Increment of Physiologically Active Compounds in Germinated Brown Rice Treated with Chitosan and its Effect on Obesity of Rat Fed a High Fat Diet (키토산을 처리한 발아현미의 기능성분 증대 및 비만에 미치는 영향)

  • This study was to investigated the changes of physiologically active components in germinated brown rice treated with chitosan (CGBR) and its anti-obesity effect in rat fed a high fat diet. Contents of physiologically active compounds such as total phenolic compounds, total dietary fiber, $\gamma$-aminobutyric acid (GABA), and total phytic acid in CGBR were significantly higher than those of traditional germinated brown rice (GBR). Lipase inhibitory activity of CGBR was higher than those of GBR and brown rice (BR). High fat diets containing CGBR, GBR, and BR were administered to three groups of male Sprague-Dawley rats for four weeks. All groups showed no significant difference in body weight, total abdominal fat, and plasma lipid levels. However, CGBR group appeared to have lower body weight gain and total abdominal fat level than other groups fed high fat diets containing GBR and BR. Total cholesterol and LDL-cholesterol contents in plasma of CGBR group were also lower than those of other groups. Thus, new germination method of brown rice using chitosan is a useful process, which utilizes plant defense responses to elevate the production of secondary metabolites and anti-obesity effect.

Effects of Extraction Methods of Medicinal Plants on Human Growth of Neuroblastoma SK-N-SH Cells (추출방법에 따른 한약재의 인체신경모세포 SK-N-SH 보호 효과)

  • Extraction methods of medicinal plants were evaluated for growth enhancing effects of human neuroblastoma SK-N-SH cells. Hot-water extraction (reflux for 5 hr), hot-water extraction post treatment (100$^{\circ}C$ or 120$^{\circ}C$, 90 min) and ethanol extraction (reflux for 5 hr) methods were applied to Angelica gigas, Rhemania glutinosa, Paeonia lactiflora and Cnidium officinale samples to extract their constituents. Cells were treated for 2 hr with various concentrations of extracts (0, 0.5, 1, 2, 4 ${\mu}g/{\mu}l}$ media) prior to $H_2O_2$ (250 ${\mu}M$) treatment for 2 hr to provide oxidative stress. Cell viability, caspase-3 expression and apoptosis were measured for cells treated with sample extracts. Hot-water extract exhibited a stronger growth enhancing and apoptosis protecting ability than other extracts. These activities were shown at less than 1 ${\mu}g/{\mu}l}$ concentration, and not greater than 2 ${\mu}g/{\mu}l}$ concentration. Hot-water extract contained more polyphenolic compounds than other extracts coming along with stronger antioxidant activity. The efficacy of antioxidant activity was stronger in the hot-water extract of Angelica gigas than other hot-water extracts of medicinal plants. These results suggest that hot-water extraction is an appropriate method to extract materials for growth enhancing and apoptosis protection of SK-N-SH cells, and hot-water extracts of Angelica gigas might be useful materials for protection from aging brain cells.

Comparison of Quality of Peeled Lotus Roots Stored in Various Immersion Liquids during Storage (침지액을 달리한 박피 연근의 저장중 품질 비교)

  • This study was carried out to investigate the efficacy of strong acidic electrolyzed acid water (SAEW; Strong acidic electrolyzed water, pH 2.76, ORP 1,128 mV, HClO 105.0 ppm) and low alkaline electrolyzed water (LAEW; Low alkaline electrolyzed water, pH 8.56, ORP 660 mV, HClO 73.8 ppm) as storing liquid for peeled lotus root. During storage at $5^{\circ}C$, it was showed that SAEW and LAEW inhibit growth of microorganisms until at least 5 days of storage. Total phenolic contents, polyphenol oxidase (PPO) activity, and color differences value $({\Delta}E)$ of peeled lotus roots stored in SAEW and LAEW were lower than that of one stored in TW (tap water). The hardness decrement of lotus roots stored in SAEW and LAEW were lower than that of lotus root stored in TW too. Contents of moisture crude protein crude fat, crude ash, crude fiber, and total sugars were gradually decreased during storage. Whereas vitamin C content of lotus root stored in 0.6% acetic acid was most rapidly decreased to 25% as compared with one of initial days of storage that of lotus roots stored in SAEW and LAEW was not decreased significantly as compared with one stored in TW. Sensory characteristic during storage was preferable on lotus root stored in SAEW to the other treatments.

