Effects of Silk Protein Hydrolysates on Blood Glucose in C57BL/KsJ db/db Mice (실크단백질 효소 가수분해물이 2형 당뇨 마우스 C57BL/KsJ db/db의 혈당에 미치는 영향)

  • This study was performed to investigate the effect of silk protein hydrolysates on blood glucose in diabetic mice (C57BL/KsJ db/db). The silk protein hydrolysates hydrolyzed by protease contains 87.52% of peptides of which molecular weight was below 2,000 dalton. The content of free amino acids was 14.80 g/100 g silk protein hydrolysates and major free amino acids were Pro, Thr, Arg and Ala. Silk protein hydrolysates were administered to the animals for 9 weeks at doses of 0.2, 0.5% and 0.8% solution. The body weight increase in the 0.5 and 0.8% fed groups were higher than control group. Food and water intake in the silk protein hydrolysates fed groups were lower than control group. The weight of liver was not different among groups, while the weight of kidney in control group was higher than silk protein hydrolysates fed groups. The blood glucose level in silk protein hydrolysates fed groups was lower than control group. In the glucose tolerance test, the blood glucose level in control group was the highest at 15 minutes after glucose injection while those in silk protein hydrolysates fed groups were the highest at 30 minutes. Results in this study suggest that silk protein hydrolysates show hypoglycemic effect in C57BL/KsJ db/db mice.

Enzymatic Preparation and Antioxidant Activities of Protein Hydrolysates from Tenebrio molitor Larvae (Mealworm) (갈색거저리 유충 단백가수분해물의 제조 및 항산화 활성)

  • The present study was carried out to evaluate the applicability of Tenebrio molitor larvae (mealworm) as a health functional food material in order to contribute to the development of the domestic insect industry and health functional food industry. Protein hydrolysates were prepared from mealworm powder by enzymatic hydrolysis using five different proteases (alcalase, bromelain, flavourzyme, neutrase, and papain), and the hydrolysates were then tested for their antioxidant activities. Based on available amino group contents and sodium dodecyl sulphate-polyacrylamide gel electrophoresis analyses, mealworms treated with alcalase ($4,781.39{\mu}g/mL$), flavourzyme ($5,429.35{\mu}g/mL$), or neutrase ($3,155.55{\mu}g/mL$) for 24 h showed high degree of hydrolysis (HD) value, whereas HD values of bromelain ($1,800{\mu}g/mL$) and papain-treated ($1,782.61{\mu}g/mL$) mealworms were much lower. Protein hydrolysates showing high HD values were further separated into > 3 kDa and ${\leq}3kDa$ fractions by a centrifugal filter system and then lyophilized, and the production yields of the low molecular weight protein hydrolysates (${\leq}3kDa$) by alcalase, flavourzyme, and neutrase were 42.05%, 26.27%, and 30.01%, respectively. According to the RC_{50} values of the protein hydrolysates (${\leq}3kDa$) obtained from three different antioxidant analyses, all three hydrolysates showed similar antioxidant activities. Thus, alcalase hydrolysates showing the highest production yield of low molecular weight protein hydrolysates were further tested for their inhibitory effects on peroxidation of linoleic acid by measuring thiobarbituric acid values, and the results show that peroxidation of untreated linoleic acid increased dramatically during 6 days of incubation. However, pretreatment with the hydrolysates ($100{\sim}800{\mu}g/mL$) significantly inhibited linoleic acid peroxidation in a dose-dependent manner over 6 days.

  • Hydroxyproline and Pro-Hyp dipeptide are the digestive products of collagen hydrolysate called collagen peptide. Some suggested that collagen peptides could improve aged or damaged skins, however, the effects of collagen peptides on the skin have not been known. In this study, we investigated the effects of digestive products of collagen peptides, hydroxyproline and Pro-Hyp dipeptide on skin quality using the UV-damaged dorsal skin of hairless mouse as a model system. Female SKH hairless mice were pre-irradiated with UV for 7 weeks, and then hydroxyproline, Pro-Hyp dipeptide were orally administered for 7 weeks with UV irradiation. Wrinkle formation (by replica image), skin elasticity, barrier status (by TEWL, transepidermal water loss), epidermis thickness, and biophysical changes in the stratum comeum (by hematoxylin & eosin staining) were examined. With the oral peptide treatment, effects such as skin barrier maintenance, anti-skin thickening, and recovery of the stratum corneum were observed. These results indicate that oral intake of collagen peptides may have beneficial effects on damaged skin cells.

