Microbiological Evaluations on the Facilities and Utilities of Korean Restaurants (한식당 설비와 기구의 미생물 평가)

  • The microbiological examinations were conducted for the hygienic evaluation on three Korean restaurants during summer season in Busan, Korea. Total one hundred and sixty swabbed samples using sponge were collected from the surface of facilities and utensils at restaurants and total and coliform counts were measured. Also thirty- six air samples were collected at inside of three restaurants for measuring total, coliform, Staphy-lococcus and mold and yeast counts. All collected samples kept in an ice-packed box were transported to the laboratory and analyzed. The results demonstrated that most swabbed samples were highly contaminated with microorganisms and coliforms. The degree of contamination depended on the sampling sites. Averages of total counts of surface swab samples were ranged from not detectable to 2.14$\times\10^{9}$/200 $cm^{2}$, while those of coliforms from not detectable to 8.34$\times\10^{7}$/200 $cm^{2}$/200 $cm^{2}$. Microorganisms also detected from most agar strips of air samples for total, coliform, Staphylococcus and mold and yeast counts. The severely contaminated sites were floor, trench, water bottle, plastic drainer, rubber gloves, shelves, and unsealed wet towel. Those sites should be focused and controlled according to control Points of sanitation standard operating Procedures. Also, periodic microbiological examination in addition to visual examination should be applied on those highly contaminated sites for reducing risk of foodborne disease outbreak at restaurants

Effect of control measures on the contamination and growth inhibition of Listeria monocytogenes in Flammulina velutipes (팽이버섯 재배 농가에서 Listeria monocytogenes 오염과 성장억제를 위한 관리기술 효과)

  • The consumption of Flammulina velutipes mushroom imported from Korea has been associated with the cases of listeriosis in the United States, Canada, and Australia. We investigated the effect of sanitizing the plastic wrapper (used in packaging F. velutipes) with slightly acidic electrolyzed water (SAEW) and ultraviolet C waterproof light-emitting diode (UVC-W-LED) on reducing the Listeria monocytogenes. Further, the effect of UVC-LED on L. monocytogenes growth in F. velutipes at different storage temperatures (2, 4, and 10℃) was determined. The combined (SAEW+UVC-W-LED) treatment for 5-10 min reduced 99.9% of bacterial population from the contaminated plastic wrapper. In addition, the UVC-LED treatment for 3 min reduced the L. monocytogenes concentration in F. velutipes by 0.47 log CFU/g. Moreover, the growth of L. monocytogenes in the treated mushrooms was slower than that of the untreated (control) ones. L. monocytogenes concentration in F. velutipes increased over 3 log CFU/g at 2℃ and 10℃ for 60 and 10 days, respectively. The growth of L. monocytogenes at the bottom of mushrooms was faster than that at the top at both the temperatures. These results indicate that the combined SAEW+UVC-W-LED treatment of plastic wrappers and the UVC-LED treatment of mushrooms can be used as potential hurdle technologies to control the risk of L. monocytogenes in mushrooms prior to packaging at farms.

Cleaning and Decontamination Method of Books for Their Sanitary Circulation (책의 위생적 유통관리를 위한 세정 소독 방법)

  • In order to find a sanitary logistic way to handle library books, papers and environmental sources contacting the books were tested in their microbial contamination load and methods to decontaminate the books were investigated. Generally bacterial load of inner book pages was very low, but increased when contaminated with liquid such as saliva. In contrast, their lateral ends showed much higher bacterial contamination presumably due to dry dust contamination on there. As operations to improve the sanitary book conditions, turbulent air blow was found to be workable for reducing dry dusty contamination and 280 nm ultra-violet (UV) light emitting diode (LED) was so for decontaminating wet surface contamination. Microbial inactivation by the UV LED could be realized with irradiation for more than 5 minutes at 2 cm distance. Air blow of 5.5 m/s for 0.5~1 minute could reduce the dusty contamination on a model book surface.

