. To this end, first, we look at the conditions that both consumers and producers can satisfy in terms of the OC curve and find out what sampling methods satisfy the desired level of producer risk and consumer risk. Next, two methods of the reliability sampling methods such as attribute and variable reliability sampling methods are examined. Specifically, the attribute reliability sampling method is a form of sampling plan where n samples are tested for a certain period of T hours and the lot is accepted if the number of failures is less than or equal to a certain number c. On the other hand, the variable reliability sampling method is a form of sampling plan where the lot is accepted if the reliability evaluation scale such as MTBF satisfies a certain standard. Both sampling plans may also use inspection tables.

Cultural characteristics on collected strains of Lentinula edodes and correlation with mycelial browning (표고 수집균주의 재배적 특성 및 갈변과의 상관관계)

  • Shiitake mushroom (Lentinula edodes) is usually cultivated on the oak log. Log cultivation of this mushroom is getting difficult to get oak logs and has a weak point of its long cultivation period. Recently sawdust cultivation is getting increase. It is important to make mycelia browning on the substrate surface. This browned surface in sawdust cultivation plays an important role like as artificial bark of the oak log, which protects the other pests and suppresses water evaporation in the substrate. The period for mycelia browning is so long that the sawdust cultivation of Shiitake mushroom can not spread well into the mushroom farms. The development of methods for the rapid mycelia browning is quite required. In this article we would like to find cultural characteristics of collected strains and to see the correlation with mycelial browning. Mycelial growth in the media was different according to kinds of media and strains. The optimal temperature on mycelial growth was $20-25^{\circ}C$. Browning patterns of mycelium under 200 Lux seemed to be used for a key to differentiate the strains for sawdust cultivation. Browning period was 30-40 days in the agar media and 70-100 days in the sawdust bag cultivation. When we considered the productivity and the other characteristics, ASI 3046 is the best for the bag cultivation. Significance between mycelial growth and browning was not accepted, but that of mycelial growth between on PDA and sawdust was accepted. Browning period on the PDA and sawdust showed a strong relationships. These results suggested that the browning habits could not be depend on the difference of media, but on their own properties. To select the strain showed fast browning can be done by using agar media for saving time.

  • 1. Objectives and Importance. Many silkworms have been damaged by nuclear polyhedrosis virus diseases thoughout the country every year causing a decease in cocoon production by approximately ten per cent per year. The damage caused by the infections virus has occured in spite of complete disinfection. In this respect, it is well known it is impossible, at the present time, to protect the silkworm from these virus infections through chemical and physical control methods. Therefore, this author has attempted to solve this urgent problem from the view point of heredity and breeding, discovering the different resistances and heritabilities among 120 stains collected from throughout the country, and selecting the ones with highest resistance for the basic materials in the silkworm breeding. 2. Results of work 1) The strains with strong resistance to the nuclear polyhedrosis virus diseases are N$_4$, N$\sub$15/, N$\sub$48/, C$\sub$55/ and Em. the log ED$\sub$50/ values of them vary between 0.799 and 1.611. The susceptible strains are N$\sub$20/, C$\sub$62/, N$\sub$76/, N$\sub$79/ and C$\sub$108/, the log ED$\sub$50/ values of them vary between 5.159 and 7.258. (Reference Table 4) The Japanese strain with a log ED$\sub$50/, value of 3.770 is the strongest, followed by the Chinese strain with a log ED$\sub$50/ value of 3.564. The weakest is the European strain with a log ED$\sub$50/ value of 3.3381. The direction coefficient of the regression equation of the susceptibility varies between 0.1 and 0.6, the uniformness of the resistance of the preserved strains of this country is comparatively low. The hereditary henomena of the resistance of each strain and the conerete method of its application for silkworm breeding main the subjects for later studies. 2) The content of water and ash in silkworm has not been correlated with the capability for resistance to the virus diseases(Reference. Table 8). but it is very significantly correlated with mortality rate (in common reaning). In the case of the silkworms which have just completed the fourth moulting the content of water and ash is not related to the mortality rate. In the case of the silkworms which have just completed the third moulting, however, the water( +0.326) and ash (+0.362) registered a high significance. The ash content in the first ($\div$0.520) and second ($\div$0.386) moults is highly significant but water content in both cases is not significant (Reference Table 7). 3) The No. 205 strain proved to be the best in character among the various F$_1$ hybrids. No. 204 was very good in strength but a little lower in cocoon character than the control. No. 212 was a little low in coccon character and mortality was average, but the cocoon harvest was the best among all the varieties offered for (Reference Table 9). 4), In short, the above mentioned strains which are known to have strong resistance to the virus disease are expected to provide basic data for breeding strong varieties. 청년기 여성의 의류상표선택과 가정환경과의 관계 연구

