Quality characteristics of rice cupcakes prepared from Korean rice varieties (품종이 다른 국내산 쌀을 이용한 쌀컵케이크의 품질 특성)

  • The physicochemical properties of rice flours and quality characteristics of rice cupcake were investigated using the domestic rice varieties Goami, Hanareum, Hanmaeum, Deuraechan, Seolgaeng, and Haiami. Protein and amylose contents were highest in. The final and setback viscosities of Haiami, Hanmaeum, Deuraechan, and Hanareum were higher than those of Goami and Seolgaeng. The specific volume of rice cupcakes was lower with Goami and Haiami than with the others. Seolgaeng cupcake had a dented surface and irregular air pockets, while the Hanareum cupcake was well shaped and had uniform air pockets. The hardness, springiness, and chewiness of Goami and Haiami cupcakes were higher than those of the others, while adhesiveness was observed only in Seolgaeng cupcake. The overall preferences scores of rice cupcakes prepared from Hanareum, Hanmaeum and Deuraechan were higher than for the other cupcakes (p<0.05). These results suggested that amylose content, and final and setback viscosities of rice flour are very important factors in rice cupcakes.

  • The objective of this experiment was to test the effect of EG F on nuclear maturation of pig immature oocytes in vitro. Basic medium used TCM-199 supplemented with 0.2 mM pyruvate, 1 ${\mu}\textrm{g}$/ml estradiol-I7$\beta$ and 25 ${\mu}\textrm{g}$/ml gentamycin, this medium treated with EGF, FSH and FBS. Experiment 1 examined to the effect according to the addition of FSH or EGF (0, 1. 10 and 100 ng EGF/ml) in oocytes maturation. Nuclear maturation rates (M ll %) of 1, 10 and 100 ng EGF/ml (83.0. 8fi.7 and H7.5%) treatments were significantly higher than those of non- and FSH-treated groups (27.3 and 60.3%, p < 0. 001). Experiment 2 examined to the interactive effects of EGF. FSH or FBS during oocytes maturation. Nuclear maturation rates (M ll %) of EGF alone, EGF plus FSH, EGF plus FBS, FSH plus FBS, and EGF plus FSH added FBS treatments (86.7, 90.2, 87.1. 89.6% and 92.6%) were significantly higher than those of non, FSH, and FBS alone treatments (22.3, 52.2 and 42.3%, p < 0.001). Also, cumulus cells expansion of oocytes maturation was examined to total treatments. Normal cumulus cells expansion was shown by FSH plus FBS, EGF or EGF with FBS combination treatments, but cumulus cells of oocyte complexes were still clumped together in EGF-treated groups although they had separated from oocytes. However, EGF showed a positive on nuclear maturation. These results conclude that EGF alone can stimulate nuclear maturation in pig immature oocytes.