  • 공기를 주 절연매질로 사용하는 전력기기의 소형화 추세속에서 제품의 절연성능을 확보하기 위해 '절연배리어'인 고체절연물이 사용된다. 고체절연물의 연면 절연성능은 운전 환경에서 발생된 수분 및 오염원 등에 영향을 받기 쉽기 때문에, 다양한 오염도 조건에서 고체절연물의 연면 절연파괴특성 분석이 필요하다. 본 논문에서는 전력기기에 폭넓게 적용되는 있는 고체절연물에 대하여 평등전계가 모의된 평판 - 평판 전극 실험 조건에서 연면거리 및 오염도를 정량화한 전기전도도에 따른 AC 연면 절연파괴특성을 분석하였다. 그 결과, AC 연면 절연파괴전계는 고체절연물 재질에 관계없이 단(短) 갭에서 연면거리에 대한 의존성을 거의 보이지 않았다. 또한, PC 연면 절연파괴전계는 BMC 및 에폭시 대비 높았으며, 이것은 재질에 따른 연면 절연파괴 형태 및 표면의 소수성 차이와 관련이 있는 것으로 사료된다.

  • 본 연구에서는 건축자재로 널리 사용되는 PVC바닥상재 표면에 내오염성과 항균성을 증가시키기 위한 광경화형 코팅액 개발에 관한 것이다. PVC 바닥상재용으로 사용되는 우레탄-아크릴 수지에 수용성 대전방지제와 항균제를 배합하여 오염 및 정전기로부터 보호하고 미생물에 의한 전염성질환을 예방할 수 있는 광경화형 내오염/항균 코팅액에 관한 실험을 수행하였다. 연구결과, 수지에 수용성대전방지제 15%와 항균제 1%를 혼합하여 제조한 코팅액을 Bar-coater No.12로 코팅한 표면이 전기저항($10^9\Omega/cm^2$), 내오염도(매직 Test, 먼지부착 Test) 및 부착력(100%) 양호, 항균성(99.99%)로 가장 좋은 물성을 보였다.

  • 해수 환경에 노출된 대부분의 금속재료는 직 간접적으로 부식의 영향을 받는다. 이와 같이 해수에 침지된 금속재료의 부식 방지에는 음극방식법이 주로 사용되고 있다. 과거의 음극방식 설계에는 양극 크기와 방식전류 밀도, 소모율 등 양극 자체에 대한 변수만이 고려되었으나, 20여 년 전부터는 해수 오염도, 수온 및 유속 등을 종합적으로 고려하여 설계하고 있다. 특히 대부분의 금속은 부식성 환경에서 생성된 부동태 피막이 해수 유동에 의해 파괴되면서 급속히 손상된다. 따라서 본 연구에서는 해수 특성을 고려한 최적의 소요방식전류밀도 선정을 위해 전기화학실험을 실시하였다. 실험 결과, 해수 온도 상승에 따라 확산속도가 빨라져 최적 방식 전류밀도가 증가하는 경향을 나타냈다.

Survival and Cross-contamination of Escherichia coli O157:H7 on Various Agricultural Product-Contact Surfaces (농산물 접촉 표면 재질에 따른 Escherichia coli O157:H7의 생존 및 상추로의 교차오염도 조사)

  • To evaluate the effect of surface contaminated with Escherichia coli O157:H7 (E. coli O157:H7) on the microbiological safety of lettuce, this study was conducted to investigate the attachment, biofilm producing, survival, and cross-contamination of E. coli O157:H7 on stainless steel and polyvinyl chloride (PVC). The attachment rate of E. coli O157:H7 on PVC was 10 times higher than that on stainless steel after exposure 1 h in cell suspension. However, there was not a difference between two types of surface after exposure for 6 h and 24h. The biofilm producing of E. coli O157:H7 was TSB > 10% lettuce extracts > 1% lettuce extracts > phosphate buffer. When two kinds of materials were stored at various conditions ($20^{\circ}C$ and $30^{\circ}C$, relative humidity (RH) 43%, 69%, and 100%), the numbers of E. coli O157:H7 at $30^{\circ}C$, RH 43% or RH 69% were reduced by 5.0 log CFU/coupon within 12 h regardless of material type. Conversely, the survival of E. coli O157:H7 at RH 100% was lasted more than 5 days. In addition, the reduction rate of E. coli O157:H7 was decreased in the presence of organic matter. The transfer efficiency of E. coli O157:H7 from the contaminated surface to lettuce was dependent upon the water amount of the surface of lettuce. Especially, the transfer rate of E. coli O157:H7 was increased by 10 times in the presence of water on the lettuce surface. From this study, the retention of E. coli O157:H7 on produce contact surfaces increase the risk cross-contamination of this pathogen to produce. Thus, it is important that the surface in post harvest facility is properly washed and sanitized after working for prevention of cross-contamination from surface.

