Somatic Embryogenesis in Immature Zygotic Embryo Cultures of Korean Soybean (Glycine max L.) Culitivars and Effect of 2,4-Dichlorophenoxyacetic Acid on Somatic Embryo Morphology (한국 품종 대두(Glycine max L) 미숙배로부터 체세포배발생과 배지의 2,4-Dichlorophenoxyacetic Acid 농도가 체세포배의 형태에 미치는 영향)

  • Immature zygotic embryos of five Korean soybean cultivars cultured on Murashige and Skoog's (MS) medium supplemented with various concentrations of 2,4-Dichlorophenoxyacetic acid (2,4-D) produced somatic embryos without forming an intervening callus. The highest frequency (up to 90%) of somatic embryo formation was obtained when zygotic embryos were cultlued on medium containing 1 to 2 mga 2, 0-D in four cultivars. However the frequency was highly variable to the cultivars. Transversely sliced primary somatic embryo halves were also capable of forming secondary embryos at frequencies of up to 70% when cultured on medium containing 0.1 to 1 mg/L 2,4-D. Somatic embryos formed on zygotic embryos cultured on medium containing 0.1 to 0.2 mg/L 2,4-D had two cotyledons more frequently than one horn-type cotyledon and those on medium containing 0.5 to 4mg/L 2,4-LD had a horm-type cotyledon at a prominently higher freequency. However somatic embryos on medium containing 10mg/L or higher concentrations of 2,4-D were usually shunted at the globular stage even after transfer to medium containing lower concentrations of 2,4-D or other growth regulators. non somatic embryos with one or two cotyledons or a hem-type cotyledon were transferred to medium containing $GA_3$, those with two cotyledons converted to plantlets at a higher frequency (25%) than the others.

Differences of Germination Rate, Degree of Solute Leakage and Water Uptake Rate of Soybean Seeds in Two Storage Periods (저장기간에 따른 콩종실의 발아율이, 용출정도 및 수분흡수율의 차이)

  • This investigation was conducted to investigate if germination rate, degree of solute leakage and water uptake rate of seeds are a meaningful information for the selection criteria among soybean genotypes and to find out their characteristics in the variable temperature and moisture conditions encounted during long-term warehouse storage. Germination rate in soybean sprouting box and petri dish differed largely among genotypes stored for twenty months but there was little difference when stored for eight months. Length of hypocotyl was shorter at twenty months of storage than at eight months. Degree of solute leakage of Paldalkong and Pulmuwon was twice higher than that of SS88038 lines at twenty months of storage. In 30 minutes after imbibition, water uptake rate in all genotypes stored for twenty months was faster than these stored for eight months, and water uptake rate in Paldalkong and Pulmuwon stored for eight months was 1.5 and 2 times faster than in SS88038 lines. In correlation coefficients, the germination ability and emergence exhibited significantly negative correlation with degree of solute leakage at twenty months of storage. Water uptake rate in soybean seeds stored for eight months had highly significant positive correlation with degree of solute leakage in 30 minutes and one hour after imbibition while for twenty months of storage, it had highly significant positive correlation with that in 30 minutes, one hour and three hours after imbibition. In eight months of storage, the number of open micropyle had positive correlation with water uptake rate at 30 minutes after imbibiton. But in seeds stored twenty months, it wasn't correlated with water uptake.

  • Soybean is very useful crop to supply vegetable protein for human. Supply of soybean is increased because it has useful ingredient. Recently, cultivation of soybean in paddy field is increasing due to the increase of rice stockpile in Korea. Hence, in this study, expression of protein was identified regarding different environment for cultivation to investigate the effect of different environment on protein expression. Two-dimensional electrophoresis was performed to investigate the expression of protein using image analysis program to measure degree of protein expression in numerical value. Hannam-kong, Beakcheon-Kong, Hwangkeum-Kong, and Danwon-Kong were used as plant material. 2-DE combined with image analysis revealed that each degree of protein expression of Hannam-Kong and Hwangkeum-Kong in upland field was higher than degree of protein expression in paddy field. However, in case of Beackcheon-Kong, the phenomenon was opposite. In Danwon-kong, the degree of protein expression was not different between up-land field and paddy field. To this end, major protein spots were not different between paddy field and upland field among all cultivars. It could be suggested that protein expression is not severely different by various environment, but different environment affects degree of protein expression.

