Probiotic Microbial Fermentation of Poncirus trifoliata Extract by Probiotic Strain Isolated from Fermented Gimchi and Antioxidant Activity (발효 김치로부터 분리한 프로바이오틱 균주에 의한 지실추출물 발효와 항산화능 분석)

  • To protect skin problems, new natural material alternative to synthetic antioxidants has been extensively selected from natural sources such as plants, animals, and microorganisms. Poncirus trifoliata of those has been widely used as treatment of allergy, chronic inflammatory diseases, and natural antioxidant. In recent days, microbial fermentation to natural products has been reported to increase feasibly their bioavailability. Accordingly, we performed the fermentation using hot water extract of Poncirus trifoliata by isolated Leuconostocs sp. strain JYK and investigated the change of antioxidative activity. Antioxidative material such as hesperidine naringine, imperatorin, and luteolin was found in hot water extract of Poncirus trifoliata. Total phenolics compounds and flavoniods in hot water extract were $71.2{\pm}4.58GAE(mg/g)$ and $25.1{\pm}4.12$ hesperidine(mg/g), respectively. After fermentation, their values were $89.2{\pm}13.47GAE(mg/g)$ and $31.0{\pm}4.06$ hesperidine(mg/g), respectively. After fermentation, ABTS[2,2-azino-bis (3-ethylbenzthiazoline-6-sulfonic acid)] and DPPH(1,1-diphenyl-2-picrylhydrazyl) radical were highly enhanced from $70.2{\pm}9.01$ to $86.2{\pm}3.72$ and $18.7{\pm}1.81$ to $26.6{\pm}4.06$, respectively. Thus microbial fermentation offers a tool to further increase the bioactive potential of Poncirus trifoliata.

Quality Changes of Peeled Potato and Sweet Potato Stored in Various Immersed Liquids (다양한 침지액 보관에 따른 박피 감자와 고구마의 품질변화)

  • The efficacy of strong acidic electrolyzed water (SAEW) at pH 2.53, with ORP of 1,088 mV and HClO concentration of 91.25 ppm, and low alkaline electrolyzed water (LAEW) at pH 8.756, with ORP of 534 mV and HClO concentration of 105.70 ppm, as storing liquids for peeled potato and sweet potato was evaluated in this study. During storage at $5^{\circ}C$, total phenolic contents and PPO activities of peeled potato and sweet potato stored in SAEW and LAEW were lower than those of control samples stored in tap racer (TW) with 0.85% (w/v) NaCl and 0.5% (w/v) sodium metabisulfite (SMS). Increment in color differences and decreases in hardness of peeled potato and sweet potato stored in SAEW and LAEW were lower than those of controls. Also, SAEW and LAEW inhibited growth of microorganisms for at least 3-6 days of storage. The sensory characteristics of peeled potato and sweet potato stored in LAEW were best during the first half of the storage period, compared to samples preserved by other methods.

Physiological Activities of $Ulmus$ $pumila$ L. Extracts (유근피 추출물의 생리활성)

  • This study was carried out for the functional investigation of the $Ulmus$ $pumila$ L. extracts for use in functional-food processing. Extracts of $Ulmus$ $pumila$ L. were obtained using distilled water and 70% ethanol, and the extracts were tested for their electron-donating ability, SOD-like activity, nitrate-scavenging ability, and anticancer (MDA and A 549 cells) activity. The extraction yields of the water and ethanol extracts were 12.7 and 12.0%, respectively; the polyphenol contents were $623.5{\pm}2.4$ and $710.5{\pm}2.1$ mg/100 g; the electron-donating ability was high in proportion with the density; and the water extract was higher than the ethanol extract (76 and 64%, respectively) at 1,000 ppm. In all the 1,000 ppm densities, the SOD-like activity of the water extract was far higher than that of the ethanol extract (53 and 38%, respectively), and the nitrite-scavenging ability of the ethanol extract was higher than that of the water extract (47 and 43%, respectively). As for the anticancer ability at 1,000 ppm, it was 62% in the water extract and 42% in the ethanol extract in the MDA cell, and 60% in the water extract and 45% in the ethanol extract in the A 549 cell. Thus, the proliferation inhibition ability of the water extract against cancer cells was found to be far higher than that of the ethanol extract (60 and 45%, respectively).

