Quality Characteristics of Pressed Ham Containing Grape Seed Oil (포도씨유가 함유된 프레스햄의 품질특성)

  • Pressed ham was manufactured to investigate the effects of grape seed oil on the quality characteristics of pressed ham. Five treatments were divided based on differences in the amount of grape seed oil added into the pressed ham. For control, 10% of back fat was only added without grape seed oil. For the first treatment, 10% of grape seed oil among the lard component added into the pressed ham was replaced. For the 2nd, 3rd and 4rd treatments, 20%, 30% and 40% of grape seed oil was respectively replaced. Pressed ham manufactured using grape seed oil was vacuum packaged and then stored for 1, 7, 14, 21 and 28 days at 4℃. Samples were analyzed for shear force value, sensory properties, TBARS and fatty acid composition. In the 1, 21 and 28 days of storage, shear force value of grape seed oil treatment (T4) was significantly lower than that of control (P<0.05). No remarkable differences were found in sensory properties among control and grape seed oil treatment groups. The TBARS value was significantly higher in control than in grape seed oil treatment group(T4) at 28 days of storage (P<0.05). The TBARS of control and grape seed oil treatment groups increased significantly as the storage period increased(P<0.05). The linoleic acid(C18:2) content of grape seed oil treatment groups was significantly higher than that of control(P<0.05). But the contents of C10:0~C20:4 were decreased significantly by grape seed oil additive (P<0.05). Saturated and monounsaturated fatty acid content of control was significantly higher than that of grape seed oil treatment groups(P<0.05). Whereas the increase level of grape seed oil additive resulted in the significantly higher polyunsaturated fatty acid content(P<0.05). Based on these findings, we conclude that the sensory properties and lipid oxidation(TBARS) of manufactured pressed ham were not affected by grape seed oil addition. Also, our results indicate that high-quality pressed ham can be manufactured with strengthen of polyunsaturated fatty acid content.

Sex Determination of Porcine IVF Embryos by Polymerase Chain Reaction (PCR) (중합효소연쇄반응을 이용한 돼지 체외수정란의 성감별)

  • This study was carried out to determine sex of porcine embryos produced by in vitro fertilization. Porcine oocyte-cumulus complexes were cultured in BSA-free North Carolina State University (NCSU) 23 medium containing porcine follicular fluid (10%), cystein (0.1 mg/ml) and hormonal supplement (10 IU eCG and 10 IU hCG per ml) for 20~22 hrs. They were then cultured in the same medium but without hormonal supplement for additional 20~22 hrs. After culture, cumulus cells were removed and oocytes were co-incubated for 6 hrs with four different concentrations (5$\times$10$^4$, 2.5$\times$ 10$^{5}$ , 5.0$\times$10$^{5}$ and l0$\times$10$^{5}$ ) of porcine sperm. After fertilization, oocytes were transferred into NCSU 23 with 0.4% BSA medium. The cleavage and blastocyst formation rates were evaluated at 48 and 144 hrs, respectively. In this study, the polymerase chain reaction (PCR) was used to determine the sex of porcine embryos in the stage of blastocyst. The PCR was performed using a set of oligonucleotide primers (5‘-TCATGGACCAGGTAGGGAAT-3', 5’-GAAAGACACGTCCTTGGA GA-3') for 491 bp fragment of porcine male-specific DNA sequence. In the flour different sperm concentration (5$\times$10$^4$, 2.5$\times$10$^{5}$ , 5.0$\times$10$^{5}$ and l0$\times$10$^{5}$ ) for fertilization condition, the cleavage rate was 55.95, 67.88, 60.18 and 47.60%, respectivety, and the development rate of blastocysts was 16.03, 20.40, 21.41 and 12.37%, respectively. At 5.0$\times$10$^4$and 2.5$\times$10$^{5}$ of sperm concentrations per ml cleavage rate and development rate of blastocyst were higher than those of 5.0$\times$10$^4$and l0$\times$10$^{5}$ of sperm concentration (P<0.01). The male of porcine embryos was detected at 491 bp by PCR, and 18 of the 31 porcine blastocysts were the male (58.1%) and the rest 13 were the female(41.9%).

