Antioxidative Activities by Water-Soluble Extracts of Morus alba and Cudrania tricuspidata (뽕나무(Morus alba)와 꾸지뽕나무(Cudrania tricuspidata)의 수용성 추출물에 의한 항산화 활성)

  • The antioxidative activities of water-soluble extracts from leaves and stem bark of Morus alba and Cudrania tricuspidata were compared in vitro experimental models. Antioxidative activities were measured by inhibition activity against lipid peroxidation of mouse liver microsome, and they were showed in the following order; stem bark of C. tricuspidata(53%)>stem bark of M. alba(43%)>leaves of C. tricuspidata(38%)>leaves of M. alba(43%). In antioxidative activities determined by thiocyanate method and TBA method, the water-soluble extract of stem bark of C. tricuspidata showed the highest antioxidative activity. The water-soluble extracts of leaves were slightly stronger than other extracts in DPPH$({\alpha},{\alpha}'-diphenyl-{\beta}-picrylhydrazyl)$ method. The concentrations of total polyphenolic compound from water-soluble extracts of leaves and stem bark of M. alba and C. tricuspidata were 1.32%, 1.28%, 1.34% and 1.30% respectively. In these results, the water-soluble extract of stem bark from Cudrania tricuspidata showed the highest antioxidative activity.

Quantitative Determination of Phenol in Water Using GC-MS after Liquid-Liquid Extraction and Acetylation (액-액 추출과 아세틸화 후 GC-MS를 이용한 물 중 phenol의 분석)

  • BACKGROUND: The phenol concentrations in water samples were determined using gas chromatography after derivatization of the analyte to phenyl acetate followed by extraction using a large volume of solvent. However, this procedure requires an additional purification step and is not analytically efficient. METHODS AND RESULTS: In this study, phenol was first extracted from an acidified water sample using ethyl acetate and then acetylated using acetic anhydride in the presence of a small amount of water and $K_2CO_3$. The derivative was extracted using 1mL of n-butyl acetate. One microliter of the extract was analyzed by GC-MS without further purification. The calibration curve showed good linearity with the $r^2$ value of 0.9968. The method detection limit and the limit of quantitation were estimated to be $0.18{\mu}g/L$ and $0.56{\mu}g/L$, respectively. Repeatability (RSD, n=3) and recovery (n=3) were 9.1%-4.3% and 90.6%-110.5%, respectively. The concentrations of phenol in a few samples of stream water were distributed in the range of $2.51-7.51{\mu}g/L$. CONCLUSION: This method is simpler and faster to implement than those currently utilized and shows high analytical reliability. It can be applied to the quantitative determination of phenol concentrations in surface water and groundwater samples.

  • 정보공유를 목적으로 제안된 웹의 활성화와 함께 유용한 정보들이 웹상에 기하급수적으로 등장함에 따라 정보공간의 확장으로 인한 검색 신뢰도의 저하 문제에 직면하게 되었다. 본 연구에서는 대용량 웹 환경하에서 사용자의 정보발견을 돕기 위해 텍스트이외의 새로운 요소들을 사용하여 특정장르문서를 발견하는 개념을 도입하였다. 먼저 사용자가 발견하고자 하는 장르의 모습을 텍스트, URL정보, 링크 정보. 문서구조 정보 등의 장르 식별요소 값을 이용해 표현한 후, 후보 문서들의 장르관련도를 측정함으로써 특정장르 문서를 검색한다. 각 장르식별요소값은 나름대로의 방법에 의해 계산되는데 $0{\sim}1$사이의 값을 가지며, 종합적인 장르관련도는 각 장르식별요소값의 증거통합 방법에 의해 구한다. 본 논문에서는 각 장르식별요소들의 역할과 장르식별요소가 장르발견에 미치는 영향을 알아보며, 최종적으로 특정 장르 문서발견에 있어서의 검색 신뢰도 향상을 보이기 위해 실험모델을 설계/구현하였다. 본 실험은 웹 문서를 대상으로 하는데, 아직까지 URL, 링크 정보를 모두 갖춘 테스트컬렉션이 없기 때문에 실험을 위해 일반적인 웹 문서로 직접 구성한 컬렉션을 사용하였다. 발견하고자 하는 장르는 "컴퓨터 분야의 컨퍼런스 홈페이지"로 정하였으며 30개의 컴퓨터 분야를 선정하였다. 비교대상으로는 일반 웹 검색 엔진인 알타비스타와 메타검색 엔진인 메타크롤러를 선택하였고. 각 질의에 대해 상위 30개의 결과를 대상으로 정확도를 평가하였다. 결과로서 각 장르식별요소들은 모두 검색 신뢰도의 향상에 기여를 하며, 제안하는 방법은 알타비스타와 메타크롤러에 비해 각각 평균적으로 67.34%, 71.78%의 검색 신뢰도 향상을 보임을 입증하였다.적응에 문제점을 가지기도 하였다. 본 연구에서는 그 동안 계속되어 온 한글과 한잔의 사용에 관한 논쟁을 언어심리학적인 연구 방법을 통해 조사하였다. 즉, 글을 읽는 속도, 글의 의미를 얼마나 정확하게 이해했는지, 어느 것이 더 기억에 오래 남는지를 측정하여 어느 쪽의 입장이 옮은 지를 판단하는 것이다. 실험 결과는 문장을 읽는 시간에서는 한글 전용문인 경우에 월등히 빨랐다. 그러나. 내용에 대한 기억 검사에서는 국한 혼용 조건에서 더 우수하였다. 반면에, 이해력 검사에서는 천장 효과(Ceiling effect)로 두 조건간에 차이가 없었다. 따라서, 본 실험 결과에 따르면, 글의 읽기 속도가 중요한 문서에서는 한글 전용이 좋은 반면에 글의 내용 기억이 강조되는 경우에는 한자를 혼용하는 것이 더 효율적이다.이 높은 활성을 보였다. 7. 이상을 종합하여 볼 때 고구마 끝순에는 페놀화합물이 다량 함유되어 있어 높은 항산화 활성을 가지며, 아질산염소거능 및 ACE저해활성과 같은 생리적 효과도 높아 기능성 채소로 이용하기에 충분한 가치가 있다고 판단된다.등의 관련 질환의 예방, 치료용 의약품 개발과 기능성 식품에 효과적으로 이용될 수 있음을 시사한다.tall fescue 23%, Kentucky bluegrass 6%, perennial ryegrass 8%) 및 white clover 23%를 유지하였다. 이상의 결과를 종합할 때, 초종과 파종비율에 따른 혼파초지의 건물수량과 사료가치의 차이를 확인할 수 있었으며, 레드 클로버 + 혼파 초지가 건물수량과 사료가치를 높이는데 효과적이었다.\ell}$ 이었으며 , yeast extract 첨가(添加)하여 배양시(培養時)는 yeast extract 농도(濃度)가 증가(增加)함

