Induction of Apomixis by Chemical Mutagen Treatment and Ovule Development in Inbreed lines of Corn (옥수수 자식계통들에서 화학적 돌연변이 유발성질 처리에 따른 apomixis 유발과 배주발생)

  • M1 plants which were produced from seed soaking in chemical mutagen, EMS or NaN$_3$, appeared wide morphorogical variations such as dwarf, albino, twisted leaf, white streaked leaf, and purpled stem. In mutants of reproductive organs, there were monoecious plants such as female-flower plant and male-flower plant, multiple spikes, and steriled plants among M1 plants. Also, barren stalk was increased significantly in M1 plants. Ear bagging at ear initiation stage prevented seed set on cob in normal plants. In spite of ear bagging, M1 plants which had cobs with seed set was 3.9-11.2% of stalks developed from seeds soaking with mutagens, but only three or four kernels could be matured on a cob. Ear bagging after mutagen injection into initiating ear produced 5.1-10% in cobs with seed set, but only 1.7-6.3 kernels could be matured. Cobs removed silk at four hours after artificial pollination increased the rate of cobs with seed set to 27%. Microscopic observation confirmed that ontogeny of kernels matured from ear bagging and mutagen treatment would be both adventitious and diplosporous apomictic reproduction. Chromosome set of M2 seedling was found to be diploid type in chromosomal counting of root tip. As M$_2$ plants showed an uniform appearence within each lines and their CV of plant height were ranged 4-6% in each lines, we concluded that they were apomictic progeny. But we could not find any marker traits combined with apomixis.

Processing Suitability of Canned Ark Shell (새고막의 통조림 가공 적성)

  • This study was carried out to process canned ark shell with highly quality by management of proper processing conditions for prevention of oxidation and discoloration by heating. Ark shell has hemoglobin as blood pigment in red blood shell which same as other cockles. Hemoglobin is easy to come oxidation and browning reaction, and it has large contents of carotenoid as meat pigment. Proximate compositions in ark shell were 76.9% of moisture, 18.1% of crude protein, 1.8% of crude lipid, 1.3% of carbohydrate and 1.6% of crude ash. And contents of carotenoid and hemoglobin were 0.67~1.02mg% and 0.98~1.64g/dl, respectively. When the living ark shell was soaked in 2% NaCl solutions, about 89% of mud was removed after 10 hours soaking, and over 91% was removed when the pH was adjusted to 7.5. Carotenoid pigment were prepared that extracted from ark shell by using acetone. And determined visible spectrum were two peak at 452nm and 687nm, and λmax were 452nm. During thermal treatment at 95$^{\circ}C$, 111$^{\circ}C$, 116$^{\circ}C$ and 121$^{\circ}C$ for 60 minutes, retention ratio of carotenoid were 71.8%, 66.8%, 64.4% and 36.5%, and after 120 minutes retention ratio were 56.6%, 30.6%, 30.3% and 17.2%, respectively. When heated at 95$^{\circ}C$, 111$^{\circ}C$, 116$^{\circ}C$ and 121$^{\circ}C$, formation of browning material were increased at high temperature and long time treatment.

  • The quality of tap water on the whole water-supply system, from a large filtration plant to a private faucet, has to be guaranteed the standards of drinking water. At this point in time, however, the supply process of the tap water has not been monitored and managed scientifically. The piped water, especially the most small-scale reservoirs(underground or overhead type) are always exposed to various contaminations and impurities. Recently monitoring systems of water-quality were spread on some large filtration plants or distributing reservoirs. In particular, the water quality monitoring method using the internet is adopted into some local government whose inhabitants can check up the water quality anytime and anywhere. The construction of this system that has to apply a large scale needs, and has a limitation on the small water-supply system, such as apartments, public facilities and small-scale underground or overhead reservoirs. In this work, we suggest the integration system of individual water-quality sensor modules that have a low price. By using the developed integration system and online monitoring program operated on the internet, the system managers of reservoirs can monitor and manage water-quality characteristic values of drinking water in online. Since the proposed system was modularized, the system can be applied easily into various reservoirs with a low cost and regardless of its scale, small or large.

