A Case with Tyrosinemia Type I Detected by Neonatal Screening Test (신생아 대사이상 선별검사 이상으로 진단된 I형 타이로신혈증)

  • Tyrosinemia type I is an autosomal recessive inborn error of tyrosine metabolism that caused a mutation. Clinical symptoms include progressive liver damage with liver failure, coagulopathy, hypophosphataemic rickets, renal tubular dysfunction and a high risk of hepatocellular carcinoma. If left untreated, the affected infants may die from liver failure within the first year of life. PharmacoloIcal therapy with 2-(2-nitro-4-trifluoromethylbenzoyl)-1,3-cyclohexanedione (NTBC) has offered an effective therapeutic option in addition to dietary restriction of tyrosine and phenylalanine. As prognosis of tyrosinemia type I is improving with early diagnosis and early treatments, it meets the criteria for a condition that would benefit from newborn screening. We report a case of tyrosinemia type I diagnosed by newborn screening and successive biochemical analysis of plasma and urine, treated by dietary restriction and NTBC.

The Molecular Study of Phosphotyrosine Protein Phosphatase (PtpA) from Streptomyces coelicolor A(3)2 (방선균이 생산하는 인산화타이로신 단백질 포스파타아제의 분자생물학적 연구)

  • The cloning and expression of Phosphotyrosine Protein Phosphatase into E. coli provides important tools of understanding of its functions and signal transduction mechanisms. The abundant soluble protein of the Phosphotyrosine Protein Phosphatase A (PtpA) and the active site mutant PtpA(C9S) were produced using the expression vector pET26 in E. coli and pIJ6021 with the thiostrepton in S. lividans. The enzyme activity of both proteins extracted by Ni-NTA column had same results from the expression vector pET26 and pIJ6021. The enzyme activity of phosphatase was found in the protein of PtpA, but not in that of C9S. The western blot detected by penta His-tag antibody resulted in the inducer, thiostrepton was not a good trigger to induce a large amount of PtpA protein. The overexpression of both proteins had no significantly different effect on the A factor cascade related to the secondary metabolite and mycelium formation between PtpA and C9S. However, overproduction of PtpA protein using pIJ6021 in S. lividans brought about a dramatic decrease in the amount of phosphotyrosine proteins (p200, p90, and p65), but no significantly phenotypic variation in S. lividans. This indicates that PtpA has an important proteome role in signal transduction mechanism of producing massive amount of phosphotyrosine protein in Streptomyces sp.

Synthesis of Self-Assembled Peptide Nanoparticles Based on Dityrosine Covalent Bonds (다이타이로신 공유결합으로 자기조립된 펩타이드 나노입자의 합성)

  • In this study, a method of self-assembly of peptides based on irreversible covalent bonds was studied by mimicking a biological covalent bond, dityrosine bond. A tyrosine-rich short peptide monomer having the sequence of Tyr-Tyr-Leu-Tyr-Tyr (YYLYY) was selected to achieve a high-density of dityrosine bond. The peptide nanoparticles covalently self-assembled with dityrosine bonds were synthesized by one-step photo-crosslinking of a peptide using a ruthenium catalyst under visible light. The effect of the concentration of each component for the size of the peptide nanoparticle was studied using dynamic light scattering, UV-Vis spectroscopy, and transmission electron microscopy. As a result, the synthesis conditions for size of the peptide nanoparticles ranging from 130 nm to 350 nm were optimized.

Effect of $\beta$-Carotene on Enzymatic Browning of Chlorogenic Acid and Tyrosine (베타-카로틴이 클로로젠산과 타이로신의 효소적 갈변화에 미치는 영향 연구)

  • The browning rate of potato juice alone and potato juice with added carrot juice or water(1:1 by volume) were studied. As the amount of carrot juice added was increased, the browning rate proportionally decreased: from 1.86 of potato juice alone to 0.36 for the potato juice and carrot juice mixture(l:1). This inhibitory effect of carrot juice on browning led us to assume that $\beta$-carotene, the major constituent of carrot carotenoids, may be involved in impeding the browning reaction of potato juice. Therefore, model system of enzymatic browning, consisting of chi orogenic acid/tyrosine and $\beta$-carotene/carrot carotenoids, were studied. The results shows the browning rates of chlorogenic acid alone and chlorogenic acid with added $\beta$-carotene. The initial browning rate of chlorogenic acid was faster($\delta$L= 1.1/min.)than that of chlorogenic acid with added $\beta$-carotene($\delta$L=0.6/min.).

