Characterization and Volatile Flavor Components in Glutinous Rice Wines Prepared with Different Yeasts of Nuruks (누룩에서 분리한 효모를 이용한 찹쌀발효주의 이화학적 특성 및 휘발성 향기성분)

  • In order to investigate the effect of different yeasts (La Parisienne (LP), Y18-2, Y54-3, Y90-2, Y90-9 and Y272-7) from nuruks on the quality of Glutinous rice wines, physicochemical properties and volatile flavor components were evaluated. Glutinous rice wines prepared with different yeasts were analyzed for ethanol, pH, total acid, amino acid, soluble solid, coloring degree, UV absorbance, reducing sugar, organic acid, free sugar and volatile compounds. After fermentation for 17 days, the ethanol contents ranged from 13.40 to 14.50%, while the total acid levels were from 0.33 to 0.44%. The amino acid contents in six samples ranged from 0.13 to 0.18%, while soluble solid contents ranged from 12.1 to $14.7^{\circ}Bx$. The glutinous rice wine prepared with LP showed the highest level of coloring degree, soluble solid and reducing sugar among six samples. Organic acid contents of the glutinous rice wine prepared with LP had the highest levels of lactic acid and acetic acid, while the glutinous rice wine prepared with Y90-9 had the highest level of succinic acid. In all glutinous rice wines tested, the most abundant free sugars were glucose followed by maltose. Volatile flavor components in the glutinous rice wines were identified by using GC-MSD. Nineteen esters, ten alcohols, eight acids, one aldehyde and one miscellaneous compound were identified in the glutinous rice wines. Using relative peak area, it was found that other than ethyl alcohol, hexadecanoic acid ethyl ester was the major component, predominantly found in the range of 2.73-10.41%. Phenylethyl alcohol, isoamyl alcohol, ethyl oleate, ethyl linoleate and tetradecanoic acid ethyl ester were some of the major volatile components present through the fermentation, respectively. Overall, it was shown that different yeast strains from nuruks greatly affected chemical and volatile characteristics of the glutinous rice wines.

Partial Purification of OsCPK11 from Rice Seedlings and Its Biochemical Characterization (벼 유식물에서 OsCPK11의 부분 정제 및 생화학적 특성 규명)

  • Calcium is one of the important secondary signaling molecules in plant cells. Calcium-dependent protein kinases (CDPK)-the sensor proteins of Ca2+ and phosphorylating enzymes-are the most abundant serine/threonine kinases in plant cells. They convert and transmit signals in response to various stimuli, resulting in specific responses in plants. In rice, 31 CDPK gene families have been identified, which are mainly involved in plant growth and development and are known to play roles in response to various stress conditions. However, little is known about the biochemical characteristics of CDPK proteins. In this study, OsCPK11-a CDPK in rice-was partially purified, and its biochemical characteristics were found. Partially purified OsCPK11 from rice seedlings was obtained by three-step column chromatography that involved anion exchange chromatography consisting of DEAE, hydrophobic interaction chromatography consisting of phenyl-Sepharose, and gel filtration chromatography consisting of Sephacryl-200HR. An in vitro kinase assay using partially purified OsCPK11 was also performed. This partially purified OsCPK11 had a molecular weight of 54 kDa and showed a strong hydrophobic interaction with the hydrophobic resin. In vitro kinase assay showed that the OsCPK11 also had Ca2+-dependent autophosphorylation activity. The OsCPK11 phosphorylated histone III-S, and the optimum pH for its kinase activity was found to be 7.5~8.0. The native OsCPK11 shared several biochemical characteristics with recombinant OsCPK11 studied previously, and both had Ca2+-dependent autophosphorylation activity and favored histone III-S as a substrate for kinase activity, which also had a Ca2+-dependence.

Metabolic Adjustments of Lactate Dehydrogenase Isozymes to the Environmental Temperature in Bluegill (Lepomis macrochirus) (환경온도에 대한 파랑볼우럭(Lepomis macrochirus) 젖산탈수소효소 동위효소들의 대사조절)

