작업환경을 위한 TVL의 근거 - PENTACHLOROPHENOL(1)

  • 펜타클로로페놀의 현재 직업적 노출기준은 $0.5\;mg/m^3$(TLV-TWA)으로 권고하고 있다. 이 수치는 눈, 점막과 상기도의 염증과 발한, 열, 위장 불만, 시각 장해, 중추 신경계와 심혈관계 장해를 유발할 수 있는 급성 독성의 가능성을 최소로 하는 것을 목적으로 설정하였다. Chloracne는 공업용 펜타클로로페놀을 사용하거나 펜타클로로페놀을 생산하는 근로자들에 대해서 보고하였다. 펜타클로로페놀은 피부를 통해서 즉시 흡수되는데, 이 물질의 조직 독성은 펜타클로로페놀을 포함한 용액이나 펜타클로로페놀로 오염된 의류와 접촉된 사람에게서 보고되었다. 따라서 피부경고주석은 적합한 것으로 판단되었다. 펜타클로로페놀의 발암성 잠재력에 대한 명확한 증거는 간세포성과 신장에 가까운 신생물이 관찰된 설치류 생물 검정에서 발견되었다. 그러므로 본 물질은 인간에게 불명확한 관련성을 가진 동물성 발암물질(A3)로 설정하였다. 충분한 자료가 확보되지 않아 감작성 (SEN) 경고주석 또는 TLV-STEL은 권고되지 않았다. 펜타클로로페놀은 생물학적 노출지수(BEIs)가 권고된 물질이다.

Effects of Proteins on the Reactivity of Various Phenolic Compounds with the Folin-Ciocalteu Reagent (다양한 페놀성 물질과 Folin-Ciocalteu 시약과의 반응성에 미치는 단백질의 영향)

  • The Folin-Denis assay using the Folin-Ciocalteu (F-C) reagent has been commonly used for analyzing the total phenolic compound content in various food products. In the present study, the effects of proteins on the reactivity of the F-C reagent with different phenolic compounds were investigated. Bovine serum albumin (BSA) or skim milk proteins showed a concentration-dependent increase in color response in the Folin-Denis assay; these proteins decreased the color response of most phenolic compounds tested. The reactivity of phenolic compounds was significantly less pronounced in the presence of BSA and this interference was greater at higher concentrations of phenolic compounds. The reactivity of phenolic compounds with the F-C reagent was reduced significantly by their oxidation; the reaction of the oxidized products with the F-C reagent was more severely affected by BSA. The interfering effects in the Folin-Denis assay might be attributable to binding interactions of phenolic compounds with proteins.

추출용매와 농도에 따른 배 페놀성물질의 추출

  • 국민생활 수준의 향상과 더불어 배의 생산은 꾸준한 증가추세를 보이고 있고 2010년까지 계속 증가될 전망이다. 한편 재배기술에 의한 미숙, 태풍에 의한 낙과, 수확 후 저장중 손상으로 약 15%의 배가 유실된다. 이러한 상품성이 낮은 배의 효율적인 활용 방안을 모색하기 위한 일환으로 배를 기능성식품 소재로 이용하고자 배의 페놀성물질 추출효율을 알아보기 위하여 추출용매와 농도에 따른 추출율을 비교하였다. 신고배 과피를 신선시료, 열풍건조시료와 동결건조시료 3가지 상태로 하여 methanol, ethanol, acetone과 물 추출물을 Folin-Denis법으로 총페놀성물질의 함량을 측정하여 비교하였다. 시료의 상태에 따라 추출효율에 약간의 차이를 볼 수 있었는데 신선시료에서 모든 농도범위에서 아세톤의 추출효율이 가장 높았고 메탄올, 에탄올과 아세톤 3용매에서 모두 60∼80% 농도범위에서 높은 추출효율을 보였다. 그리하여 3가지 용매에서의 최적추출농도를 알아보고자 60∼80% 범위내에서 5%농도 간격으로 페놀성물질을 추출하여 그 함량을 측정하였다. 결과 메탄올에서는 70%와 75%에서, 에탄올에서는 70%에서, 아세톤에서는 65%, 70%, 75%에서 유의적으로 높은 함량을 나타내었다. 따라서 신선시료에서 메탄올, 에탄올과 아세톤으로 페놀성물질 추출 시 약 70% 농도로 추출하는 것이 가장 효율적이라고 판단된다. 또한 물 추출은 가장 낮은 효율을 보였다. 열풍건조시료에 대한 각용매의 농도별 추출효율은 아세톤의 농도가 높을수록 추출효율이 떨어졌고 특히 무수 아세톤에서는 그 효율이 물 추출효율보다 더 낮게 나타났다. 20∼80% 범위에서 아세톤의 추출효율이 가장 높았고 다음은 에탄올, 메탄을 순이었다. 아세톤은 40∼60% 범위에서, 에탄올은 60∼80% 범위에서, 메탄올은 60∼99% 범위에서 높은 추출효율을 보여 각각 이 농도범위에서 다시 총함량을 측정하였다. 결과 각농도에 따른 추출함량에 유의적인 차이를 나타내지 않았다. 동결건조시료에서 아세톤에 의한 추출효율은 열풍건조시료에서와 비슷한 결과를 보였고 또한 20∼80% 농도범위에서 메탄올의 추출효율이 가장 낮았고 다음은 에탄올, 아세톤의 순으로 높았다. 물 추출효율은 20% 메탄을 추출효율과 비슷한 수준이었다. 메탄올에서는 60∼80%, 에탄올과 아세톤에서는 40∼60% 범위에서 높은 추출효율을 보였다. 마찬가지로 이 농도범위에서 최적의 추출농도를 확인하고자 각 용매와 농도별로 페놀성물질을 추출하여 그 함량을 측정한 결과 메탄올에서는 5% 농도차이가 그 추출효율에 유의적인 영향을 주지 않는 것으로 나타났다. 에탄올에서는 40%에서 가장 높은 함량이 측정되었고 아세톤에서는 50%에서 측정되었다. 따라서 시료의 상태와 상관없이 배 과피의 페놀성물질 추출용매로는 40∼70%의 함수 아세톤이 적합한 것으로 사료된다.