  • 지구의 평균기온이 지속적으로 상승함에 따라 지구온난화로 인한 기후변화는 지구에 분포되어 있는 극지방의 빙하들을 녹임으로써 지구의 물 순환시스템을 교란시켜 집중호우와 태풍, 가뭄, 낙뢰 등 예측을 불허하는 극한 기상변화를 발생시키고 있다. 기후변화로 인해 바다의 수온이 상승함에 따라 빙하가 녹거나 바닷물이 팽창하여 해수면이 상승하게 되는 바, 1990년 대비 2000년대에 동해바다에서는 상승률이 0.07cm/yr이던것이 0.20cm/yr로 나타났으며, 서해바다에서는 0.14cm/yr이던 것이 0.18cm/yr로 나타났고, 남해바다에서는 0.32cm/yr이던 것이 0.34cm/yr 로 평균상승률이 1990년대에 비해 증가하는 것으로 관측되었다. 본 연구에서는 기후변화로 인한 자연재해 피해를 최소화하기 위해 소방방재청의 현행 업무를 중심으로 재해를 예방하기 위한 풍수해 업무별 추진해야할 연구과제들을 조사 제시하고자 하였다. 기후변화에 따른 재난분야의 정책과 관련된 연구적인 측면의 분야를 제시하기 위하여 최근에 나타난 자연재난 피해현상에 대한 원인 및 대책을 기초로 하여, 재난관리분야에서 추진하고 있는 업무를 계승 발전시킴으로써 기후변화로 인한 피해를 예방 또는 최소화 할 수 있는 방향으로 제시하고자 하였다. 기후변화에 따른 소방방재청의 풍수해 재난관리 분야 종합계획을 제시하기 위해 본 연구에서는 기후변화 관련 최근 국내 외의 동향을 먼저 살펴보았다. 1977년부터 2006년까지 우리나라 최근 30년간의 재해연보에 제시되어 있는 시설물별 피해액을 조사하여 시설물 중 피해액이 많은 순으로 주요피해 시설물을 파악하였다. 여기에서 주요피해 시설물로는 하천, 도로, 소하천, 수리, 농경지, 사방 등의 순으로 나타났다. 이러한 주요시설물에 대한 피해현황을 파악하기 위하여 대규모 풍수해 피해에 대한 현황, 원인분석 및 대책이 제시되어 있는 각종 피해조사 보고서, 연구보고서 및 전문 학술지 기사들을 수집 분석하였으며, 수집된 자료를 토대로 각각의 재해피해현상에 대하여 시설물의 피해현상, 원인 및 대책을 분류하여 분석하고자 한다. 재난관리 분야 중 우수유출저감시설 관련 제시된 업무방향을 보면 침투 저류를 위한 우수유출저감시설의 개발연구, 침수위험지구의 지정기준 등급별 방재대책 방안연구, 유역별 재해위험 저감능력의 평가기준 개발, 단위구역별 우수유출저감시설의 확보기준 연구, 우수유출저감시설의 국내 표준화 방안 연구, 우수유출저감시설 설치자에 대한 인센티브 도입방안 연구, 피해지역의 매입을 통한 저류지화 방안 연구, 우수유출저감시설 설치효과의 교육 홍보 및 우수유출저감시설의 국제 표준화 기준 제정 추진 등이 필요할 것으로 조사되었다. 여기에서 제시된 재난관리 업무분야별 많은 연구과제들이 향후 연구할 수 있는 재원확보로 이어져 재난관리의 업무발전에 도움이 될 수 있도록 하여야 하겠으며, 주요 결론으로는 다음과 같다. 첫째, 우리나라는 기후변화에 대해서는 기존에 소극적으로 대응하였으나 기후변화대책기획단을 만들어 적극적으로 대처하고 있으므로 기후변화와 관련된 여러분야가 활성화 될 것으로 판단된다. 둘째, 국외의 기후변화 대응사례에서 보면 시설물의 규모를 볼 때 큰 규모의 예산을 투입하는 것으로 판단되는바, 이는 향후의 불확실한 기후변화에 대비하는 선진적인 판단으로 검토되어야 할 것이다. 셋째, 풍수해 관련 주요업무 8가지에 대하여 추진해야할 업무방향 48개를 선정 제시하였다.

Electrochemical Characteristics of Silicon/Carbon Composites with CNT for Anode Material (CNT를 첨가한 Silicon/Carbon 음극소재의 전기화학적 특성)

  • Silicon/Carbon/CNT composites as anode materials for lithium-ion batteries were synthesized to overcome the large volume change during lithium alloying-de alloying process and low electrical conductivity. Silicon/Carbon/CNT composites were prepared by the fabrication processes including the synthesis of SBA-15, magnesiothermic reduction of SBA-15 to obtain Si/MgO by ball milling, carbonization of phenolic resin with CNT and HCl etching. The prepared Silicon/Carbon/CNT composites were analysed by XRD, SEM, BET and EDS. In this study, the electrochemical effect of CNT content to improve the capacity and cycle performance was investigated by charge/discharge, cycle, cyclic voltammetry and impedance tests. The coin cell using Silicon/Carbon/CNT composite (Si:CNT=93:7 in weight) in the electrolyte of $LiPF_6$ dissolved in organic solvents (EC:DMC:EMC=1:1:1 vol%) has better capacity (1718 mAh/g) than those of other composition coin cells. The cycle performance of coin cell was improved as CNT content was increased. It is found that the coin cell (Si:CNT=89:11 in weight) has best capacity retension (83%) after 2nd cycle.