Farm Animal Mortality Management Practices in Sunchon-si (순천시의 폐사가축 처리실태에 관한 연구)

  • Disposal methods of managing carcass in Korea livestock production systems include burying, digesting, rendering, carcass dumping to manure pile, dead animal disposer and mini-incinerator. Burying was usually the most practical method of carcass disposal in our livestock farms. Burying, carcass dumping to manure pile, dead animal disposer and mini-incinerator may have environmental regulatory and economic liabilities when used as a means of carcass disposal. In many cases in this survey, these disposal methods offer a poor choice for the producer due to individual site conditions, geology, cost, air emissions, rendering plants. A survey questionnaire that addressed the issues to livestock producers was prepared. The questionnaire addressed two main topics as follows: 1) types of livestock and generation amounts of carcass 2) Number of breeding animals and disposal methods of livestock mortality. A total of 36 livestock producers were interviewed. The results of obtained in this survey were summarized as follows: The number of breeding poultry, swine, beef cow and dairy cow was 251,000, 2,600, 142 and 92 heads per year and the generation amounts of annually carcass was 0.46, 15.32, 0.36, 1.36 tons per year of each poultry, swine, beef cow and dairy cow farms, respectively. The disposal methods of carcass were burying (42%), carcass dumping to manure pile (36%), rendering (8%), incineration (6%), digesting (6%), carcass disposer (2%), respectively. These results can be used as basic information to establish the standard of carcass composting facility.

  • 한국농어촌공사 산하의 농업용저수지 중 3786개소에 대한 수질조사를 '19년도에 실시한 결과, TOC 기준 4등급 초과 저수지 비율은 약 20%로써, 도심 근교 저수지에서 녹조현상 빈발로 인해 수질, 악취, 미관 등의 환경문제 개선 민원이 다수 발생하고 있다. 현재 녹조 발생 사후관리를 위해 주로 사용되고 있는 대형 조류제거선은 저수심 수변부에서의 적용성에 한계가 있고, Al 기반의 응집제를 사용하여 조류를 수거해서 폐기하고 있는 실정이다. (주)이엔이티는 농어촌연구원, (주)코레드, (주)삼호인넷과 함께 호소나 정체하천의 수변지역에 적용될 수 있는 저에너지형 유해조류 포집시스템 개발과, 수거된 조류부산물을 무독화하여 농업재료로 재활용하는 방안을 연구하고 있다. 저수지나 정체수역의 녹조는 바람, 수면유동 등에 의해 수변에 집적되는 특성이 있어, 인공지능 기술로 녹조현상을 감시하여 조류 밀집구간에 접근할 수 있는 자율이동식 수상이동장치를 개발 중이다. 수상이동장치는 조류포집장치를 탑재하기 위한 부력체, 원격 운전이 가능한 무인항법장치, 수변식생대 및 저수심지역 이동을 고려한 수차방식 추진체, 전체 장치의 전원 공급을 위한 고성능 배터리 등으로 구성하여 상세 도면 설계를 진행하고 있다. 조류포집장치에는 표층에 주로 분포하는 남조류를 선택 흡입하는 포집 부표를 적용하였고, Al계 응집제 사용을 배제한 분리막 실험을 통해 침지형 막분리조 및 가압형 농축조를 설계하였다. 유해조류 포집 및 농축은 수상에서 이동체에 탑재하여 이뤄지고, 육상에서는 자원 회수가 가능하도록 회분식 응집공정으로 구분하였다. 조류 밀집지역에서 수거된 조류의 무독화 및 농업재료 자원화 타당성 평가를 위해 특용 버섯균주를 활용한 시료별 분석항목을 선정하고 실험 매트릭스에 따라 실증실험을 수행하였다. 수거조류를 전처리하여 성분 및 발열량을 분석하고 버섯재배 전후의 마이크로시스틴 독소(LR, RR, LR)를 포함한 성분 분석을 수행하여, 고체연료, 비료 및 사료로 활용방안을 검토하였다. 무인자율이동 조류포집장치는 실증화 규모로 제작하여 기선정된 테스트베드에서 현장적용성 평가를 수행할 예정이다. 본 연구를 통해 개발된 유해조류 포집 시스템은 기존의 녹조제거 방안을 보완하여 정체수역의 생태계 복원 및 친수공간의 환경개선 등에 적용되며, 무독화가 입증된 유해조류의 농업재료 자원화 기술은 고부가 상품 개발 및 환경폐기물 감축에 활용될 것이다.

