Development of Analytic Methods for Veterinary Drug Residue in Animal Products by Liquid Chromatography-Mass Spectrometry (축산물 중 잔류 성장보조제 분석을 위한 액체크로마토그라피-질량분석법 개발 및 적용)

  • In this study, an analytical method was developed for estradiol-17��, testosterone, and progesterone, which are growth promoters as veterinary drug residues in animal food. The analytes were separated using liquid chromatography, and was injected into a mass spectrometer through the electrospray ionization(ESI) process and detected in multiple reaction monitoring(MRM) mode. As the method was validated by CODEX CAC/GL 71-2009 guideline, it met the acceptable. The analysis of beef, pork, and chicken distributed in Korea was conducted with an established method to confirm the applicability of the actual sample. It was confirmed that the developed method can be quickly and reliably in the analysis for the growth promoters identified in domestic distributed livestock products. Through subsequent research, a highly utilized analysis method will be completed if the number of growth promoters is expanded based on the method and the simultaneous analytical method is established by including all of it.

Isolation of Angiotensin Converting Enzyme Inhibitors from Pig Blood (돼지혈액으로부터 항고혈압물질의 분리)

  • Angiotensin converting enzyme (ACE) inhibitors were isolated and purified from pig blood plasma. Pig blood plasma was obtained after removing blood cells by centrfugation, followed by the addition of anticoagulant to whole pig blood. To precipitate plasma proteins, pig blood plasma was treated with 4% trichloroacetic acid (TCA) as a final concentration. ACE inhibitors were isolated from plasma protein hydrolysates and TCA supernatant, using ultrafiltration, gel permeation chromatography, and reverse-phase high pressure liquid chromatography. ACE inhibitors isolated from plasma hydrolysates and TCA supernatant had $IC_{50}$ values of $23\;{\mu}M$ and $2\;{\mu}M$, pentapeptide, respectively.

Effects of Germination on Antinutritional Oligosaccharides of Mung Beans (녹두발아시 항영양 과당류의 변화)

  • To examine changes of antinutritional oligosaccharides, particularly raffinose and stachyose, of mung beans during germination, mung beans (Phaseolus aureus) were germinated at $25^{\circ}C$ for 7 days after soaking for 15 hr. The 80% ethanol extracts of mung bean sprouts were analyzed by Kieselgel thin-layer chromatography using a solvent system, ethylacetate, acetic acid and water (3:1:1, v/v). The moisture level was increased greatly after soaking followed by stabilization along to the 3rd day of germination. The level was increased slightly on the 4th day and thereafter stabilized. Antinutritional oligosaccharides, raffinose and stachyose was diminished significantly on a thin-layer chromatogricadtly on the 1st and 2nd day of germination, and disappeared almost thereafter.

  • The study was attempted to isolate taurine from the oyster shucking juice known as one of the by-products of oyster processing using ion exchange column chromatography. Three hundred grams of the oyster shucking juice were loaded onto a column packed with 300 ml of the Dowex 50W $H^+$ form. And taurine-rich fractions were further purified in columns packed with 150 ml of Dowex 2 OH form and the 150 ml of Amberite IRA-410 OH form consecutively. The purity and the yield of taurine recovered from the oyster shucking juice by this method were 94.7% and 84.8%, respectively.

  • The fatty acid composition of thiocyanate utilizing Bacillus brevis isolated from carbonization wastewater was determined by Gas Chromatography and the results were analyzed. In addition to the saturated and unsaturated straight chain fatty acids this B. brevis strain contained a hydroxy fatty acid. The hydroxy fatty acids in general are shown to be interesting chemotaxonomic markers of bacteria. Cyclopropane fatty acids are totally absent in this strain. A comparison of the fatty acid composition of this strain with B-33 and B-34 strains of Bacillus brevis shows that there are deviations among these strains. The deviation in Bacillus brevis could be due to the stress effect of thiocyanate. This result supports that fatty acid synthesis depends highly on the environment.

  • 발파후에 2차연소 또는 폭발(이하, 2차연소라 한다. )이 일어났다는 사실은 폭약이 폭발후에 어떤 가연성가스가 발생하고 그 가연성가스가 잔존하는 폭발열 또는 기타의 점화원에 의해 연소되었음을 의미한다. 폭약이 폭발하였을 때, 발생 가능한 가연성물질은 유리탄소, 일산화탄소, 수소 등으로 추정할 수 있는데 실험결과에서는 가연성물질의 주성분이 수소인 것으로 나타났다. 본 연구에서는 에멀존계 함수폭약이 산소평형, 알루미늄함량, 알루미늄형태와 크기 그리고 포장지의 두께에 따라 수소가 발생되는 양을 가스크로마토그라피를 이용하여 측정하였다. 상기의 열거한 요인들은 모두 수소발생량과 관계가 있는데, 이중에서도 가장 중요한 요인은 산소평형과 알루미늄의 함량인 것으로 나타났다. 한 예로 알루미늄이 15%가 포함되고 산소평형이 -10인 에멀존계 함수폭약은 폭발후에 19.4%의 수소를 함유하고 있는 후가스를 발생시켰으며 이 가스를 포집하여 공기중에 방출시키면서 성냥불을 가까이 하였더니 연소가 되었다. 따라서 에너지를 높이기 위하여 알루미늄의 함량을 높이고 산소평형을 지나치게 마이너스로 설계한다면, 2차연소는 언제든지 발생할 가능성이 있다고 판단된다. 알루미늄의 함량을 가능한 적게, 산소평형을 가능한 0에 가깝게 설계해야 만이 2차연소 현상을 방지할 수 있을 것이며 ㄸ한 최적의 설계뿐만이 아니라 정확한 제조와 품질검사도 2차연소 현상을 방지하는데 중요한 몫을 할 것으로 판단된다.

  • Less-volatile organic components in leaves from aromatic tobaccos of different varieties, both Oriental and Korean types were isolated and concentrated using a simple apparatus with fewer manipulations. Each less-volatile concentrate was then subjected to spectrophotometric recording in the visible range, to thin-layer chromatographic group separation, and high-performance liquid chromatographic profile analysis. The methods allow detection of significant quantitative differences in visible absorption spectra, TLC patterns, and high resolution HPLC profiles among varieties.

Production of T-2 Toxin and Its Metabolites by Fusarium sporotrichioides Isolates from the Corn Producing Area in Korea (우리나라 옥수수산지에서 분리한 Fusarium sporotrichioides 균주들에 의한 T-2 독소 및 관련 대사물의 생성)

  • Four isolates of Fusarium sporotrichioides obtained from the corn producing area were tested for their toxicities by feeding the crude cultures to rats. Three out of four isolates were highly toxic and killed all rats within 3-4 days after feeding. The chemical analyses of toxic cultures by thin layer chromatography and gas chromatography-mass spectrometry revealed that two isolates from Jeongsun district produced T-2 toxin and its related trichothecenes. This is the first report that F. sporotrichioides isolates produce T-2 toxin in Korea.
