Cell Viability and Antioxidant Enzyme Activity in the Cell of Ginseng (Panax ginseng C.A. Meyer) Treated with Soil Extracts (인삼재배지의 토양추출물이 종자 발아와 세포의 항산화효소 활성에 미치는 영향)

  • One hundred-eighty extracts of soil collected from ginseng (Panax ginseng C.A. Meyer) fields were subjected to lettuce germination test, electrolyte leakage, cell viability and antioxidant enzyme activity test. Regardless of various cultivation periods, there was no significant difference in soil pH, the content of organic matter and available phosphate in ginseng fields. Based on lettuce seed germination test, six soil extracts showing inhibition of germination and/or seedling growth were selected for further study. Selected soil extracts markedly inhibited cell viability of ginseng cultured cells but leakage of electrolytes were not affected by the treatment. Enzyme activity of superoxide dimutase in ginseng cultured cells was not affected by the treatment with the soil extracts. However, those of peroxidase and catalase were significantly inhibited by the treatment with soil extracts which showed inhibition of lettuce seed germination and seedling growth.

세포능식촉진인자(MPF)의 특이적 억제제 생산 토양균주의 검색 및 억제제의 분리와 특성 규명에 관한 연구

  • 본 연구에서는 토양균이 생성하는 2차 대사산물로부터 세포증식 촉진인자 (Maturation Promoting Factor. MPF)인 cdc2/cdk2-cyclin의 복합체를 특이적으로 억제하는 물질을 분리하고 그 물질의 생리활성을 조사하였다. 토양으로부터 순수분리된 300여개의 토양균에서 생성된 배양액을 취하여 MPF의 특이적인 기질인 합성 peptide (CSH103 :HATPPKKKRK)를 사용하여 인산화 활성을 측정하였다. 그중 MPF 활성 억제능이 90% 이상인 19개의 균주를 1차적으로 선정한 후 각각에 대하여 열/pH에 대한 안정성, 각종 용매에 대한 추출성 등 이화학적 성질을 규명하였으며 이중 균주 LPL 931로부터 MPF 활성 억제제를 분리하고자 하였다. 예비실험의 결과로부터 토양균 LPL 931을 대량배양하여 열처리하고, 비이온성 수지인Amberlite XAD-2에 결합시키고 70% acetone으로 용출시켰다. 이 추출물로부터 ethylacetate와 n-butanol을 사용하여 MPF 억제 활성물질을 추출하였다. 이 추출액을 실리카겔 관 크로마토그래피, 분취 TLC, 분취 HPL를 하여 MPF 활성 억제 분획을 분리하였다.

Effects of Aqueous Extracts from Naturalized and Korean Wild Plants on Seed Germination and Seedling Growth of Zoysiagrass (자생식물과 귀화식물의 수용 추출액이 Zoysiagrass의 발아와 유근생장에 미치는 영향)

  • The soil pH favored by Korean wild plants is 5.33~7.20, while naturalized invader species prefer soils of pH 3.95~6.48. The germination rate of Zoysia japonica was inhibited sharply, up to 60%, in extracts of naturalized plants of concentrations over 50%. Erigeron canadensis extract most strongly inhibited germination, while the Korean wild species, Cassia mimosoides var. nomame increased germination rates in concentrations of 30% and 50%. The seedling growth of Zoysia japonica in extracts of Korean wild species and naturalized species did not show differences, but was inhibited strongly, up to 60%, in the extract of Cassia mimosoides var. nomame concentration of over 15%. Analysis of the extract from Cassia mimosoides var. nomame identified procatechuic acid, $\rho$-hydroquinone, $\rho$-coumaric acid and ferulic acid at 254nm; and vanillic acid, hydroquinone, benzoic acid and cinnamic acid were discovered at 284nm. The seed germination and seedling growth of Zoysia japonica were investigated under different concentrations of phenolic compounds. Ferulic acid and vanillic acid were associated with an increased germination rate, while $\rho$-coumaric acid appeared to inhibit seedling growth.