Effects of Hepcidin Hormone on the Gene Expression of Ferroportin and Divalent Metal Transporter 1 in Caco-2 Cells and J774 Cells (Caco-2 소장세포와 J774 대식세포에서 Hepcidin 호르몬이 철분 수송체 Ferroportin과 Divalent Metal Transporter 1의 유전자 발현에 미치는 영향)

  • Hepcidin is a peptide hormone produced by the liver, of which secretion is closely related to iron status in the body. However, little is known about the molecular mechanism(s) by which this peptide regulates body iron homeostasis. The purpose of this study was to determine the effects of hepcidin treatment within the physiological concentration range on the expressions of two different iron transporter proteins-ferroportin (FPN) and divalent metal transporter 1 (DMT1). Differentiated Caco-2 intestinal cells and macrophage J774 cells were treated with either synthetic hepcidin or hepcidin-rich fraction separated from human urine at the concentration of 10 nM and 100 nM for 24 hours. Results show that hepcidin treatment in differentiated Caco-2 cells or in J774 cells did not change the level of either FPN mRNA or DMT1 mRNA. On the other hand, hepcidin treatment at the dose of 100 nM significantly decreased the FPN protein levels and DMT1 protein levels in differentiated Caco-2 cells. Similarly, urinary hepcidin treatment (10 nM & 100 nM) also significantly decreased the levels of FPN and DMT1 proteins in J774 macrophage cells. These results showed that hepcidin might play an important role in the regulation of iron homeostasis by lowering the protein levels of iron transporter FPN and DMT1 both in enterocytes and in macrophage cells.

Fermentation Characteristics of Cheese Slurry prepared from Caseinates (카세인을 이용한 치즈곤죽의 발효특성)

  • To shorten the processing of cheese slurry, four different slurries, ie, Control, Cheddar 1 and 2, and Italian-type that were made of Na-caseinates, cream, trace elements, lactic culture, and enzymes were fermented at $30^{\circ}C$ for 7days with daily stirring. PH, titratable acidity, soluble nitrogen, viable cell count, active SH groups, total volatile fatty acid, free fatty acid, electrophoretic patterns of degraded caseins, and viscosity were analyzed to investigate physicochemical properties of fermented slurries. Acid production was accelerated in the cheese slurries with protease than that without the enzyme and PH of the former was decreased after three days of fermentation to 4.90. The Change of titratable acidity agreed to PH patterns. Soluble nitrogen of the Control slurry was increased slowly for four days and then rapidly to 40% of total nitrogen while those containing protease to 70%. The protease of lactic cultures used (Streptococcus lactis and Streptococcus cremoris) broke down as-casein more rapidly than $\beta$-casein and most proteins were degraded to peptides and amino acids after three days of fermentation. Total volatile fatty acids were increased by added lipase and free fatty acids composition analyzed by GLC in cheddar slurry with 0.00001% lipase was similar to that of commercial cheddar cheese, while that in Italian-type slurry was a half of that in commercial Italian cheese. Active SH groups were increased in the cheese slurries with glutathione from fourth day of fermentation. The viscosity of slurries decreased very rapidly by addition of protease.

