Effect of Milk Peptide on Bone Metabolism (우유단백질 유래 펩타이드가 골 대사에 미치는 영향)

  • Bone undergoes continuous remodeling throughout the life and bone health is governed by the balance of bone resorbing osteoclast and bone forming osteoblast. Bone resorption is reflected in tartrate resistant acid phosphatase, pyridinium cross link and collagen telopeptide, whereas bone formation activity can be expressed as bone specific alkaline phosphatase, osteocalcin and procollagen I extension peptide. Milk basic protein and lactoferrin have been reported as active proteins to modulating bone metabolism. In addition to these proteins, some bioactive milk peptides released during lactic fermentation may provide beneficial effect on bone metabolism. The effects of fermented products of Lactobacillus casei ATCC 393 on bone metabolism were investigated using a variety of biochemical markers in osteoblastic MC3T3-E1 cells and ovariectomized rats. Based on the results, the fermented products of Lactobacillus casei ATCC 393 played an functional role in bone metabolism by suppressing bone resorption and by increasing bone formation.

  • Tissue engineering has been enhanced by advance in biomaterial nature, surface structure and design. In this paper, I report specifically vertically aligned titania ($TiO_2$) nanotube surface structuring for optimization of titanium implants utilizing nanotechnology. The formation, mechanism, characteristics of titania nanotubes are explained and emerging critical role in tissue engineering and regenerative medicine is reviewed. The main focus of this paper is on the unique 3 dimensional tubular shaped nanostructure of titania and its effects on creating epochal impacts on cell behavior. Particularly, I discuss how different cells cultured on titania nanotube are adhered, proliferated, differentiated and showed phenotypic functionality compared to those cultured on flat titanium. As a matter of fact, the presence of titania nanotube surface structuring on titanium for dental applications had an important effect improving the proliferation and mineralization of osteoblasts in vitro, and enhancing the bone bonding strength with rabbit tibia over conventional titanium implants in vivo. The nano-features of titania nanotubular structure are expected to be advantageous in regulating many positive cell and tissue responses for various tissue engineering and regenerative medicine applications.

Effects of Samkieumgamibang Extract on Osteoclast Differentiation and Osteoblast Function (삼기음가미방(三氣飮加味方)이 파골세포의 분화 및 조골세포의 활성에 미치는 영향)

  • Objectives: This study was performed to evaluate the effect of Samkieumgamibang (SKG) on osteoporosis. Methods: The osteoclastogenesis and gene expression were determined in RANKL-stimulated RAW 264.7 cell. And, osteoblastogenesis was also determined in rat calvarial cell. Results: SKG decreased the number of TRAP positive cell in osteoclast. It also decreased the expression of Cathepsin K, MMP-9, TRAP, c-fos, NAFTc1 and JNK1 in osteoclast. SKG increased the expression of iNOS in RANKL-stimulated in osteoclast. Otherwise, SKG inhibited TRAP activity in osteoclast. SKG increased cell proliferation, ALP activity, bone martix protein, collagen and nodule in osteoblast. Conclusions: It is concluded that SKG might decrease the bone resorption resulted from decrease of osteoclast differentiation and it's related gene expression. And, SKG might increase the bone formation resulted from increase of osteoblast function.

Effect of water extract and distillate from the mixture of black goat meat and medicinal herb on osteoblast proliferation and osteoclast formation (흑염소와 약용식물 복합 증탕추출액 및 증류액이 조골세포 증식과 파골세포 형성에 미치는 영향)

  • Purpose: The effects of water extract and distillate from the mixture of black goat meat and medicinal herb on MG-63 osteoblast proliferation and mouse bone marrow derived osteoclast formation were investigated. Methods: Proximate composition, volatile basic nitrogen (VBN), mineral content, free amino acid composition and free fatty acid composition in black goat meat were determined. Water extract and distillate were prepared with three groups; goat meat only (BG-E, BG-D), six herbs added group (BG-E6, BG-D6), and eight herbs added group (BG-E8, BG-D8). Osteoblast proliferation, mineralization and calcium uptake activity of MG-63 cells were measured and tartrate resistant acid phosphatase activity of osteoclasts was analyzed. Results: Black goat meat had remarkably low fat and high level of calcium. Glutamic acid was the most abundant amino acid. Herbs added extract groups (BG-E6 and BG-E8) showed increased MG-63 cell proliferation in a concentration dependent manner, while all the distillates did not show the effect. All extracts and distillates showed significantly increased osteoblast mineralization depending on the concentration. In particular, herb added extract, BG-E6, increased 170.3% of control and the distillate of BG-D and BG-D6 increased up to 168.5% and 159.8%, respectively. Calcium uptake activities of all water extracts showed remarkable increase of BG-E6 and BG-E8 up to 615.5% and 628.1% of control, respectively. Ditillates had no effect except BG-D6. All water extracts significantly reduced the activity of tartrate-resistant acid phosphatase (TRAP) in osteoclasts derived from mouse bone marrow. Conclusion: Combination of black goat meat and medicinal herb increased the MG-63 cell proliferation and effectively inhibited osteoclast differentiation in both water extracts and distillate of them, which implies that they could be used as potent functional food materials for bone health.

