Seasonal Variation of Heterotrophic Bacteria of the Marine Ranching Ground of Tongyeong Coastal Water, Korea (통영 바다목장 해역의 종속영양세균의 계절적 변화)

  • Surface and bottom sea water samples were harvested from the 5 stations in the marine ranching ground of Tongyeong coastal water from year 2000 to 2002. Cell number of heterotrophic bacteria was determined by using the plate counting method to explore the variation of the cell population of heterotrophic bacteria. Sea water samples collected in summer (in July and August) contained much larger number of heterotrophic bacteria than those harvested in spring, autumn and winter. Heterotrophic bacteria were usually more abundant in surface sea than in bottom sea water. However the reverse was true for sea water collected in December 2001 and February 2002 due to suspended solids accumulating more abundantly in seabed area because of the slower convective current of the sea water in winter. Number of heterotrophic bacteria did not have a strong relationship with frequency of typhoon indicating that the path and powerfulness of the typhoon, localized torrential downpour and temperature variation accompanying the typhoon should be considered all together at the same time as well as the frequency of typhoon to explain clearly the variation of cell number of heterotrophic bacteria. The dominant species isolated from the marine ranching ground of Tongyeong were identified to be Pseudomonas fluorescens, Pseudomonas stutzeri, Acinetobacter lwoffii and Sphingomonas paucimobilis.

Marine Environmental Characteristics in the Coastal Area Surrounding Tongyeong Cage-Fish Farms (통영 가두리 양식장 인근 해역에서의 해양환경 특성)

  • To assess environmental characteristics of the aquaculture area in Tongyeong, pH, dissolved oxygen (DO), dissolved inorganic nitrogen (DIN), dissolved inorganic phosphorous (DIP) and chemical oxygen demand (COD), and acid volatile sulfur (AVS) were measured in seawater and sediment samples collected from 10 locations of Tongyeong coastal area from July to December in 2013. The quality of the seawater may be affected by seasonal variation rather than the distance from fish farm. However, sediment was contrary to seawater; the distance from fish farm may be a main factor to affect COD and AVS in sediment than season and other parameters. It is expected that contaminated organic sediments of fishery located in semi-closed bay are rapidly dispersed into surrounding waters due to fast current.

A Study on Furniture Making based on Hojangjul (호장줄을 이용한 가구 제작에 관한 연구)

  • This paper presents research on furniture making based on Hojangjul, which is an engraving technique that highlights the wooden patterns of traditional furniture. Somokbang, one of the 12 workshops in Tongyeong, has used Hojangjul as an engraving technique since Imjinwaeran. It was inherited by the late Cheon Sang-won, who was No. 59 of National Important Intangible Cultural Properties, and developed into Samhojang, Ohojang, Seongtoinoimun, Hotguinoimun, and Gyeopguinoimun. It was then spread to Masan, Ulsan, and Gwangyang via marine routes and further into the inland areas via land routes, developing in various patterns and forms. The present study investigated and examined the making techniques and characteristics of Hojangjul engraved in joinery furniture in Tongyeong, Masan, and Ulsan in the Gyeongsang region and Gwangyang and Dongbok in the Jeolla region, thus shedding new light on the traditional techniques that have been gradually disappearing and promoting the transmission, inheritance, and development of the traditional techniques.

Investigation on the Cause of Bad Natural Seed Collection of the Pacific Oyster, Crassostrea gigas: Relationships between the Conditions of Mother Shell and the Viability of the Released Eggs and Larvae Based on the Pathological and Embryological Survey (참굴 채묘 부진 원인 구명에 관한 연구 -병리 발생학적 조사를 통한 참굴 모패의 건강도와 난 및 유생의 생존율과의 상관관계 구명 -)