The Environmental Characteristics and Factors on the Cultured manila clam (Ruditapes philippinarum) at Hwangdo and Jeongsanpo of Taean in the West coast of Korea (서해 연안 황도와 정산포 바지락 양식장의 환경특성)

  • To assess the effect of environmental factors on the sustainability of cultured production manila clam (Ruditapes philippinarum), we investigated the habitat characteristics of tidal flat (Hwangdo and Jeongsanpo in Taean). We measured the physiochemical parameters (temperature, salanity, pH, dissolved oxygen and nutrients) and the characteristics (mean size, chemical oxygen demand, ignition loss, C/N ratio and C/S ratio) of surface sediments. The C/N ratio of Hwangdo and Jeongsanpo were 9.0, 5.3 and the C/S ratio was 0.162, 0.159. The concentration of chlorophyll a at Hwangdo was higher than that of Jeongsanpo and species of micro algae were 102 and 100. The enrichment factor (Ef) and index of accumulation rate (Igeo) of the metals showed that those research areas can be classified as heavily polluted, heavily to moderately polluted, or more or less unpolluted, respectively. So two survey area of cultured clams in the tidal flat have been effected by the various environmental conditions, there are needed an improvement methods and continuous research for increasing the production of manila clam (Ruditapes philippinarum).

Effects of Heating Temperatures and Times on Anthocyanin Pigments in Grape Juice (열처리 조건이 포도 주스내 anthocyanin 색소에 미치는 영향)

  • The influence of temperature and heating time on the stability of anthocyanin pigments in grape juice were investigated. There was no significant differences in soluble solids, acidity and pH of grape juice under various temperature and heating time. Residual total anthocyanin of grape juice was decreased by heating temperature over 100$^{\circ}C$ for 1 min. Delphinidin-3-glucoside and cyanidin-3-glucoside were rapidly decreased by heating treatment, but petunidin-3-glucoside and malvidin-3-glucoside were more stable than others. These results suggests that the optimum quality of grape juice was heated at 90 $^{\circ}C$ for 1 min.

Evaluation of resistance to Pierce's disease among grapevine cultivars by using the culture filtrates produced from Xylella fastidiosa (Xylella fastidiosa의 배양여액을 이용한 포도나무 피어스병 품종 저항성 검정)

  • This study investigated whether culture filtrates produced by Xylella fastidiosa can be used to determine the varietal susceptibility to Pierce's disease in grapevines (Vitis spp.) as a substitute for pathogen inoculation or field screening. A bioassay of grape leaves with culture filtrates from the pathogen showed that their phytotoxicities were active and host-selective. Ethyl acetate extracts from them also showed toxicities and host selectivity in both bunches of grapes and muscadine grapes. The sensitive range of plants to the culture filtrates and their ethyl acetate extracts was consistent with the host range of the Pierce's disease pathogen. Susceptible cultivars are sensitive to even highly diluted culture filtrates, while resistant cultivars were not affected even at their original culture filtrates. Susceptible cultivars were more sensitive to the undiluted culture filtrate than were highly diluted culture filtrates, and the younger leaves were the most sensitive to the culture filtrates in grapes. Although some European grape cultivars showed moderately susceptibility in this study, the determination of varietal resistance to Pierce's disease by the treatment of culture filtrates of pathogens could provide valuable information for the preliminary selection of genetic resources and seedlings from hybridization in a disease resistant grape breeding program.

