Recombinant Saccharomyces cerevisiae strains harboring various copy numbers of hirudin gene were developed to study dependency of hirudin expression level on its gene copy number. A linear relationship between the copy number of hirudin expression cassette and hirudin expression level was observed up to 10 copies. A double <5-integration vector truncated wi 디 1 the unnecessary bacterial genes before yeast transformation showed a four-fold increase in transformation efficiency and a 1.3-fold enhancement in hirudin expression level compared with a single <5 system. Gratuitous hirudin expression strain was developed by disrupting the GALl gene of S. cerevisiae. Glucose that was fed in a limited manner effectively supported cell growth and hi겨din expression by the gratuitous strain. Effects of methanol concentrations on hirudin production in recombinant Hansenula polymorpha were investigated in continuous and fed-batch cultures. At a steady-state of continuous culture, an optimum methanol concentration of 1.7 g/L was determined at a dilution rate of 0.18 $h^{-1}$ with 1.8 mg/L ${\cdot}$ h hirudin productivity.

Acid-stable ${\alpha}-amylase$ was partially purified from Paecilomyces subglobosum by Sephadex G-150 gel filtration. About 7.7-fold purification was obtained and the partially purified preparation has 5.0 U of ${\alpha}-amylase$ activity per mg of protein. Using this partially purified ${\alpha}-amylase$, general properties were studied and it showed the maximal activities at the conditions of pH 4.0 and $38^{\circ}C$. High stability of the acid-stable ${\alpha}-amylase$ in acidic condition was observed, whereas thermal stability was similar to the conventional ${\alpha}-amylase$.

A calf suffering from diarrhea was admitted to the Animal and Plant Quarantine Agency for diagnostic evaluation. Postmortem examination revealed that the mesenteric lymph node was enlarged and small intestine wall was thin. Microscopically, a large number of small round organisms were attached to the small intestine villi. Villous atrophy and proprial neutrophil infiltration were also observed. Based on modified Ziehl-Neelsen staining, electron microscopy, and ELISA results, the calf was diagnosed with fatal cryptosporidiosis.

Lakatos's mathematical philosophy implies that the mathematical knowledge is quasi-empirical and provides the context where mathematics grows and develops. So, it is educationally significant. But, it is not easy to apply Lakatos's methodology to teaching elementary mathematics, because Lakatos's logic of the mathematical discovery is based on the proofs and refutations but elementary mathematics does not contain any proof. This study is to develop the schemes that apply Lakatos's methodology to teaching elementary mathematics and to provide the teaching examples. I devised the teaching process and the curriculum development method. And I developed the teaching examples.

The purpose of this study was to examine how the Lakatos method works in the elementary teacher education program. Elementary preservice teachers were given a task in which they examined the Pick's theorem. The finding revealed that Lakatos method was usable in the elementary teacher education. They produced initial conjecture and found counterexamples, and finally made improved conjectures. These experience encourage them to change their belief of teaching and learning mathematics and to find alternative ways of teaching mathematics.

This study investigated the quality characteristics of muffins prepared with tagatose. The effect of tagatose were evaluated in terms of height, volume, weight, specific volume, baking loss rate, moisture contents, colorimeter, sensory evaluation, and texture of muffins during storage (1, 3, 5 days) and texture was measured using a texture analyzer. As the ratio of tagatose increased, the volume, specific volume, baking loss rate, and height decreased, whereas moisture content increased. During storage, control without tagatose showed the highest springiness lowest hardness by TPA, whereas hardness of muffins increased and springiness decreased as the contents of tagatose increased. Lightness of crust and crumb decreased significantly as contents of tagatose increased. According to the sensory evaluation, the control group showed the highest score in terms of texture but was not significant with 2% tagatose (T2). For appearance, color, flavor, taste, and overall acceptance, T2 showed the best result.

The endowed privilege to an established profession acts as an important motivation related to professionalization of occupations. The established profession enjoys cognitive, legal, and economic privilege. This study analyzes the professionalization process of interior design. Specifically, the study evaluates the suitability of the body of knowledge and professional ethos(i.e., protection of citizen's health, safety, and welfare), while analyzing the professionalization of interior design in the United States. The research aims at proposing a reference point for the professionalization of Korean interior design. Utilized data include research paper, court sentencing, and reports published by related professional organizations. The study concludes that 3E's are a means of professionalization of interior design, while separating interior design from interior decorating. In addition, there is no evidence to the protection of citizen's HSW. Rather, it increases service costs and restricts the job opportunities. When the public is not persuaded, the argument related to the necessity of license for the protecting citizen's HSW can be consumed as a mere rhetoric, concealing intentions for formulating a cartel. Thus, this study proposes that the professionalization of interior design may focus on the establishment of identity and body of knowledge, which are specific to interior design.

We study on administration system and teaching of R &E(Research and Education) in Korea Science Academy(KSA) laying stress on Mathematics project No.7. We analyze in detail administration system of R&E in KSA(for example, aim, human constitution, practical execution), and draw some meaningful suggestions in order to receive successful results in R&E of KSA. And we describe mathematical topics, problems, and results which are received by students of KSA in the process of R&E(project No.7).

The purpose of this study is to analyze the music of the Moombaton genre and to find out the characteristics of Jamaican music in it. I analyzed Lewis Poncy's and BTS's rhythmically, melodically and instrumentally to find the characteristics of Jamaican music. As a result, the rhythm features a Dembow rhythm and reggae rhythm derived from traditional Jamaican music. and As a feature of the melody, toasting, a unique Jamaican expression method, was used. Instrumental features include Latin American percussion, South American melodic instruments, and the use of tr-808. This study is meaningful in creating another fusion genre by finding the elements of Jamaican music in the current popular genre of EDM.

In this paper, wear characteristics of PVD coatings were compared on the die surface for cold stamping of MART1470 steel sheet with the finite element analysis and the pin-on-disc wear test. Three types of PVD coatings (CrN, TiAlCrN, and MoS2TiCr(W)N) were considered for the tool surface made of STD11 material. The stamping process of an auto-body part was analyzed with the finite element method. Ranges of process variables for the wear test such as contact pressure, relative speed, and sliding distance were predicted from analysis results. In order to quantitatively analyze wear characteristics of each coating, the amount of wear was measured and compared according to process variables with the pin-on-disc wear test. The influence of each process variable was investigated and the wear characteristics of the three coating layers were quantitatively compared. It was confirmed that the wear characteristics of MoS₂TiCr(W)N coating were better than those of CrN and TiAlCrN. It was noted that the proposed prediction approach could predict and respond to the wear phenomenon occurring in the stamping process.
