Immunohistochemical identification of porcine reproductive and respiratory syndrome virus antigen in the lungs of naturally infected piglets (돼지 생식기 호흡기 증후군 바이러스에 자연감염된 포유자돈의 폐장에서 면역조직화학법을 이용한 바이러스 항원의 확인)

  • 돼지 생식기 호흡기 증후군 바이러스의 nucleocapsid와 반응을 하는 SDOW17 단크론항체를 이용하여 중성 포르말린에 고정시킨 자연감염된 포유자돈의 폐장에서 면역조직화학법을 이용하여 돼지 생식기 호흡기 증후군 바이러스 항원을 확인하였다. 서울대학교 수의과대학 병리학교실에 의뢰된 포유자돈들 중에서 병리조직학적으로 폐장에서 간질성 폐렴이 관찰된 포유자돈 7두를 임의로 선택하여 본 실험을 실시하였다. 간질성 폐렴의 병변으로 많은 수의 대식세포 침윤을 동반한 폐포벽 두께의 증가와 제II형 폐포세포의 비후가 관찰되었다. 검사한 7두 포유자돈중에서 6두에서 돼지 생식기 호흡기 증후군 바이러스에 대한 항체를 enzyme-linked immunosorbent assay에 의해 확인하였다. SDOW17 단크론항체를 이용한 면역조직화학염색과 간질성 폐렴의 대식세포에서 돼지 생식기 호흡기 증후군 바이러스의 항원을 검출하였고, 항원은 (주로)대식세포의 세포질에서만 진한 갈색의 양성반응이 관찰되었다. 이상 검사결과 돼지 생식기 호흡기 증후군 바이러스는 폐장의 간질과 폐포강에 분포되어 있는 대식세포에서 주로 증식하는 것으로 판명되었다. 본 실험에서 사용한 면역조직화학법은 돼지 생식기 호흡기 증후군 바이러스 감염여부를 바이러스 분리 또는 혈청검사 없이 진단하는데 사용할 수 있는 유용한 진단방법으로 판명되었다.

  • Cytomegalovirus (CMV) pneumonia is a serious opportunistic infection in an immunocompromised host such as an AIDS patient or transplant recipient undergoing immunosuppressive therapy. Diffuse alveolar hemorrhage (DAH) is a relatively uncommon condition and it occurs most often in patients with systemic autoimmune disease. However, various types of infectious pneumonia with Mycoplsma hominis, Stenotrophomonas maltophilia and Pneumocystis jirovecii have been reported to be associated with the development of DAH. The association of CMV infection with the development of DAH has rarely been reported. We experienced a case of DAH associated CMV pneumonia and the patient was successfully treated with the use of antiviral agents and steroids.

Effect of Ginseng Saponin Fractions on Phagocytosis and Chemotaxis of Murine Macrophages (대식세포의 식세포활동과 화학주성에 대한 인삼분획물의 영향)

  • The phagocytosis and chemotaxis of murine macrophages after treated with saponin fractions are investigated. Phagocytic appearance against yeast was photographed by dying with Wright-Giemsa. Phagocytic activity of peritoneal macrophage was invreased in diol saponin treatment by 48% and was decreased in total saponin treatment by 35%. The ingestion of alveolar macrophage was increased by 50% maximally. Peritoneal chemotactic activity was shown in 17% increases and only diol saponin had effect in alveolar macrophage by 16%. According to SDS-PAGE method the contents of actin did not show any alterations.

  • In acute lung injury (ALI) by lipopolysaccharide (LPS), the underlying cause of infiltration and migration of neutrophils into the alveoli is considered to be from increased production of platelet-activating factor (PAF) in the pulmonary surfactant lining the alveolar lumen. In this study I partially confirmed this concept. LPS increased lung leak and the infiltration of neutrophils in the lung of rats given LPS intratracheally. The migration of neutrophils into the lung, which had caused oxidative stress, was also morphologically identified. I verified that the metabolism of the pulmonary surfactant was affected and that there was increased production of PAF in the pulmonary surfactant, both of which are considered to contribute to ALI by LPS in rats.