  • Infection rates of oyster ovarian parasite, Marteiliodes chungmuensis and productivity of the oyster shellstock infected with the parasite were investigated at the main seed collection areas in the southern coast of Korea where the extreme bad seed collection of oyster occurred in 1992 and 1993 to evaluate the cause of the bad seed collection. Additionally, the bacterial flora of the sea water and oyster lana were examined to identify the shellfish larva pathogenic bacteria like Vibrio sp. and Pseueomonas sp. In August 1992 to September 1993, infection rate of oyster ovarian parasite, M. chungmuensis at Tongyong, Kyongsangnam province, and Yosu, Chollanam province where the bad seed collection occurred, were $11.8\~100\%$ and $14.3\~100\%$, respectively. But the parasite was not detected in the shellstock collected at Daechon, Chungchongnam province. While a virus-like particle was identified in the cytoplasm of the egg infected by the parasite. The parasite infected egg was not able to fertilize completely. Uninfected egg in the gonad contaminated by the parasite could be able to fertilize but showed an abnormal development till D-shaped larva and then, died of necrosis after D-shaped lana. And some lana developed from low lipid content egg could not develop to the spat and died after the early umbo stage. The predominant bacteria in the oyster lana collected at bad seed collection areas were Pseudomonas sp. and Pseudomonas like bacteria and the occupancy rates were $53.3\~87.1\%$.

Sanitary Characteristics of Seawater and Sediments in Tongyeong Harbor (통영항의 해수 및 저질의 위생학적 특성)

  • The bacteriological and physiochemical analysis of sea water and sediments in Tongyeong harbor was conducted to evaluate sanitary conditions. The samples were collected at 8 stations established once a month from June, 2008 to May, 2009. During the study period, the range of temperature was from 6.7 to $25.2^{\circ}C$, transparency ranged from 1.2 to 2.6 m, chemical oxygen demand ranged from 1.90 to 2.92 mg/L, dissolved oxygen ranged from 6.2 to 10.5 mg/L, dissolved nitrogen ranged from 0.052 to 0.098 mg/L, phosphate ranged from 0.044 to 0.065 mg/L, respectively. Seafood, if eaten raw, carries the risk of food poisoning. Seafood poisoning is often cause by pathogenic microorganism originating from fecal contamination, such as Salmonella sp., Shigella sp. and norovirus. Fecal coliforms are an important indicator of fecal contamination. Therefore, data on fecal coliform are very important for evaluating the safety of fisheries in coastal areas. So, we investigated the sanitary indicate bacteria. The coliform group and fecal coliform MPN's of sea water in Tongyeong harbor were ranged from < 1.8~22,000/100 mL (GM 164.9 MPN/100 mL) and < 1.8~7,900 MPN/100 mL (GM 33.7 MPN/100 mL), respectively. Total coliform were detected 97.0% in 96 of samples and 68.9% of total coliforms were fecal coliforms. These results similar to another seawater detection ratio of total coloforms and fecal coliforms. The Vibrios was isolated and identified with VITEK system. Four hundred eighty strains that were obtained from sea water samples in Tongyeong harbor Detection ratio Vibrio alginolyticus, 34.2%, Vibrio parahaemolyticus, 13.8%, Vibrio vulnificus 10.0%, and V. mimicus 12.5% respectively. Vibrio cholerae O1, was not detected. During the study period, the ranges of water content, ignition loss, COD, and acid volatile sulfates in sediments in Tongyeoung harbor were 41.0~57.4%, 7.8~10.5%, 6.51~9.30 mg/g, 0.04~0.09 mg/g, respectively. Heavy metals in sediment of Tongyeoung harbor were Cd, $0.10{\pm}0.05$; Cu, $4.79{\pm}8.20$; As, $1.95{\pm}0.17$; Hg, $0.10{\pm}0.07$; $Cr^{6+}$, $0.34{\pm}0.12$; Zn, $125.33{\pm}16.40$; Ni, $16.43{\pm}1.93$ mg/kg.

