Antimicrobial and Antioxidative Effects of Alismatis Rhizoma Extract (택사 추출물의 항균 및 항산화 효과)

  • Antimicrobial and antioxidative effects of Alismatis Rhizoma which has been used as a galenic for diuretic and antiphlogistic were investigated. In a paper disc test, water extract of Alismatis Rhizoma showed antimicrobial activities against almost all of the microbial species tested, while their activities of methanol and ethanol extract was low than the activity of water extract, and the antimicrobial activity of water extract was most prominent against Vibrio parahaemolyticus. Antioxidative activity against linoleic acid was not observed in methanol extract. In contrast to methanol and ethnol and ethanol extract, water extract showed a antioxidative activity against linoleic acid. However, the antioxidative of water extract was lower than that of 0.1% BHT.

  • 엘라스타제는 폐의 탄성 섬유를 파괴하여 폐기종과 폐고혈압을 유발하는 역할을 한다. 이에 본 연구는 택사(澤瀉) 약침액(藥鍼液)의 엘라스타제 억제효능과 항산화효능을 측정하여 폐기종과 폐고혈압 치료의 가능성을 확인하고자 실시 하였다. 방법 : 택사(澤瀉) 약침액(藥鍼液)의 엘라스타제에 대한 억제효능과 di(phenyl)-(2,4,6-trinitrophenyl) iminoazanium (DPPH) 및 nitric oxide (NO) 자유기 소거능을 측정하였다. 결과 : 택사(澤瀉) 약침액(藥鍼液)은 엘라스타제 활성을 통계적으로 유의하게 억제하였고, DPPH 및 NO 자유기 소거능이 있음이 통계적으로 유의하게 확인되었다. 결론 : 본 연구를 통해 택사(澤瀉) 약침액(藥鍼液)의 엘라스타제 억제효능과 항산화 효능이 확인되었다. 본 연구 결과를 근거로 시험관내 실험이외의 추가적인 연구를 통해 폐기종과 폐고혈압 치료제로서 택사(澤瀉) 약침액(藥鍼液)의 활용을 고려해 볼 수 있을 것으로 기대한다.

Proper Harvesting Time and Drying Temperature for Improving the Alisma plantago Quality and Yield (택사 수량과 품질 향상을 위한 적정 수확시기와 건조 온도)

  • The characteristics associated with the Alisma plantago quality and the factors as the proper harvesting time and drying temperature of Alisma plantago were examined from 2004 to 2006 at Sunchon, Southern region. The Alisma plantago cultivars tested were Sunwol and Yongjun. The highest yields of root and good quality, color and luster were obtained when harvested 22 days after first frost date and dried with $35-40^{\circ}C$ of a weeks at the drying oven for Sunwol and Yongjin cultivars. Considering from our results, optimum harvesting time were most likely to be harvesting time of 22 days after first frost date and drying temperature of $35-40^{\circ}C$ of a weeks at the drying oven.

Isolation and Quantitative Analysis of Alisol B 23-Acetate from the Rhizome of Alisma orientale (택사에서 Alisol B 23-Acetate의 분리 및 함량분석)

  • Alismatis Rhizoma is an oriental medicine originated from the rhizome of Alisma orientale or Alisma plangtago-aquatica var. orientale (Alismataceae). As an standard compound of this plant, alisol B 23-acetate was isolated from the dichloromethane fraction of Alisma orientale and identified by the spectroscopic evidences. A Quantitative analysis of alisol B 23-acetate using HPLC method showed that the average content was 0.47$\pm$0.11% in 33 samples throughout the various regions of Korea.

Effect of Dietary Supplementation of Alisma canaliculatum (Alismatis Rhizoma) and Viscum album (Mistletoe) on Growth Performance and Immunity in Broiler Chickens (택사와 겨우살이 급여가 육계의 생산성 및 면역성에 미치는 영향)

  • This study was conducted to investigate the effect of dietary supplement of Alisma canaliculatum and Viscum album on the growth performance and immune response of broiler chickens. One hundred and forty Ross broiler chicks (1 day old) were assigned to 4 treatments in 5 replications with 7 birds per replication. The dietary treatments were NC group (without antibiotics), PC group (basal + 50 ppm Oxytetracycline), AC group (basal + Alisma canaliculatum 0.5%), and VA group (basal + Viscum album 0.5%). The results revealed that body weight gain and feed intake were not significantly different among the treatments. Addition of AC to the diet showed significantly higher feed conversion ratios than the VA addition diet and other treatments (p<0.05). The concentration of linolenic acid, ${\alpha}$-linolenic acid and arachidonic acid in the AC and VA treatment were relatively higher than that in the antibiotic treatment, while there was no difference in the concentration of SFA, USFA, and USFA/SFA. The serum IgG concentration in chickens tended to increase in the AC and VA treatment groups compared to NC group. The IL2 concentration in AC and VA groups was significantly higher than in other group (p<0.05). IL6 was not detected, however. GOT, GPT, and T-billirubin in the VA group were significantly higher than in other groups (p<0.05). Thus, the VA additives can result in the damage of liver and kidney. In conclusion, the dietary supplement of a 0.5% of AC and VA showed a similar growth performance and increased immunity response comparable to supplementing a 50 ppm Oxytetracycline. Therefore, it can be said that the AC and VA are possible alternates of antibiotics.

