  • 본 분석방법은 토양중에 방향성화합물을 함유한 유류 존재를 스크리닝하는 방법으로 ASTM(The American Society for Testing and Materials)에 등록되어 있으며, 수분 제거제를 이용하여 토양수분에 의한 정량 오차를 줄이고 UV-Spectrophotometer로 정량한다. 오염된 유류의 종류를 알고 있으나 동일한 유류를 구하지 못할 경우에는 일반적으로 동일종에서 얻어진 계수로 농도를 추정하며, 오염된 유류의 특성을 모를 경우에는 본 방법으로 오염여부 판단만이 가능하다. 그러나 토양에 오염된 것과 동일한 유류로 검량선을 작성할 경우에는 대략적인 정량도 가능하며, 분석방법을 요약하면 다음과 같다. 일정량의 토양시료에 CaO를 첨가하여 토양수분에 의한 영향을 제거한 뒤 이소프로필알콜(isopropyl alcohol)로 추출하여 여과한다. 여과액을 254nm의 UV파장에서 흡광도를 측정하여 오염된 것과 동일한 유류로 작성된 검량선을 이용하여 정량한다.

Behaviour of the Soil Residues of the Acaricide-Insecticide, [$^{14}C$]Acrinathrin;I. Behaviour during Crop(Maize) Cultivation (살비살충제 [$^{14}C$Acrinathrin 토양 잔류물의 행적 규명;I. 농작물(옥수수) 재배시의 행적)

  • In order to elucidate the fate of the residues of the pyrethroid acaricide-insecticide, acrinathrin in soil, maize plants were grown for one month on the specially-made pots filled with two different types of soils containing fresh and one-month-aged residues of [$^{14}C$]acrinathrin, respectively. The mineralization of [$^{14}C$]acrinathrin to $^{14}CO_2$ during the one-month period of aging and of maize cultivation amounted to $23{\sim}24%$ and $24{\sim}33%$, respectively, of the original $^{14}C$ activities. At harvest after one-month growing, the shoots and roots contained less than 0.1% and 1% of the originally applied $^{14}C$ activity, respectively, whereas the $^{14}C$ activity remaining in soil was $65{\sim}80%$ in both soils. Three degradation products with m/z 198(3-phenoxybenzaldehyde), m/z 214(3-phenoxybenzoic acid), and m/z 228(methyl 3-phenoxybenzoate) besides an unknown were identified from acetone extracts of both soils without and with maize plants after treatment of [$^{14}C$]acrinathrin, by autoradiography and GC-MS, and those with m/z 225(3-phenoxybenzaldehyde cyanohydrin) and m/z 198 (3-phenoxybenzaldehyde) from acetone extract of the Soil A treated with 50 ppm acrinathrin and grown with maize plants for 30 days were identified by mass spectrometry. These results suggested that the hydrolytic cleavage of the ester linkage adjacent to the $^{14}C$ with a cyano group, forming 3-phenoxybenzaldehyde cyanohydrin. The removal of hydrogen cyanide therefrom leads to the formation of 3-phenoxybenzaldehyde as one of the major products. The subsequent oxidation of the aldehyde to 3-phenoxybenzoic acid, followed by decarboxylation would evolve $^{14}CO_2$. Solvent extractability of the soils where maize plants were grown for 1 month and/or [$^{14}C$]acrinathrin was aged for 1 month was less than 31% of the original $^{14}C$ activity and over 95% of the total $^{14}C$ activity in soil extracts was distributed in the organic phase. Accordingly, acrinathrin turned out to be degraded rapidly in both soils and be bound to soil constituents as well, not being available to crops.

The Effects of pH Change in Extraction Solution on the Heavy Metals Extraction from Soil and Controversial Points for Partial Extraction in Korean Standard Method (용출액의 pH 변화가 토양내 중금속 용출에 미치는 영향과 그에 따른 국내 토양 오염 공정시험방법의 문제점)