  • 본 연구의 목적은 청년기 여성의 국내 및 외국 의 류상표선태과 가정의 가치지향성 및 물리적 가정환 경과의 관계를 조사하며. 상표선택에 따른 상표결정 동기의 차이를 파악하고 의복구매 후 만족도에 영향을 주는 특성을 조사하는 데 있다. 연구방법은 질문지법을 사용하였으며, 조사대상은 서울시내 여고생과 여자대학생 408명이었다(여고생 224명, 여대생 184명). 가정환경변인 측정도구는 정원식의 '가정환경진단검사'중에서 가정의 가치지향 성을 사용하였으며. 사회상승주의, 물질주의, 전통주 의에 관한 내용을 포함하였다. 상표선택은 최근에 구 입한 캐주얼웨어의 상표에 대한 내용과 구입희망 상 표를 조사하였으며, 상표결정동기는 5점 척도의 10 문항을 사용하였다. 가정의 물리적 환경은 부모의 학력, 사회계층, 주거공간을 조사하였으며, 인구통계적인 변인으로 학년, 연령, 용돈을 조사하였다. 조사기 간은 2000년 4월이었다. 본 연구의 결과는 다음과 같다. 첫째, 여고생 및 여대생이 구입한 의류상표는 국 내상표가 24.0%. 외국 상표가 37.5%. 상표명을 기억 하지 않는 상표무관심 집단이 38.5%였다. 구입희망 상표는 외국상표가 48.8%. 국내상표가 26.0%. 무관 심집단이 25.2%로서 구입한 상표보다 구입희망상표 의 경우 외국상표가 더 높은 비율을 나타냈다. 둘째, 가정의 가치지향성은 국내 및 외국의류상표 구입집단에 따라 유의한 차이가 없었다. 그러나 가정 의 가치지향성 중에서 사회상승주의는 구입희망 집 단에 따라 유의한 차이가 있어, 외국상표 구입희망 집단은 가정의 사회상승주의가 높았고, 상표무관심 집단은 사회상승주의가 낮았다. 따라서 사회적 지위 상승을 추구하는 가치관을 지닌 가정의 청소년들은 부모의 가치관에 영향을 받아 외국상표 의류를 희망 하여 의복을 통해 신분상승을 추구하려는 정도가 높은 것으로 해석된다. 셋째, 국내 및 외국 의류상표 구입집단운 부모의 학력, 가정의 사회계층 및 주거공간과 유의 한 관계가 있었다. 아버지와 어머니의 학력이 대졸 이상의 경우 는 외국상표를 더 많이 구입하였고, 부모띄 학력이 고졸인 청소년은 상표 무관심집단이 많았다. 또 상류 층은 외국상표 및 국내상표를 구매한 비율씨 비슷하 게 높았으나 하류층은 상표 무관심집단이 더 많은 편이었다. 구입회망상표와 사회계층 및 주거공간과 는 유의한 관계가 없었다. 따라서 청년기 여성은 가정의 사회계층과 관계없이 외국상표의 구입을 희망 하는 정도가 높다고 할 수 있다. 넷째, 외국상표 구입 집단은 국내상표 구입 집단 보다 상표를 미리 결정하는 경우가 더 많았고. 백화 점에서 구입하는 정도가 더 높았다. 또한 여대생은 의류상표선택과 용돈이 유의한 관계가 없었으나 여고생은 유의한 관계가 있었다. 다섯째. 외국상표 구입집단은 국내상표 구입집단 보다 주위 사람의 상표와 상표의 명성이 더 높은 구입동기로 작용하였으나. 품질, 유행, 할인가, 디자인, 가격, 착용감은 유의한 차이가 없었다. 여섯째, 상표결정동기 중 할인가 구매는 가정의 사회상승주의 및 물질지향주의와 유의한 정적 관계 가 있었고, 적절한 가격도 물질지향주의와 정적 관계 를 나타냈다. 가정의 전통주의는 의복의 디자인과 부 정적인 관계, 상점의 위치와는 정적관계가 있었다. 따라서 전통주의적인 가치관을 지닌 가정의 자녀는 편리한 상점의 위치가 상표결정동기로 작용하는 데 영향을 주는 특성임을 알 수 있다. 일곱째, 의복만족에 동시에 영향을 주는 요인은 디자인, 면안성, 타인의 착용( - ). 상표의 명성, 외국 상표선택, 의복품질로 나타났으며. 6개 변인을 통한 설명력은 24.4%였다. 즉 상표를 결정할 때 디자인, 편안성, 상표의 명성, 의복품질을 고려하며. 주위사람들이 착용한 상표를 고려하지 않을수록 만족도가 높았으며, 외국상표를 선택한 집단일수록 만족도가 높았다. 종합적으로 볼 때 가정의 사회상승주의가 높은 청년기 여성은 외국상표 구입을 희망하는 비율이 높고 부모의 사회경제적 지위가 높을수록 외국상표 구입 빈도가 높았으며. 외국상표를 구입한 여성은 의복구 매 후 만족도가 높았다. 또한 청소년들이 외국 의류 상표를 구매하는 데에는 상표의 명성과 타인의 옷차 림이 영향을 주었으나, 타인의 착용에 영향을 받는 정도가 높을수록 의복 구매 후 만족도는 낮아진다고 할 수 있다.