Cluster Analysis of Korean Domestic Soybeans based on Yields Characters in Paddly Field Cultivation (논 재배 시 한국 콩 품종의 수량성 관련요인분석에 의한 품종군 분류)

  • In order to select the suitable soybean cultivars for paddly field cultivation, growth response and yields characters were investigated with 37 Korean domestic soybean cultivars, in Yeoncheon, Gyeonggi province. The results can be summarized as follows ; When the soybeans were cultivated in paddy field, significant differences were observed in growth responses at each growth stage and yields among the cultivars. Yields of six cultivars, such as Danbaekkong, Paldokong, Pungsannanulkong, Myeongjunamulkong, Saeolkong, and Namhaekong, were less than 200kg/10a, however, seven cultivars, such as Shinpaldalkong 2, Sohokong, Dormikong, keumkangkong, Bukangkong, Dajangkong, and Geomjeongkong 2, showed highest yields ranged from 301~385kg/10a. At V5 stage, highly positive correlation(r=091$^{{\ast}{\ast}}$) was observed between the top dry weight and leaf area while negative correlation was existed between T/R ratio and root dry weight. However, at R2 age, leaf area was correlated with both top and root dry weight while T/R ratio was correlated with leaf area and top dry weight. Thirty-seven soybean cultivars used in this experiments were classified into 3 different clusters based on 19 important agronomic characters, and genotypic properties of the cultivars were similar in each cluster. In results, high yields could be achieved by selected cultivation of suitable soybean cultivars such as Shinpaldalkong 2 which showed early maturation date and highly ripening ratio.

Screening of Antimutagenic Activities from Cereals and Beans Including Rice (쌀을 포함한 곡류 및 두류의 항변이원 활성의 검색)

  • We have established the quantitative method for assay antimutagenic activity from natural products using SOS chromotest technique. Establishment of the method in this study makes it possible to numerize antimutagenic activities from samples in term of the sample amount required for 50% inhibition to mutagenic activity induced by the chemical mutagen under the standard assay condition. Antimutagenic activities of rices from different cultivars as well as other cereals were assayed through this method. The results revealed that antimutagenic activities of mutant cultivar, Suwon 393(Hyangdo) and Sanghaehyanghyulla(Jado), were higher than Chuchung which mainly consumed for steamed rice, and also indicated that antimutagenic activities of cereals, such as job'tear, buckwheat, small red bean, black bean were generally higher than that of brown rice.

Correlation between Leaf Size and Seed Weight of Soybean (콩의 잎 크기와 종실 무게와의 상관)

  • This study was carried out to examine whether the leaf size is likely to be used as a selection criterion for large seed genotype in soybean (Glycine. max (L.) Merr.) breeding program. Two hundred twenty nine soybean germplasms which had collected in Korea, United States, China and Japan were used in this experiment. The area of unifoliate leaf, middle leaflet of first trifoliate and third trifoliate leaf ranged from $3.2cm^2$ to $33.8cm^2$, 9.2 to $29.5cm^2$, and 7.2 to $58.9cm^2$, respectively. One hundred seed weight also showed great variation from 2.7 to 39.0 gram. The average leaf area of unifoliate, middle leaflet of first trifoliate and third trifoliate leaf were $15.7cm^2$, $18.1cm^2$ and $32.7cm^2$, respectively, and that of seed average weight was 17.2 gram per one hundred seed. Significantly positive correlations were observed between seed weight and leaf area of unifoliate (r=$0.80^{**}$), first trifoliate (r=$0.75^{**}$) and third trifoliate (r=$0.67^{**}$), respectively. Both the leaf length and leaf width of unifoliate, middle leaflet of first trifoliate and third trifoliate leaf were significantly positively correlated with seed weight and both the correlations of unifoliate were higher than the other leaves. The correlations of leaf width in soybean leaflet were higher than those of leaf length. Leaf length/width (L/W) ratio of upper leaf was higher than that of lower leaf in the leaf size. Both the leaf area and leaf width of unifoliate leaf are the most suitable predictive characteristics of early selection in related to seed weight for soybean breeding program.

