Antioxidant and Physicochemical Changes in Salvia plebeia R. Br. after Hot-air Drying and Blanching (열풍건조 및 데침 처리에 따른 배암차즈기(Salvia plebeia R. Br.)의 항산화 및 이화학적 특성 변화)

  • The aim of this study was to investigate physicochemical changes in Salvia plebeia R. Br. (SPA) upon hot-air drying and blanching. After hot-air drying and blanching, total polyphenol and flavonoid contents were reduced from 96.64 mg and 48.40 mg gallic acid equivalent/g to 29.70 mg and 22.10 mg quercetin equivalent/g, respectively. DPPH radical scavenging activities at $25{\mu}g/mL$ of SPA were 94.5% for ascorbic acid, 84.3% for hot-air dried SPA, and 59.7% for blanched SPA and there was no significant difference between those of hot-air dried SPA and ascorbic acid as a positive control. Total sugar contents were 7.187% and 6.104% for hot-air dried SPA and blanched SPA, respectively. During the blanching process, sucrose and maltose contents decreased, whereas glucose and fructose contents increased. Glucose and fructose were converted into citric acid in blanched SPA, whereas sucrose and maltose were converted into tartaric acid, malic acid, and succinic acid. Fourteen kinds of amino acids were found, but methionine, lysine, glycine, histidine, and cysteine were absent. The proportion of essential amino acids was 61.76% upon hot-air drying, which was a relatively high amount. In addition, hot-air drying resulted in 1.40 mg/100 g of norvaline as well as 39.00 mg/100 g of GABA, which are non-amino acids. Therefore, Salvia plebeia R. Br. can be used not only as vegetables but also as highly useful and various health functional foods with antioxidant effects and excellent nutrition.

Antioxidant Activities of Ethanol Extracts from Different Parts of the Black Raspberry (Rubus occidentalis) Obtained Using Ultra-sonication (초음파 처리에 의한 검정라즈베리 부위별 에탄올 추출물의 산화방지 활성)

  • This study was carried out to investigate the antioxidant effects of different parts (stems, leaves, and seeds) of the black raspberry for utilization as food materials. Different parts of the black raspberry were subjected to extraction via ultra-sonication extraction methods using water and ethanol at various concentrations (25, 50, 75, and 100%). Antioxidant capability of the extracts were determined by amounts of phenolic compounds, with flavonoid contents, radical scavenging activity, and reducing power. Irrespectively of ethanol concentration, extracts of stem showed the highest total phenolic compounds and antioxidant activities among different parts of black raspberry. The total phenolic compounds extracted from the black raspberry stem using 25 and 50% ethanol showed $348.21{\pm}5.40$ and $343.39{\pm}5.94mg/g$, respectively. Fifty percent ethanol extracts of the black raspberry stem showed the highest DPPH ($EC_{50}$ value: $60.89{\mu}g/mL$) and ABTS radical scavenging activities ($EC_{50}$ value: $82.57{\mu}g/mL$). Further, 25% ethanol extacts of the black raspberry stem ($0.263{\pm}0.004$) was found to have the highest reducing power. The highest antioxidant activity of black raspberry stem indicates that black raspberry stem may be useful source for functional food.

The anti-oxidant, whitening and anti-wrinkle effects of Castanea crenata inner shell extracts processed by enzyme treatment and pressurized extraction (효소처리 및 가압추출 공정을 이용한 율피 추출물의 항산화, 피부 미백 및 주름개선 효과)

  • In this study, the anti-oxidant, whitening, and anti-wrinkle effects of Castanea crenata inner shell extracts processed by enzyme treatment and pressurized extraction were investigated. The Castanea crenata inner shell was first hydrolyzed using celluclast, viscozyme, or hemicellulase. Then, it was subjected to pressure extraction for different durations (30, 60, and 120 min). The yields of the Castanea crenata inner shell extracts processed by different enzyme treatments followed by pressurized extraction for different times are in the range of 12.42-29.80%. The total polyphenol, flavonoid, and tannin contents of the C30m (celluclast enzyme and autoclave extracts at 30 min) extract were 15.48, 10.82, and 15.82 g/100 g, respectively. The total sugar content of the H120m(hemicellulase enzyme and autoclave extracts at 120 min) extract is 61.07 g/100 g. The 1,1-diphenyl-2-pycrylhydrazyl (DPPH) and 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activities of the C30m extract at $1,000{\mu}g/mL$ are 89.20, and 81.96%, respectively. The superoxide radical scavenging and ferric-reducing antioxidant power of the C30m extract at $1,000{\mu}g/mL$ are 67.63% and $1,324.79{\mu}M$, respectively. Further, the tyrosinase and elastase inhibition activity of the C30m extract at $1,000{\mu}g/mL$ are 61.32, and 61.06%, respectively. Our results indicate that the Castanea crenata inner shell extracts processed by enzyme treatment followed by pressurized extraction could have beneficial effects on facial skin and they should be considered for use in new functional cosmetics.