Research Trends for Soil-Related Algal Toxicity (토양 관련 조류독성 연구동향)

  • Soil ecological risk assessment requires terrestrial toxicity data based on trophic levels including plants, earthworms, nematodes, and springtails. To expand the trophic levels, it is needed to consider primary producer algae, nearly distributed in terrestrial environment, as representative terrestrial test species. In this study, we collected research cases focused on soil-related test species and exposure media from SCI papers, and analyzed exposure media, test species, test chemicals, and other test methods, for reviewing research trends of soil-related algal toxicity. Up to now, in the soil-related algal toxicity, test species were 8 cases (Pseudokirchneriella subcapitata, Chlorella vulgaris, Scenedesmus bijugatus, Chlorococcum infusionum, Scenedesmus subspicatus, Nostoc linckia, Synechococcus elongatus, and Chlorococcum sp.) and endpoints were cell count or photosynthetic pigment content. Also, 5 of exposure media were liquid medium, soil extracts, porewater, agar medium, and soil. Most of papers used algae isolated from natural soils or soil extracts. There were only one case for assessing algal toxicity in soil medium. More researches regarding algal toxicity in soil environments need to be conducted consistently.

Antimicrobial Activities of Extracts from Several Native and Exotic Plants in Korea (수 종의 한국자생식물과 귀화식물 추출액이 토양미생물에 미치는 항균활성)

  • The soil pH favored by several native plants in Korea ranges 5.33∼7.20, while a more acidic range of pH 3.95∼6.10 is acceptable to exotic plants. Ethanol extracts of native and exotic plants in Korea were investigated for antimicrobial activity against Bacillus sphiaericus 2362, Bacillus thuringiensis var. subtilis and Bacillus thuringiensis var. cereus and Actinomycetes. Higher antimicrobial activity was observed from the extract of exotic plants than those of native plants. The ethanol extract of Ambrosia artemisiifolia var. elatior was observed to have the highest antimicrobial activity against 4 species of soil microbes. Especially, antimicrobial activity of Ambrosia artemisiifolia var. elatior showed the largest clear zone of 48mm in Actinomycetes. Larger clear zone was formed in the order of caffeic acid, benzoic acid and ρ -coumaric acid among the nine chemical compounds. Accordingly, the antimicrobial activity of Ambrosia artemisiifolia var elatior against Actinomycetes was found to be due to the synergetic effect of chemical compounds.

비소 오염 토양, 하천 퇴적물 및 광미의 복원을 위한 토양 세척 공정 개발에 대한 연구

  • 비소로 오염된 토양, 하천 퇴적물 및 광미의 복원할 때, 토양 세척 공정에서 중요한 인자인 비소의 화학적 결합형태와 세척제에 따른 용출특성과 고효율 세척 및 세척액의 재활용도를 높이기 위한 공정을 바탕으로 토양세척장비를 설계하였다. 화학적 결합형태에 있어서 토양은 잔류 결합형태가 주되고, 퇴적물의 경우는 철산화물과의 결합형태가 강하며, 광미는 황화물과의 결합에 따른 잔류형태와 철산화물과의 결합형태가 상당부분을 차지한다. 세척제에 따른 용출특성으로부터, 철산화물과 황화물과 결합하고 있는 비소의 화학적 결합형태를 파괴하면서 비소를 추출할 수 있는 용제로 HCl, Oxalate, EDTA, M$_2$O$_2$를 사용하였다. 추출 결과, 비소가 철산화물과 결합한 형태가 비중이 높을수록 EDTA 나 Oxalate가 효율이 높으며, 황화물에 대해서는 HCl과 $H_2O$$_2$이 상대적으로 높은 추출 효율을 보였다. 구성된 세척조는 밀폐실린더형과 스크류이송형 세척조로 구성되어 각각 혼합교반에 의한 세척과 토양입자 분급에 따른 세척이 가능하다. 세척 공정중 최적 산도 조절이 중요한 인자가 되며, 세척액의 재활용도를 높일때, 세척수에 용해되어 있는 비소 및 중금속과 미립자의 동시 제거를 위한 응집 침전조에서 응집제에 의해서 미립자와 함께 제거하는 응집, 침전 및 분리공정을 배치하였다.

