Production of Bio-ethanol from Brown Algae by Physicochemical Hydrolysis (물리화학적 가수분해에 의한 갈조류 바이오 에탄올 생산)

  • In this study, the productivity of bio-ethanol obtained from various brown-algae raw materials was examined. Brown-algae polysaccharide, consisting of alginate and laminaran, is usable for the effective production of bio-ethanol if it is hydrolyzed to monomer unit. The objective of this study is the production of bio-ethanol from brown-algae using a heat-treatment and acid-treatment. Bio-ethanol was produced by Saccharomyces cerevisiae KCCM1129 and Pachysolen tannophilus KCTC 7937 strains. Laminaran japonica was higher than Sagassum fulvellum and Hizikia fusiformis, Laminaran japonica optimum pre-treatment is used to derive the ethanol production of Saccharomyces cerevisiae KCCM1129 and Pachysolen tannophilus KCTC 7937 respectively 9.16 g/L, 9.80 g/L. The maximum output of Sargassum fulvellum and Hizikia fusiformis was very low as 0.22 g/L.

광양만의 환경오염과 해조군집 변화 연구

  • 광양만 지역 수직군집구조는 최상부는 해조류가 거의 분포하지 않으며, 상층 하부부터 Ulva spp.가 우점하고 있으며, 저서동물은 최상부지역에는 따개비류가 우점하며, 하부로 갈수록 Oysters가 우점하였다. 그리고 층별군집구조는 상층부는 Biofilm이, 중ㆍ하층부는 Ulva spp.가 우점하며, 하층부로 갈수록 Turf algae의 밀도가 높다. 따개비류는 상층부와 중층부에서 우점하고 있으며, 중하층부로 갈수록 Oysters가 우점하며 총알고동은 상층부에서 중하부에서는 삿갓조개류의 밀도가 높았다. 남해 서상지역의 수직군집구조는 8월 조사결과 상부는 Brown crustose algae가 중부부터는 지층이, Ulva spp. 등이 하부에서는 유절산호조류, 개서실 등이 우점하였으며, 10월 조사에서 상부에는 해조류가 거의 발견되지 않았으며, 중부이하에서 지층이, 개서실, 유절산호조류 등이 하부에는 가시명불 등이 우점하였다. 저서동물은 말미잘, 삿갓조개류, 고둥류 등이 높이와 관계없이 균일한 밀도를 보였다. 그리고 층별군집구조는 상층부는 지층이, 무절산호조류 등이, 중층부는 개서실, ULva spp. 유절산호조류, 지층이 등이, 하층부는 개서실, 유절산호조류, 가실덤불 등이 우점하였다. 저서동물은 전층에서 따개비류, 말미잘 등이 우점하였다. 상주지역의 수직군집구조는 상부는 해조류가 거의 대부분 분포하지 않으며, 중부는 Biofilm, 우뭇가사리가, 하부에서는 지충이, 패, Brown crustose algae, 무절산호조류, 톳 등이 밀도가 높게 나타났다. 저서동물은 상부에는 총알고둥과 따개비류, 중ㆍ하부는 삿갓조개류, 말미잘이 우점하였다. 그리고 수평군집구조는 Biofilm이 상층부의 일부 지역에서 조사되었으며, 중층부는 Biofilm, Brown crustose algae, 무절산호조류, 패, 지층이 등이 우점하며, 하부는 지충이, 톳, 유절산호조류, 무절산호조류, Brown crustose algae 등이 우점하였다. 저서동물은 상층부는 총알고둥이 중층부는 삿갓조개류, 말미잘, 군부, 해면류가 하층부는 해면류, 삿갓조개류, 말미잘 등이 우점하였다.

Antitumor Activity of Seaweeds toward Sarcoma-180 (해조류의 Sarcoma-180에 대한 항암효과)

  • Protein-polysaccharides(PP) extracted from four species of gulfweed, fusiforme, sea-mustard and sea-tangle were examined for antitumor activity toward sarcoma-180 implanted in mice, ICR. Polysaccharides in protein-polysaccharides extracted from gulfweed, fusiforme, sea-mustard and sea-tangle were 61.48, 55.61, 34.06 and 30.28%, respectively. Monosaccharides of four seaweeds consisted of glucose, galactose, mannose, fructose and xylose, and major amino acids consisted of glutamic acid, aspartic acid, cystein, valine and glycine. Antitumor activity of the protein-polysaccharides extracted from sea-mustard showed the highest inhibition ratio of 69.76% when PP was injected to the mice at the dose of 100mg/Kg/day. PP of sea-tangle showed the maximum survival ratio of 25.22% when injected at the dose of 100mg/Kg/day.