  • The aim of this study was to examine the metabolic adjustment of lactate dehydrogenase (EC 1.1.1.27, LDH) isozymes to the environmental temperature in bluegill (Lepomis macrochirus). This study included three groups of bluegill collected in April (group Ⅰ), May (group Ⅱ), and September (group Ⅲ). The LDH activities of skeletal muscle, heart, and brain tissues were higher in group Ⅲ than in groups Ⅰ and Ⅱ. The citrate synthase (EC 4.1.3.7, CS) activity was higher in skeletal muscle but lower in heart and brain tissues of group Ⅱ as compared to group Ⅰ. In contrast, the CS activity was lower in skeletal muscle and higher in heart and brain tissues in group Ⅲ than in group Ⅱ. Furthermore, the LDH/CS activity ratio was higher in the skeletal muscle and brain in group Ⅲ than in groups Ⅰ and Ⅱ. Accordingly, anaerobic metabolism was increased in group Ⅲ. LDH A4, A2B2, and B4 isozymes were expressed in skeletal muscle, heart, liver, and brain tissues. The LDH C hybrid was detected in brain tissue. The LDH A4 isozyme was successfully purified by affinity chromatography. The molecular weight of the purified LDH A4 isozyme was 136 kDa and its optimal pH for enzymatic activity was 8.0. The KmPYR values of LDH in skeletal muscle were 0.161-0.227 mM using pyruvate as a substrate. These kinetic properties of LDH in skeletal muscle are consistent with the fact that bluegill is a cold-adapted species. These results may be useful for predicting the habitat use of this fish.

Quality Characteristics of Yulmoo Mul-kimchi Containing Saltwort (Salicornia herbacea L.) (함초 분말 첨가 열무 물김치의 품질특성)

  • Saltwort (Salicornia herbacea L.), as a natural additive for regulating Mul-kimchi fermentation, was assessed for physicochemical and sensory characteristics of Yulmoo Mul-kimchi during storage. Saltwort in the form powder was directly added to the Yulmoo Mul-kimchi preparation at 0 (control), 1, 3, 5, and 7% (w/v) per weight of salt to evaluate their physicochemical, sensory, and microbiological characteristics in storage at $10^{\circ}C$ for 30 days. The pH values of all treatments were high, indicating a less acidic environment in all treatments compared to those of the control sample throughout the preservation period. Total acidity increased with storage time as is usually seen with normal kimchi fermentations, whereas the increases were more gradual in the 3 and 5% treatments. The increases in total vitamin C continued until days 6 (control) to 13 (7%), and were different according to the amount of added saltwort and then they decreased after each peak. The fluctuation in reducing sugars fol owed a similar trends of total vitamin C content by showing an initial increase, followed by a decrease based on the saltwort concentration and storage day. The 5% treatment was most effective for suppressing the increase in turbidity among all treatments. The 3 and 5% treatments showed the highest lactic acid bacteria counts during the entire preservation period. In the sensory evaluation results, adding saltwort at more than 3% concentration attained an overall higher scores of acceptability with respect to color, smell, taste, fresh taste, sour taste, crisp, and overall acceptability characteristics. In conclusion, adding saltwort, particularly at concentrations of 3 and 5% extended the preservation period of Yulmoo Mul-kimchi by retarding fermentation effectively.

Effects of Diffusibility of Bubbling Tablet Herbicide Formulations for Paddy Rice (수도용 발포성정제 제초제의 확산성에 미치는 영향)

  • A series of studies involving formulation processes, bubbling activity test, diffusibility test and biological efficacy test was undertaken to develop Jumbo herbicide formulations in paddy rice field. Gas evolution speed from the tablets prepared by different organic acids was in the order of oxalic acid, malonic acid, citric acid, and tartaric acid. The total volume of evolved gas from the tablet and diffusibility of the active ingredient in the submerged water were increased with increase of water temperature; the volume from 1 g of tablet at 10, 15, 25 and $30^{\circ}C$ for 5 minutes after immersion into water was 20, 25, 28, 45, 57 mL, respectively. The concentration of halosulfuron-methyl and pyriminobac-methyl in submerged water at 5, 15, 20 and $30^{\circ}C$ at the 2.4 m distance from the applied spot of the tablet was 20, 48, 85, and 97% of the concentration of treated spot, respectively. The evolved gas volume from the tablets was not affected by pH of submerged water. The concentration of halosulfuron-methyl in different sizes of submerged water within 24 hours after treatment of the tablet was maintained 0.16 ppm, which is ideal concentration at standard dosage regardless of the submerged water area. The concentration of pyriminobac-methyl was also uniformly dispersed in the water within 24 hours after applying it into the submerged water. The wind velocity of 5 m $sec^{-1}$ on concentration distribution of halosulfuron-methyl and pyriminobac-methyl in the submerged water 24 hours after treatment was not influenced; an equal concentration in the up the wind and down the wind from the applied spot was maintained. Spot treatments of one tablet formulations(5 g) including 4 times higher dosage at 4 different spots resulted in even concentration distribution of active ingredient in the water 24 hours after applying it into the submerged water.