Increment of Physiologically Active Compounds in Germinated Brown Rice Treated with Chitosan and its Effect on Obesity of Rat Fed a High Fat Diet (키토산을 처리한 발아현미의 기능성분 증대 및 비만에 미치는 영향)

  • This study was to investigated the changes of physiologically active components in germinated brown rice treated with chitosan (CGBR) and its anti-obesity effect in rat fed a high fat diet. Contents of physiologically active compounds such as total phenolic compounds, total dietary fiber, $\gamma$-aminobutyric acid (GABA), and total phytic acid in CGBR were significantly higher than those of traditional germinated brown rice (GBR). Lipase inhibitory activity of CGBR was higher than those of GBR and brown rice (BR). High fat diets containing CGBR, GBR, and BR were administered to three groups of male Sprague-Dawley rats for four weeks. All groups showed no significant difference in body weight, total abdominal fat, and plasma lipid levels. However, CGBR group appeared to have lower body weight gain and total abdominal fat level than other groups fed high fat diets containing GBR and BR. Total cholesterol and LDL-cholesterol contents in plasma of CGBR group were also lower than those of other groups. Thus, new germination method of brown rice using chitosan is a useful process, which utilizes plant defense responses to elevate the production of secondary metabolites and anti-obesity effect.

Comparison of Quality of Peeled Lotus Roots Stored in Various Immersion Liquids during Storage (침지액을 달리한 박피 연근의 저장중 품질 비교)

  • This study was carried out to investigate the efficacy of strong acidic electrolyzed acid water (SAEW; Strong acidic electrolyzed water, pH 2.76, ORP 1,128 mV, HClO 105.0 ppm) and low alkaline electrolyzed water (LAEW; Low alkaline electrolyzed water, pH 8.56, ORP 660 mV, HClO 73.8 ppm) as storing liquid for peeled lotus root. During storage at $5^{\circ}C$, it was showed that SAEW and LAEW inhibit growth of microorganisms until at least 5 days of storage. Total phenolic contents, polyphenol oxidase (PPO) activity, and color differences value $({\Delta}E)$ of peeled lotus roots stored in SAEW and LAEW were lower than that of one stored in TW (tap water). The hardness decrement of lotus roots stored in SAEW and LAEW were lower than that of lotus root stored in TW too. Contents of moisture crude protein crude fat, crude ash, crude fiber, and total sugars were gradually decreased during storage. Whereas vitamin C content of lotus root stored in 0.6% acetic acid was most rapidly decreased to 25% as compared with one of initial days of storage that of lotus roots stored in SAEW and LAEW was not decreased significantly as compared with one stored in TW. Sensory characteristic during storage was preferable on lotus root stored in SAEW to the other treatments.

Effective menu retrieval for electronic information system (전자정보 시스템의 효율적 메뉴검색)

  • Hitel 과 같은 전자정보 시스템은 사용자가 원하는 정보를 체계적으로 얻을 수 있도록 하기 위하여 메뉴들을 적당히 계층적으로 구성하여 제공하고 있다. 그러나, 보통 이 메뉴들의 계층이 정확한 분류법에 기초하여 만들어지지 않았을 뿐 아니라 그 양도 엄청나게 방대하여, 이 메뉴 계층을 이용하여 사용자가 원하는 정보를 얻기가 쉽지 않다. 실험적으로 보통 Hitel을 자주 이용하는 사람들도 자신이 주로 이용하는 메뉴들의 구성만 이해하고 있을뿐, 사용하지 않는 부분의 메뉴들의 구성은 잘 알지 못하는 것이 일반적이었다. 따라서 Hitel을 자주 이용하는 사용자도 자신이 이용해 보지 않은 정보를 얻기 쉽지 않으며, 더더욱 초보자에게는 이 메뉴계층을 이용하여 원하는 정보를 얻기가 쉽지 않은 실정이다. 본 연구에서는 정보검색 기술을 이용하여 Hitel과 같은 전자정보 시스템에서 사용자가 쉽게 자신이 원하는 정보를 얻을 수 있는 보조 시스템을 개발하고자 한다. 본 시스템은 사용자가 메뉴계층을 이용하기 전에 간략한 자연어로 입력을 주면, 여기에 적합한 메뉴나 실제 정보를 검색해 낸다. 따라서 사용자는 이 메뉴정보를 이용하여 메뉴계층을 쉽게 따라갈 수 있을 뿐 아니라, 경우에 따라서는 원하는 실제 정보를 검색하기 때문에 메뉴계층을 탐색할 필요가 없다. 본 연구에서는 자연어 입력을 최장 일치 방법으로 의미있는 명사들을 추출하여 불리한 질의어로 만든 후, 명사들 사이의 관계가 표현된 시소러스를 이용하여 이 질의어를 확장시킨다. 다음에 이 질의어들을 메뉴들과 부분/정확부합을 통하여 관련된 메뉴들을 찾아낸 후, 이들의 계층과제를 고려하여 최종 메뉴들을 검색한다. 본 시스템은 현재 C언어로 만들어져 구동중이며, 정확한 실험은 아직 하지 않았지만 높은 검색율을 보이고 있다. industrialized, was improved by introducing pressure in cooling procedure for both carbon and iron thermistors.er>$CHCl_3$>Hexane층 순으로 높은 활성을 나타냈다. 5. 아질산염소거능은 끝순, 들깨잎, 콩나물이 우수하였고 그중 들깨잎이 저해율 72%로 가장 높았으며, 용매분획 중에는 BuOH과 water추출물의 활성이 가장 높았다. 6. Study on the Application of Indigenous Pigmented Rice for Garaedduk Adapted with Mechanically Impacting Technology (기계적 충격기술이 토종 유색미 함유 가래떡 노화지연에 미치는 효과)