Bacteriological and Physiochemical Water Quality of Seawater in Tongyeong Harbor, Korea (통영항 해수의 세균학적 및 이화학적 수질)

  • The bacteriological and physiochemical analysis of seawater in Tongyeong harbor was conducted to evaluate sanitary conditions, The samples were collected at 6 stations established once a month from January to December, 2000. During the study period, the ranges of temperature, transparency, chemical oxygen demand, dissolved oxygen, dissolved nitrogen, phosphate and chlorophyll-a were $6.8\sim25.2^{\circ}C,\;1.0\sim2.5\;m,\;1.79\sim2.41\;mg/L,\;5.7\sim10.1\;mg/L,\;6.59\sim10.53{\mu}g-at/L,\;0.56\sim1.01{\mu}g-at/L\;and\;1.21\sim9.54\;mg/m^3$, respectively, The viable cell counts of seawater in Tongyeong harbor ranged from $3.0\times10^4CFU/mL\;to\;6.9\times10^6CFU/mL$. The coliform group and fecal coliform MPN's of the samples were ranged $23\~4,600\;MPN/100\;mL$ (means 540 MPN/100 mL) and $11\~1,600\;MPN/100\;mL$ (means 210 MPN/100 mL), respectively, The coliform group was classified with IMViC reactions and pathogenic vibrios were analyzed. Two hundred eighteen strains that were obtained from seawater samples in Tongyeong harbor represented Escherichia coli group, $66.1\%$; Citrobacter freundii group, $11.0\%$; Enterobacter aerogenes, $9.6\%$; and unknown, $13.3\%$, respectively. During the study period, infectious bacteria such as Vibrio cholerae O1, Salmonella sp. and Shigella sp. were not detected from the samples, but detection ratios of V. parahaemolyticus, V cholerae non-O1 and V. vulnificus were $10.0\sim30.1\%$.

SNP-based Genetic Diversity and Relationships Analysis of the Korean Native Black Goat and Crossbred Goat (SNP 정보를 활용한 재래흑염소와 교잡종 염소의 유전적 다양성 및 유연관계 분석)

  • This study was conducted to investigate the genetic diversity and genetic taxonomic relationships between Korean native black goat (KNBG) populations and crossbred goats. The 45,658 common single nucleotide polymorphisms present in the KNBG strain and crossbred goat were used for the analysis. The expected and observed heterozygosity (which can be indicators of genetic diversity) were in the order of crossbred, Gyeongsang National University, Jangsu, then the Tongyeong strains. The variance component represents the degree of genetic diversity between groups. The highest variance (19.98 %) was between the Dangjin and Gyeongsang National University strains. The lowest variance (8.87 %) was between the Jangsu and Tongyeong strains. In addition, the genetic distance between the populations showed that Jangsu and Tongyeong formed one branch (they were very similar genetically). The Dangjin and the Gyeongsang National University strains appeared to form a second branch. Furthermore, the crossbred formed one branch with the Dangjin and the Gyeongsang National University strains. Therefore, the results of this study can be used as basic data to reduce unnecessary inbreeding and genetic resource flow between the KNBG populations. The basic data indicates the uniqueness of the genetic resources of the domestic lineage. These findings provide a basis for differentiating KNBG and Crossbred goats to use to improve the desirable characteristics of this species.

STUDY ON THE MORPHOLOGICAL VARIATIONS OF MUSSEL MYTILUS CORUSCUS GOULD (담치의 형태변이에 관한 연구)

  • The followings are the results of study about morphological variation of mussel, Mytilus coruscus collected respectively from Sanyang-myeon Tongyeong-goon Gyeongnam on 13th January, Yeonwha-do Yokji-myeon Tongyeong-goon Gyeongnam on 15th May and down below the Yeong-do Bridge Busan on 20th August, 1964. The biggest mussel of the above three areas have been from Yeong-do Bridge, whose shell height is 164.1 mm, shell length 77.8mm, shell breadth 52.2mm, total weight 291,9g and shell length is as follows : from Yeong-do Bridge L=0.4954H + 1.9516, from Sanyang-myeon L=0.3718H + 14.145, from Yeonwha-do L=0.4074H + 9,6610 The relationship between shell height and shell breadth is as follows : from Yeong-do Bridge B=0.3426H + 0.2052, from Sanyang-myeon B=0.3084H + 3.6183, from Yeonwha-do B=0.3507H + 0.8028 In view of the above relationship, it is concluded that the slope value of mussel, nearer to the inshore from the off-sea, is similar to that of M. edulis, from which are can presume that M. coruscus could be changed in shell form according to its environment.
