  • To gain a better understanding of the germination and growth of Scirpus maritimus, pot and field experiments were conducted to observe the development of new rhizomes and tubers, the growth rate of culms. and the germination behaviour of tubers of this perennial weed. Tuber placement, water depth, salinity and pH were also observed for their possible influence on the growth of S. maritimus.

  • We investigated the optimal cultivation system and dry processing method of rhizomes from Curcuma longa L. in Suncheon, the southern area of Korea. The experimental results are summarized as followings. Curcma longa rhizomes were transplanted on Apr. 30th, May 10th, and May 20th. The plant height, stem diameter, number of leaves and yield were highest in transplanted on May 10th. The fresh rhizome weight of Curcma longa was the heaviest in November 20 and the optimal dry temperature was $40^{\circ}C$. In sensory evaluations, there were significant differences among the Curcuma longa pills in terms of flavor, color, color and texture and overall acceptability, and honey added groups received significantly higher scores than the other groups.

The isolation of Bacillus subtilis KYS-10 with antifungal activity against plant pathogens (식물 병원균에 대한 항진균 활성을 갖는 Bacillus subtilis KYS-10의 분리)

  • This study was investigated for the purpose of the isolation and identification of antagonistic bacteria with antifungal activity against plant pathogens. This bacteria denominated Bacillus subtilis KYS-10 and the optimum growth condition were 4% sucrose, 1% yeast extract, 0.2% $K_2HPO_4$, pH 7, 150 rpm, $30^{\circ}C$, 8 day. The antifungal activities against nine plant pathogens determined inhibition zone size by diffusion methods. The results, G. zeae (scab) 70 mm and P. grisea KACC 40439 (blast), P. capsici KACC 40177 (phytophthora blight) and C. destructans KACC 41077 (root rot of ginseng) 40~43 mm, and C. gloeosporioides KACC 43520 (ripe rot), C. gloesporioides KACC 40003 (anthracnose), S. shiraiana KACC 41065 (stem rot) and S. shiraiana (mulberry sclerotial disease) 35~39 mm and F. Oxysporum KACC 44452 (bulb rot of ginseng) 28 mm. From these experiment results, author suggest that Bacillus subtilis KYS-10 would be developed as a biological control agent thorough the field experimet in the near future.

  • In this study, established soil-borne Bacillus sp. FF-9 with strong antifungal activity was isolated for identification and to determine optimal culture condition. By using 16s rDNA sequencing method, FF-9 of the selected bacteria was identified as genus Bacillus sp., Bacillus sp. FF-9 was cultured at $30^{\circ}C$, for 24 h in the LB medium. Cell growth increased quickly after 6 h and the highest cell growth was indicated at 12 h. The most antifungal activity against Rhizoctoina solani AGII-II appeared at 18 h and the optimal temperature and pH were 30 and pH 8.0, respectively. A testing of carbon and nitrogen sources showed the highest antifungal activity at 1% lactose and 1% yeast extract Furthermore an addition of salt showed the most antibiotic activity in the 0.15% $K_2HPO_4$.

  • The cultural conditions of the osmophilic yeasts for higher concentration of NaCl selected in the previous report were examined and the results obtained were as follows. 1) The strain $T_3\;and\;T_8$ were grown exceedingly well on the media containing 15 percent of NaCl and $T_5\;and\;T_9,\;T_{10}\;and\;T_{11}$ on the media containing 5 percent of NaCl. 2) The optimum temperature for growth of the strain $T_3\;and\;T_5$ was $30^{\circ}C,\;T_8\;T_{10}$ and $T_{11}\;was\;25^{\circ}C\;and\;T_9\;was\;35^{\circ}C.$ 3) Their lethal temperature was $60^{\circ}C$ (treatment for 10 minutes). 4) The optimum pH for growth of the strain $T_3\;and\;T_8$ was pH 4.0, $T_5$ was pH 6.0 and $T_9\;T_9\;and\;T_{11}$ was pH 5.0, respectively.

Optimum Conditions for the Protoplast Formation of Lactobacillus plantarum and Leuconostoc mesenteroides (Lactobacillus plantarum과 Leuconostoc mesenteroides의 Protoplast 생성의 최적 조건)

  • Protoplasts of both strains were produced by lysozyme digestion at $30^{\circ}C$ for 180min. Both strains were treated with $40{\mu}g$/ml of lysozyme in 30mM Tris-HCl buffer(pH 7.5) containing 10% sucrose at the late logarithmic growth phase. It was found that the efficiency of protoplast formation was high at $30^{\circ}C$ and pH 7.5 by measuring the decrease in absorbance. Optimum concentrations of sucrose $Ca^{2+}, \;Mg^{2+}$ for protoplast formation were determined to be 15%, 20mM and 6mM, respectively. Hydrolysis of cell wall and protoplast formation efficiency for L. plantarum showed better results than those for Leu. mesenteroides. The resistances to antibiotics erythromycin and chloramphenicols were chosen as the selection marker for the fusant between L. plantarum and Leu. mesenteroides. Production phase of protoplast in Leu. mesenteroides was also compared with L. plantarum in this paper.

