To develope natural food preservatives, ethanol and water extracts were prepared from the Lentinus edodes and antimicrobial activities were examined against 10 microorganisms which were food borne pathogens and / or food poisoning microorganisms and food-related bacteria and yeasts. Ethanol extract exhibited antimicrobial activities for the microorganisms tested, but not on lactic acid bacteria and yeast Especially, minimum inhibitory concentration(MIC) for Escherichia coli were as low as 0.5 mg/mL. Antimicrobial activity of the ethanol extract was stable by the heating at 121$^{\circ}C$ for 15 min and not affected by pH. The ethanol extract of Lentinus edodes exhibiting high antimicrobial activity. The highest antimicrobial activity adjust bacteria tested was found in the ethylacetate fraction.

The aim of this study was to investigate the anti-obesity effects of Lentinus edodes water extract powder (LEP) in mice fed a high fat diet (HF, 45% kcal fat). Mice were administrated a HF diet supplemented with 1%, 3%, or 5% LEP for 12 weeks. Consumption of HF diet caused increases in body weight, serum lipid profiles, and adipose tissue weights. Serum TC and TG levels in the LEP-supplemented groups were lower than those in the NC group. Supplementation with 5% LEP significantly suppressed body weight gain and reduced the weight of subcutaneous adipose tissue compared to the HF group. HF diet ingestion resulted in higher lipid content and increased lipid peroxidation in the liver. However, LEP supplementation inhibited accumulation of hepatic lipids induced by HF diet, considerably decreased MDA levels, and elevated total antioxidant activity in the livers of mice in the 5% LEP group. Histopathological analysis indicated that the livers of mice fed HF diet developed hepatic steatosis, whereas LEP-treated groups showed small fat droplets. These results suggest that long-term supplementation with LEP may also have an ameliorating effect on HF-induced obesity.

Purpose: The purpose of this study was to investigate antifungal activity of shiitake mushroom yeast and hyphal type of Candida albicans. Materials and Methods: The extract from shiitake mushroom was collected by drying the supernatant after soaking shiitake mushrooms in water or ethanol. The antifungal activity of the extracts against yeast type of C. albicans was investigated by the susceptibility assay using microplate. C. albicans biofilm was formed on 12-well plate using Ham's F-12 medium in CO2 incubator and treated with the ethanol extract. Furthermore, C. albicans biofilm was formed on denture base resin disk and treated with or without the ethanol extract in the presence of denture cleanser. Live C. albicans in biofilm was counted by cultured colony forming unit value after inoculated on agar plate. Results: Ethanol extract from shiitake mushroom showed stronger antifungal activity against yeast type of C. albicans compared to its water extract. The ethanol extract significantly reduced count of C. albicans in hyphal biofilm (P < 0.05). Also, the ethanol extract showed synergistically antifungal effect with denture cleanser on candidal biofilm on denture base resin disk (P < 0.05). Conclusion: The ethanol extract of shiitake mushroom may be a candidate for preventing candidal stomatitis as well as denture-related stomatitis.
The total phenolic and flavonoid contents of Lentinus edodes extracts were determinated by spectrophotometrical .method, and antihepatotoxic activity was detected on glutathione S-Transferases(GST). The total phenolic contents was highest water extract than solvent(ethanol, methanol) extracts, but flavonoid content was appear on opposite. GST activity was the highest appears in water extract. This fact verified of anticancer effect indirectly of Lentinus edodes.
In Korea, shiitake mushroom, Lentinula edodes, is cultivated on artificial medium containing oak sawdust and wheat bran. The annual production of spent mushroom substrate (SMS) of shiitake, a byproduct of the mushroom industry, is estimated to reach over 50,000 tons per year. This study aimed to improve the use of SMS as a novel bioresource. Hot water extracts of SMS after the first and third harvest were prepared and their bioactivities evaluated. Hot water extracts of uninoculated medium and shiitake were used as controls. Extracts of SMS showed higher radical scavenging of DPPH anions, ABTS cations, nitrites, and a higher reducing power than those of shiitake or medium extracts. After the first and third harvests at 0.5 mg/disc, SMS extracts showed no antibacterial or antifungal activities against the pathogenic and food-spoilage bacteria and fungi. However, they showed good inhibitory activities against ${\alpha}$-glucosidase at 0.5 mg/ml. In addition, SMS extracts had strong anti-coagulation activities via their inhibition of thrombin, prothrombin, and blood coagulation factors without platelet aggregation activity. Our results suggested SMS should no longer be perceived as a useless byproduct but should be understood as a novel bioresource, the extracts of which could be developed as antioxidant, antidiabetic, and antithrombosis agents.
Physiological functionality of browned oak mushroom was evaluated with focusing its electron-donating ability to DPPH radicals, scavenging ability to superoxide radicals and hydroxyl radicals, and inhibitory activity on lipid peroxidation. The results showed that overall antioxidative activities of browned oak mushroom were superior to those of raw oak mushroom with marketable quality, implying possible involvement of resultant browning reaction products in an increment of antioxidativity. The increased radical-scavenging ability was suggested to mainly be exerted by direct quenching of both superoxide and hydroxyl radicals, not by inhibition of xanthine oxidase activity and $Fe^{2+}$ chelation, respectively. Collectively, these results indicate a possible use of unmarketable browned mushroom as a material for manufacturing various processed functional foods.