Quality and Sensory Characteristics of Sweet Persimmon Distilled Spirits (단감 증류주의 품질 및 관능 특성)

  • The objective of this study was to investigate the quality and sensory characteristics of sweet persimmon mashes and spirits fermented by three different yeasts [Saccharomyces species, labeled as LB (Lalvin BM $4{\times}4$), LD (Lalvin DV10), and UC (Uvaferm CM)]. In the production of sweet persimmon wines, UC had the highest alcohol content of 10.20% (v/v) after 9 days of fermentation (P<0.05). The range of pH after fermentation was 3.63~3.75. The total aerobic bacteria and yeast contents increased until day 3 and then continuously decreased to approximately 8.60 log CFU/mL and 8.20 log CFU/mL, respectively. Reducing sugar contents were 3.37% in UC, 3.91% in LD, and 4.05% in LB after fermentation. Total sugar contents were 4.89% in UC, 6.24% in LD, and 5.47% in LB after fermentation. Two-stage single-pot distillation was conducted to produce sweet persimmon spirits. While conducting second distillation, spirits were collected every 100 mL fraction. The alcohol contents gradually decreased as more fractions were collected. The amounts of acetaldehyde were 226 mg/L for LD, 225 mg/L for LB, and 310 mg/L for UC in the first fraction. LD produced the highest volume (677.8 mL) of alcohol in its body part in comparison with LB (408.0 mL) and UC (445.4 mL). In the descriptive analysis, UC had stronger intensities of sweet aroma, sour aroma and taste, and persimmon flavor (P<0.05). Persimmon characteristics seemed to be well characterized by UC. In conclusion, LD was the most efficient in terms of production cost, whereas UC would be used to produce a more flavorful sweet persimmon spirit.

