Luminol-H2O2-Cu(II) 시스템을 이용한 방향족 아미노산의 화학발광법적 정량 (Determination of aromatic amino acids by chemiluminometric assay with Luminol-H2O2-Cu(II) system)

  • Luminol-$H_2O_2$-Cu(II) 시스템을 이용한 화학발광법을 사용하여 방향족 아미노산인 트립토판, 타이로신 및 페닐알라닌의 정량분석을 하였다. 세 종류의 방향족 아미노산(트립토판, 타이로신, 페닐알라닌)을 luminol-$H_2O_2$-Cu(II) 시스템에 첨가하였을 때 아미노산이 존재하지 않을 때보다 화학발광세기가 더욱 증가하는 현상을 관찰하였으며, 이러한 현상을 이용하여 각 방향족 아미노산을 정량분석 하였다. 방향족 아미노산의 최적분석 조건을 조사하기 위하여, Cu(II) 이온 촉매에 의한 루미놀과 과산화수소의 화학발광반응에 미치는 아미노산의 반응속도론적 영향을 조사하였고 과산화수소와 Cu(II) 이온의 농도 그리고 pH와 완충용액의 영향을 조사하였다. 루미놀 화학발광 시스템의 최적 분석조건 하에서 수용액 중의 방향족 아미노산 정량분석을 위해 얻은 검정곡선에서 직선성이 성립하는 농도 범위는 각각 트립토판은 $1.0{\times}10^{-6}{\sim}2.0{\times}10^{-5}\;M$, 타이로신은 $1.0{\times}10^{-6}{\sim}2.0{\times}10^{-5}\;M$ 그리고 페닐알라닌은 $2.0{\times}10^{-6}{\sim}2.0{\times}10^{-5}\;M$이었으며, 이 구간에서 각 아미노산에 대한 상대표준편차(n = 4)는 순차적으로 3.21%, 2.64% 그리고 2.48%이었다. 그리고 각 아미노산의 검출한계($3{\sigma}/s$)는 트립토판 $6.8{\times}10^{-7}\;M$, 타이로신 $5.7{\times}10^{-7}\;M$, 페닐알라닌 $9.6{\times}10^{-7}\;M$이었다.

트립토판 열분해물 유발 변이원성에 대한 현미 및 백미의 억제 효과 비교 (A Comparison of Inhibitory Effects in Brown and White Rice ( Oryza sativa L. ) against Mutagenicity Induced by Tryptophan Pyrolysates)

  • The inhibitory effect of rice(Oryza sartiva L., illpumbyeo) against mutagenicity induced by tryptophan pyrolysates were investigated using Salmonella typhimurium reversion assay. Both methanol extracts of obtained from brown and white rice were found to possess strong activites of inhibiting the mutagenicities of 3-amino-1,4-dimethyl-5H-pyriod[4,3-b]indol(Trp-P-1) and 3-amino-1-methyl-5H-pyrido[4,3-b]indol(Trp-P-2) on Salmonella typhimurium reversion assay. As the concentration of methanol extract increased, inhibitory effect on mutagenicity increased but reached at steady state as inhibition rate of 90% when the concentration was above 10mg/plate. There was no significant difference(p>0.05) in inhibitory effect of methanol extracts between brown and white rice against tryptophan pyrolysates.

트립토판 보강식이를 섭취한 마우스에서 serotonin 대사와 morphine 진통기작 관련성에 대한 연구(II) (A Study on the Serotonin Metabolism and the Morphine-related Analgesic Mechanism in Mice Fed Tryptophan Supplemented Deit (II))

  • In this study we fed control diet and tryptophan supplemented diets containing 0.35% tryptophan to ICR mice for 2 weeks. The concentrations of serotonin and 5-HIAA were changed by injection of the serotonin synthesis inhibitor, p-CPA and the serotonin precursor, serotoninP and the change of brain serotonin concentration negatively correlated with that of pain sensitivity, and p-CPA and serotoninP also changed the analgesic effect of morphine. The injection of naloxone, the opiate antagonist, resulted in an increase in the writhing frequency, but its antagonistic effect was not significant. The concentration of 5-HIAA elevated in mice brain at least 3hr after administration of morphine hydroxide indicates that the changes in brain serotonin metabolism may be associated with the acute effects of morphine analgesia. In short, these results not only suggest that tryptophan supplemented diet suppress pain sensitivity in mice, but also indicate that at least in part analgesic mechanism of serotonin may be associated with morphine analgesia.