Purification and Characterization of Hemagglutinating Protein from Rhizome of Alisma orientale (택사(Alismatis Rhizoma) Hemagglutinating Protein의 정제와 특성)

  • Lectin was purified by using $(NH_4)_2SO_4$, DEAE-cellulose ion-exchange chromatography and Sephadex G-150 column chromatography from Alismatis Rhizoma(AR). The specific activity of AR lectin was 50, 441units/mg, and purification folds were 114. The AR lectin agglutinated human erythrocytes of all types(A, B, O, AB). The molecular weight of AR lectin was estimated about 90, 500 daltons by gel filtration and each subunits were 42,000, 27,000 and 22,500 daltons on SDS-PAGE respectively. The hemagglutinating activity of the lectin was inhibited by sialic acid, glucose, ribose, galactose, sucrose, and lactose. It was also inhibited by cations such as $Hg^{++},\;Fe^{++},\;Cu^{++}\;and\;Pb^{++}$.

Effects of Butanol Fraction of Alisma canaliculatum with Vitamin E on Plasma Levels of Glucose and Lipid in Streptozotocin-Induced Diabetic Rats (택사 butanol 분획물과 vitamin E의 투여가 streptozotocin 유발 당뇨 흰쥐의 혈당 및 혈장 지질수준에 미치는 영향)

  • The effects of butanol (BuOH) fraction of Alisma canaliculatum (Ac) with vitamin E in streptozotocin (STZ)-induced diabetic rats were determined. Sprague-Dawley rats were divided into 5 groups: normal, STZ-control, and 3 diabetic-experimental groups. Diabetes mellitus was induced by injection of STZ (45 mg/kg) into the tail vein. The BuOH fraction of Ac and vitamin E were administrated orally in experimental rats for 21 days: Ac group (400 mg/kg), Ac-VE group (Ac 400 mg/kg & vitamin E 10 mg/kg), and VE group (vitamin E 10 mg/kg). The body weight losses were seen in all groups except normal, and the decrements in experimental groups were less than that in diabetic-control group. The plasma glucose levels were significantly lower in Ac group compared to STZ-control group on 21 day (p<0.05). The plasma level of insulin was slightly higher in AC-VE group than other diabetic groups. The plasma cholesterol levels of diabetic-experimental groups were significantly lower than those of STZ-control group on 14 day (p<0.05). ALT and AST activities of diabetic-experimental groups were significantly lower than that of STZ-control group (p<0.05). The results suggested that the BuOH fraction of Ac might possess hypoglycemic properties in STZ-induced diabetic rats and no synergistic effect of vitamin E was seen during the experimental period.

Effects of $H_2O$ Fraction of Alisma canaliculatum with Chromium on Blood Glucose Levels and Lipid Metabolism in Streptozotocin-Induced Diabetic Rats (택사 $H_2O$ 분획물과 Chromium의 투여가 Streptozotocin 유발 당뇨 흰쥐의 혈당 및 혈중 지질함량에 미치는 영향)

  • Hypoglycemic effects of $H_2O$ fraction of Alisma canaliculatum (Ac) with chromium (Cr) in streptozotocin (STZ)-induced diabetic rats were investigated. Sprague-Dawley rats were divided into five groups: normal, diabetic-control, and three diabetic experimental groups. Diabetes mellitus was induced by injection of STZ (45 mg/kg) into tail vein, and $H_2O$ fraction of Ac and Cr were administrated orally to experimental rats for 14 days: Ac group (1,000 mg/kg), Cr-Ac group (Ac 1,000 mg/kg & Cr 100 mg/kg), and Cr group (100 mg/kg). Body weight losses were observed in all groups except the normal group, while the losses in experimental groups were significantly less than in diabetic-control group. The plasma glucose levels were significantly lower in Cr, Ac-Cr, and Ac groups compared to STZ-control group. Plasma cholesterol and TG levels of Ac-Cr group were significantly lower than those of diabetic-control group. Plasma HDL-cholesterol levels of three experimental groups were not significant compared to diabetic-control group, while the level of Cr group decreased more than diabetic-control group. The results suggest that there was no synergistic effect in the treatment of $H_2O$ fraction of Ac in combination with Cr, an indication that $H_2O$ fraction of Ac with Cr might possess some hypoglycemic and hypotriglyceridemic properties in STZ-induced diabetic rats.