  • The present study was performed to expand the application of mechanically impacting technology reported in our previous studies. Several parameters were analyzed to research Korean indigenous pigmented rice. The proximate composition of Korean indigenous rice was the following; 12.0-13.4% moisture, 74.1-77.3% carbohydrate, 6.8-8.7% crude protein, 1.68-3.25% crude lipid, 0.78-1.38% crude ash, with a 6.6-7.07 pH value. The phenolic compound content of red rice was twice that of black rice. Moreover, antioxidative activity was estimated by DPPH radical-scavenging activity (IC50 = 0.15-1.04 mg/mL). Anthocyanin and chlorophyll in each pigmented rice type were detected by spectrophotometer. Garaedduk samples were prepared with non-waxy rice, salt, 24% water, and/or 15% indigenous pigmented rice(red, green and black, w/w of total swelling non-waxy rice) and adapted with or without mechanically impacting technology. Hardness, as a main texture profile parameter of Garaedduk was maintained up to 3 days under storage at $20^{\circ}C$. The results suggest that Korean indigenous pigmented rice can be applied for functional Garaedduk adapted with mechanically impacting technology.

Prediction of Optimal Leaching Conditions for Green Tea (녹차의 음용을 위한 최적 침출조건 예측)

  • Tea was known to have different taste and flavor with leaching temperature and time. This study was designed to optimize leaching condition for green tea bag that has been popular to consumers. Soluble solid, total phenolics, and total flavonoid content increased as leaching temperature and time increased. The ranges of maximum leaching condition for these components were $96.1{\sim}99.0^{\circ}C\;and\;5.7{\sim}6.8\;min$. As leaching temperature decreased and leaching time increased, however, electron donating ability (EDA) increased and showed the highest values at $65.3^{\circ}C$ and 7.2 min. The overall acceptability was maximal at $70.2^{\circ}C$ and 3.1min. Based upon the physicochemical and organoleptic properties, it was predicted that the optimal range of leaching conditions for green tea bag was $73{\sim}83^{\circ}C\;for\;5.3{\sim}6.3\;min$.

A Study of the Nutritional Composition of Aralia continentalis Kitagawa and Aralia continentalis Kitagawa Leaf (땅두릅과 땅두릅잎의 영양성분에 관한 연구)

  • In this study, we assessed the functional characteristics of Aralia continentalis Kitagawa (AcK) and Aralia continentalis Kitagawa leaf (AcK-leaf), including crude saponin contents, antioxidant activity, nutritive elements, dietary fiber, and chlorophyll contents. The results of our analysis of nutritive elements in AcK, the contents of the general components, inorganic components, and vitamins were all relatively high. The contents of inorganic components were also high in the following order: K>Ca>P>Na>Mg. Among vitamins, $\beta$-carotene and vitamin C contents were all high. The crude saponin content was 63.7 mg/g in the AcK and 63.5 mg/g in the AcK leaf, and the antioxidant activity was determined to be relatively high. With regard to the dietary fiber content, the total dietary fiber content was 2.13% (soluble 0.40, insoluble 1.72) in the AcK and 5.98% (soluble 1.06, insoluble 4.89) in the AcK leaf, and the total chlorophyll content was 92.58 mg in the AcK and 147.25 mg/100 g in the AcK-leaf.

Antioxidative Activities by Water-Soluble Extracts of Morus alba and Cudrania tricuspidata (뽕나무(Morus alba)와 꾸지뽕나무(Cudrania tricuspidata)의 수용성 추출물에 의한 항산화 활성)

  • The antioxidative activities of water-soluble extracts from leaves and stem bark of Morus alba and Cudrania tricuspidata were compared in vitro experimental models. Antioxidative activities were measured by inhibition activity against lipid peroxidation of mouse liver microsome, and they were showed in the following order; stem bark of C. tricuspidata(53%)>stem bark of M. alba(43%)>leaves of C. tricuspidata(38%)>leaves of M. alba(43%). In antioxidative activities determined by thiocyanate method and TBA method, the water-soluble extract of stem bark of C. tricuspidata showed the highest antioxidative activity. The water-soluble extracts of leaves were slightly stronger than other extracts in DPPH$({\alpha},{\alpha}'-diphenyl-{\beta}-picrylhydrazyl)$ method. The concentrations of total polyphenolic compound from water-soluble extracts of leaves and stem bark of M. alba and C. tricuspidata were 1.32%, 1.28%, 1.34% and 1.30% respectively. In these results, the water-soluble extract of stem bark from Cudrania tricuspidata showed the highest antioxidative activity.
