Effects of Chitosan Treatment on Changes of Soyasaponin Contents in Soybean Sprouts (키토산 처리가 콩나물의 Soyasaponin 함량변화에 미치는 효과)

  • Elicitors are defined as substances that induce defense responses in plants, which include an increased synthesis of secondary metabolites. Saponin, one of the secondary metabolites, has various physiological effects such as anticancer, antioxidant, cholesterol-lowering activities, etc, in human. This study was carried out to find whether a treatment of soybean sprouts with chitosan as an elicitor, increases saponin contents. Saponin contents in soybean sprouts increased by the chitosan treatment during cultivation, reached the peak on the sixth day, and then decreased. A biosynthesis of group B soyasaponin appeared to be regulated differently. The content of soyasaponin I, a member of group B saponin, was the highest in 250 ppm chitosan-treated soybean sprouts, while the contents of soyasaponin II, III and IV were the highest in 1,000 ppm chitosan-treated soybean sprouts. The content of soyasaponin V changed little in soybean sprouts that had been treated with various concentration of chitosan.

Flavor Compounds and Physicochemical Properties of Low-fat Functional Sausages Manufactured with Chitosans during Refrigerated Storage (키토산을 첨가한 저지방 기능성 소시지의 향미 성분 분석과 냉장 저장 중의 이화학적인 특성)

  • The objective of this study was to evaluate the physico-chemical properties and flavor compounds of sausages with various levels and molecular weight (MWs) of chitosans, during storage at $4^{\circ}C$. Various MWs (Low: 1.5 kDa; Medium: $30{\sim}50$ kDa; High: 200 kDa) and two levels (0.3 and $0.6\%$) of chiosans were dissolved and measured the viscosity at $4^{\circ}C$, pH values were not affected (p>0.05) by either MWs or levels of chitosans. The addition or high MWs or chitosan into the pork salt soluble protein (SSP) increased the viscosity, whereas no differences were observed in low and medium MWs of chitosan. Textural profile analysis (TPA) was affected by the addition of medium or high MWs of chitosan. As a result, the addition of medium of chitosan increased the hardness, gumminess, chewiness, cohesiveness and springiness values, whereas increased level of chitosan didn't affect TPA values, except few cases. Approximately twenty-nine flavor compounds were identified in the low-fat and regular-fat sausages, however the addition of chitosans didn't impair the flavor composition of the sausages, These results indicated that the addition of chitosans didn't affect the flavor profiles, but affected the textural properties in the sausages, especially MWs higher than 30 kDa.

Studies on the Improvement of Shelf-life and Quality of Vacuum-Pckaged Seasoned Pork Meat by Added Chitosan During Storage (키토산 첨가 양념돈육의 저장성 및 품질 개선에 관한 연구)

  • This study was carried out to investigate the effect of chitosan on shelf-life and quality of vaccum-packaged seasoned pork. Chitosan was used 120 kDa and various concentration such as 0.05 - 1.0%. The total bacterial counts, pH, oxidation of lipid, surface color and water holding capacity of the sample were determined during storage periods. The total bacterial counts of seasoned pork without chitosan were increased the latter period of storage, but that of seasoned pork with chitosan was decreased such as inhibition of bacterial growth effectively. Effect on shelf-life of seasoned porks when added 0.100/0, 0.50% and 1.00% of chitosan, respectively were maintained pretty well during 10 days of storage. Content of TBARS in seasoned pork without chitosan was increased than that of seasoned pork with chitosan during period of storage. And also effect of self-life and inhibition of lipid oxidation were increased with following concentration of chitosan. The variation of pH was low and stable in seasoned pork with chitosan during periods of storage. The change of color such as lightness(L^*), redness(a^*), yellowness(b^*) in seasoned pork with chitosan was detected higher than that seasoned pork without chitosan. Water holding capacity of seasoned pork with low in the early period of storage. These colors and water holding capacity were very stable untill 10 days of storage.

Product Quality of Low-Fat Sausages Formulated with Two Levels of Chitosan (키토산의 함량을 달리하여 첨가한 저지방 소시지의 품질 특성)

  • Physicochemical and textural properties, and sensory evaluation of low-fat sausages (LFSs) manufactured with 0.3 and 0.6% chitosan were investigated and compared with those of regular-fat sausages (RFSs). Although the batter pH was not changed with the addition of chitosan (p>0.05), product pH was reduced (p>0.05) with the addition of chitosan up to 0.6%. Proximate composition and cooking loss (%) were not affected by the addition of chitosan. Expressible moisture 1r91ues (EM, %) of LESs were lower (p<0.05) than those of RFSs, but not affected by the addition of chitosan. The addition of chitosan in the sausage formulation became harder, springier and more cohesive (p<0.05), whereas, no differences were observed (p>0.05) in these values of sausages containing between 0.3 and 0.6%. Batter viscosity was not affected by the addition of chitosan, but LESs with or without chitosan had lower viscosity than RFSs due to high moisture (%). Sensory evaluation results showed that LFSs containing 0.3% chitosan had most parameters similar to those of low-fat control. These results indicated that chitosan at the level of 0.3% can be used for the manufacture of LFSs without quality defect.