  • A structured kinetic model is proposed to describe cell growth and secondary metabolite, flavone glycosides, synthesis in batch suspension culture of Scutellaria baicalensis G. The model has been developed by representing the physiological state of cell described as the activity and viability which can be estimated based on the culture fluorescence. In the model, three type of cells are considered; active-viable, nonactive-viable and dead cells. Viable cell weight could be determined based on the relative fluorescence intensity. The flavone glycosides could be produced by both active-viable and non-active viable cells with a different production rate. And the model includes the cell expansion due to glucose concentration and death phase which accounts for the release of intracellular secondary metabolite into medium. Dependent variables include substrate concentration(glucose), cell mass(dry cell weight and fresh cell weight), product concentration(flavone glycosides), activity and viability. Satisfactory agreement between the model and experimental data is obtained from shake flask culture of Scutellaria baicalensis G. The proposed model can predict the cell growth and flavone glycosides synthesis as well as intermediate materials.

Effect of Phosphate on Rheological Properties of Rice Starch Suspension (인산염이 쌀전분 현탁액의 리올로지에 미치는 영향)

  • Rheological properties of 5% starch suspension were investigated using a capillarity tube viscometer. Nonglutinous and glutinous rice starch suspensions at $30-60^{\circ}C$, and $30-55^{\circ}C$, respectively, had no yield stress and showed dilatant flow behavior in the absence or presence of phosphate. However, flow behavior of nonglutinuous starches at $65^{\circ}C$ and glutinous starch at $60^{\circ}C$ was pseudopalastic. Flow activation energies for rice starch suspensions were 0.88-1.45 kcal/mole at $30-50^{\circ}C$ and about 45-73 kcal/mole $60-65^{\circ}C$. Flow activation energy in the presence of phosphate was 90-182 kcal/mole at $60-65^{\circ}C$. Akibare starch had the highest activation energy, and glutinous starch the lowest.

A Study for the Safety on the Flame Exposure of the Propane Cylinder (소형 프로판 용기의 화염 노출에 대한 안전성 연구)

  • To evaluate the safety of propane cylinder, the flame test was performed by the flame exposure scenario of propane cylinder. The cylinder which was exposed in a flame was rapidly occurred to increase the internal pressure by liquid expansion, if so it cause of physical explosion. Therefore, the cylinder which was applied with thermal pressure relief device sholud be not bursted and the propane should be discharged to outside safely. The flame average temperature that was around of cylinder is $600^{\circ}C$, and then it increased $700^{\circ}C$ by discharged propane. The result of flame test, the cylinder was deformed, but it was not bursted. The regulations of flame exposure test for propane cylinder were not restricted, this paper can be applied to restrict the flame test if the cylinder is possible to expose the flame. Also, the results is expected as reference for estimation of the pressure cylinder performance.

Cryopreservation of Bovine IVM/IVF/IVC Hatched Blastocysts (체외생산된 소 완전탈출 배반포기배의 동결보존)

  • This study was to test whether the viability of bovine hatched blastocysts (HBs) can be maintained after vitrification and thawing. The HBs were produced in vitro at Day 9 and Day 10 after IVF, and they were classified to small (S-HBs; ø$\leq$300 ${\mu}{\textrm}{m}$) and large(L-HBs; ø>300 ${\mu}{\textrm}{m}$) on the basis of embryo diameter using eyepiece micrometer. As freezing solution, we used EFS35 which consisted of 35% ethylene glycol (EG), 18% ficoll, 0.3 M sucrose and 10% FBS added in mDPBS. Vitrification was taken by two-step method, the HBs were equilibrated in 10% EG for 5 minutes and then shortly exposed in EFS35 and plunged into L$N_2$for 30~45 sec. After thawing, the survival rates were assessed by the re-expansion of the blastocoel during 2 h and 16 h of culture. The results obtained in these experiments were summarized as follows; 1) When the blastocysts(40.8%) recovered at Day 8 after IVF were further cultured for 24 h(Day 9 after IVF) and 48 h(Day 10 after IVF), the rates of HBs were 20.5% and 6.7%, respectively. Also, the total cell number of HBs on Day 9 was significantly higher than that of HBs on Day 10 (p<0.01). 2) When the effects of freezing solution to the survival of Day 9 L-HBs were examined, the rate of vitrified group (75.7%) was significantly lower than 100% of control and exposed group(p<0.05). 3) When the survival rates of vitrified HBs according to size and developmental age were examined, the data of L-HBs (75.5%) and S-HBs(63.6%) on Day 9 were slightly higher than those of L-HBs(64.3%) and S-HBs(60.7%) on Day 10. 4) Also, when the in vitro survival of Day 9 HBs was evaluated under different culture condition after thawing, the result in culture medium only (79.3%) was significantly higher than 43.2% in co-culture group (p<0.05). These results demonstrated that bovine HBs can be successfully cryopreserved by two-step vitrification method using EFS35.