Physicochemical characteristics of Sengmaksan added with Liriope platyphylla roasted for different times (덖음 처리 시간을 달리한 맥문동을 첨가한 생맥산의 이화학적 특성)

  • This study investigates, the physicochemical characteristics of Sengmaksan (SM) prepared with Liriope platyphylla (LP) that had been roasted for different times (0, 30, 60, and 90 min, denoted as S-0, S-30, S-60, and S-90, respectively) The Hunter's color values such as lightness (L), redness (a), and yellowness (b) were the highest in S-0, while the lowest was found in S-90. The amount of soluble solid and reducing sugar content of S-60 were higher than the others. None of the samples exhibit significant differences in, their pH and acidity. The total content of phenolic compounds increased with the LP roasting time, but the total flavonoid and total anthocyanin contents of the SM decreased at the same time. The total ginsenoside (Ro, Rb2, Re, Rf, Rg1, Rg2, Rg3, Rh1, and Rh2) content did not show significant differences. The DPPH and ABTS radical scavenging activities increased according to the concentration, as well as with the LP roasting time. The ferric reducing antioxidant power (FRAP) showed trends similar to the radical scavenging activity, but it was more sensitive to the LP roasting time. From these results, the active ingredient in S-60 was higher, and the antioxidant activities of SM increased along with the roasting time of LP.

Analysis of antioxidant activities, beta-glucan, and nutritional contents by different strains of Volvariella volvacea (풀버섯 균주별 항산화 활성, 베타글루칸 및 영양성분 함량 분석)

  • The hot-water extracts of four strains of Volvariella volvacea [Vv (KMCC04386), Vv-Chi (KMCC04382), V9-21 (KMCC04380), and VG-19 (KMCC05115)] were prepared to determine their antioxidant activities, ��-glucan content, and nutritional content. Among the four V. volvacea strains, Vv strain showed the highest DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity (23.7%). The highest total polyphenol and total flavonoid contents (8.17 mg y/g and 3.46 mg QE/g, respectively) were observed in the Vv-Chi strain. The ferric reducing antioxidant power (FRAP) and reducing power were significantly higher in the Vv-Chi strain compared to those in the other V. volvacea strains (p<0.05). There were no significant differences in the nitrite scavenging activity among the four different strains (p<0.05). The ��-glucan content in the four V. volvacea strains ranged from 15.13-16.07%, and the VG-19 strain had the highest ��-glucan content (15.73%). The VG-19 strain also had the highest total amino acid (986.8 mg/kg) and essential amino acid (369.3 mg/kg) contents among the four V. volvacea strains. The results of this study showed that the Vv-Chi strain exhibited the highest antioxidant activity, while the ��-glucan and nutritional contents were higher in the VG-19 strain compared to those in the other strains of V. volvacea.

Analysis of Ingredients and DPPH, ABTS Activity for the Development of Cosmetic Raw Materials using 5 Kinds of Plants Native to Mt. Jiri (지리산 자생식물 5종의 화장품 원료개발을 위한 성분 및 DPPH, ABTS 활성분석)

  • Five species of plants (Clerodendrum trichotomum Thunb., Angelica dahurica (Fisch. ex Hoffm.) Benth. & Hook. f. ex Franch. & Sav., Caryopteris incana (Thunb. ex Houtt.) Miq., Lonicera japonica Thunb., and Parasenecio auriculatus var. matsumurana Nakai) native to the clean area of Mt. Jiri were selected. The collection period was from May to September 2021, and the five species plants were collected in their native habitats with flowers in full bloom. The collected plants were extracted with 70% EtOH, and 17 kinds of polyphenol components were analyzed. Next, flowers, leaves, stems, and roots were separated from plants, extracted with 70% EtOH for each part and experiments were conducted on DPPH, ABTS, total polyphenols, and total flavonoids. The results are as follows. 1. It was found that there were a total of 8 kinds of polyphenols contained in 5 species of plants that are native to Mt. Jiri. Among the polyphenol components, chlorogenic acid was contained in 4 species of plants, and caffeic acid was contained in 2 species of plants. 2. As a result, the DPPH radical scavenging activity was the best in the stem of P. auriculata and the C. trichotomum was good regardless of the specific part. It was found that the activity-scavenging activity was good in the flowers of A. dahurica and the leaves of L. japonica. 3. The highest ABTS radical scavenging activity was C. trichotomum Thunb., whose EC50 value was 38.73~66.28ppm. Next, the leaves and stems of L. japonica Thunb., A. dahurica and P. auriculata, and the leaves and stems of C. incana appeared in that order. 4. The highest total polyphenol content was 154.83mg GAE/g in the leaves of C. trichotomum, followed by about 130mg GAE/g in the flowers of C. trichotomum and P. auriculata. The lowest was 26.27mg GAE/g in the stems of A. dahurica.