  • Heavy metals are extracted from Chonju stream sediment, roadside soils and sediments along Honam expressway, soils and tailings from mining area using three different methods (partial extraction in Standard Method, partial extraction method with maintaining 0.1 N of extraction solution and Sequential Extraction Method). In samples having buffer capacity against acid, pH 1 (0.1 N HCl) of extraction solution can not be maintained and pH of extraction solution increases up to 8.0 when partial extraction in Standard Method is used. The averages and ranges of HPE(heavy metals extracted using partial extraction in Standard Method)/HPEM(heavy metals extracted using partial extraction method with maintaining 0.1 N of extraction solution) values are 0.479 and 0.145~0.929 for Cd, 0.534 and 0.078~0.928 for Zn, 0.432 and 0.041~0.992 for Mn, 0.359 and 0.011~0.874 for Cu, 0.150 and 0.018~0.530 for Cr, 0.219 and 0.003~0.853 for Pb, and 0.088 and 1.73${\times}$10$^{-5}$~0.303 for Fe. These data indicate that the difference between HPE and HPEM is large in the order of Fe, Cr, Pb, Cu, Mn, Cd and Zn. The amounts of heavy metals extracted decreases in the follow order; Sum III(sum of fraction I, II, III in sequential extraction)>HPEM>Sum III (sum of fraction I and II)>HPE for Zn, Cd and Mn and Sum III>HPEM>HPE for Cr and Fe. In the case Cr, Sum II is lower than HPEM and higher than HPE. In case of Cu, extracted heavy metals is large in the order Sum IV>HPEM>Sum III HPE. HPE/HPEM value decreases with increasing the amount of HCl used for maintaining 0.1 N of extraction solution. For samples with high buffer capacity, HPE/HPEM value in all elements is lower than 0.2. On the other hand, for samples with low buffer capacity, HPE/HPEM value are over 0.2 and many samples have values higher than 0.6 for Zn, Cd Mn and Cu due to the small difference between Sum II and Sum III, and relatively higher mobility. However, for Fe and Cr, HPE/HPEM value is below 0.2 even for samples with low buffer capacity due to their low mobility and big difference between Sum II and Sum III. This study indicates that the partial extraction method in Korean Standard Method of soil is not suitable for an assessment of soil contamination in area where buffer capacity of soil can be decreased or lost because of a long term exposure to environmental damage such as acidic rain.

Screening for Antagonistic Plants for Control of Phytophthora spp. in Soil (토양중(土壤中)의 Phytophthora spp. 방제(防除)를 위한 길항식물(桔抗植物)의 탐색)

  • Among 100 species in 54 families of plants tested, leaf extracts from Allium cepa, Allium sativum, Malussieboldii, Reynoutria japonia and Rheum coreanum were inhibitory on mycelium growth of phytophthora spp.. Especially, Allium sativum and Malus sieboldii were strongly inhibitory. Allium sativum, Malussieboldii and Rhem coreanum were strongly inhibitory on zoosporangium germination of P. capsici. Malus sieboldii only were strongly inhibitory on zoosporangium germination of P. nicotiana and P. infestans.And Malus sieboldii were strongly effected on disease control of these pathogens. These results indicate the possibility of finding antagonistic plants in the nature for the control of certain pathogens in soil.

  • The extraction methods of PBBs from environmental samples by soxhlet extraction for solid phase and liquid-liquid extraction for liquid phase were compared. After extraction, silicagel and florisil as column clean up methods were used for the comparison of cleanup efficiency with different solvent system. Also, the analytical instruments were used GC/MSD. The 22 kinds of PBBs standards were used to establish the analytical methods to perform the experiments of recoveries and detection limits in water and soil. The detection limits of PBBs represented 5~10 ng/L for water and 0.5~3.5 ng/g for soil samples, respectively. The recoveries of water samples represented 96~107% in n-hexane, dichloromethane and toluene for water, and 60~80% in combined solvent with n-hexane and acetone. The recoveries of soil sample were surveyed 67~97% in soxhlet extraction and 64~76% in ultrasonic extraction with selected solvents.

Change in the non-extractable bound residue of TCAB as a function of aging period in soil (Aging 기간에 따른 TCAB의 추출불가 잔류물의 토양중 변화)

  • In order to elucidate the possible change in the non-extractable bound residue of TCAB(3,3' 4,4' - tetrachloroazobenzene) in soil as a function of aging period, uniformly ring-labelled $^{14}C-TCAB$ was treated to soil(organic matter : 1.8%), and aged for 3, 6, 9, 12 and 15 months at $21{\pm}1^{\circ}C$, respectively. $^{14}CO_2$ evolution and volatilization loss during the aging were negligible. The amounts of non-extractable bound residue of TCAB increased gradually from 7.55% in 3-month aging to 19.32% in 15-month aging. Partition data suggested no formation of polar groups in the chemical structure of TCAB. Most of $^{14}C-radioactivity$ of bound residues was present in humin in the range of 50.52 to 58.93%. The fact that the number of microorganisms in soil decreased relative to the control suggested no chance of their involvement in the formation of non-extractable bound residues. Accordingly, the increase in the non-extractable bound residue of TCAB in soil with aging period is believed to be due to the transformation of the trans isomer to the cis one which is more polar and more adsorptive than the former.