Aspergillus nidulans내에서 Aspergillus awamori의 Glucoamylase 유전자 발현 (Expression of Aspergillus awamori Glucoamylase Gene in Asperillus nidulans)

  • A awamori 의 glucoamylase 유전자와 A. nidolans 의 trpC maker 유전자를 가진 A. nidulans 의 expression vector 를 만들었다. 이재조합 plasmid 를 트립토판 영양요구주의인 A. nidulans B17 에 형질전환시켰다. Southern blotting 으로 vector DNA 가 A. nidulans 의 chromosomal DNA 에 integration 된 것을 확인하였다. Northern blotting 의 결과, glucoamylase 유전자는 induction 조건에서 mRNA 를 합성하였다. glucoamylase 의 역가가 형질전환체에서 증가하였으며, nondenaturing polyacrylamide gel 에서 glucoamylase 의 활성이 나타났다.

  • 12종의 암 조직을 분석한 결과 지용성 아미노산인 트립토판이 대조군에 비해서 8배나 많았고 황과 인 및 철은 $80{\sim}1,400$배나 많았으며 구리와 아연은 아주 적게 들어 있었다. 이들 영양성분들이 암 조직 단백질의 거대화와 비정상적인 조직증식에 관여할 가능성이 있을 것으로 추정된다.

트립토판이 스트레스 받은 쥐 뇌의 세로토닌과 오피에이트 수용체 결합에 미치는 영향 (Effect of Tryptophan on Serotonin and Opiate Receptor Binding in Stressed Rats)

  • Brain serotonin and its utilization was investigated on stressed rats after feeding high tryptophan diet for a month. High tryptophan fed rats displayed significantly higher level of serum tryptophan, brain tryptophan, serotonin (5-hydroxytryptamine; 5-HT) and 5-hydroxyindoleacetic acid (5-HIAA) than the control diet fed rats. When rats were treated with 3 hour immobilization (IMMB) stress, serotonin turnover was slightly increased, but not statistically significant, in control diet group rats. However in high tryptophan diet rats, 3 hr IMMB stress resulted in statistically significantly (p<0.05) decreased the serum tryptophan, brain tryptophan and 5-HT level. The concentration of 5-HIAA was significantly increased indicating accelerated utilization of the brain 5-HT of the high trp. fed rat. The utilization pattern of the serotonin was found to be similar among young and adult rats. Rats on a tryptophan enriched diet displayed higher coping ability to the stress as they exhibited smaller increment of corticosterone level. A possble involvement of opioid system was suggested in serotonin utilization by measuring total $^{3}$[H]-naloxone binding in brain.

유전자 조작한 Klebsiella pneumoniae로부터 L-트립토판 생산을 위한 발효배지 조건 (Media Optimization for L-tryptophan Production by Genetically Engineered Klebsiella pneumoniae)

  • The optimum medium composition for the production of L-tryptophan with Klebsiella pnuemoniae pheA tyrA trpE trpR/pSC 101-trp$^{+}$ and the effect of precusors in the optimum medium were studied. The specific growth rate in the optimum medium was almost the same as that in the basal medium, the former showing 1.01 and the latter 1.07 hr $$^{-1}$, but the produced tryptophan was increased 45% in the optimum medium. The maximum amount of produced tryptophan was 159 mg/l within 14 hours. Tryptophan production was ceased by casamino acid addition over 4 g/l in medium, but cell maSS increased with its addition. Indole and anthranilate as precusors had toxic effect on growth and tryptophan production at experimented concentration range (over 20 mg/l), but L-serine had good effect on tryptophan production, resulting in 175 mg/l tryptophan within 14 hours.

  • The effect of cations on the formation of protein aggregates was examined by in vitro refolding of mutant tryptophan synthase $\alpha$-subunit in which Pro 24 was replaced by Leu. $NH^{4+},; K{^+}; and; Na^{+}$ and no effect, but $Mg^{2+}; and; Ca^{2+}$stimulated the formation of protein aggregates in dose-dependent manner. It is suggested that $Mg^{2+} and Ca^{2+}$ may be implicated in the formation of protein aggregates in vivo.

  • Objective : Photodynamic therapy (PDT) using topical aminolevulinic acid (ALA) has increasingly been used for the treatment of acne vulgaris and several studies have shown its clinical efficacy. However, ALA-PDT needs a relatively long incubation period and is frequently associated with adverse effects. Triptophan and Riboflavin has been introduced as a new photosensitizer for the treatment of acne recently. Method : 11 patients with acne vulgaris were received PDT with tryptophan and riboflavin four to six times over two to three months. The photos of each patient were taken before and after treatment. Result and conclusion : PDT with tryptophan and riboflavin may be an effective and safe treatment for the Acne vulgaris patients.