  • An epulis was occurred on gingiva of 11-year old female dog, Yorkshire terrier. Our case had feature of ossifying epulis but there were a few multi-nucleated giant cells (MGCs). MGCs had osteoclast-like appearance and giant cell epulis usually appears at the site of tooth extraction. Therefore, we suggest that appearance of MGCs in our case may be due to phagocytosis pre-formed osteoid/bone or our case may be mixed epulides of ossifying and giant cell epulis by mixed stimulation of chronic gingivitis and trauma and in flammation by tooth extraction. Thus, MGCs have possibility enough to appear in ossifying epulis, but ossifying epulis accompanying MGCs has not been reported. Therefore, our case may deserves an attention as an unique case and will be helpful to study pathogenesis of giant cell containing lesion of the jaw.

The Stimulatory Effect of Dexamethasone on RANKL-induced Osteoclastogenesis (Dexamethasone에 의한 RANKL 유도성 파골세포 분화 촉진 효과)

  • We explored the effects of dexamethasone on osteoclast precursors using BMMs. Dexamethasone inhibited the proliferation of BMMs. Furthermore, it stimulated the osteoclast formation via NFATc1 activation in the presence of RANKL. Since dexamethasone targeted the early stage of osteoclast formation, we investigated its effect on mRNA expression of GR and $IFN-{\beta}$. Whereas dexamethasone had no effects on GR expression in the presence of RANKL, it reduced the expression of $IFN-{\beta}$, suggesting that dexamethasone increased RANKL-induced osteoclast formation by modulating $IFN-{\beta}$.

  • To determine the regulatory roles of PDE4 inhibitor on LPS-induced osteoclastogenesis, we investigated the effect of a PDE4 inhibitor on osteoclast formation in the presence of LPS. A specific PDE4 inhibitor, rolipram, increased LPS-induced osteoclast formation in cocultures. To verify that whether rolipram acts indirectly on osteoblasts, we investigated the TRANCE and COX-2 mRNA expression levels in osteoblasts. Treatment of rolipram increased the expression of TRANCE and COX-2 mRNA in osteoblasts stimulated by LPS. On the contrary, rolipram did not augment the number of osteoclasts differentiated from bone marrow cells by LPS. In conclusion, the stimulation of LPS-induced osteoclast formation by the PDE4 inhibitor are attributable to its indirect effect on osteoblasts, not to their direct effect on bone marrow-derived osteoclast precursors.

  • We investigated the role of L-type $Ca^{2+}$ channel in receptor activator of nuclear factor-kappaB ligand (RANKL)-induced osteoclast formation. BayK 8644, a L-type $Ca^{2+}$ channel agonist, was shown to increase the RANKLinduced osteoclastogenesis and actin ring formation in mouse bone marrow-dereived macrophage (BMM) culture system. BayK 8644 stimulated RANKL-induced extracellular signal-regulated kinase (ERK) and p38 MAP kinase (MAPK) activation, which leads to increased nuclear factor of activated T cells (NFAT)c1 expression. Taken together, these data indicate that L-type $Ca^{2+}$ channel regulates osteoclast formation possibly through ERK- and p38-mediated NFATc1 expression.

Effect of Myricetin in Osteoclast Differentiation and Bone Resorption (파골세포 분화와 골 흡수에 myricetin의 효과)

  • Osteoclasts are bone-resorbing giant cells that differentiate from hematopoietic cells of the monocyte/macrophages. Excessive osteoclast differentiation leads to gradual loss of bone mass causing fracture of the skeleton. The aim of this study was to develop a drug candidates for the treatment of osteoporosis. RANKL-induced osteoclast differentiation was dose-dependently inhibited by myricetin. Myricetin inhibited the expression of c-Fos, NFATc1, and TRAP in BMMs treated with RANKL. Myricetin disrupted the structure of actin ring and suppressed osteoclastic bone resorption. Also, myricetin induced apoptosis in mature osteoclasts. Myricetin inhibited the phosphorylation of ERK in mature osteoclasts treated with M-CSF. The activation of caspase-9 and caspase-3 was increased by myricetin treatment. Our results suggest that myricetin may be an effective agent to prevent bone diseases such as osteoporosis.