Characteristics of Soybean Soaking Water after Heat Treatment (대두 열처리 담금 용액의 특성)

  • Soybeans released proteins when immersed in water at $50{\sim}60^{\sim}C$. We investigated the changes in the characteristics of soybean when soaked in water at different temperatures and studied the electrophoretic properties of soy proteins in recommended Korean soybean varieties after heat treatment. Soybean seeds were heated in soaking water at temperatures of 30, 40, 50, 60, $70^{\circ}C$ for 90 min, and also from 10 to 150min at $60^{\circ}C$. The pH value of the water decreased with heating time at $60^{\circ}C$, and the amount of soluble solids increased with temperature and heating time. The protein concentration of the solution increased with temperature and time. From SDS-PAGE of the proteins in soaking water, we detected two new bands of 16 kDa- and 31 kDa-proteins from the Korean soybean varieties on heat treatment.

  • Lipoxygenase activity of soybean seeds of approximately 507 genotypes as well as its inheritance and selection efficiency in early breeding generation was measured in the Department of Agronomy, Seoul National University to facilitate breeding for low lipoxygenase activity of soybean. Average seed lipoxygenase activity of 507 cultivars and strains was 350 unit (unit: $\Delta$0.01 /min. /mg at 234nm) and ranged from 50 to 670 unit. There was no difference in mean lipoxygenase activity according to apparent seed characters such as seed coat and embryo color. But early mature soybean genotypes had fairly low lipoxygenase activity. Lipoxygenase activity was inherited quantitatively, in which additive effect was greater than dominant one and proportion of gene with positive effects was similar to that with negative ones. Estimated narrow- and broad-sense heritabilities were 0.78 and 0.86 for lipoxygenase activity, respectively. Heritability measured from selection in early breeding lines for high or low lipoxygenase activity was 64~76% or 54~62%, respectively. And selection for high lipoxygenase activity increased by 29.7~44.7%, whereas that for low ones decreased by 21.8~27.3%, respectively, when compared to random population. Clear effect in selecting of lipoxygenase activity was present in early generation.

Effects of Soil Inoculation on the Nodule Formation, Growth and Yield of Soybean (Glycine max L. Merrill) (종토접종이 대두 근류 형성과 생육 및 수량에 미치는 영향)

  • The crop rotation is very important in the organic agriculture and the soybean crop should be included in the rotation. Even though the inoculation of the root can be performed by the soil inoculation in the soybean crop, the effect of the soil inoculation should be proved in the organic soil. For the research of the effects in the formation of the nodule of soybeans, the growth and the quantity through the inoculation into the soil, the field experiment was conducted in 2006 at Ipjang Experimental Farm of Dankook University. The number of nodule in the testing varieties of the inoculated soil plot was $1.4{\sim}2$ times larger than those in the non-inoculated soil plot. The number of nodule was decreased in the non-inoculated soil plot after R2 stage, but it was increased in the inoculated before R4 stage and it was decreased after then. The periodical change of weight of the nodule was shown as same as the number of nodule. The dry weight of inoculated was increased than non-inoculated in Daewonkong, but it was decreased than non-inoculated in Shinpaldalkong2 and SS2-2. The amount of chlorophyll of the inoculated soil plot was more than the non-inoculated soil plot by 9% in Daewonkong, but the inoculated soil plot was less than the non-inoculated soil plot by 13%, 33% each in Shinpaldalkong2 and SS2-2. The number of pods, the number of grains and the weight of grains in the yield characteristics of the inoculated soil plot were more than in the non-inoculated soil plot in Daewonkong, but the number of pods, grains and the weight of grains in the yield result in the inoculated soil plot was less than in the non-inoculated soil plot in Shinpaldalkong2 and SS2-2. The mean yield(per 10a) of Daewonkong was increased 11% in the inoculated soil plot, but the mean yeil(per 10a) of Shinpaldalkong2 was decreased 4% and SS2-2 was also decreased 23%.

Variation of Major Characters in Soybean Varieties II. Effects of Day Length (대두품종의 주요특성 변이 II. 일장조건의 영향)

  • These experiments were conducted to investigate the effect of day length on some chracteristics and seed weight in the soybean cultivars. Those were classified into three type's of seed weight with delaying seeding date in the previous paper. Flowering was delayed as the longer day length was employed. When the length became longer. the number of leaves at flowering greatly length of main stem and the number of nodes at maturity greatly increased. The seed weight decreased when the day length was longer than 14 hours. Negative correlation was obtained between seed weight and day length. There was negative correlation between seed weight and the number of days required to flowering, and length of main stem and the number of nodes at maturity. Type I cultivars decreased in seed weight when the day length was longer than 16 hours, but this trend was found to be clear with type II cultivars. Type III cultivars decreased in seed weight with increasing the day length.