  • This study was to examine whether Hanwoo IVM/IVF/IVC blastocyst can be successfully survived in vitro/in vivo after vitrification and one-step dilution. For vitrification, blastocysts were serially exposed in glycerol (G) and ethylene glycol(EG) mixtures[10% (v Iv) G for 5 min., 10% G plus 20% EG (v/v) for 5 min., and 25% G plus 25% EG (v/v) for 30 sec.] which is diluted in 10% FBS added D-PBS. And then they were loaded in the straw, placed in cold nitrogen vapor for 3 min. and plunged into L$N_2$(-196$^{\circ}C$). One-step dilution within the straw was done in $25^{\circ}C$ and 36$^{\circ}C$ water for about 5 min. and 3 min., respectively. Recovered embryos after one-step dilution were cultured in cumulus cell mono-layered drop for 48 h or were transferred into recipient cows. When the embryo survival in vitro was assessed to re-expanded and hatched rates at 24 hand 48 h after one-step dilution, the results of vitrified group (85.4, 43.8%) was high, although these results were significantly lower than normal development (100.0, 63.3%) of control group, respectively (P<0.001, P<0.05). When in vitro survival of vitrified groups according to developmental stage was compared, the results of fast developed embryos (expanded blastocyst and early hatching blastocyst stage) were significantly higher than those of delayed developed one (early blastocyst stage) after one-step dilution (early hatching: 88.0, 48.0%: expanded: 81.1, 45.3%; early: 66.7, 14.3%) (P<0.05). Also, in case of in vitro survival of vitrified groups according to embryo age (day 7, 8 and 9), when embryo age was younger, in vitro survival was significantly higher (day 7: 67.3, 34.5%; day 8: 76.9, 40.7%; day 9: 60.9, 23.9%)(P<0.05). Finally, when in vivo development potential of vitrified and one-step diluted Hanwoo blastocysts was examined, 4 of 8 recipient (50%) cows became confirmed pregnant. These results demonstrated that our vitrification and one-step dilution technique can be applied easily and effectively on field trial without the equipment and embryological skills required for conservative dilution and transfer.

Quality Changes of Commercial Kimchi Products by Different Packaging Methods (소포장 김치의 포장방법별 품질특성 변화)

  • The effect of various packaging methods on kimchi quality was investigated in order to develop the packaging techniques for preventing commercial kimchi products from inflation and explosion, due to fermentative gas evolved during storage and distribution. Kimchi was packaged in different methods; 1) atmospheric packaging(AP), 2) check valve packaging(CV), 3) double packaging(DP), and 4) vacuum packaging(VP). The quality of kimchi during storage at $10^{\circ}C$ was evaluated in terms of gas composition, free volume, pH, titratable acidity color index and sensory properties. The gas composition inside packages showed different curves according to the packaging methods. Due to fermentative gas accumulation in both AP and CV, $CO_2$ concentration increased by 2 stepwise pattern, while $O_2$, concentration decreased exponentially. In DP, $O_2$ concentration remained constant, but $CO_2$ concentration increased by 2 stepwise pattern and then decreased. In contrast, VP produced low $O_2$ and high $CO_2$ concentrations only at the end of storage. The free volume in both AP and CV showed typical sigmoidal curves similar to $CO_2$ concentration changes. It remained constant in DP, but started to increase at the late stage of storage in VP. There was no significant effect of packaging methods on pH changes of kimchi. In titratable acidity, DP maintained relatively higher than others. Regarding to the color change of crushed kimchi juice in all packages, L and b values decreased exponentially but a value remained constant during storage. Color index(L b/a) of crushed kimchi juice decreased exponentially and remained constant at the end of storage. The growth of lactic acid bacteria was VP, CV, AP, DP in increasing order. In sensory test, the sourness scores of DP were fairly higher than those of others, but the texture was not significantly affected by the packaging methods. The preference for kimchi showed VP>AP, CV>DP in order of score. In this study, it could be proposed to employ DP and VP method as the effective packaging techniques for preventing commercial kimchi products from inflation.