  • Nitrogen fertilizers such as urea are readily hydrolyzed in soils to produce ammonium ions which pass through nitrification and denitrification processes. These serial processes have drawn attention due to nitrogen losses, eutrophication, blue baby syndrome, and ozone depletion problems. The purpose of this study was to test the inhibitory effects of hot-water extract and organic solvent fractions of Artemisia asiatica leaves on soil urea hydrolysis and nitrification. In addition, the effects of organic solvent fractions on urease activity and ureolytic bacterial population were also investigated. First, hot-water extract of Artemisia asiatica leaves inhibited soil nitrification substantially with a marginal stimulatory effect on soil urea hydrolysis. Soils treated with hot-water extract of Artemisia asiatica leaves showed significant decreases in the accumulation of soil $NO_3-N$ (~68% decrease) compared with the control soil without the treatment of hot-water extract. In contrast, $CHCl_3$/MeOH fraction and basic aqueous layer of Artemisia asiatica leaves inhibited soil urea hydrolysis very strongly, causing 5.8 and 4.3-fold higher accumulation in amounts of remaining urea-N compared with the non-treated soil. Meanwhile, non of the organic solvent fractions showed any significant effects on soil nitrification inhibition. The inhibition of ureolytic bacterial activity by $CHCl_3$/MeOH fraction and aqueous basic layer of Artemisia asiatica leaves without any effects on urease activity itself led us to conclude that the inhibitions of soil urea hydrolysis were caused by the antagonistic effects on ureolytic bacterial activity.

Properties of Suppressive and Conducive Soils to Ginseng Root Rot (인삼 근부병 억제토양 및 유발토양의 특성)

  • Biological, physical and chemical characteristics of suppressive and conducive soils to ginseng root rot were investigated. Population of antagonistic microorganisms to Fusarium solani was much higher in suppressive soils than in conducive soils, whereas the numbers of Fusarium species were smaller in suppressive soils. Mycelial growth and chlamydospore formation of Fusarium solani were inhibited in suppressive soils. In the water extract of suppressive soils, lysis of germination tube and macroconidia of F. solani was occurred by antagonistic microorganisms at 4 hours after treatment. There were no significant differences in physical and chemical characteristics between supressive soils and conducive soils to ginseng root rot, however, clay content of suppressive soils was a little higher than that of conductive soils.