Whitening Effect of Hizikia fusiformis Ethanol Extract and Its Fractions (톳(Hizikia fusiformis) 에탄올 추출물 및 분획물의 미백활성)

  • Melanin synthesis is catalyzed by tyrosinase. To investigate the whitening effect of Hizikia fusiformis, fractions from ethanol extract of H. fusiformis were prepared by a systematic fractionation procedure with solvents such as methanol, hexane, butanol, and $H_2O$. The ethanol extract and its fractions were then subjected to evaluate the inhibitory effects on the tyrosinase activity and melanin synthesis in murine B16F10 melanoma cells. The ethanol extract and aqueous fraction exhibited a whitening effect with no cytotoxicity. The ethanol extract showed the highest whitening effect among the samples. The inhibitory effect of $100{\mu}g/ml$ of ethanol extract was higher than that of $10{\mu}g/ml$ of arbutin, but it was lower than that of $10{\mu}g/ml$ of kojic acid. Furthermore, the inhibitory effects of $100{\mu}g/ml$ of methanol, hexane, butanol, and aqueous fractions were similar to those of $10{\mu}g/ml$ of arbutin. The antioxidant activities were examined by comparing the results with that of ascorbic acid as a positive control. The ethanol extract and aqueous fraction showed relatively higher DPPH radical-scavenging activities compared with the other samples. Furthermore, $500{\mu}g/ml$ of ethanol extract and aqueous fraction diminished LPS-induced iNOS expression to 82 and 80%, respectively. These results suggest that ethanol extract and aqueous fraction of H. fusiformis could be used as cosmetic ingredients for whitening and skin protection effects.

STUDIES ON THE EXTRACTION OF SEA WEED PROTEINS 2. Extraction of NaCl and Alcohol Soluble Proteins (해조단백질 추출에 관한 연구 2. 식염가용성 및 알콜가용성 단백질의 추출)

  • In present study, the effects of various factors including the solvent concentration, extraction time and temperature, the ratio of sample vs extraction solvent, (w/v) and pH upon the extractability of the NaCl and alcohol soluble proteins of marine algae were investigated. Eight species of fresh algae, the major ones in consumption as food, namely Porphyra suborbiculata, Undarie pinnatifida, Hizikia fusiforme, Sargassum, fulvellum, Enteromorpha linza, Sargassum kjellmanianum, Codium coarctatum, and Ulva pertusa were used for the extraction of NaCl soluble protein and dried materials of four species, Perphyra suborbiculata, Undaria pinnatifida, Enteromorpha linza and Sargassum fulvellum were used for the extraction of alcohol soluble protein. The frozen and mascerated samples were prepared by the same method described in previous paper (Ryu, 1977). And the dried materials were moistened with alcohol solution before freezing. The effect of solvent concentration on the extractability of NaCl soluble protein differed from species. The extractability of Undaria Pinnatifide, Hizikia fusiforme, Perphyra suborbiculata, Enteromorpha linza, and Ulva pertusa reached maxima at 0.25M NaCl solution while the 1.0M for Sargassum fulvellum, Saygassum kjellmanianum and Codium coarctatum. In case of alcohol soluble proteins, it was shown at $20\%$ ethanol solution for Porphyra suborbiculata, Undaria pinnatifida, Enteromorpha linza, and Sargassum fulvellum. Variation of the ratio of sample vs solvent gave slight effect upon the extractability, but the ratio of 1:30(w/v) seemed most efficient for the extraction of NaCl soluble proteins and 100 ml solvent added to 1 g dried sample was effective in case of alcohol soluble proteins. Extraction time has a minimal effect upon the extraction of alcohol soluble protein, and approximately 21 to $43\%$ of algal protein was extracted within 1 hour. But in case of NaCl soluble protein extraction, the effect of time revealed differently from species to species resulting in that the extraction for 1 hour gave a maximum extractability in Ulva pertusa and Enteromorpha linza, 2 hours in Porphyra suborbiculata, Codium coarctatum and 3 hours in Undaria pinnatifica, Hizikia susiforme, Sargassum fulvellum and Sargassum kjellmanianum. When the NaCl soluble protein of Undaria pinnatifida and Enteromopha linza was extracted at various temperature, the most effective extraction temperature was $40^{\circ}C$ while the temperature was $50^{\circ}C$ for Undaria pinnatifida and $60^{\circ}C$ for Hixikia fusiforme, Sargassum fulvellum, Sargassum kjellmanianum and Codium coarctatum. Bus in case of alcohol soluble extraction, the optimum temperature was $30^{\circ}C$ for Enteromorpha linza and $40^{\circ}C$ for Undaria pinnatifida, Sargassum fulvellum and Porphyra suborbiculata. In the effect of pH on extractability, the maximum extractability of NaCl soluble proteins was obtained at pH 7to 8 and pH 8 to 9 for alcohol soluble protein.