Effects of Forest Environments on Growth and Active Compound Contents of Ligusticum chuanxiong Hort. among Different Forest Sites (기후대별 산림환경에 따른 토천궁의 생육 및 유효성분 특성)

  • Ligusticum chuanxiong Hort. is included in Umbelliferae family, it is one of the Korean traditional medicinal plants as the roots have been used to treat diseases. In this study, the growth characteristics and active compound contents of L. chuanxiong were compared among the different forest sites. As a result, root diameter and root length of L. chuanxiong was the highest in Jeongseon. Also, the fresh weight and dry weight of L. chuanxiong were the highest in Jeongseon. The total content of active compound was 23.27 mg/g the highest in Bonghwa, and 21.59 mg/g in Jeongseon, 15.87 mg/g in Hamyang was accumulated. In this study compares three forest site for cultivating of L. chuanxiong in different climate zone that the best site to product yield were Jeongseon. In this sites were located in higher altitue and lower temperature than other sites, also there were shown that lower soil moisture contents and well-drained soil. It was shown yield and active compound contents of L. chuanxiong was influenced by micro-environment conditions like as altitude, temperature, soil conditions.

Optimal Conditions for Groundcovers Cultivation of Korean Native Lythrum salicaria L. (자생 털부처꽃의 지피용 재배를 위한 최적 조건)

  • 본 연구는 자생 털부처꽃의 실용적인 산업화를 위해 지피용 재배조건을 조사하고자 수행되었다. 실험재료는 털부처꽃의 종자를 2019년 4월말에 파종하여 8주 후 얻어진 유묘를 사용하였다. 실험은 식재용기(2, 3, 4치 비닐포트), 토양종류(원예상토 단용 및 마사토:피트모스 혼용토(v/v; 2:1, 3:1), 파종용기(162, 200, 288셀 플러그트레이) 및 파종립수(1, 2, 4립)를 달리하였으며, 정식 후 처리구간의 생육특성을 조사하였다. 또한 마사토와 피트모스 혼용토는 9L당 퇴비 1L를 혼합하여 원예상토와 비교하였다. 대조구는 원예상토가 충진된 3치 비닐포트에 정식된 유묘와 셀당 1립이 파종된 200셀 플러그트레이의 유묘를 사용하였다. 유묘는 처리별로 정식한 다음 충북대학교 유리온실에서 12주간 재배되었으며, 매일 2회(5분/1회) 미스트 관수되었다. 연구결과, 털부처꽃의 유묘가 정식된 비닐포트는 포트의 볼륨이 클수록 생육이 우수한 경향을 나타냈다. 특히 4치 비닐포트는 동일한 재배기간 동안 경직경(3.61 mm), 측지수(28.2 ea), 엽수 (311.6 ea), 근장 (22.1 cm) 및 생체중(7049.9 mg)이 가장 많이 증가하였다. 토양종류는 원예상토를 단독으로 사용한 처리구의 생육이 가장 우수하였다. 반면 마사토와 피트모스 혼용토는 재배가 지속되면서 생육이 저조하여, 농업용 퇴비로는 양분이 부족한 것으로 보인다. 파종립수가 증가할수록 각 개체의 생육은 감소하였으나, 4립 파종구의 유묘는 정식 후 풍성한 볼륨을 나타냈으며, 지피용 재배목적에 부합하였다. 플러그트레이는 볼륨이 작아질수록 초장과 생체중이 일부 감소하는 경향을 나타냈으나, 전반적으로 생육은 유사한 경향을 나타냈다.

Effect of External Light Environment and Growing Degree Days on Strawberry Production (외부 광환경 및 생육도일온도가 딸기 생산량에 미치는 영향)

  • In this study, strawberries were grown during the two cultivation periods (first: 2020-2021, second: 2021-2022) to analyze the effect of the external light environment and growing degree days (GDD) on crop production. The temperature and humidity during day in a greenhouse in each cultivation period were similarly managed. At night, there was a statistical difference in temperature and humidity in the greenhouse between two periods. The accumulated solar radiation during the first cultivation period was high in September and October. Since January, the accumulated solar radiation during the second cultivation period was high. In the second cultivation period, the initial yield was small because the accumulated solar radiation and GDD was small. But accumulated yields and potential maximum yields in second cultivation period were larger than yields in the first cultivation period as the accumulated solar radiation and GDD increased. The sugar contents of strawberry decreased as GDD increased.

Effects of Red/Blue Light Ratio and Short-term Light Quality Conversion on Growth and Anthocyanin Contents of Baby Leaf Lettuce (적색/청색광의 비율 및 수확 전 광질변환이 어린잎상추의 생육 및 안토시아닌 함량에 미치는 영향)

  • To establish the optimum artificial light illumination method for baby leaf lettuce in closed plant factory system, the effects of red/blue light quality and short-term light quality conversion on growth and anthocyanin content were investigated. The growth of 'Hongha' lettuce was most favorable under red single wavelength LED light after 23 days of treatment, sequentially followed by the growth under red/blue mixed light, blue light, and fluorescent light. Total anthocyanin content in the mixed red/blue light (R57-B43) was 4.1-fold and 6.9-fold increased compared to the red LED and fluorescent light, respectively. With increasing the blue light ratio to 43%, the growth of lettuce was significantly decreased, while the relative chlorophyll content and Hunter's $a^*$ value was increased, indicating that the red/blue light ratio inversely affects on growth and anthocyanin pigment development. By changing light quality from red to red/blue mixed light source (R57-B43) for 9 days before harvest, the growth rate decreased compared to the continuous red light illumination, while the anthocyanin content dramatically increased compared to either red LED or fluorescent light. Whereas, when the light source was changed to red light, the growth rate was increased but anthocyanin content was reversely decreased. The result demonstrated that both growth and anthocyanin expression could be effectively regulated by shifting of light quality between red and red/blue mixed light source at a specific growth stage of lettuce in a plant factory.