혐기성 슬러지를 이용한 디젤오염토양의 분해

  • 생물학적 토양오염 복원기술은 산소와 영양염류를 오염토양에 공급하여 호기성 미생물의 대사작용을 자극함으로써 유류를 생분해 하는 방법으로 널리 사용되고 있다. 유류에 오염된 토양은 혐기성 상태인 경우가 대부분이기 때문에, 호기성 미생물을 이용하기 위해서는 충분한 산소를 공급하여야 하므로 운전비가 많이 드는 단점이 있다. 최근에는 혐기성 미생물을 이용하여 유류오염 토양을 정화하는 방법이 보고되고 있다. 혐기성 생분해 방법은 다소 분해 속도는 느리지만 산소를 공급하지 않기 때문에 경제적인 유류오염토양 복원 방법으로 주목받고 있다. 본 연구에서는 디젤을 사용하여 인위적으로 10000 mg/kg.TPH soil의 농도로 오염시킨 토양 50g을 100$m\ell$ 용적의 vial에 주입하고 하수처리장의 혐기성 소화조 슬러지를 15$m\ell$, 30$m\ell$을 주입하여 배양하였으며 TPH의 분해량과 CH$_4$$CO_2$ 발생량을 측정하였다. vial의 기상을 $N_2$가스로 치환함으로써 혐기성 상태가 되도록 하였으며, 35$^{\circ}C$에서 90일간 배양하였다. 그 결과, 슬러지를 주입하지 않은 대조군의 경우에는 TPH의 분해가 거의 없었지만, 슬러지를 주입한 경우에는 TPH(Total Petroleum Hydrocarbon)농도가 55% 제거된 것으로 나타났다. TPH의 분해는 CH$_4$$CO_2$ 발생량과 밀접한 상관관계를 보였다. 본 연구의 결과로부터 하수처리장의 혐기성 소화조 슬러지를 이용한 유류오염 토양의 복원 가능성을 확인할 수 있었다.양에서 유동 가능성이 있는 중금속만을 추출하였다. 분석실험은 토양의 Cd2+ 와 Pb2+를 대상으로 행하여졌으며, 여러 토양에서 추출 분석한 결과를 EDTA분석결과와 비교하였다. 실험결과, 중금속은 매우 신속하게 고분자 자성체와 결합하였고, 그 후 자성체를 외부 자장으로 모은 후 산으로 용해시키고, 결합된 중금속은 Graphite furnace AAS로 분석함으로써 빠르고 효율적으로 분석실험을 수행할 수 있음을 알 수 있었다. 한편, 실험에서 나타난 수치들을 비교 검토한 결과 토양 분석시 sandy soil에서는 자성체를 이용한 분석이 EDTA에 의한 방법보다 더 높은 추출도를 보인 반면, silt 함량이 많은 토양의 경우에서 EDTA분석에서 더 높은 중금속 추출도를 보였다.s 중에서 490nm와 555nm의 복합밴드를 포함하는 OC2 알고리즘(ocean color chlorophyll 2 algorithm)을 사용하는 것이 OC2 series 및 OC4 알고리즘보다 좋은 추정 값을 도출할 수 있을 것으로 기대된다.환경에서는 5일에서 7월에 주로 이 충체의 유충이 발육되고 전파되는 것으로 추측되었다.러 가지 방법들을 적극 적용하여 금후 검토해볼 필요가 있을 것이다.잡은 전혀 삭과가 형성되지 않았다. 이 결과는 종간 교잡종을 자방친으로 하고 그 자방친의 화분친을 사용할 때만 교잡이 이루어지고 있음을 나타내고 있다. 따라서 여교잡을 통한 종간잡종 품종육성 활용방안을 금후 적극 확대 검토해야 할 것이다하였다.함을 보이고 있다.X> , ZnCl$_{3}$$^{-}$같은 이온과 MgCl$^{+}$, MgCl$_{2}$같은 이온종을 형성하기 때문인것 같다. 한편 어떠한 용리액에서

Allelopathic Inhibition by Extracts and Volatiles from Leaf and Seed of Sicklepod (Cassia tora L.) (결명자(決明子)(Cassia tora L.) 엽(葉)과 종자(種子)의 추출물(抽出物)과 휘발성분(揮發成分)의 타감성(他感性) 생육저해작용(生育沮害作用))

  • The growth of weeds and some other plants has been considered to be inhibited by sicklepod(Cassia tora L.) sharing the habitat. The study was conducted, for the first time, to propose that this phenomenon is not only due to competition for physical and nutritional conditions but also due to allelopathy. In addition, autotoxicity of sicklepod was examined. The results are summarized as follows. 1. Rice sheath length reduced progressively from 18 % to 36 % with increasing the concentration of treating aqueous extracts of sickle pod seeds, but rice germination was not affected. In contrast, radish hypocotyl length was not reduced by the aqueous extract treatment but radish germination was significantly reduced by 66 % at 1 : 10 and 1 : 5 treatment. 2. Total chlorophyll contents in rice seedling decreased from 50 % to 65 % by treatment of seed aqueous extracts diluted from 1 : 50 to 1 : 5 ratio. 3. Aqueous extracts of sicklepod leaves significantly reduced hypocotyl length and fresh weight in radish and germination in rice, but mung bean was slightly affected by aqueous extracts only in fresh weight. 4. Volatiles from fresh, immature seeds with husk reduced the radish germination and seedling growth and radish root growth appeared to be more sensitive to the exposure to volatiles from fresh immature seeds than both germination and hypocotyl growth. 5. Volatiles from sickepod leaves inhibited germination and growth of radish, rice and mung bean, and seedling growth was more sensitive to volatiles from leaves than germination. 6. Volatiles from sicklepod leaves reduced germination and radicle length of sicklepod itself. 7. Collectively, it is concluded that there are water-soluble and volatile substances responsible for allelopathy in sicklepod.