Quality Characteristics of Sulgidduk Added with Hizikia fusiformis Powder (톳가루를 첨가한 설기떡의 품질특성)

  • This study was performed to examine the optimum addition amount of Hizikia fusiformis powder to be used with rice flour in the preparation of Hizikia fusiformis Sulgidduk (traditional rice cake). Hizikia fusiformis powder was added to rice powder at ratios of 5, 10, 15 and 20% (w/w). The moisture content of Sulgidduk was increased with the addition of Hizikia fusiformis powder, whereas the pH content was decreased. The addition also lowered the lightness and yellowness values but increased the greenness value. Mineral contents were increased. Hardness, springiness, and brittleness were decreased, and cohesiveness was increased as the added powder content also increased. In the sensory evaluation of Hizikia fusiformis Sulgidduk, color, flavor, taste, softness, chewiness, moistness, cohesiveness, and overall acceptability characteristics were all highest at the 10% addition level. Thus it can be proposed that this is the ideal level of Hizikia fusiformis addition in terms of the sensory properties of the cooked product.

남해산 해조류의 항산화 활성

  • 대사작용의 개선, 정장작용과 유독 물질의 제거, 혈압과 콜레스테롤치의 정상화, 항균 및 항암효과 등 다양한 생리작용을 갖는 것으로 알려진 해조류의 기능성 규명을 위한 연구의 일환으로 총 6종(진두발, 갈래곰보 꼬시래기, 돌가사리, 고장초 톳)의 해조류의 에탄을 추출물에 대한 항산화 효과를 실험하였다. 해조류의 환원력은 시료의 첨가농도에 비례하는 경향이었으며, SOD 유사활성은 고장초의 경우 가장 높은 활성을 나타내었는데 첨가농도 500 $\mu\textrm{g}$/$m\ell$와 1000 $\mu\textrm{g}$/$m\ell$에서 각각 35.43%와 56.71%의 활성을 나타내었다.   • This study was performed to investigate the change of chemical components and antioxidant activities in Hizikia fusiformis(Harvey) OKAMURA with blanching times. For chemical composition, the contents of minerals (K, Ca, Na, Mg, Fe, Cu, Mn and Zn), vitamins (vitamin C, ${\beta}$-carotene and ${\alpha}$-tocopherol) and total polyphenol were analyzed. The antioxidant activity was determined through free radicals (DPPH radical, superoxide anion radical, hydroxyl radical and hydrogen peroxide) scavenging activity and linoleic acid peroxidation inhibitory activity. As the blanching time increased, the contents of all minerals, vitamin C and total polyphenol decreased. However ${\beta}$-carotene and ${\beta}$-tocopherol were not affected by the blanching time. In addition antioxidant activities decreased with increasing blanching time.

참전복, Haliotis discus hannai 부착치패의 먹이 선택성과 성장

  • 우리나라에서 건강식품으로 널리 알려져 있는 참전복은 불로장수의 식품으로 널리 애용되고 있을 뿐만 아니라 발암억제 물질인 paolin을 함유하고 있어, 세계적으로 상품가치가 인정되고 있다. 이러한 중요성에도 불구하고 참전복 부착치패의 성장 및 생존율 제고는 아직까지도 해결되어야 할 문제로 남아 있는 실정이다. 참전복 부착치패의 먹이선택성 및 유인성에 관한 기존의 연구를 보면 Viana et al. (1994)은 평균 각장 4.2 cm인 치패를 이용하여 배합사료의 9가지 원료에 대한 유연성을, Harada and Kawasaki (1982)는 각장 2.4 cm 이상인 치패를 대상으로 해조류 3종(미역, 톳, 파래)의 유인효과에 관하여 보고하였다. 그러나 이와 같은 보고들은 전복 종묘생산 현장에서 보다 빠른 시기에 박리하여 해조로 먹이 전환을 하기 위한 노력과는 어느 정도의 거리가 있다고 판단된다. 따라서 본 연구에 있어서는 평균 각장 5.0$\pm$1.0 mm인 치패를 박리하여 Navicula spp., 해조 2종(다시마, 파래) 및 자체 제조한 사료를 공급하여 먹이별 유인효과를 관찰하였다.