A Study of Heavy Metal-Contaminated Soil Remediation with a EDTA and Boric acid Composite(I): Pb (EDTA와 붕산 혼합용출제를 이용한 중금속으로 오염된 토양의 처리에 관한 연구(I): 납)

  • To choose a organic acid and in-organic acid composite which is the most effective in soil-flushing process cleaning lead-contaminated sites, lead removal rates were investigated in the experiments with some organic acids; 0.01M of EDTA showed the highest lead-extraction rate ($69.4\%$) compared to the other organic acids. Furthermore, the lead removal rates were measured with 0.01M of EDIA and 0.1M of in-organic acid ; a EDTA and boric acid composite showed the highest lead-extraction rate ($68.8\%$) at pH5 compared to the other composites. As the concentration of boric acid was increased from 0.1M to 0.4M in a 0.01M of EDTA and boric acid composite, lead removal rate was decreased from $68\%\;to\;45\%$. But as the concentration of EDTA was increased from 0.01M to 0.04M in a EDTA and 0.1M of boric acid composite, permeability was decreased from $6.98{\times}10^{-4}cm/sec$ (0.01M of EDTA) to $5.99{\times}10^{-4}cm/sec$ (0.04M of EDTA). However, permeability was increased from $4.41{\times}10^{-4}cm/sec$ (0.03M of EDTA) to $6.26{\times}10^{-4}cm/sec$ (0.03M of EDTA and 0.1M of boric acid composite). indicating EDTA could increase lead dissolution/extraction rate and decrease permeability. In this system, lead remediation rate is the function of lead dissolution rate from soils and permeability of the composite into soils, and the optimized [EDTA]/[Boric acid] ratio is [0.01M]/[0.1M].

Effect of the Mixing Ratio of Pot Media on the Germination and Early Growth in Vegetable crops (배양토 조성비율이 채소작물의 발아 및 초기 생장에 미치는 영향)

  • This study analyzed physical and chemical characteristics of peat soil to use peat soil as the materials fur pot media and investigated seedling quality of horticultural plants in order to use peat soil as the raw materials fur pot media. The summary of the results is as follows; The chemical characteristics of peat soil, which is main ingredient of pot media are as follows; pH was 4.9, EC was less than $2.0ds{\cdot}m^{-1}$, which interferes the growth of the plant and organic ingredient was 33%. When looking into the germination characteristics of plants according to the mixture of pot media, red pepper showed 54.2% speed of germination and 97% germination rate in peat soil single treatment. Therefore the peatsoil was appropriate for the pot media for red pepper. In case of cucumbers, in the 50 : 50 treatment of main ingredient (peat soil) and auxiliary ingredients (vermiculite, peat moss and perlite) they showed 100% speed of germination and 100% germination rate. Therefore 50 : 50 treatment was appropriate fur the pot media for cucumbers. In case of chinese cabbage, peat soil, perlite and peat moss mixture (50 : 25 : 25) treatment showed the highest speed of germination (77.5%), while the germination rate was a little lower (92.15%) than comparative soil. However, it was appropriate for the pot media for chinese cabbage. In case of watermelon, germination was bad because of the influence of EC when the teat soil ingredient is over 80%. However, in the mixture of peat soil and vermiculite (50:50) treatment, they showed 91.6% speed of germination and 100% germination rate. Therefore it was appropriate for the pot media for watermelon. When looking into the growth of the plants according to the mixture of ingredients, peat soil and perlite (50:50) mixture showed excellent seedling quality for cucumbers, peat soil and perlite (50:50) mixture showed excellent seedling quality and it was proven to be appropriate for the pot media for cucumbers. In case of watermelon, peat soil, peat moss and perlite (80 : 10 : 10) mixture showed excellent seedling quality and it was proven to be appropriate for the pot media for watermelon.

Isolation and Characterization of a Marine Bacterium, Pseudomonas sp. YJ-1 with Anti-Methicillin Resistant Staphylococcus aureus Activity (항 Methicillin Resistant Staphylococcus aureus 활성을 가지는 해양미생물 Pseudomonas sp. YJ-1의 분리와 특성)