  • Cell growth and production of phenolic compounds by callus cultures of Panax ginseng C. A. Meyer were investigated under various phytohormones concentrations and inoculum size. The results indicated that the cell growt was improved by a MS medium supplemented with 2 mg/L of CPA. The maximum cell yield was obtained at inoculum size of 1 g/flasd. The production of phenolic compounds in the callus cultures was higher than those in the ginseng root. Especially, one cell line (20601) showed the highest content of phenolic compounds and antioxidant activity.

  • 혼합상태에서 유기화합물들이 토양에 흡착하는 성질에 관해서 5개의 유기화합물(페놀,2,4-디크로로페놀, 2,4,6-트리크로로페놀, 부루신, 따이오유리아)과 2종류의 토양을 이용하여 연구하였다. 일반적으로 따이오유리아를 제외하고는 한가지 물질이 혼합상태에 있는 다른물질이에 의 흡착력이에 제약을 받아서 단독으로 있을때 보다 흡착율이 낮았다. 따이오유리아는 단독인 경우보다 혼합상태일 때 더 많은 흡착을 보였는데 이는 혼합상태에서 오히려 흡착이 증가할수도 있음을 나타낸다. 혼합이 흡착에 비치는 영향은 페놀의 혼합상태와 단독상태에서의 흡착상태를 여러가지 형태로 비교하여 검토하였는데 산성이고 유기물질이 많은 토양보다 혼합으로 인한 흡착의 억제 효과가 뚜렷하였다.

Analysis of Phenolic Substances Content in Korean Plant Foods (국내산 식물성 식품중 페놀성 물질의 함량 분석)

  • The phenolic substances contents of 45 plant foods in Korean diet were determined by different methods. Total phenolics contents by Folin-Denis method were $0.1{\sim}5.8%$ (dry matter basis), in which persimmon leaf, chestnut's inner skin, Chinese quince, walnut, sunflower seed and arrowroot exhibited the higher levels above 2%. Condensed tannin contents by vanillin method were $0{\sim}48%$, in which Chinese quince and chestnut's inner layer gave very high levels. Protein-precipitable phenolic substances ranged from 0.4% to 2.2%, in which chestnut's inner layer, walnut and Chinese quince had the highest content. The ability of phenolics to form precipitate was higher with pepsin and albumin than with trypsin. Among different phenolics content, total phenlolics correlated significantly with protein-precipitable phenolics (r=0.65) and condensed tannin (r=0.56). Chinese quince, chestnut's inner skin and sorghum showed a relatively lower degree of polymerization, as expressed by vanillin/FolinDenis ratio. Processed foods from buckwheat, acorn, mugwort and arrowroot showed a lower content of phenolic substances, suggesting a negligible adverse effect on the bioavailability of food proteins, if any.