Studies on the Improvement of Storage Property in Meat Sausage Using Chitosan- II Difference of Storage Property by Molecular Weight of Chitosan (키토산 첨가에 의한 축육 소시지의 보존성 개선에 관한 연구- II 키토산의 분자량에 따른 보존성의 차이)

  • This study was examined for antibacterial effect of various molecular weight of chitosan against spoilage bacteria in emulsion sausage. Four different kinds of chitosan, molecular weights (M.W.) of 1 kDa, 5 kDa, 30 kDa and 120 kDa, wee used. The more molecular weight of chitosan is high, the more storage property of sausage is good during storage at $30^{\circ}C$. Storage properties of sausages between added 0.5% of M.W. 120 kDa chitosan and 150 ppm of sodium nitrite were about the same. Effect of growth-inhibitory of spoilage bacteria was not detected 0.2% of M.W. 1kDa chitosan 0.2% of M.W. 5kDa chitosan have growth-inhibitory effect over 80% against only 3 strains among bacteria isolated from spoiled emulsion sausage. But, 0.2% of M.W. 30 kDa chitosan have growth-inhibitory effect of 80% against all strains of bacteria related to spoilage of emulsion sausage, except S. typhimurium, Especially, 0.2% of M.W. 120 kDa chitosan inhibited over 80% growth against all strains used in this study. The antibacterial activity was increased with their molecular weight.

A Study on Synergisitic Effect of Chitosan and Sorbic Acid on Growth Inhibition of Escherichia coli O517:H7 and Staphylococcus aureus (E. coli O517:H7 과 Staphylococcus aureus의 증식억제에 대한 키토산과 소르빈산의 상승효과에 관한 연구)

  • This study was performed to investigate the synergistic effect of chitosan and sorbic acid as a new food preservative. So it was performed to investigate inhibitory effect on growh of E. coli 0157:H7, gram negative pathogenic food borne disease bacteria and of S. aureus, gram positive food borne disease bacteria in chitosan, sorbic acid and combination of chitosan and sorbic acid. Minimun Inhibitory Concentration (MIC) of chitosan in E. coli 0157:H7 was 500 ppm at pH 5.0, 250 ppm at pH 5.5, 500 ppm at pH 6.0, and 2000 ppm at pH 6.5, while in Staph. aureus 31.25 ppm at pH 5.0 and 62. 5 ppm at more than pH 5.5. also, MIC of sorbic acid in E. coli 0157:H7 was 500 ppm at pH 5.0, 1500 ppm at pH 5.5, and 2000 ppm at more than pH 6.0, while in Staph. aureus 1500 ppm at pH 5.0 and more than 2000 ppm at more than pH 5.5. Due to the effect of pH in E. coli 0157:H7, MIC of combined chitosan and sorbic acid was 500 ppm of chitosan with 500 ppm of sorbic acid at pH 6.5, but 250 ppm of chitosan with 31.3 ppm of sorbic acid at pH 5.0. In Staph. aureus, there was great effect of chitosan, but neither effect of pH nor sorbic acid. When E. coli 0157:H7 were treated with 500 ppm of chitosan with 500 ppm of sorbic acid and 250 ppm of chitosan with 250 ppm of sorbic acid at pH 6.5, they were inhibited. But, they were increased at the initial concentration of bacteria at 1000 ppm of chitosan in 18 hours, at 500 ppm of chitosan in 36 hours. There was no effect of growth inhibition with sorbic acid but great effect with chitosan on Staph. aureus. The correl~tions between MICs of chitosan and sorbic acid in E. coli 0157:H7 accoding to pH were higher than those in Staph. aureus. R values in E. coli 0157:H7 were 0.95 (p<0.01), 0.99 (p<0.01), 0.97 (p<0.01), and 0.99 (p<0.01) at pH 6.5, 6.0, 5.5, and 5.0 respectively. The synergistic effect of chitosan and sorbic acid in E. coli 0157:H7 could be confirmed from the result of this experiment. Therefore, it was expected that the food preservation would increase or maintain by using sorble acid together with chitosan, natural food additive that did no harm to human body.

Utilization of Chitin Prepared from the Shellfish Crust 2. Effect of Chitosan Film Packing on Quality of Lightly-Salted and Dried Horse Mackerel (갑각류부산물을 이용한 키틴의 제조 및 이용에 관한 연구 2. 키토산필름포장이 반염건(半鹽乾) 전갱이의 가공 및 저장 중 품질에 미치는 효과)

  • The preservative effect of chitosan film packing on quality of lightly-salted and dried horse mackerel was studied. In preparation of chitosan film, blue crab shell chitosan was dissolved in dilute acetic acid$(1.0\%,\;v/v)$, filtered, and spreaded on plastic plate and dried at $50\pm2^{\circ}C$. The chitosan film thus obtained was neutralized with 1.0N NaOH for 2 hrs and dried at room temperature after washing several times with distilled water. The lightly-salted and dried horse mackerel product was prepared by drying for 4 hrs at $40\pm2^{\circ}C$ in hot air dryer after packing with the chitosan film. During storage at $5.0\pm0.5^{\circ}C$, moisture content of the product was higher than that of the reference, but contents of VBN(volatile basic nitrogen) , amino nitrogen, and TMA of the product on dry basis were lower than those of the reference. Viable cell count, TBA value, and peroxide value of the product were also lower than those of the reference. Judging from the result of sensory evaluation, the chitosan film packing in the storage of lightly-salted and dried horse mackerel was remarkably elongated shelf-life of the product. From the results of chemical and sensory evaluation, it was concluded that chitosan film packing was an effective method for retaining the quality of lightly-salted and dried horse mackerel.