Activation of Phospholipase D2 through Phosphorylation of Tyrosine-470 in Antigen-stimulated Mast Cells

  • The mechanism of activation of phospholipase D2 (PLD2) remains undefined although mechanisms have been described for the activation of PLDI. By expression of mutated forms of haemaglutinnin-tagged PLD2 in a mast cell (RBL-2H3) line, we show that PLD2 is phosphorylated at tyrosines -11, -14, and -470 and that tyrosine-470 is critical for activation of PLD2 by antigen. Studies were performed with mutated-DNA constructs for haemaglutinnin-tagged PLD2 in which codons for tyrosine -11, -14, -165, and -470 were mutated to phenylalanine either individually or collectively. Transient expression of these constructs showed that mutation of tyrosine -11, -14, -470, or all tyrosines (all-mutated PLD2) suppressed antigen-induced tyrosine phosphorylation of PLD2 but only the tyrosine-470 mutant failed to be activated by antigen as assessed by in vitro assay of immunoprepitated PLD2 or by assay of PLD in intact cells. The critical role of tyrosine-470 was confirmed in studies with add-back mutants (phenylalanine back to tyrosine) of the all-mutated PLD. The findings provide the first description of a mechanism of activation of PLD2 in a physiological setting.

Preparation of Gold-Peptide Hybrid Nanoparticles and Its Applications in Catalytic Reduction of Methylene Blue (금-펩타이드 하이브리드 나노입자의 제조와 메틸렌 블루의 촉매 환원 응용)

  • In the present work, we studied a method for the synthesis of uniform gold-peptide hierarchical superstructures using tyrosine rich peptide, Tyr-Tyr-Leu-Tyr-Tyr (YYLYY). Peptide nanoparticles self-assembled by dityrosine bonds were synthesized through the photo-crosslinking reaction of the peptide, and gold-peptide hybrid nanoparticles were synthesized using biomineralization properties of tyrosine in a green synthetic manner. The synthesized gold-peptide hybrid nanoparticles were then characterized by transmission electron microscopy, scanning electron microscopy, dynamic light scattering, UV-vis spectroscopy, scanning transmission electron microscopy-energy dispersive X-ray spectroscopy, and X-ray diffraction. Furthermore, the catalytic activity of gold-peptide hybrid nanoparticles was confirmed by the reduction reaction of methylene blue where the catalytic reaction rate constant was 13.4 × 10-3 s-1.

Effects of Tyrosine Hydroxylase Levels by Lithium in Herbal Medicine (한약내의 리튬이 타이로신 가수분해효소에 미치는 영향에 대한 연구)

  • Lithium, a simple monovalent cation, is the mainstay in the treatment of manic-depressive illness, but despite extensive research, its mechanism of action remains to be elucidated. Because lithium requires chronic administration for therapeutic efficacy and because its beneficial effects last well beyond its discontinuation, it has been postulated that lithium may exert major effects at the genomic level. We have previously shown that Yungmijihwangwon (YM) increase TH protein expression through antioxidant activity and unknown effects in vivo. In the pressent study, we have sought to determine if lithium in herbal medicine also increases the expression of endogenous gene known to be regulated by AP-1 and have therefore investigated the effects of herbal medicine on tyrosine hydroxylase (TH) levels. Male mice were treated with LiCl and herbal medicine for 30 days, and TH levels and dopamine level were measured in striatum using immunoblotting and ECD-HPLC. Herbal medicine treatment resulted in statistically nonsignificant increase in TH levels in mouse striatum. The precise therapeutic relevance of these effects is presently unknown.

Antibiotic Uptake by Plants from Soil Applied with Antibiotic-Treated Animal Manure (가축분뇨 시용 토양에서 식물체의 축산용 항생물질 흡수양상)

  • Food contamination through antibiotic uptake by plants has been one of the major concerns regarding animal manure amendment to agricultural land. Antibiotic uptake by plants was tested with three veterinary antibiotics such as chlortetracycline (CTC), tylosin (TYL), and sulfamethazine (SMT) and three plants such as lettuce, tomato, and hairy vetch. Antibiotic-treated swine slurry was applied to a greenhouse soil before transplanting or sowing the plants. The treated antibiotic concentrations to the swine slurry were 22.9 mg CTC $L^{-1}$, 27.8 mg TYL $L^{-1}$, and 32.4 mg SMT $L^{-1}$. Lettuce was harvested on 64 days after transplanting and 94 days for tomato and hairy vetch. Concentrations of CTC were 3.4 ng $g^{-1}$ for lettuce and 0.7 ng $g^{-1}$ for tomato on a fresh weight basis. For TYL, 20.1 ng $g^{-1}$ from lettuce leaves and 3.0 ng $g^{-1}$ from whole hairy vetch were detected. Sulfamethazine showed greatest concentration among the three antibiotics, 63.3 ng $g^{-1}$ in lettuce leaves, 30.2 ng $g^{-1}$ in tomato fruits, and 20.9 ng $g^{-1}$ in hairy vetch. The results imply that antibiotic uptake by plants may be dependent on antibiotic type and plant type.
