Recent Progress in Mussel-inspired Catechol-conjugated Chitosan Hemostats (홍합 모사 카테콜기가 도입된 키토산 지혈제 연구 동향)

  • Since it was reported that the unusual amino acid DOPA in synergy with lysine and histidine residues found in mussel adhesive proteins plays a pivotal role in mussel adhesion in underwater environments, there has been a burgeoning development of various catecholamines-based adhesives for biomedical applications. Among these, catechol-conjugated chitosan, containing catecholamine, featuring multiple catechol groups within its aminerich chitosan backbone, has found versatile utility in fields, such as tissue adhesion, wound dressing, tissue healing, hemostats, drug delivery systems, and tissue engineering scaffolds. Significantly, chitosan-catechol is a mussel-inspired material approved by both US Food and Drug Administration (FDA) and KR Ministry of Food and Drug Safety (MFDS) for its effectiveness in hemostasis. This review focuses on 1) general aspects of catechol-conjugated chitosan, highlighting catechol group integration into chitosan backbones, 2) examination of proposed mechanisms of hemostasis, and 3) exploration of diverse physical forms, including solution, hydrogels, patches, and thin films with practical applications inapplicable to hemostasis.

Effects of Dietary Chitosan Supplementation with Chitosan Feeding Levels and Feeding Forms on Growth Performance and Carcass Characteristics of Thigh Muscular in Broiler (키토산의 급여량과 급여 방법에 따른 육계의 성장 및 닭 다리육의 품질에 미치는 영향)

  • This study was conducted to determine the effects of dietary supplementation with chitosan feeding levels and feeding forms on the performance, proximate composition, pH, thiobarbituric acid reactive substance (TBARS), water holding capacity (WHC), shear force, meat color, and fatty acid of chicken thigh meat. Two hundred broilers (Arbor Acre Broiler, male) were randomly assigned to five groups and were fed for five weeks and slaughtered. Thigh muscle was evaluated in this experiment. The amounts of proximate composition, crude fat of control and T3 for the chitosan-treated groups were significantly higher as compared with T2 (P<0.05), but no significant effects were detected on moisture, crude protein, and crude ash. By comparison, pH was significantly increased when chitosan was included at 2% into diet or more than 1% into drinking water. TBARS was significantly lower at chitosan treated groups it was decreased with increasing chitosan level in the diets (P<0.05). Therefore, chitosan had the possibility to improve shelf life of chicken meat. Higher chitosan levels and feeding additive increased high density lipoprotein cholesterol (HDL-C) and reduced total cholesterol and low density lipoprotein cholesterol (LDL-C) (P<0.05). $L^*$ and $a^*$ of chitosan treated group was higher than the control. As the dietary chitosan levels increase, the composition of palmitic acid and oleic acid levels was increased, however, those but the rates of linoleic acid and arachidonic acid were decreased. It is concluded that dietary chitosan has a positive effect on increasing HDL-C and oleic acid and decreasing total-C, LDL-C and TBARS values. Therefore, the treatment with the most significant effects in the current study was the high level of chitosan.

Product Quality and Extension of Shelf-life of Low-fat Functional Sausages Manufactured with Sodium Lactate and Chitosans during Refrigerated Storage (젖산나트륨과 키토산을 첨가한 저지방 기능성 소시지의 냉장 저장 중의 품질 및 저장성 증진 효과)