Effects of Various Physical and Chemical Factors on the Death of Trouble Seaweed Ulva australis (구멍갈파래(Ulva australis) 해조류 사멸에 미치는 여러 물리화학적 요인들의 영향)

  • Green tides, which was mainly caused by Ulva spp., have been increasing in severity and frequency globally, and have negatively affected on marine ecosystems. This study was conducted to investigate effects of various physical and chemical factors on the death of Ulva australis (ULAUS) and to consider a practical measures useful for alleviating Ulva bloom. Soaking of ULAUS thalli in pure water for 8 hr didn't induce a death, but incubation in 1.0-1.5% salinity for 7 d inhibited sporulation by about 70%. Desiccation gave rise to a serious damage when more than 40-50% of initial fresh weight was lost. ULAUS growth was sensitive to temperature and seriously inhibited from more than $30^{\circ}C$. At $35^{\circ}C$, $40^{\circ}C$, $45^{\circ}C$ and $50^{\circ}C$, treatment time required for 90-95% death of ULAUS thalli was 1 d, 10 min, 30 sec, and 1 sec, repectively. ULAUS growth was seriously inhibited at lower than pH 6.0 and completely dead at pH 4.0. Several compounds for ULAUS control was selected and the chemcals causing a rapid death were oxidants such as hydrogen peroxide and sodium percarbonate. Taken together, our results suggest that low salinities, dryness, pH, high temp. and compounds could be selected for Ulva bloom control, and high temperature and compounds seems to be useful for a development of practical control methods.

Effects of Fire Retardant Treatment on Mechanical Properties and Fire Retardancy of Particleboard and Complyboard (내화처리(耐火處理)가 파아티클보오드와 콤플라이보오드의 기계적성질(機械的性質) 및 내화도(耐火度)에 미치는 영향(影響))

  • This research was conducted to examine the feasibility of developing fire retardant particleboard and complyboard. Particleboard were manufactured using meranti particle(Shorea spp.)made with Pallmann chipper, and complyboard meranti particle and apitong veneer (Dipterocarpus spp.). Particles were passed through 4mm (6 mesh) and retained on 1mm (25 mesh). Urea formaldehyde resin was added 10 percent on ovendry weight of particle. Face veneer for complyboard was 0.9, 1.6 and 2.3mm in thickness and spread with 36 g/(30.48 cm)$^2$ glue on one side. Veneers were soaked with 10 percent solution of five fire retardant chemicals (diammonium phosphate, ammonium sulfate, monoammonium phosphate, Pyresote and Minalith), and particles with 5, 10, 15 and 20 percent solution of five chemicals. Particleboard and complyboard were evaluated on physical and mechanical properties, and fire retardancy. The results obtained were summarized as follows. 1. Among five fire retardant chemicals treated to particleboard and complyboard, the retention of ammonium sulfate in 5 percent solution showed the lowest as 1.39 kg/(30.48 cm)$^3$ exceeding the minimum retention of 1.125 kg/(30.48 cm)$^3$ recommended by Forest Products Laboratory and Koch. 2. Particleboard and complyboard treated with diammonium phosphate showed higher modulus of rupture (MOR), modulus of elasticity (MOE), internal bond strength and screw holding power than those with the other chemicals. 3. MOR and MOE of complyboard treated with fire retardant chemicals were greater than those of fire retardant particleboard. 4. Thickness swelling of fire retardant complyboard was lower than that of fire retardant particleboard. 5. The moisture content of the boards treated with Pyresote and Minalith increased and with monoammonium phosphate reduced. 6. Fire retardant particleboard showed no ignition, and fire retardant complyboard started ignition, but time required to ignite was prolonged comparing the controlboard. Complyboard with only shell veneer treated showed ignition and lingering flame, but lingering flame time was shorter than controlboard. Complyboard with treated both core and veneer showed ignition but not lingering flame. 7. Flame length, carbonized area and weight loss were smaller than controlboard but had no significant difference among chemicals treated. 8. Temperature of unexposed surface of fire retardant particleboard was lowered with the increasing concentration of five chemicals. 9. Temperature of unexposed surface of fire retardant particleboard was lowered with the highest in Pyresote and the lowest in Minalith. 10. Temperature of unexposed surface of fire retardant complyboard was lower than that of controlboard.