Monitoring of Drying Conditions for Gastrodia elata Blume by Response Surface Methodology (반응표면분석에 의한 천마의 건조조건 모니터링)

  • The Chunma (Gastrodia elata Blume) requires a simple pre-treatment, due to inconvenience or preservation, toxicities, and pungencies in intact state. Various drying conditions were examined to establish the physicochemical properties of Chunma by the response surface methodology (RSM). Weter activity and total phenolic acid contents of Chunma dried extracts were optimal at $79.56^{\circ}C$ and $79.64^{\circ}C$ for 7.17 hr and 8.74 hr, respectively, for their characteristics. Electron-donating ability and nitrite-scavenging activity were optimal at 79.79 and $7.99^{\circ}C$ for 8.57 and 8.07 hr. We generated a regression equation for each variable and then superimposed the optimal condition in acquiring values for each frying process factor, physicochemical properties, resulting in that the optimal drying condition was at $75{\sim}80^{\circ}C$ for $8{\sim}9 hr$.

Comparison of Quality Characteristics of Korean Cabbage Kimchi Added with Germinated Brown Rice Extracts and Korean Cabbage Kimchi on the Market (발아현미 추출물 첨가 배추김치와 시판배추김치의 품질특성 비교)

  • This study was designed to investigate quality characteristics of Korean cabbage kimchi added with germinated brown rice extracts (GBRC; (A) and CBREP; (B)) and Korean cabbage kimchi on the market mom the result pH and total acidity were shown to be similar in all samples. In the case of Korean cabbage kimchi added with (A) and (B), content of total sugar and reducing sugar were higher than those of Korean cabbage kimchi on the market. Color values (L, b) were shown to be similar in all samples in which a value was lower than that of Korean cabbage kimchi on the market. The amounts of total amino acid and free amino acid of Korean cabbage kimchi added (A) and (B) were higher than those of Korean cabbage kimchi on the market. But the amount of $\gammma$-aminobutyric acid (GABA) in main functional components of germinated brown rice was shown to be low. Total phenol content and other antioxidant and anticoagulant activities of Korean cabbage kimchi fortified with (A) and (B) were higher than those of Korean cabbage kimchi on the market. In conclusion, Korean cabbage kimchi added with (A) and (B) were shown to be similar in fermentation tendency compared to Korean cabbage kimchi on the market and, the functional properties could be enhanced by the addition of (A) and (B).

Quality characteristics of Sikhye prepared by different ratio of Seomaeyaksuk (Artemisia argyi H) extract (섬애약쑥 추출물의 첨가 비율에 따른 식혜의 품질특성)

  • This study was conducted to increase the functionality of Korean traditional beverage, Sikhye. The Sikhye were prepared by supplementation up to 20% of Seomaeyaksuk extract (SE) and saccharification for five hours (h). During the saccharification, turbidity of SE-added Sikhye (SES) was the highest at two hours of saccharification and then lowered gradually. L vlaue of SES did not show significant difference feduring saccharification, and both a and b value of Sikhye were increased as saccharification time passed and SE addition ratio was increased. The pH before saccharification was not significantly different by SE addition ratio but it was increased to 5.68-5.73 at 5 h saccharification. Soluble solid, total sugar and reducing sugar contents were enhanced as saccharification time and SE addition ratio were increased. The total phenolic compounds contents and DPPH radical scavenging activity showed a similar trend by saccharification time and SE addition. In sensory test, the evaluations of 5% SES and control were not significant different, but 20% SES group showed significant lower evaluation than other groups because strong color and mugwort flavor. From all of these results, physicochemical characteristics and antioxidant activity of SES was improved by SE addition but sensory characteristics was lower. Acceptable addition ratio of SE in Sikhye is determined to be within 15%.