  • This study was peformed to evaluate physico-chemical and textural properties, and shelf-life effect of low-fat functional sausages(LFFS) manufactured with sodium lactate(SL), lac color and various molecular weights of chitosans(low=1.5 kDa, medium=30∼40 kDa and high=200 kDa) during storage at 4$^{\circ}C$ for 8 weeks. LFFS had a pH range of 6.39∼6.50, 76∼78% moisture, <2% fat, 14∼15% protein. The combination of SL and low molecular weight(MW) of chitosan improved water holding capacity(WHC), however those of SL and medium MW of chitosan reduced WHC. Vacuum purge(VP) reduced with increased MW of chitosans during refrigerated storage. The addition of chitosans reduced the lightness and yellowness, but increased the redness values, which was comparable to the sodium nitrite concentration between 75 and 150 ppm. LFFS containing SL and medium MW of chitosan increased most texture profile analysis(TPA) values, as compared to controls with 75 and 150 ppm. The addition of SL in LFFS retarded the microbial growth for Listeria monocytogenes, however no synergistic effect with the addition of chitosans were observed. E coli O157:H7 and Salmonella typhimurium reduced during refrigerated storage, regardless of SL and chitosan treatments. Increased storage time increased values for VP, yellowness and textural properties. These results indicated that the combination of SL and various MW of chitosans affected the functional and textural properties, and inhibited the microbial growth for LM effectively. In addition, 0.5% lac color as a replacer for sodium nitrite improved the color development, resulting in similar hunter color values, which was comparable to the sodium nitrite concentration between 75 and 150 ppm.

Effects of Chitosan with Different Molecular Weight and Nitrite Addition on the Residual Nitrite Contents and Self-life of Emulsified Sausage during Cold Storage (분자량이 다른 키토산과 아질산염 첨가가 유화형 소시지의 냉장 저장 중 아질산염잔존량 및 저장성에 미치는 영향)

  • The objective of this study was to determine the effects of residual nitrite contents, chitosan with different molecular weight and nitrite addition on emulsified sausage during cold storage. Six types of sausages were evaluated: control, 0.5% 50 kDa chitosan (T1), 0.5% 200 kDa chitosan (T2), 150 ppm nitrite (T3), 0.5% 50 kDa chitosan+150 ppm nitrite (T4), and 0.5% 200 kDa chitosan+150 ppm nitrite (T5). Each type of sausage was tested in triplicate and assigned to one of four storage periods: 0, 10, 20 and 30 days. As the storage time increased, the presence of chitosan and nitrite resulted in decreased residual nitrite value and increased pH (in control and T2), TBARS (thiobarbituric acid reactive substance) values, and total plate counts (TPC). Values for pH, TBARS, residual nitrite and total plate counts decreased significantly in response to the addition of chitosan and nitrite relative to the control (p<0.05). T5 was redder than the control (higher CIE$a^*$) at 30 d; however, no difference in the CIE $L^*$ and $b^*$ values was observed. T5 was significantly (p<0.05) more effective at delaying lipid oxidation when compared to the other treatment groups. T5 presented TPC that was significantly lower (p<0.05) than the other groups after three days of storage. In addition, the use of chitosan and nitrite in combination had much better antioxidant and antimicrobial effectiveness than other treatment groups. In conclusion, this study demonstrates that the addition of 0.5% 200 kDa chitosan and 150 ppm nitrite in combination with emulsified sausages tended to improve antioxidative and antimicrobial effects during storage when compared to other treatment groups.