Effects of Weed Control on Pseudocercospora vitis and Fruit Characteristics in Campbell Early Vineyard (캠벨얼리 포도원에서 잡초관리 방법이 갈색무늬병 및 과실특성에 미치는 영향)

  • This study was conducted to evaluate the effect of weed control on disease incidence, plant growth and fruit quality, and to provide effective method to farmers at the vineyard. At the raincoat greenhouse cultivating 'Campbell Early', several weed controlling methods were compared to the control. Mean temperature of plots between soil covering (C.B., W.F., S.N., P.F. and R.H.) and non soil covering (M.W.2, M.W.3 and U.T.) were similar with $26.2^{\circ}C$ (July) and $25.8^{\circ}C$ (Aug.), but humidity of soil covering was sharply decreased in late-July under 40%. Soil water were all higher in soil covering plots than that of non-soil covering, however, soil temperature was recorded lower in soil covering plots. Disease incidence of Peudocercospora vitis according to soil coverings had effect on black plastic film mulching by decreasing grapevine leaf spot to 85% in July, 69% in August, compared to the control, with showing the smallest lesion incidence in FMB. Among growth characters, internode length and internode width of black plastic film mulching was higher than that of the control. SPAD value of black plastic film mulching was recorded the highest. It was not significant among the treatments, but brix was recorded highly in black plastic film mulching with $14{\sim}15^{\circ}Brix$.

Berry Production Using Secondary Shoots in 'Campbell Early' Grapevines (포도 캠벨얼리 품종의 2차지를 이용한 과실생산)

  • For the production of second crop in 'Campbell Early' grape, the primary shoots were pruned at 3rd, 6th or 9th nodes from the shoot bases on 13 days, 23 days and 33 days after full bloom date on 7th June. Secondary shoots were sprouted 7~8 days after the pruning, and it took 19~25 days for the flowering on the secondary shoots. The flower cluster number on secondary shoots were 2.8 for 13 days after full bloom, and 3.2 for 23 days and 33 days after full bloom, meaning little effect by pruning time. The 3rd node pruning produced 2~2.4 flower clusters with flower cluster length of 9.3~10.4 cm, while the 6th or 9th node pruning produced 3.1~3.8 flower clusters with flower cluster length of 12~14.9 cm, showing superior flower cluster length for the 6th or 9th node pruning. The secondary shoots developed from the buds pruned 13 days after full bloom with pruning bud positions of 6th nodes demonstrated superior fruits with higher soluble solids and lower acidity than the rest of the pruning times and positions.

Analysis of Phytosterol and Fatty Acid Compositions of Grape Seeds Produced in Korea (품종별 포도씨의 식물성 스테롤 및 지방산 조성 분석)

  • In this study, phytosterol content and fatty acid composition of grape seeds from fourteen different cultivars produced in Korea were determined. Although the total phytosterol contents depended upon the cultivars, the major phytosterol was $\beta$-sitosterol (64.9$\sim$119.3 mg/100 g) in all samples. The fatty acid composition of the grape seeds were analyzed as palmitic (16:0, 6.8$\sim$16.8%), palmitoleic (16:1, 0.1$\sim$0.2%), stearic (18:0, 4.7$\sim$5.2%), oleic (18:1, 16.6$\sim$26.1%), linoleic (18:2, 16.6$\sim$20.1%), linolenic acid (18:3, 0.3$\sim$0.5%) and arachidic acid (20:0, 0.3$\sim$0.5%). Campbell early, the mainly produced and consumed cultivar in Korea, contained relatively high phytosterol content compared to other samples and high proportion of unsaturated fatty acids. Therefore, the seeds from Campbell early might be used for functional oil production and as food ingredients.

The Effects of Preservation of Ovaries, Incubation Time and Oocytes with and without Cumulus Cells on Zona Penetration by Canine Sperm (개 난소의 보존, 난자의 배양시간 및 난구세포 부착 유무가 난자내 정자의 침입율에 미치는 영향)