  • Curcumin is a natural phenolic yellow curry spice, derived from the tumeric, which has been used for the treatment of diseases associated with oxidative stress and inflammation. Curcumin is known to have both anti-oxidative and anti-inflammatory properties. These properties can be beneficial to protect the brain from the neurodegenerative diseases. We now report the neuroprotective effects of curcumin pretreatment in primary hippocampal neurons to glutamate-induced excitotoxicity. Pretreatment of embryonic mouse hippocampal cell cultures with low does of curcumin protected neurons against glutamate-induced death, however, this neuroprotection was not correlated with the modulation of oxidative stress. Interestingly, high dose of curcumin showed the cytotoxicity in primary cultured hippocampal neurons. Immunoblot analyses showed that levels of stress response. protein HSP70 were significantly elevated in neurons exposed to low dose of curcumin, whereas levels of cleaved PARP were increased in neurons exposed to high dose of curcumin. These findings show that curcumin can modulate neuronal responses to glutamate, and suggest possible use of curcumin and related compounds in the prevention and/or treatment of neurodegenerative disorders.

Effect of Germinated Brown Rice Extract Powder on Free Amino Acid Content, Antioxidant and Nitrite Scavenging Ability of the Korean Cabbage Kimchi (동결건조 발아현미 추출분말이 배추김치의 유리아미노산 함량, 항산화성 및 아질산염 소거작용에 미치는 영향)

  • This study investigated the effect of the addition of germinated brown rice extract powder (0, 0.67, 3.35 and 6.70%) to the fermentation of Korean cabbage kimchi. During the fermentation of Korean cabbage kimchi with GBREP, content of total free amino acid and essential amino acid was showed higher value by increasing the addition of GBREP. In particular, ${\gamma}$-aminobutyric acid (GABA) content of Korean cabbage kimchi, prepared with 6.70% GBREP was 2 times higher than control Korean cabbage kimchi. By increasing the content of GBREP, total phenol content was showed high value and the content was more increased during fermentation except control Korean cabbage kimchi. By increasing the content of GBREP, electron donating activity and superoxide radical scavening activity were showed higher value and then showed maximum scavening activity on palatable period. Nitrite-scavenging ability of Korean cabbage kimchi with GBREP was higher than control Korean cabbage kimchi and showed the highest scavenging ability at pH 1.2.

Effects of steeping condition and salinity stress on quality properties in germinated black soybean (발아시 수침 및 염 스트레스에 따른 발아 검정콩의 품질특성)

  • This study investigated the effects of the steeping condition and salinity stress on the quality properties of germinated black soybean. The absorbed water content increased drastically in six hours with the increase in the steeping time and the temperature. The pH decreased as the steeping temperature increased. A gradual increase in the dissolution of the anthocyanin was observed with the steeping time. The adequate steeping conditions were found to have been six hours at $25^{\circ}C$, which showed the needed absorbed water content within a short time. The germination percentage of the black soybeans decreased with the increase in the NaCl concentration. The pinitol and total phenolics contents showed the highest values under the concentrations of 0.7% and 0.3-0.5% NaCl, respectively. The flavonoid content was highest at the 0.1% NaCl concentration. Consequently, the salinity stress during the germination of the black soybeans enhanced the pinitol content and lowered the blood sugar, which gives it an advantage as a functional food material.

Effect of the Extruded Ginseng on Antioxidant Activity (압출 성형 인삼의 항산화 활성 증강 효과)

  • This study was conducted in order to evaluate the antioxidant activity of extruded ginseng in different extracted fractions. Each of the fractions obtained from extruded ginseng and ginseng (control) were extracted with 80% ethanol, and then the lipophilic components were removed with ether while the hydrophilic components were separated with water-saturated butanol. Each of the 80% ethanol/butanol/water layers were collected and evaporated to acquire samples for tests of saponin content and antioxidant activity. The antioxidant activity of extruded ginseng fractions and ginseng fractions were determined via the oxygen radical absorbance capacity (ORAC) assay. Overall, the extruded ginseng samples harbored saponin contents of 2.2 (Rg1), 2.3 (Re), 1.2 (Rc), 1.3 (Rb2), and 2.2 (Rd) times that measured in the ginseng prior to extrusion. Antioxidant capacity was also higher, not only in the 80% ethanol/butanol which harbor a significant quantity of saponin, but also in the water fractions, which harbor relatively low quantities of saponin as compared to the control samples.