Hot-water Extraction Condition of Gastrodia elata Blume by Response Surface Methodology (반응표면분석법을 이용한 천마의 열수 추출조건 설정)

  • In order to reduce the damage of effective component during processing, we carried out hot-water extraction monitering in Gastrodia elata Blume by response surface methodology. The extraction con야tions for vanillyl alcohol, vanillin and coumarin were optimized at 91.07, 93.73 and $96.30^{\circ}C$, for 2.64, 2.58 and 3.84 hr, at the concentration of 15.16, 21.56 and 19.88 mL/g, respectively. The optimal condition for extraction of total phenolics was acheved at $98.14^{\circ}C$, 3.20 hr and 18.63 mL/g. This study elicited regression formula for each variable, and superimposed the total optimal conditions of extraction for effective compounds and optimal condition for antioxidant characteristics dividing processing factors. It is shown that the predicted result under the extraction condition as follows: extraction temperature for $90{\sim}100^{\circ}C$, extraction time for $3{\sim}4hr$, and solvent volume of $15{\sim}20mL/g$ of sample.

Physicochemical Characteristics and Antioxidant Activities of Luffa cylindrica (L.) Roem (수세미오이의 이화학적 특성과 항산화활성)

  • Physiochemical characteristics of Luffa cylindrica (L.) Roem and antioxidant activities of its four extracts were analyzed. The chemical composition of L. Roem contained 93.69% moisture, 1.64% crude fiber, 0.52% crude protein, 0.27% crude fat, and 0.06% crude ash. The major minerals were potassium, phosphrous, and calcium. We also analyzed the major organic acids, acetic acid and succinic acid. In free amino acids, ${\beta}$-aminoisobutyric acid showed the highest concentration (100.74 mg/100 g), followed by phosphoethanolamine, urea, asparagine, and valine. Glutamic acid was the highest hydrolyzed amino acid with 1,039.99 mg/100 g followed by aspartic acid, lysine, and arginine. Four extracts from L. Roam, hot-water (LCH), cold-water (LCC), 80% ethanol (LCE), and methanol (LCM), were prepared. Total phenolic and flavonoid levels of LCE exhibited higher than three extracts. The antioxidant potential of extracts from L. Roem were investigated using DPPH, ABTS, and NBT assays. Of the four extracts, LCE had relatively high antioxidant capability on DPPH and superoxide anion radical scavenging activities (12.13%, 16.88%, and 26.61%). Based on the above results, it is suggested that an 80% ethanolic extract from L. Roem was a natural antioxidant material for health food and should be a good ingredient for functional food.

Analysis of Nutritional Compositions and Antioxidative Activities of Korean Commercial Blueberry and Raspberry (국내 시판 블루베리와 라즈베리의 영양성분 분석 및 항산화 활성)

  • The nutritional compositions and antioxidative activities of Korean commercial blueberry and raspberry were investigated. The proximate compositions were 10.47% and 22.67% in moisture, 2.66% and 2.64% in crude protein, 2.04% and 1.67% in crude fat, 81.36% and 70.19% in nitrogen free extracts, 1.48% and 0.85% in crude fiber, and 1.99% and 1.98% in ash of blueberry and raspberry, respectively. Total phenolics content were higher in blueberry (9.028 mg/g) than in raspberry (5.340 mg/g). Major elements of blueberry and raspberry were Ca (451.34 and 97.48 mg/100 g), K (355.40 and 215.20 mg/100 g), P (321.10 and 294.04 mg/100 g), and Na (137.58 and 137.67 mg/100 g). The total amino acid contents of blueberry and raspberry were 2,011.44 mg /100 g and 2,098.82 mg/100 g, respectively. Amino acid were mainly composed of glutamic acid, aspartic acid and leucine. The DPPH and ABTS radical scavenging activities of the 80% methanol extract from blueberry and raspberry were 88.67% and 62.77%, 76.34% and 30.53% at a concentration of 5 mg/mL. The 80% methanol extract from blueberry and raspberry showed considerable antioxidative activity against reducing power in dose-dependent manner. Antioxidative activities using $\beta$-carotene-linoleate and FTC method were twice higher in blueberry than raspberry.