  • 본 연구는 개의 불임해결과 체외수정란을 생산할 목적으로 난소의 보존 및 난구세포의 부착 여부가 신선 및 동결 개 정자를 이용한 투명대 반응에 미치는 영향을 조사하였다. 1. 적출한 난소를 4$^{\circ}C$ 와 salt에 각각 48시간 보존 후 회수한 난구세포 부착 난자와 나화난자의 정자침입율은 각각 62.5%, 37.5% 및 42.5% 및 22.4%로서 난소를 적출 후 곧 바로 회수한 난구세포 부착 및 나화난자 내 정자침입율인 93.3%와 56.7%에 비해 현저히 낮게 나타났다. 2. 적출한 난소를 4$^{\circ}C$ 에 각각 4시간, 14시간 및 48시간 보존 후 회수한 난구세포부착 난자의 정자침입율은 각각 92.5%, 90.0%, 85.0%로서 신선 난소 난자의 정자침입율 93.3%에 비해 유사하거나 약간 낮게 나타났다. 3. 적출한 난소를 salt에 각각 4시간, 14시간 및 48시간 보존 후 회수한 난구세포부착 난자의 정자침입율은 각각 85.0%, 77.5%, 72.5%로서 신선난소로부터 회수한 난자의 정자침입율 93.3%에 비해 낮게 나타났다. 4. 적출한 난소를 4$^{\circ}C$ 와 salt에 각각 24시간 보존 후 회수한 난구세포부착 난자의 체외수정율과 체외발생율은 각각 50.0%, 22.5% 및 40.0%, 15.0%로서 신선난소로부터 회수한 난구세포부착 난자의 체외수정율과 체외발생율 72.5%와 32.5%에 비해 낮게 나타났다.

체외성숙시간이 돼지 단위발생능에 미치는 영향

  • 난자성숙 중 MPF활성은 MI 과 MII에서 최대로, MII로의 성숙 직후 난자를 핵이식에 이용하는 것은 핵과 세포질 사이의 reprogramming에의 효율을 높일 수 있을 것이다. 일반적으로 돼지의 IVF나 핵이식은 체외성숙후 44h에 이뤄지고 있다. 이는 aging을 낳을 수 있고 aging은 또한 발달능을 떨어뜨릴 수 있다. 따라서, 본 실험은 돼지의 체외성숙시간에 따른 체외성숙율과 단위발생후 체외발달율을 비교하여 최적의 체외성숙시간을 찾고자 실시하였다. 돼지 난포란을 10% pFF, 0.1mg/ml cysteine, 10IU/ml PMSG, 10IU/ml hCG, 10ng/ml EGF가 첨가된 TCM-199배양액에서 22, 30, 44시간 동안 배양하여 성숙을 유도하였다. 체외성숙이 야기된 난자는 난구세포를 제거한 후 전기자극(2.0kv/cm, $30 \mu s$) 후 5분 동안 TCM-199에서 세정하고 다시 4시간 동안 6-DMAP에서 배양된 후 4mg/ml BSA가 첨가된 NCSU-23에 넣어 $39^{\circ}C$, 5% $CO_2$배양기에서 각각 6-7일 동안 배양을 실시하였다. 체외성숙 22시간 성숙난자는 M I 44.3%(35/79)와 A I -T I 36.7%(29/79)로 81%가 M II로의 성숙에는 아직 미치지 못했다. 30시간 성숙난자는 46.3%(56/121)가 M II로 성숙하였고, M I과 A I -T I 은 각각 25.6%(31/121), 27 3%(33/121)이었다. 44시간 성숙난자는 78%(71/91)가 M II로 성숙하였고, M I 과 A I -T I 은 각각 12.1%(31/121), 7 7%(33/121)이었다. 단위발생율은 22시간에 난할율은 35.4%(75/137)이었고, 배반포 발달은 없었다. 30시간에 난할율은 51.8%(145/280)이었고, 배반포 발달율은 5.5%(8/145)이었다. 44시간에 난할율은 80.0%(244/306)이었고, 배반포 발달율은 14.3%(35/144)이었다. 본 연구결과 핵 및 세포질의 완전한 성숙은 44h에 이루어지고, 이에 따른 배발달율도 뒤따름을 알 수 있었다. 난자가 핵성숙의 완성에도 불구하고 완전한 활성을 위한 발달능은 갖지 못함을 알 수 있었으며, 질 좋은 배반포 생산을 위해 핵과 세포질 성숙의 synchronous가 중요하다 사료된다. 체외성숙을 위한 배양기술의 진전과 더불어 체외성숙 시간에 따른 세포질적인 기전에 대한 연구가 필요하다.