키토산 분자량 변화에 따른 수중의 여러 중금속들의 제거에 관한 연구

  • Autoclaving처리를 하지 않은 키토산과 autoclaving처리한 키토산과의 중금속 흡착실험을 해 본 결과 다음과 같은 결과를 도출 할 수 있었다. 1) 먼저 중금속간의 흡착능을 고찰하기 위하여 Langmuir와 Freundlich 흉착등온식에 적용시켜 본 결과 Freundlich 흡착등온식 보다는 Langmuir 흡착등온식이 보다 적합한 것으로 나타났다. 2) 시간에 따라서 autoclaving 처리한 키토산의 중금속 제거 가능성과 그 효율을 검토하기 위하여 각 중금속간의 Langmuir 흡착등온식을 이용하여 $q_{max}$를 나타냈다. 15 min > 60 min > 0 min 순으로 모든 중금속 제거 실험에서 15 min 동안 autoclaving 처리한 키토산의 중금속 흡착량이 가장 높은 것으로 나왔다. 그러므로 기존의 키토산 보다는 15 min동안 autoclaving 처리한 키토산이 중금속 제거에 더 좋은 흡착제로써 역할을 할 수 있을 것으로 판단된다. 3) 키토산을 이용한 중금속 제거에서는 $Pb^{2+}$ > $Cd^{2+}$ > $Cu^{2+}$ > $Cr^{3+}$순서로 제거가 되었다. 여러 연구자들의 실험 결과를 종합해 볼 때 Pb$^{2+}$가 중금속 중에 제거가 잘 된다는 연구 결과가 많이 발표 되었으며, $Cd^{2+}$, $Cu^{2+}$, $Cr^{3+}$의 경우에는 흡착제의 종류에 따라서 제거되는 순서가 다르다는 연구 결과들이 보고 되고 있다. 그러나 어떠한 이유로 중금속의 제거에 차이가 있는지에 대해서는 명확한 결론이 내려져 있지 않는 실정이다. 이러한 중금속간의 경쟁적인 관계에 대해 더 많은 세밀한 연구가 이루어져야 할 것 같다.는 0.52mg/$\ell$~0.99mg/$\ell$~의 범위이었다. 신천에 금호강물을 혼합한 이후에도 부유물질, 생화학적산소요구량, 암모니아태 질소, 총인 등의 농도가 개선되지 않았다. 즉 금호강물의 혼합은 신천수질환경사업소에서 배출되는 방류수에 함유되어 있을 2차 오염물질의 희석이라는 이점외의 수질개선효과는 확인되지 않았다.l years and a new type of transfer crane has been developed. Application of Various Hydrophobic Moiety-modified Chitosan Nanoparticle as a Drug Delivery Carrier (다양한 소수성 물질이 개질된 키토산 나노입자의 약물전달체로서 응용성 고찰)

  • Natural polymer chitosan has been widely applied to medical fields due to its biochemical activities such as anticancer, antibacterial and lowering cholesterol in addition to biocompatibility and biodegradability. Currently, researches are being actively conducted to develop various drug-encapsulated chitosan nanoparticles for curing different diseases by applying chitosan to a drug delivery system. The free amine ($-NH_2$) group present in chitosan can bind to various hydrophobic groups by physical and chemical modification and the chitosan with hydrophobic groups can form shell-core nanoparticles by self-assembly when dispersed in water. In addition, an insoluble drug can increase the solubility against water when it was encapsulated in the core of chitosan nanoparticles. Also, the therapy effect can be maximized by minimizing side effects of drugs such as proteins, anticancer drugs and vaccines when they were encapsulated in the core of chitosan nanoparticles. Moreover, it is possible to control the particle size and release rate according to the hydrophobic group introduced to chitosan, so that it can be applied to a wide range of medical fields. The purpose of this review is to discuss the preparation and property of chitosan nanoparticles modified with various hydrophobic groups, and the application to drug delivery systems according to their property.

Thermomechanical Characteristics of Poly(vinyl alcohol)/Chitosan Films and Its Blend Hydrogels (폴리(비닐 알코올)/키토산 블렌드와 블렌드 수화젤의 열특성)

  • Films of poly(vinyl alcohol)(PVA)/chitosan blends and its blend hydrogels were prepared by the solution casting method. The state of miscibility of the blends and blend hydrogels were examined over the entire composition range by differential scanning carorimetry (DSC), thermogravimetry (TGA), and dynamic mechanical analysis (DMA). DSC analysis shows the depression of melting point of PVA in the blends and the decrease of crystallization temperature of PVA in the blends were observed with increasing chitosan content in the blends. TGA analysis indicates that chitosan was thermally more stable than PVA and the thermal stability of PVA in the blends was higher than that of pure PVA, due to some interactions between two component polymers in the blend. The glass transition temperature $(T_g)$ of the chitosan and of PVA, measured by DMA, were at 160 and $90^{\circ}C$, respectively. The $T_g$ of the blends was changed with the content of chitosan in the blends. The results of thermal and viscoelastic analysis indicate some miscibility between component polymers in the blend exists. Moisture and cross linking in the blend and blend hydrogel, which strongly change thermal and physical properties of hydrophilic polymers, affected the miscibility of chitosan and PVA to a small extent.