Factors Affecting Reactivity of Various Phenolic Compounds with the Folin-Ciocalteu Reagent (다양한 페놀성 물질과 Folin-Ciocalteu 시약의 반응성에 미치는 영향 요인 평가)

  • The Folin-Ciocalteu (F-C) reagent has been extensively used for quantifying total phenolic contents in many different types of food materials. Since several different procedures of the assay methods using the F-C reagent have been applied, we investigated changes in reactivity of various phenolic compounds with the F-C reagent under three different assay conditions and factors affecting reactivity. Among 10 standard compounds tested, compounds with high hydroxyl density (number of -OH/molecular weight) showed a largely different response according to addition sequence of the F-C reagent or $Na_2CO_3$. Preincubation in $Na_2CO_3$ significantly reduced the reactivity of the phenolic compounds bearing galloyl moiety (e.g. gallic acid, tannic acid, and epigallocatechin-3-gallate) with the F-C reagent, while monophenol compounds including ferulic acid and sinapinic acid were more stable as compared to diphenols. There was little change in response to the F-C reagent of all phenolic compounds incubated in acidic pH; their reactivity except ferulic acid was reduced significantly when incubated in neutral or alkaline pH. Changes in reactivity of gallic acid incubated in $Na_2CO_3$ or neutral/alkaline pH conditions were the most prominent. $H_2O_2$ generated from phenolic compounds did not affect the reaction with the F-C reagents. The present results suggest that reactivity of different phenolic compounds with F-C reagent was affected considerably by different procedures of the assay, and the total phenolic contents could be fluctuated according to standard compounds and assay scheme.

Accumulation of Crude Lipids, Phenolic Compounds and Iron in Rusty Ginseng Root Epidermis (적변삼 외피에서 지질, 페놀성물질 및 철 성분의 축적에 관한 연구)

  • The study was conducted to investigate the tissue and chemical characteristics of rusty root epidermal cells. In histological study, the rusty symptoms were frequently observed in the epidermis of ginseng root and to be yellow under microscopic observation. Disks of the epidermal cell tissue of the rusty root were usually 2 and 3 times greater in the number of cell layer and thickness of cell wall than the healthy root, respectively. The color degree of methanol extracts from the rusty root epidermis was 5.5 times higher than that of the healthy root. And the extracts of rust matter in the root epidermis were easily dissolved in polar solvents compared to nonpolar solvents. UV-absorption spectra of methanol extracts in various fractions of phenolics showed a maximum peak between 275∼280 nm. The crude lipids and phenolic compounds such as acid insoluble bound phenolics, acid insoluble esterified phenolics, acid insoluble condensed phenolics, insoluble bound phenolics and free phenolics were also more in the rusty root epidermis than in the healthy one. Fe content in the rusty root epidermis was 2.7 times higher than that of healthy one. It was presumed that the phenolic compounds(precursor of the rusty) in association with lipid and iron in the root epidermis might defence the root when ginseng root was depressed by the unfavorable conditions in soil and/or portions of a root system were subjected to anoxic conditions.

Degradation of Phenol by "TiO2 Ceramic Membrane+UV+H2O2" AOP ("TiO2 촉매막+UV+H2O2" 고도산화법(AOP)을 이용한 페놀 분해)

  • Photocatalytic oxidation conditions of reactant recirculation flow rate 275 mL/min, aeration rate 2 LPM and $UV+TiO_2+H_2O_2$(500 mg/L) proved to be appropriate for water including organic materials treatment. With increasing turbidity and suspended solids concentration, at turbidity 10 NTU-suspended solids concentration 29 mg/L the phenol degradation efficiency increased, which in turn decreased at turbidity 50 NTU-suspended solids concentration 170 mg/L, however no significant differences were observed, demonstrating similar results with those obtained at zero turbidity and suspended solids concentration. The degradation efficiency of phenol decreased with increasing influent phenol concentrations. The $UV+TiO_2+H_2O_2$ photocatalytic advanced oxidation process conducted is considered to be possibly applied to the drinking water treatment, and the post-treatment process of biological wastewater treatment.

  • Chemical structures of phenolic antioxidative components of defatted Terminalia chebula R. were elucidated by used MS and H-NMR. The results showed that the phenolic antioxidative components were identified as ferulic acid, vanillic acid, p-coumaric acid in free-acid extracts, and caffecic acid, vanillic acid and p-coumaric acid in soluble-acid extracts, and caffeic acid, phloroglucinol and pyrogallol in insoluble-bound extracts, respectively.