Purpose: The purpose of this study was to investigate antimicrobial activity of extracts from shiitake mushroom against periodontopathogens and its cytotoxicity for human gingival fibroblast. Materials and Methods: Shiitake mushroom was soaked in water and acetone, and the supernatant was dried to collect its extract. The susceptibility of periodontopathogens for the extracts was investigated. Human gingival fibroblast was treated with the extracts, and the cell viability was measured CCK-8 solution. Results: The water extract from shiitake mushroom significantly reduced the growth of periodontopathogens at 2.5 mg/ml (P < 0.05). The acetone extract significantly inhibited the growth of Porphyromonas gingivalis and Tannerella forsythia at 0.32 mg/ml and Treponema denticola growth at 0.64 mg/ml (P < 0.05). The cytotoxicity of the extract was shown at a concentration of 2.5 mg/ml. The extracts with a concentration of 1.25 mg/ml appeared to be reduce cell viability after 4 h. Conclusion: The extracts of shiitake mushroom have antimicrobial activity against periodontitis-causing bacteria and relieving inflammation. Therefore, the extracts may be a candidate for preventing and treating periodontal disease.

Shiitake mushroom (SM; Lentinus edodes) are cultivated and consumed in many Asian countries including Vietnam, China, Japan, Korea, and Thailand. In Asia, SM are mainly dried and used as flavoring. The aim of this study was to compare the effects of SM created with different drying processes, such as oven-dried and sun-dried, on the antioxidative and antigenotoxic effects. Raw and dried SM were extracted with acetone, ethanol, methanol, and hot water. The antioxidant effects of SM were evaluated by determining total phenolic content, DPPH radical scavenging activity (RSA), an ORAC assay, and a cellular antioxidant capacity (CAC) assay. The inhibitory effect of SM on oxidative stress-induced DNA damage in human leukocytes was evaluated by a Comet assay. The total phenolic content of raw SM extracted with methanol and of that extracted with water were significantly higher than the dried SM. Among the water extracts, the $IC_{50}$ for DPPH RSA of raw and sun-dried SM were significantly higher than that of oven-dried SM. Sun-dried SM showed the most potent ORAC value at 50 g/mL. The CAC against $AAPH^-$ induced oxidative stress in HepG2 cells, and $H_2O_2$ induced DNA damage were effectively protected against by all SM extracts. These results suggest that unprocessed SM are the best antioxidants, and that the sun-dried method would be the best option to use in terms of antioxidant activity and the antigenotoxic effect.

The aim of this study was to investigate the antioxidant and antimicrobial activities of various solvent (acetone, ethyl acetate, and ethanol) extracts from Lentinus edodes. The antioxidant activities were evaluated by measuring total polyphenol and flavonoid contents, 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity. Total polyphenol content and ABTS radical scavenging activity were highest in ethanol extract. ABTS radical scavenging activity of ethanol extract showed the highest value (98.5%), which was similar to that of ascorbic acid (95.7%). The ethyl acetate extract from Lentinus edodes showed relatively high total flavonoid content and DPPH radical scavenging activity. Negative correlations were found between total polyphenol contents and DPPH radical scavenging activities in Lentinus edodes extracts. Antimicrobial activities of the extracts were determined against Bacillus subtilis, Staphylococcus aureus, Micrococcus luteus, Escherichia coli, Pseudomonas aeruginosa, and Enterobacter cloacae by the disc diffusion method. The acetone and ethanol extracts showed moderate antimicrobial activities against almost all tested microorganisms except E. coli and S. aureus, respectively. The ethyl acetate extract showed a significant growth inhibition effect against E. coli, Ent. cloacae, and B. subtilis.

This study evaluated the characteristics and antioxidant properties of cookies prepared with different amounts (0%, 1%, 3%, 5%, and 7%) of Lentinus edodes water extract (LEWE). The baking loss rate of cookies increased as the amount of LEWE increased (p<0.05). The pH of cookies significantly decreased when amounts of LEWE increased (p<0.05), but spread factor, leavening rate, and moisture content showed no significant difference among the samples. L value and b value of cookies significantly decreased when amounts of LEWE increased (p<0.05), but a value of cookies significantly increased when amounts of LEWE increased (p<0.05). Hardness was higher in the 7% LEWE cookies. Chewiness and brittleness increased with increasing LEWE concentration, but cohesiveness and springiness showed no significant differences among the samples. Total polyphenol content and DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity significantly increased upon the addition of LEWE (p<0.05). In a sensory evaluation, the appearance, color, and taste of the cookies were rated higher in groups containing 0% LEWE, but the flavor, texture, and overall acceptability scores were highest for cookies with 1% LEWE added. These results suggest that cookies prepared by adding 1-3% LEWE achieve the best sensory evaluation and antioxidant activity.