Effects of Dietary Liquid Chitosan on the Performance of Broiler Chickens (액상 키토산의 급여가 육계의 생산성에 미치는 영향)

  • This study were conducted to evaluate effects of drinking concentrated liquid chitosan on performance of broiler. A total of 240 broiler chicks at 7 days of age were fed the commercial diet and water until 47 days of age, drinking water divided into three treatments, 0 ppm (control), 200 ppm (200) and 400 ppm (400) of concentrated liquid chitosan. The body weight gain and feed conversion ratio were not different by drinking concentrated liquid chitosan until 28 days of age, but body weight gain and feed conversion ratio were improve by drinking concentrated liquid chitosan over 28 days of age. Both body weight gain and feed conversion ratio were tend to improve and significantly improved (p<0.05) in 200 and 400 than control at 29 to 35 days of age, respectively. While body weight gain was tended to improve and significantly improved (p<0.05) in 400 and 200 than control at 36 to 47 days of age, respectively. There was a decrease in the microflora population of E coli. in the cecum contents and feaces of broilers by drinking concentrated liquid chitosan. This effect is higher in 400 than 200. The digestibility of dry matter and crude fat of feed were tend to increased in broiler by drinking concentrated liquid chitosan. Digestibility of crude protein of feed of broiler in 200 and 400 was significantly improved (p<0.05) as compared with those in control. These results indicated that when broiler chicks drinking concentrated liquid chitosan of 400 ppm, the improvement of the body weight gain, feed conversion ratio and feed nutrients digestibility, decreasing of microflora population of both E coli. and Salmonella in the cecum contents and feaces.

Studies on the Mass-production System for Making Biodegradable Film Based on Chitosan/gelatin Blend (키토산/젤라틴 블랜드 폴리머를 이용한 생분해성 필름의 대량생산 시스템에 관한 기초 연구)

  • To mass-product useful biopolymer films, chitosan/gelatin blend films were prepared by solution casting method. Effect of mixing ratio, tensile strength(TS), elongation(${\Delta}E$) at break, total color difference(E), water vapor permeability(WVP) and oxygen permeability(OP) on chitosan/gelatin blend films properties were investigated. TS, ${\Delta}E$, E, WVP and OP values of chitosan/gelatin blend films were 43.43-38.30 MPa, 9.02-15.09%, 1.28-3.81, $0.8420-0.9673ng{\cdot}m/m^2{\cdot}s{\cdot}Pa$ and $1.5472{\times}10^{-7}-1.5424{\times}10^{-7}mL{\cdot}{\mu}m/m^2{\cdot}s{\cdot}Pa$, respectively. TS of the blend films decreased, while E and E of the blend films increased with increasing chitosan content. WVP and OP of the blend films did not show any significant relationship with mixing ratio and thickness of the blend films. OP of the blend films were lower than those of low density polyethylene and oriented polypropylene.

Preparation of Chitosan-coated Magnetite Nanoparticles (키토산이 피복된 나노 크기의 자성체 분말 제조)

  • Magnetic nanoparticles can be used for a variety of biomedical applications. They can be used in the targeted delivery of therapeutic agents in vivo, in the hyperthermic treatment of cancers. in magnetic resonance (MR) imaging as contrast agents and in the biomagnetic separations of biomolecules. We have synthesized magnetite $(Fe_3O_4)$ nanoparticles using chemical coprecipitation technique with sodium oleate as surfactant. Nanoparticle size can be varied from 2 to 8nm by controlling the sodium oleate concentration. Magnetite phase nanoparticles could be observed from X-ray diffraction. Magnetic colloid suspensions containing particles with sodium oleate and chitosan have been prepared. Nanoparticles, both oleate-coated and chitosan-coated, have been characterized by several techniques. Atomic farce microscope (AFM) was used to image the coated nanoparticles. Magnetic hysteresis measurement were performed using a superconducting quantum interference device (SQUID) magnetometer at room temperature to investigate the magnetic properties of the magnetite nanoparticles. The SQUID measurements revealed superparamagnetism of nanoparticles.