  • 계산을 통한 단백질 상호작용 예측 기법의 중요성이 제기되면서 많은 단백질 상호 작용 예측 기법이 제안되고 있다. 하지만 이러한 기법들이 일반 사용자가 손쉽게 사용할 수 있는 서비스 형태로 제공되고 있는 경우는 드물다. 본 논문에서는 현재까지 알려진 단백질 상호작용 예측 기법 중 예측 기법의 완성도가 높고 상대적으로 예측 정확도가 높은 것으로 알려진 도메인 조합 기반 단백질 상호 작용 예측 기법을 PreSPI(Prediction System for Protein Interaction)라는 서비스 시스템으로 설계하고 구현하였다. 구현된 시스템이 제공하는 기능은 크게 도메인 조합 기반 단백질 상호 작용 예측 기법을 서비스 형태로 만들어 제공하는 기능으로 입력 단백질 쌍에 대한 상호작용 예측이 중심이 된 핵심기능과, 핵심 기능으로부터 파생되는 기능인 부가 기능, 그리고 주어진 단백질에 대한 도메인 정보검색 기능과 같이 단백질 상호작용에 관하여 연구하는 연구자에게 도움이 되는 일반적인 기능으로 구성되어 있다. 계산을 통해 단백질 상호 작용을 예측하는 시스템은 대규모계산이 요구되는 경우가 많아 좋은 성능을 갖추는 것이 중요하다. 본 논문에서 구현된 PreSPI 시스템은 서비스에 따라 적절히 그 처리를 병렬화 함으로써 시스템의 성능 향상을 도모하였고, PreSPI 가 제공하는 기능을 웹 서비스 API 로 Deploy 하여 시스템의 개방성을 지원하고 있다. 또한 인터넷 환경에서 변화되는 단백질 상호 작용 및 도메인에 관한 정보를 유연하게 반영할 수 있도록 시스템을 계층 구조로 설계하였다. 본 논문에서는 PreSPI 가 제공하는 몇 가지 대표적인 서비스에 관하여 사용자 인터페이스를 중심으로 상술함으로써 초기 PreSPI 사용자가 PreSPI 가 제공하는 서비스를 이해하고 사용하는 데에도 도움이 되도록 하였다.있어서 자각증상, 타각소견(他覺所見)과 함께 이상(異常)은 확인되지 않았으며 부작용도 없었다. 이상의 결과로부터, ‘펩타이드 음료’는 경증고혈압 혹은 경계역고혈압자(境界域高血壓者)의 혈압을, 자각증상 및 혈액${\cdot}$뇨검사에도 전혀 영향을 미치지 않고 저하시킨다고 결론지었다.이병엽을 염색하여 흰가루 병균의 균사생장과 포자형성 등을 관찰한 결과 균사가 용균되는 것을 볼 수 있었으며, 균사의 용균정도와 분생포자형성 억제 정도는 병 방제효과와 일치하는 경향을 보였다.을 의미한다. IV형은 가장 후기에 포획된 유체포유물이며, 광산 주변에 분포하는 석회암체 등의 변성퇴적암류로부터 $CO_{2}$ 성분과 다양한 성분의 유체가 공급되어 생성된 것으로 여겨진다. 정동이 발달하고 있지 않으며, 백운모를 함유하고 있는 대유페그마타이트는 변성작용에 의한 부분용융에 의해 형성된 멜트에서 결정화되었으며, 상당히 높은 압력의 환경에서 대유페그마타이트의 결정화작용 과정에서 용리한 유체의 성분이 전기석에 포획되어 있다. 이때 용리된 유체는 다양한 성분을 지니고 있었으며, 매우 낮은 공융온도와 다양한 딸결정은 포유물 내에 NaCl, KCl 이외에 적어도 $CaCl_{2},\;MgCl_{2}$와 같은 성분을 포함하고 있음을 지시한다. 유체의 용리는 적어도 $2.7{\sim}5.3$ kbar 이상의 압력과 $230{\sim}328^{\circ}C$ 이상의 온도에서 시작되었다.없었다. 결론적으로 일부 한방제와 생약제제는 육계에서 항생제를 대체하여 사용이 가능하며 특히 혈액의 성분에 유의한 영향을 미치는 것으로 사료된다. 실증연구가 필요할 것으로 사료된다.trip과 Sof-Lex disc로 얻어진 표면은 레진전색제의 사용으로 표면조도의 개선

  • Lentils (Lens culinaris) are a nutritious and popular food throughout the world. This review provides an overview of the nutritional and functional properties of lentils. Lentils have high nutritional value: high protein, high fiber, low fat, and a variety of minerals. The addition of lentils to other foods could enhance their health benefits. Additionally, there is no decrease in the amino acid content in lentils when cooked; rather, the amino acids increased and the phytic acids decreased. Lentils are generally known to have biomedical functions including antioxidative, anti-cancer, anti-inflammatory, antihypertensive, and thrombolytic properties. The data presented here show that fermentation would increase the amount of bioactive substances in the beans. Therefore, fermentation techniques could be a new approach to produce lentil-based foods. They are also eco-friendly, cost-effective, and feasible processes. This review has suggested a future development of new lentil-related foods using traditional fermentation technology.