Comparison of Rhizosphere Environments in Soils Suppressive and Conducive to Ginseng Root Rot (인삼근부병 억제토양 및 유발토양의 근권환경 비교)

  • Soils suppressive and conducive to ginseng root rot were studied by examining the mycelial growth of Fusarium solani, Phytophthora cactorum, and Sclerotinia sp. on extracts of each type soil. Rhizosphere environments of the two soils were also compared. Mycelial growth of all root rot fungi used was more severely restrained on the suppressive soil extract agar than that of conducive one. However, when heated at 100C for 30 minutes, mycelial growth of F. solani and Sclerotinia sp. was not affected, regardless of type soil used, whereas R. solani and P. cactorum grew better on conducive soil extract. Mycelial growth of all fungi used was stimulated as the treated temperature became higher. No significant differences between the two types of the soil were found in propagules of F. solani. The numbers of total fungi and total bacteria and the ratioes of total fungi to Fusarium and total bacteria to Fusarium were higher in the suppressive soils than in the conducive ones. Higher amount of clay existed in the suppressive soils, Mg and Na contents were lower in those soils than the conducive ones.

Remediation of Heavy Metal Contamination in OBOD Site with Soil Washing : Selection of Extractants (토양세척법에 의한 중금속오염 폐탄처리장 토양의 정화 : 세척액의 선정)

  • The efficiences of mineral acid (HCl), neutral salts ($CaCl_2$), and chelating agent (citric acid and $Na_2$-EDTA) were tested for extracting heavy metals from open burning and open detonation (OBOD) site soil. The extraction efficiencies of Cd, Cu, Pb and Zn from soil for various extractants were in the order of HCl > citric acid > $Na_2$-EDTA > $CaCl_2$, HCl (1.0 M) extracted effectively 82%, 86%, 80%, and 46% of initial total concentrations of Cd, Cu, Pb, and Zn, respectively. Significant negative correlations were observed between pH of extractant and amount of extracted heavy metals. Initially, examined heavy metals were predominantly bound to carbonate and Fe, Mn-oxide fraction. Though the significant amount of carbonate and Fe, Mn-oxide bounded metals were removed but a significant amount remained metals shifted to exchangeable (more mobile) fraction by HCl and citric acid extraction. The increased mobility of remaining metals could be problematic for water resources, thus careful management is needed to control the movement of heavy metals.

  • These studies was conducted to investigate the influence of ALLELOPATHIC effects of upland weeds on crops and soil microorganisms. The aqueous extract of weed residues were used to identify physiological controll of plant and microorganisms. The results obtained are as follows : 1. The aqueous extract of weeds controlled germination and growth of plant, especially the 5%-extract of Potulaca oleracea suppressed the growth of soybean about 8~12%. 2. The 5%-extract of Potulaca oleracea inhibited effect of phytosynthesis of soybean about 60%. 3. The move the concetration of phenolic compounds in soil, the less a number of actinomycetes and bacteria. 4. The number of soil microorganism(fusarium oxysporum spp.) were decresed as to the concentration of vanillic acid was increased.

A study on the washing remediation of tailing waste and contaminated surrounding soil of a bandoned metal mines (폐금속광산 광미 및 주변 오염토양 세정에 관한 연구)

  • This study has been carried out to examine the feasibility of washing technique for reducing the heavy metal contamination level of tailing wastes and agricultural soil surrounding abandoned metal mines. Some organic acids with low molecular weight were used as washing solution. Initial contamination levels of copper and lead for some soil samples were found to exceed the standard levels of countermeasure and concern, and those of cadmium to approach the standard level of countermeasure. Experimental results using sequential extraction method revealed that more than half of copper and lead existing in tailing wastes are adsorbed forms available for plants. There are some proportional relationships between metal concentrations determined by using 0.1N HCI solution and those determined by sequential extractions. Citric acid was turned out to be superior to oxalic acid and acetic acid with low molecular weight in washing above three metals. When citric acid is used for washing heavy metals from soil, it is desirable to operate at pH less than 5.5 for better washing effect. Metal removal effect by citric acid solution has been proved to depend upon solution concentration and the mass ratio of solution to soil. Addition of SDS(Sodium Dodecyl Sulfate) to citric acid improved the washing effect of cadmium among three metal most significantly. while copper removal did not change. Washing technique using citric acid for removal of heavy metals from agricultural soil or tailing wastes is recognized to be an effective remediation method.