Effects of different cooking methods on folate retention in selected mushrooms (다양한 조리법에 따른 버섯류의 엽산 리텐션)

  • This study was performed to investigate the effects of different cooking methods (boiling, roasting, stir-frying, and deep-frying) on folate retention in 6 kinds of mushrooms (Beech-, button-, Juda's ear-, oak-, oyster-, and winter-mushrooms) frequently consumed in Korea. In order to assure reliability of analytical data, trienzyme extraction-L casei method was verified and analytical quality control was also evaluated. Folate contents of mushrooms varied by 6.04-64.82 g/100 g depending on the type of mushrooms. and were significantly affected by cooking methods. Depending on cooking methods, folate contents of mushrooms decreased by 22-48%, 2-31%, and 17-56% for Juda's ear-, oak- and oyster-mushrooms, respectively, while 17-90% of folate was increased in Beech mushroom. Overall, the largest weight loss was found in boiled mushrooms, but the lowest one in deep-fried samples. True folate retention rates considering processing factor were less than 100% for all cooked mushrooms except for Beech samples. Overall, folate loss was the largest by boiling with water but the smallest by deep-frying. Both accuracy and precision of trienzyme extraction-L-casei method were excellent based on a recovery close to 100% and coefficient variations less than 3%. Quality control chart of folate analysis (n=26) obtained during the entire study and an international proficiency test (z-score=-0.5) showed that trienzyme extraction-L casei method is reliable enough for production of national folate database.

  • Although sawdust cultivation of shiitake (Lentinula edodes) is becoming more common, it is insufficiently competitive in spring and autumn, the best time to breed shiitake. Thus, it is urgently needed to develop a technique for all year round cultivation of shiitake using mushroom growing beds. In the present study, the temperature changes according to the location of shiitake cultivation facilities were investigated. We confirmed that a refrigerator, an air conditioner, triple membranes, shiitake cultivation beds, fog nozzles which were installed in the shiitake cultivation facilities play an important role in keeping the low temperature. Bag cultivation of shiitake was tested in temperature variation from $14^{\circ}C$ to $29^{\circ}C$ with a $3^{\circ}C$ interval to know its cultivating temperature range in hot summer season. In summary, the sawdust cultivation of shiitake is possible when the temperature difference between top and bottom is maintained below $1^{\circ}C$. And the temperature of the shiitake cultivation facilities should be maintained below $23^{\circ}C$ in the induction period for fruitbody formation.

Measurement of Radiation Using Tissue Equivalent Phantom in ICR Treatment (자궁강내 근접방사선조사시 인체조직등가 팬톰을 이용한 방사선량 측정)

  • This study is to compare A point doses in human pelvic phantom by film dosimetry, computer planning and manual calculation by using of along-away table. We developed tissue equivalent human pelvic phantom composed of four pieces of cylindrical acryl tubes with water, to simulate intracavitary radiation (ICR) in patients with cervix cancer. When the phantom assembled from 4 pieces, it has a small space for inserting Fletcher-Suit-Delclos applicator like a human vagina. Fletcher-Suit-Delclos applicator inserted into the space was packed tightly with furacin gauzes, and three $^{137}Cs$ sources with radioactivity of $15.7mg\;Ra-eq$ were inserted into the tandem. For the film dosimetry, two pieces of X-OMAT V film (Kodak Co.) of which planes include point A, were arranged orthogonally in the slits between phantoms. A point dose and iso-dose curves were measured by means of optical densitometer. A point doses by film dosimetry, RTP system and manual calculation by using of along-away table were compared, and iso-dose curves by film dosimetry and computer planning were also compared. The dose of A point was 51.2cGy/hr by film dosimetry, 46.7cGy/hr by RTP system and 47.9 cGy/hr by along-away table. A point dose by computer planning was similar to the dose by calculation using of along-away table with acceptable accuracy $({\pm}3%)$, however, the dose by film dosimetry was different from two others with about 10% error. Since most clinical beams contains a scatter component of low energy photons, the correlation between optical density and dose becomes tenuous. In addition, film suffers from several potential errors such as changes in processing conditions, interfilm emulsion differences, and artifacts caused by air pockets adjacent to the film. For these reasons, absolute dosimetry with film is impractical, however, it is very useful for checking qualitative patterns of a radiation distribution. In future, solid state dosimeter such as TLD must be used for the dosimetry of ionizing radiation. When considerable care is used, precision of approximately 3% may be obtained using TLD.