  • Although the typical CT findings of Pneumocystis jiroveci pneumonia (PJP) include diffuse or multifocal areas of ground-glass opacities in both lungs, it can also rarely manifest as multiple pulmonary nodules. We report a rare case of atypical PJP in an immunocompromised patient with multiple myeloma, presenting as widespread ground-glass opacities and multiple necrotic subpleural nodules in both lungs on CT, which proved to be granulomatous PJP on percutaneous transthoracic needle biopsy.

PCR in diagnosis of pneumocystosis of rats (중합반응을 이용한 흰쥐 페포자충증의 진단)

  • Polymerase chain reaction (PCR) is a powerful technique to detect scanty amount of DNA from living organisms. The present study intended to develope specific primers for PCR diagnosis of pneumocystosis and to evaluate diagnostic efficacy by preparation of template DNAs from invasive BAk fluid and also to screen serum or blood as a non-invasive specimen. Albino rats of Wistar or Fischer strains were experimentally infected by Pneumocwstis ccrinii. Extracted DNAs or cell Iysates of their blood, bronchoalveolar lavage fluid, and lung homogenate were used as the tenlplate DNA. Primers were synthetic oligonucleotides among 16s rDNA sequences. All of the primer combinations gave PCR products, but the primer pair of #24 and #27 gave best quality product of 666 bp. The sensitivity of PCR with Iysates of BAk fluid was 57.7% but it increased to 84.6% with extracted DNAs. None of BAL Iysate or DNA was positive among 13 microscopically negatives. The serum DNAs were positive only in 2 cases out of 20 morphologically positive rats. DNAs of human, rat, other parasites, yeast, and microorganisms were negative. The findings suggest that the present primers are specific but simple Iysate of BAL fluid is not sensitive. PCR may be used as a routine diagnostic method of pneumocystosis if simple and rapid preparation of non-invasive clinical specimens are available.

The Harmful Effects of Prolonged Strenuous Treadmill Exercise on Bronchoalveolar System in Rats (장시간의 고강도 트레드밀 운동이 기관지 폐포계에 미치는 유해한 효과)

  • We designed this study to investigate the effects of continuous strenuous aerobic exercise on the respiratory system in a rat model. After exercise for 8 weeks, rats' weights were higher in the exercise groups than in the Control group (non-exercise). Rats in Exercise-120 min group (Ex-120 group) had the lowest weights. Total leukocyte counts in bronchoalveolar lavage fluid (BALF) were higher in exercise groups than in Control group. The Ex-30 and Ex-120 groups had higher neutrophil counts, whereas that in the Ex-60 group was lower than in the Control group, and that in the Ex-30 group was the highest. Lymphocyte and monocyte counts were higher in all exercise groups than in the Control group, and those in the Exercise-120 min group were the highest. Interleukin-6 (IL-6) level was the highest, while IL-10, interferon-$\gamma$ and nitric oxide (NO) levels were the lowest in the Ex-120 group when compared to the Control and other exercise groups. These findings suggest that strenuous aerobic exercise for short periods (30 min) may have a beneficial effect on decrease in body weight, whereas prolonged-strenuous aerobic exercise (>1 hr) may be adverse to leukocyte and immune levels in the bronchoalveolar system, as well as result in an increased production of oxygen free radicals.

Effect of Thermal Environment by Green Roof and Land Cover Change in Detached Housing Area (옥상녹화 및 토양피복 변화가 단독주택지 외부 열환경에 미치는 영향 분석)

  • Used as foundation resources for environment improvement and preservation of single-housing residential area by practicing classification of biotope with the concept of ecological area rate applied and performing urban thermal environment prediction simulation. Biotope is classified as seven types according to classification of biotope which is carried out with the concept of ecological area rate applied. The classification is listed below in descending order: building biotope(48.16%), impervious pavement biotope(39.75%), greenspace biotope(6.23%), crack permeable pavement biotope(3.26%), whole surface permeable pavement biotope(2.51%), parts permeable pavement biotope(0.04%). As a result of analysing prediction of variation and characteristics of thermal environment of single-housing residential area, land surface temperature per types of biotope are evaluated as listed below in descending temperature order: impervious pavement biotope > building biotope > greenspace biotope > permeable pavement biotope. In case 2 where vegetated roof hypothetically covers 100% of the roof area, temperature is predicted to be $33.58^{\circ}C$ Max, $23.85^{\circ}C$ Min, and $27.74^{\circ}C$ Avg. which is Approximately $5.19^{\circ}C$ lower than a non-vegetated roof. Average outdoor temperature for case 2 is studied to be $0.18^{\circ}C$ lower than case 1.