Microbiological Contamination Levels in the Processing of Korea Rice Cakes (떡류의 제조공정별 미생물학적 오염도 평가)

  • This study was conducted to evaluate microbial contamination levels of Korea traditional rice cakes such as Sirutteok, Garaetteok and Gyeongdan in the manufacturing process and environment. The microbial contamination levels such as total aerobic bacteria, fungi, coliforms, Escherichia coli, Staphylococcus aureus, Bacillus cereus and Clostridium perfringens of rice cake products were analyzed. The contamination levels of total aerobic bacteria, coliforms, fungi and B. cereus in raw materials were in the range of 2.4~4.5, ND~1.9, 1.2~2.1 and 1.0~2.1 log CFU/g, respectively. The microbial contamination levels of total aerobic bacteria, coliforms, fungi and B. cereus in manufacturing process of rice cakes were increased in the soaking and grinding steps and were decreased in steaming step. E. coli, S. aureus and C. perfringens were not detected in any manufacturing process and environment. The microbial contamination levels of raw materials and final products of rice cake were suitable for microbial safety standard in Korea. However, the manufacturing environment such as equipments and employee's sanitation were in trouble for microbial safety. The results of this study suggest that safety educatio n for personal hygiene and safetymanagement in processing environment are continuously required to assure safety in working environment and employee's individual hygiene.

  • It is going to select one X-ray generating device for diagnosis in a radiography laboratory at K university in Gangwon-do to detect bacteria on the surface contamination of tables, IP cassettes, and lead gowns for medical radiation shielding and to inform students of the need for proper disinfection control and hand hygiene. Then disinfection was carried out with tissue, tissue cleaner and 70% alchol and immediately collected with sterile cotton swabs to assess the contamination distribution status and disinfection effects of the surface. The results of measuring the degree of contamination on the surface showed that the largest number of bacteria were detected in Apron, and the evaluation of the disinfection effects according to surface contamination showed a noticeable effect at 70% Alcohol in IP Cassette, and the disinfection effect was the same for Apron. Therefore, in order to prevent bacterial infections among students, basic hand washing and regular disinfection should be performed before the practice to prevent infection.

A Study on the Removal of Cu and Fe Impurities on Si Substrate (Si 기판에서 구리와 철 금속불순물의 제거에 대한 연구)

  • As the size of the integrated circuit is scaled down the importance of Si cleaning has been emphasized. One of the major concerns is abut the removal of metallic impurities such as Cu and Fe on Si surface. In this study, we intentionally contaminated Cu and Fe on the Si wafers and cleaned the wafer by cleaning splits of the chemical mixture of $\textrm{H}_2\textrm{O}_2$ and HF and the combination of HF treatment with UV/$\textrm{O}_3$ treatment. The contamination level was monitored by TXRF. Surface microroughness of the Si wafers was measured by AFM. The Si wafer surface was examined by SEM. AES analysis was carried out to analyze the chemical composition of Cu impurities. The amount of Cu impurities after intentional contamination was abut the level of $\textrm{10}^{14}$ atoms/$\textrm{cm}^2$. The amount of Cu was decreased down to the level of $\textrm{10}^{10}$ atoms/$\textrm{cm}^2$ by cleaning splits. The repeated treatment exhibited better Cu removal efficiency. The surface roughness caused by contamination and removal of Cu was improved by repeated treatment of the cleaning splits. Cu were adsorbed on Si surface not in a thin film type but in a particle type and its diameter was abut 100-400${\AA}$ and its height was 30-100${\AA}$. Cu was contaminated on Si surface by chemical adsorption. In the case of Fe the contamination level was $\textrm{10}^{13}$ atoms/$\textrm{cm}^2$ and showed similar results of above Cu cleaning. Fe was contaminated on Si surface by physical adsorption and as a particle type.