Purification and Characterization of the Bacteriocin Produced by Lactococcus sp. KD 28 Isolated from Kimchi (김치에서 분리한 Lactococcus lactis가 생산하는 박테리오신의 정제 및 특성)

  • The bacterial strain isolated from Kimchi showed antibacterial activity against Micrococcus luteus IAM 1056. The selected strain was identified as Lactococcus lactis by 16S rRNA nucleotide sequence analysis and named as Lactococcus sp. KD 28. The treatment of culture supernatant with proteinase K removed antibacterial activity, indicating its proteinaceous nature, a bacteriocin. This bacteriocin was sensitive to hydrolytic enzymes such as ${\alpha}$-chymotrypsion, trypsin, proteinase K, lipase, ${\alpha}$-amylase and subtilisin A. The bacteriocin was highly thermostable and resistant to heating at $80^{\circ}C$ for up to an hour but 50 % of the total activity was remained at $100^{\circ}C$ for 30 min. The pH range from 2.0 to 8.0 had no effect on bacteriocin activity and it was not affected by solvents such as acetonitrile, isopropanol, methanol, chloroform and acetone up to 50% concentration. The bacteriocin showed antibacterial activity against M. luteus IAM 1056, Lactobacillus delbrueckii subsp. lactis KCTC 1058, Enterococcus faecium KCTC 3095, Bacillus cereus KCTC 1013, B. subtilis KCTC 1023, Listeria ivanovii subsp. ivanovii KCTC 3444, Staphylococcus aureus subsp. aureus KCTC 1916, B. megaterium KCTC 1098 and B. sphaericus KCTC 1184. The bacteriocin was purified through ammonium sulfate concentration, SP-Sepharose chromatography and RP-HPLC. The molecular weight was estimated to be about 3.4 kDa by tricine-SDS-PAGE analysis.

  • Differentiation of cells can be induced through modulation of endogenous regulators using exogenous factors. Useful transfection systems to transport a specific exogenous regulator into cell have been tried but still there are many obstacles to overcome. In this study, we examined the transfection efficiency of cell permeable peptides (CPPs) in mouse embryonic stem cell under the various conditions. To identify the CPP-mediated translocation of a protein, we employed recombinant CPP-enhanced green fluorescent protein (EGFP). Viability of R1 cells was different between experimental groups depending on the kind of CPP and the concentration of CPP-EGFP. Translocation of CPP-EGFPs into the R1 cells was not detected until 30 min after CPP-EGFPs treatment in all groups. After 1 hr, translocation of pEP-1-EGFP-N was detected, but it could not be detected in the other group. Transfection of pEP-1EGFP-N was independent on its concentration. The time course did not show saturation even after 24 hr in pEP-1-EGFP-N. These results showed that the permeability depended on the kind of CPP and the location of His-tag in the case of examined CPPs, and did not need biological energy. On summary, the efficiency of transfection of CPP-EGFP depends on the CPP sequences but the culture time is not a key factor in transfection for the mouse embryonic stem cell. For the future studies to improve the efficiency of translocation of protein into embryonic stem cells, it is needed to develop modified CPP or mediator. The studies would be very useful to induce the differentiation of embryonic stem cells.

Isolation and Characterization of Bacillus Species Having Antifungal Activity Against Pathogens of Ginseng Damping Off (인삼모잘록병원균에 항균활성을 갖는 Bacillus 균의 분리 및 특성조사)

  • This study was performed to select potentially available biological control agent from soil bacteria for prevention of ginseng damping off. More than five hundred strains were isolated from ginseng rhizosphere soil. By testing antifungal activity, we have selected three soil bacteria strains and their ability to produce antibiotics and lytic enzymes such as cellulase, protease and pectate lyase was examined. Also, the presence of genes for biosynthesis of lipopeptide such as fengycin, bacillomycin D, surfactin, iturin A, and zwittermicin A was investigated in selected strains. All three strains produced cellulase, protease, and xylanase. Moreover, these strains had gene for biosynthesis of bacillomycin D, surfactin, and iturin A. ES1 and ES3 strains were identified Bacillus methylotrophucus and ES2 was confirmed Bacillus amyloliquefaciens using phylogenetic analysis on the basis of 16S rRNA gene sequences. In field test, control value of ES1, ES2 and ES3 treatment was 32.4%, 46.8% and 36.7%, respectively. This results indicate that antagonistic microbes with high ability of antifungal and lytic enzyme activity can be used as a useful biological control agent to control ginseng damping off.