  • The aim of this study was to isolate and identify marine bacterium with anti-methicillin-resistant Staphylococcus aureus (MRSA) activity, and to purify the anti-MRSA compound, as well as to determine its activity and synergistic effects. Among the marine bacteria isolated in this study, the YJ-1 isolate had the strongest anti-MRSA activity. The YJ-1 isolate was identified on the basis of its biochemical characteristics and an analysis of 16S rRNA gene sequences. The YJ-1 isolate showed over 99.2% homology with Pseudomonas stutzeri, and was designated as a Pseudomonas sp. YJ-1. The optimal culture conditions were $25^{\circ}C$ and initial pH 7.0. For the purification of the anti-MRSA compounds, the YJ-1 was cultured in Pa PES-II medium, and the culture filtrates were extracted by ethyl acetate, hexane, and 80% MeOH. The 80% MeOH fraction was separated by a $C_{18}$ ODS column, silica gel chromatography and a reverse phase HPLC, to yield three anti-MRSA agents, the MR1, MR2, and MR3 compounds. When the MR1 compound of $250{\mu}g\;mL^{-1}$ concentration was applied to the MRSA cells, over 95% of bacterial cells was killed within 48 hr. Compared with vancomycin and ampicillin, the MR1 compound showed significant anti-MRSA activity. In addition, the anti-MRSA activity was increased by dose and time dependent manners. Furthermore, the combination of an MR1 compound with vancomycin produced a more rapid decrease in the MRSA cells than did the MR1 compound alone. Taken together, our results suggest that the Pseudomonas sp. YJ-1 and its anti-MRSA compounds could be employed as a natural antibacterial agent in MRSA infections.

Production of Medium-chain-length Poly (3-hydroxyalkanoates) by Pseudomonas sp. EML8 from Waste Frying Oil (Pseudomonas sp. EML8 균주를 이용한 폐식용류로부터 medium-chain-length poly(3-hydroxyalkanoates) 생합성)

  • In this study, to reduce the production cost of poly(3-hydroxyalkanoates) (PHA), optimal cell growth and PHA biosynthesis conditions of the isolated strain Pseudomonas sp. EML8 were established using waste frying oil (WFO) as the cheap carbon source. Gas chromatography (GC) and GC mass spectrometry analysis of the medium-chain-length PHA (mcl-PHAWFO) obtained by Pseudomonas sp. EML8 of WFO indicated that it was composed of 7.28 mol% 3-hydrxoyhexanoate, 39.04 mol% 3-hydroxyoctanoate, 37.11 mol% 3-hydroxydecanoate, and 16.58 mol% 3-hydroxvdodecanoate monomers. When Pseudomonas sp. EML8 were culture in flask, the maximum dry cell weight (DCW) and the mcl-PHAWFO yield (g/l) were showed under WFO (20 g/l), (NH4)2SO4 (0.5 g/l), pH 7, and 25℃ culture conditions. Based on this, the highest DCW, mcl-PHAWFO content, and mcl-PHAWFO yield from 3-l-jar fermentation was obtained after 48 hr. Similar results were obtained using 20 g/l of fresh frying oil (FFO) as a control carbon source. In this case, the DCW, the mcl-PHAFFO content, and the mcl-PHAFFO yields were 2.7 g/l, 62 wt%, and 1.6 g/l, respectively. Gel permeation chromatography analysis confirmed the average molecular weight of the mcl-PHAWFO and mcl-PHAFFO to be between 165-175 kDa. Thermogravimetric analysis showed decomposition temperature values of 260℃ and 274.7℃ for mcl-PHAWFO and mcl-PHAFFO, respectively. In conclusion, Pseudomonas sp. EML8 and WFO could be suggested as a new candidate and substrate for the industrial production of PHA.

Study on water quality prediction in water treatment plants using AI techniques (AI 기법을 활용한 정수장 수질예측에 관한 연구)

  • In water treatment plants supplying potable water, the management of chlorine concentration in water treatment processes involving pre-chlorination or intermediate chlorination requires process control. To address this, research has been conducted on water quality prediction techniques utilizing AI technology. This study developed an AI-based predictive model for automating the process control of chlorine disinfection, targeting the prediction of residual chlorine concentration downstream of sedimentation basins in water treatment processes. The AI-based model, which learns from past water quality observation data to predict future water quality, offers a simpler and more efficient approach compared to complex physicochemical and biological water quality models. The model was tested by predicting the residual chlorine concentration downstream of the sedimentation basins at Plant, using multiple regression models and AI-based models like Random Forest and LSTM, and the results were compared. For optimal prediction of residual chlorine concentration, the input-output structure of the AI model included the residual chlorine concentration upstream of the sedimentation basin, turbidity, pH, water temperature, electrical conductivity, inflow of raw water, alkalinity, NH3, etc. as independent variables, and the desired residual chlorine concentration of the effluent from the sedimentation basin as the dependent variable. The independent variables were selected from observable data at the water treatment plant, which are influential on the residual chlorine concentration downstream of the sedimentation basin. The analysis showed that, for Plant, the model based on Random Forest had the lowest error compared to multiple regression models, neural network models, model trees, and other Random Forest models. The optimal predicted residual chlorine concentration downstream of the sedimentation basin presented in this study is expected to enable real-time control of chlorine dosing in previous treatment stages, thereby enhancing water treatment efficiency and reducing chemical costs.