Shelf-life and Quality Characteristics of Tofu Coagulated by Calcium Lactate (젖산칼슘을 응고제로 한 두부의 품질특성과 저장성)

  • To investigate the utilization of calcium lactates (CaL) as coagulants for tofu manufacture, the quality characteristics and shelf-life of tofu made by CaL-P (black snail powder) and CaL-A (black snail ash) were investigated and compared to calcium chloride (CC), magnesium chloride (MC), calcium sulfate (CS ) and standard calcium lactate (CaL-S). And also, total microbe and turbidity of the tofu were determined during storage at 1$0^{\circ}C$. Coagulation ability of CaL-A was the highest, and the ability of CaL-P was higher than that of CaL-S. Yield of CaL-A tofu was similar to those of CS and CC tofu, while the yield of CaL-P tofu was 50% compared to that of CC. L* value of CaL-P tofu was lower, but a* and b* values were higher than those of other tofus. The hardness of tofu showed in the order of CaL-S>CS>CC>CaL-P>MC>CaL-A, while the cohesiveness showed in the order of MC>CaL-S>CC>CS>CaL-P>CaL-A. Calcium contents were 57 mg% in MC tofu, 174 mg% in CS tofu, 116 mg% in CaL-S tofu, 95 mg% in CaL-A tofu and 172 mg% in CaL-P tofu. From the results of microscopic observations, the lower hardness showed the more soft and the smaller particle. The particle of CaL-A tofu was small and uniformity but the size of CaL-P and CC tofu showed coarse. Sensory quality of CaL-P and -A tofu were better than the other tofu evaluated by texture, springiness, flavor and overall taste. The shelf-life estimated by total microbe was 4∼6 days in CC, MC, CS, CaL-S and CaL-A tofu, but 8 days in CaL-P tofu at 1$0^{\circ}C$. From the above results, the CaL-P and -A may believe to use as coagulant for tofu manufacture due to its softened taste and enhanced shelf-life, and higher calcium content which has higher absorbability in human body.

  • This experiment was carried out to study on methods of processing the fruit base which is to be used for preparing the fruit yoghurts. Common fruits were compared and peach was chosen among them for this purpose by determing the rate of decrease in hardness during the process of cooking. The maturity, temperature of cooking and methods of peeling were tested to maintain the desirable hardness during the process, and the effects of metallic ion addition on the hardness of the flesh were also studied. The results obtained were as follows. 1. The peaches were keeping the most stable texture during cooking, among common fruits, however the hardness of apples were decreased markedly. Rate of decreases in the flesh hardness were high during the initial stages of cooking then slowered in all fruits tested. 2. Flesh hardness of the peaches were largely affected by the temperature of cooking. And the texture of freeze-stored fruits was severly damaged during thawing, so that could be used only for the limited purposes such as for stirred-type yoghurt. 3. Divalent matallic ions, especially calcium ion, were effective for supporting the flesh hardness of peaches during cooking. 4. The optimal concentration of calcium chloride addition was 0.3%, and this resulted in its final concentration in the product under the legal dosage without any subsquent changes in colour and flavor. 5. The effects of calcium chloride addition were greater in the froms of dices than in those of slices, and in overmature fleshs than in immature ones. 6. Treatment of calcium chloride by soaking the slices or dices of lye-peeled peaches in 0.3% solution was found to be adaptable for the practical process.

On the Oxidation of Rice Lipid Fractions Extracted from the Whole Grain by Immersion (용매침지(溶媒浸漬)에 의하여 추출(抽出)된 쌀알의 유지획분(油脂劃分)의 산화(酸化)에 대(對)하여)

  • Oxidative changes in the rice lipids under accelerated condition were studied by measuring the changes in weight gain, total carbonyl compound, malonaldehyde, and fatty acid composition. Rice lipids were prepared by extraction with either n-hexane or ethanol from polished rice grain and purified by Folch's method. The lipid preparations were either incubated in dark at $40^{\circ}C$ or irradiated with ultra-violet light for a period of 55 days. Weight gain by oxygen absorbed sharply increased within 3 days in the rice lipids under UV light irradiation. However, with the rice lipids at $40^{\circ}C$ incubation a moderate increase in weight was observed only after 45 days storage. Their induction periods were one day (hexane extracted, under UV light), 2 days (ethanol extracted, under UV light), 30 days (hexane extracted, at $40^{\circ}C$), and 40 days (ethanol extracted, at $40^{\circ}C$) respectively. Oxidative rancid odor appeared at the end of the induction period. Total carbonyl compound and malonaldehyde markedly increased within 7 days, and decreased in the rice lipids under ultra violet light irradiation, while at $40^{\circ}C$ incubation they were continued to increase slowly through out the storage. The hexane extracted lipid were less stable than ethanol extracted lipid on the basis of oxygen absorption, malonaldehyde and other carbonyl compound formation. With the hexane extracted lipid during 55 days incubation at $40^{\circ}C$, the contents of linoleic and linolenic acids decreased, while the oleic, stearic, and palmitic acids increased.