Development of Optimal Cultivation Conditions and Analysis of Antioxidant Activities of Arctium lappa Sprout Vegetables (우엉 새싹채소의 재배환경 구축 및 항산화 활성 탐색)

  • This study was conducted to develop functional sprout vegetables with antioxidant effects using seeds of Arctium lappa. The seeds germinated vigorously under light at $25^{\circ}C$, reaching germination rate of 82% within 4 days. Germinated seeds were placed under darkness at various temperatures to force growth in length, and it was demonatrated that $20^{\circ}C$ was optimum temperature. Greening treatment reduced growth in length, but promoted growth of cotyledons. Harvested A. lappa sprout vegetables maintained freshness longer at $10^{\circ}C$, rather than $4^{\circ}C$. Ventilation holes in storage containers had no effects on storage periods. Antioxidant activity of vegetable that received greening treatment for 1-3 days was investigated, and it was shown that free radical scavenging effects and ferrous ion chelating effects was higher than those of commercially available brocoli, cauliflower, pea and bean sprout. Contents of total polyphenol and flavonoid were also higher, especially by 3 day greening. The longer the treatment, the more the inhibition on peroxidation of linoleic acid. Sprout vegetable of A. lappa had higher antioxidant activity compared with adult plant. In conclusion, sprout vegetable of A. lappa has great potentiality for use as one of sprout vegetables.

Study on the Quality Improvement of Acidic Citrus Juices, Citrus natsudaidai and Citrus grandis, by Bipolar Membrane Electrodialysis (전기투석용 bipolar 막을 이용한 하밀감 및 당유자 주스의 품질개선에 관한 연구)

  • Acidic citrus juices obtained from C. natsudaidai and C. grandis were electrodialyzed with bipolar and organic acid selective membrane (ion exchange membrane) cartridges. The pH levels of the acidic citrus juices gradually increased to 14.5% (C. grandis) and 25.2% (C. natsudaidai) by electrodialysis with the bipolar membranes, while levels remained consistent when organic acid selective membranes were applied. The total acidity levels decreased more with the organic selective membrane than with the bipolar membrane. Conductivity rose with the bipolar membranes while the value continued to fall rapidly with the organic selective membranes. Sugar and flavonoid contents remained relatively unchanged, without any significant differences before and after electrodialysis with each membrane. Also, ion contents were almost unchanged with the bipolar membranes and the electrolyte, $K_2SO_4$, as compared to rapid changes in sodium and potassium levels with the organic selective membranes and the electrolyte, $K_2SO_4$. In summary, the use of bipolar membranes provided juice with better sensory quality than that of the organic acid selective membranes.

Antioxidant and Antimicrobial Activities of Artemisia capillaris Thunberg (사철쑥의 항산화성과 항균성)

  • This experiment was carried out to obtain the basic information on processing for product of high quality goods in Artemisia capillaris. We investigated antioxidant and antimicrobial activities by harvesting date and plant parts in Artemisia capillaris. Contents of total polyphenol compounds and flavonoids were the highest in leaf, followed by capitulum and stem. Leaf on June 30 contained 76.7 mg/g DW total phenolic compounds and 78.2 mg/g DW flavonoids. As $RC_{50}$ value, that was, the concentration of sample required for 50% reduction of DPPH (2,2-diphenyl-l-picrylhydrazyl) absorbance, was very low as $5.42\;{\mu}g$ in leaf on June 30, antioxidant activity was the highest. In addition, $RC_{50}$ of BHA, BHT and ${\alpha}-tocopherol$ were $3.09\;{\mu}g$, $24.30{\mu}g$ and $2.87{\mu}g$, respectively. And capitulum had antimicrobial activities against Bacillus subtilis, Staphylococcus aureus, Enterobacter aerogenes, Citrobacter freundii, Vibrio vulnificus, Pseudomonas aeruginosa, Saccharomyces cerevisiae. By the way, leaf and stem rarely had antimicrobial activity. Antimicrobial activities of capitulum according to harvesting date were very various. Capitulum on August 30 had the highest antimicrobial activities against Bacillus subtilis, Staphylococcus aureus, Vibrio vulnificus, Saccharomyces cerevisiae and on July 30 against Enterobacter aerogenes, Citrobacter freundii, Pseudomonas aeruginosa.

Comparison of Antioxidant Effects by Different Extraction Methods in Flowers of Aster scaber, Aster maackii, Coreopsis lanceolata and Coreopsis tinctoria (참취, 좀개미취, 큰금계국 및 기생초 꽃의 추출방법에 따른 항산화 활성 비교)

  • Antioxidant substances were extracted from flowers of Aster scaber, Aster maackii, Coreopsis lanceolata and Coreopsis tinctoria using 2 types of extraction methods- ultrasonic wave and reflux, and antioxidant effects were compared. Higher yield of extract was obtained by ultrasonic wave method in all 4 species. Extraction time was different depending on species, but in all 4 species DPPH and ABTS free radical scavenging and ferrous ion chelating activity were higher or similar by ultrasonic wave than reflux method. Content of total polyphenol and flavonoid was mainly higher by ultrasonic wave than reflux method. Longer the treatment, the more the antioxidant activity and bioactive substances. Anti-lipid peroxidation of ultrasonic and reflux extracts obtained from flowers of Aster scaber and Coreopsis lanceolata were compared. Flower extract of Aster scaber obtained by reflux method showed the highest inhibition effect against peroxidation of linoleic acid, but extract of Coreopsis lanceolata obtained by 15-minute ultrasonic extract showed the highest effect. Ultrasonic wave extraction was more economical and efficient method than reflux extraction.