  • Background: An acute lung injury(ALI) is characterized by the recruitment, activation, and apoptosis of inflammatory cells, numerous products released by inflammatory cells such as reactive oxygen species, inflammatory mediators, and a variety of proteolytic enzymes. It was reported that bacterial infections in diabetics showed impaired PMN functions such as reduced PMN respiratory burst and decreased microbicidal activity in inflamed tissue. However, the effect of the proteinase - inhibitor (MMP-9 vs TIMP-1) in ALI in diabetics is unclear. This study evaluated the differences in the expression of MMP-9 and TIMP-1 after the stimulation of endotoxin in a rat model. Methods: Six-week-old male Sprague-Dawley rats were classified into normal, DM, LPS and DM+LPS groups. The peripheral blood, BAL fluids, and lung tissues were obtained from individual rats. The MMP-9 activity was measured by gelatin zymography and the TIMP-1 level was measured by Western blotting. Results: The total BAL cells of the DM-LPS groups were significantly lower than the LPS groups (p < 0.01). The MMP-9 activities in the serum were higher in the DM+LPS groups than in the other groups. The MMP-9 activities in the BAL fluids were significantly higher in the DM+LPS group than in the normal and diabetic rats (p < 0.05). TIMP-1 expressions in the BAL fluids were significantly lower in the DM+LPS group than other groups (p < 0.05). The ratio between MMP-9 and TIMP-1 in the BAL fluids was significantly higher in the DM+LPS groups (p < 0.05). Conclusion: In ALI in diabetics the higher MMP-9 activity and lower TIMP-1 level are believed to prolonged and intensify the course of inflammation.

The Radioprotective Effect and Mechanism of Captopril on Radiation Induced Lung Damage in Rat (방사선조사에 의한 쥐 폐손상에 방사선보호제로서 Captopril의 역할에 관한 연구)

  • Purpose : It was reported that Captopril (angiotensin converting enzyme inhibitor) had an effect to reduce the pneumonitis and pulmonary fibrosis induced by radiation in rat. We peformed this study to investigate the radioprotective effect and mechanism of Captopril. Methods and Materials : The comparison was made between the radiation only group and the combined Captopril and radiation group by examining histopathologic findings and immunohistochemical stains $(TNF\alpha\;and\;TGF\beta1)$ at 2 and 8 weeks after irradiation. Each group has 8 to 10 rats (Sprague-Dawley). 12.5 Gy of X-ray was irradiated to the left hemithorax in a single fraction. Captopril (50 mg/kg/d) mixed with water was given per oral and continuously from 1 week prior to irradiation up to 8th week of the experiment. Result : In the combined Captopril and radiation group, the histopathologic changes which were hemorrhage into alveolar space, changes of alveolar epithelium, bronchial epithelium and blood vessels, and perivascular edema were less severe than in the radisation only group at 2 weeks. At 8 weeks, the alveolar epithelial changes and perivascular edema were less prominant in the combined Captopril and radiation group. At 2 weeks, the $TNF\alpha$ expression of the combined Captopril and radiation group was markedly decreased at the alveolar epithelium (p<0.01), lymphoid tissue (p=0.06) and the macrophage of alveolar space (p<0.01) compared with the radiation only group. Furthermore the $TGF\beta1$ expression was significantly prominant at the alveolar epithelium (p<0.02) and the macrophage in alveolar space (p<0.02). At 8 weeks, the expression of $TNF\alpha\;and\;TGF\beta1$ of most sites, except $TGF\beta1$ of the macrophage of alveolar space (p=0.09), showed no significant difference between 2 groups. Conclusion : This study revealed that early lung damage induced by irradiation was reduced with the addition of Captopril in the latent and early pneumonitis phase. The expression of $TNF\alpha\;and\;TGF\beta1$ at 2 weeks and $TGF\beta1$ at 8 weeks was further decreased in the combined Captopril and radiation group than the radiation only group. From these results, it may be concluded that the proinflammatoy cytokine $(TNF\alpha)$ and fibrogenic cytokine $(TGF\beta1)$ probably play the role of the radioprotective mechanism in Captopril.
