Purification and Characterization of Anti-complementary Polysaccharide from Phellinus linteus Mycelia (상황버섯(Phellinus linteus) 균사체로부터 항보체 활성 다당류의 정제 및 특성)

  • We have isolated an anti-complementary polysaccharide from the hot water extracts of Phellinus linteus mycelia. Anti-complementary polysaccharide, PL-5-IIIa, was purified by ultrafiltration, gel permeation chromatography using Sepharose CL-4B. GPC (Sepharose CL-4B) and its homogenicity was demonstrated by HPLC. Using gel permeation chromatography with standard dextrans, its molecular weight was determined as about 800,000 dalton. The purified PL-5-IIIa was identified as a protein bound polysaccharide comprising of 29.6% protein and 64.2% carbohydrate which was composed of fucose(15.8%), galactose(43.1%) and mannose(40.6%).

  • The putative endochitinase gene, yheB of Escherichia coli K-12 is not expressed under lab culture conditions. The endochitinase gene was amplified by PCR and subcloned into pET28c vector and pQE9 vector, respectively. The endochitinase produced in E. coli harboring pET28c containing yheB or pQE9 vector containing yheE was partly released into the growth medium. The overproduced endochitinase was partially purified by His affinity column chromatography and DE-52 column chromatography. The apparent molecular weight of the endochitinase determined by SDS-polyacrylamide gel electrophoresis was about 97,000. The purified E. coli endochitinase showed maximal chitinolytic activity at pH 6 and $40^{\circ}C$.

  • The textural properties of cooked rice and the molecular weight distribution of amyloses of three rice cultivars different in cooking quality were investigated by the method involving rheometry and gel chromatography he tested rice cultivals were Chunmabyeo (short grain), Yongmunbyeo (medium grain) and Mahatma (long grain) The cooked rice prepared with Mahatma was higher in hardness and lower in adhesiveness than those prepared with the other two cultivars. The molecular wright distribution of amylose from Mahatma was found to be higher than those from the other cultivars.

  • The retention mechanism and separation of various aromatic sulfonic and carboxylic acids on reversed-phase liquid chromatographic column were studied in the mobile phase containing dodecyltrimethylammonium bromide (DTAB). The retention mechanism was found to be followed the ion-interaction model where the DTAB occupies a primary layer at the stationary phase while the sample anions and other co-anions in the system compete for forming the secondary layer. The capacity factors of samples were influenced by the several factors such as pH, concentration of various organic solvents, co-anions in the mobile phase and functional groups in sample molecules. Some mixtures of organic samples were attempted to separate under optimum condition.

  • As the surfactants that were used in micellar reaction, emulsion polymerization and phase-transfer reaction etc. have the problems, the cleavable surfactant that was converted to inactive compound after such as the reaction was synthesized to above 90% yield. And this surfactant and intermediates were separated through thin layer chromatography and column chromatatography and their molecular structures were confirmed from IR. $^{1}H$-NMR and elementary analysis spectra. And its surface-active properties and acid hydrolysis will be serialized in II.

  • An ion-association chromatography was applied for the separation and determination of individual rare earth elements (REE) contained in mineral monazite. Prior to the determination, the group separation of REE was achieved by a cation exchange column of Dowex 5OW-X8 resin. The quantitative recovery of REE by the resin column, free from coexisting elements in monazite, was confirmed with radioactive tracers as well as with ICP-MS. Individual REE at ppm level was separated on reversed-phase column ($\mu$-Bondapak $C_{18}$) using gradient elution from 0.05 to 0.3 M $\alpha$-hydroxyisobutyric acid at pH 4.6. The individual REE was detected at 546 nm following post-column reaction with PAR (4-(2-pyridylazo)-resorcinol monosodium salt).

  • This research was conducted to analyze the effects of temperature on coking characteristics of kerosene. The kerosene was heated to 600 K, 700 K, and 800 K, and the cooled samples were collected. The used copper tubes were replaced according to the temperature conditions. The liquid and copper specimens were analyzed by gas chromatography-mass spectrometry and scanning electron microscopy equipped with an energy dispersive x-ray spectrometer, respectively. The results of the analysis confirmed that a carbon deposit was formed from the coking of fuel on the inner surface of the copper specimen at a relatively high temperature (800 K) of the copper tube.

  • In fields of operating or handling a hydrogen isotope facility, and of the technology for nuclear fusion source management, gas chromatography has been used as one of the practical techniques lot separation and enrichment of hydrogen isotopic gases including tritium. Chromatographic separation experiments of the hydrogen isotope mixture (hydrogen, deuterium and tritium) were carried out by use of a commercially available gas chromatograph. An aliquot of gas sample was injected by a specially designed vacuum sampler into the stream of inert carrier gas which went through the separation column under liquid nitrogen temperature. The complete separation of hydrogen isotopic molecules was observed with an alumina adsorbent partially deactivated by coating with 10% manganese chlorine. In addition, fairly good separation conditions were obtained without any appearance of nuclear spin isomers with shorter retention time, which would be available for the practical applications of the hydrogen isotope separation and enrichment.

Purification of Streptomyces Phospholipase D by Immunoaffinity Chromatoghraphy using Peptide Antibodies (Streptomyces phospholipase D의 정제를 위한 면역친화 크로마토그래피의 개발)

  • An immunoaffinity chromatography for the specific binding of Streptomyces somaliensis phospholipase D (PLD) that is considered as an industrially potential enzyme was developed. By using the protein structure prediction programs and the X-ray crystal structure of a Streptomyces PLD, 5 different epitopes with high antigenicity that are predicted to locate on the surface of the S. somaliensis PLD were selected and then synthesized for the preparation of antipeptide antibodies. Each purified rabbit IgG was coupled with NHS-activated Sepharose to prepare the immunoaffinity resins. After one-step purification of the culture concentrate on the antipeptide IgG-coupled Sepharose column, SDS-PAGE and the Western blot analysis of the purified samples showed that purification of PLD on the affinity columns was satisfactory, indicating that the peptide design using the structural information of Streptomyces PLDs was rational. However, the purified PLD in the solution aggregated rapidly, which resulted in poor specific activity and low purification yield.

  • The simultaneous analysis of alkylphenols, chlorophenols and bisphenol A in biota samples was performed by gas chromatography-mass spectrometry-selected ion monitoring mode. The phenols were extracted from sample with organic solvent and Forisil and Silica columns for clean-up procedure were compared. Recovery studies were performed at 1-ppm level of phenols added to each biota sample. Their recoveries ranged between 83 and 116% with coefficient of variations of 2.4-11.9%. To improve the detection limits of phenols, trimethylsilyl (TMS) derivatization was applied. The gas chromatographic properties of free phenols and TMS derivatized phenols were also investigated.