Prediction of Optimal Leaching Conditions for Green Tea (녹차의 음용을 위한 최적 침출조건 예측)

  • Tea was known to have different taste and flavor with leaching temperature and time. This study was designed to optimize leaching condition for green tea bag that has been popular to consumers. Soluble solid, total phenolics, and total flavonoid content increased as leaching temperature and time increased. The ranges of maximum leaching condition for these components were $96.1{\sim}99.0^{\circ}C\;and\;5.7{\sim}6.8\;min$. As leaching temperature decreased and leaching time increased, however, electron donating ability (EDA) increased and showed the highest values at $65.3^{\circ}C$ and 7.2 min. The overall acceptability was maximal at $70.2^{\circ}C$ and 3.1min. Based upon the physicochemical and organoleptic properties, it was predicted that the optimal range of leaching conditions for green tea bag was $73{\sim}83^{\circ}C\;for\;5.3{\sim}6.3\;min$.

Comparison of Physiological Activities of Rubus coreanus Miquel during Maturation (복분자딸기의 성숙 단계별 생리활성 비교)

  • Total polyphenolic compounds, DPPH radical scavenging activity, superoxide dismutase (SOD), vitamin A, C, E, flavonoid and anthocyanin of Rubus coreanus Miquel during maturation were investigated to expand the utilization of Rubus coreanus Miquel as functional food material. Physiological activities of unripened, middle-ripened and ripened fruit of Rubus coreanus Miquel were examined. The ripened fruit included higher contents of total polyphenolic compound compared with those of the other sample groups, unripened and middle-ripened fruit. SOD of unripened, middle-ripened and ripened fruit were 59.15, 80.05 and 95.03%, respectively. Scavenging concentration ($SC_{50}$) of unripened and middle-ripened fruit for 0.4 mM DPPH (1,1-diphenyl 2-picryl hydrazyl) was 2.3 mg/mL by weight, but only 1.6 mg/mL for ripened fruit. The vitamin A content of the Rubus coreanus Miquel was found to be lower as the fruit ripened; however, vitamin E showed the reverse pattern. In addition Rubus coreanus Miquel is a fruit which has a high vitamin C content regardless of maturity. The flavonoid contents of Rubus coreanus Miquel were in order of ripened fruit ($49{\mu}m/mg$)>middle-ripened fruit ($16.4{\mu}m/mg)$>unripened fruit ($15.4{\mu}m/mg$). Also, the contents of anthocyanin in ripened fruit were $394.50{\mu}m/mg$, which were higher than those of middle-ripened ($12.43{\mu}m/mg)$ and unripened fruit ($7.89{\mu}m/mg$).

Antioxidative Activities by Water-Soluble Extracts of Morus alba and Cudrania tricuspidata (뽕나무(Morus alba)와 꾸지뽕나무(Cudrania tricuspidata)의 수용성 추출물에 의한 항산화 활성)

  • The antioxidative activities of water-soluble extracts from leaves and stem bark of Morus alba and Cudrania tricuspidata were compared in vitro experimental models. Antioxidative activities were measured by inhibition activity against lipid peroxidation of mouse liver microsome, and they were showed in the following order; stem bark of C. tricuspidata(53%)>stem bark of M. alba(43%)>leaves of C. tricuspidata(38%)>leaves of M. alba(43%). In antioxidative activities determined by thiocyanate method and TBA method, the water-soluble extract of stem bark of C. tricuspidata showed the highest antioxidative activity. The water-soluble extracts of leaves were slightly stronger than other extracts in DPPH$({\alpha},{\alpha}'-diphenyl-{\beta}-picrylhydrazyl)$ method. The concentrations of total polyphenolic compound from water-soluble extracts of leaves and stem bark of M. alba and C. tricuspidata were 1.32%, 1.28%, 1.34% and 1.30% respectively. In these results, the water-soluble extract of stem bark from Cudrania tricuspidata showed the highest antioxidative activity.

Study on Anti-oxidant and Anti-inflammatory Activity of Eggplant-cheongyeolsodokum (가지-청열소독음(淸熱消毒飮)의 항산화 및 항염 효능에 관한 연구)

  • The purpose of this study is verification of the anti-oxidant effect and anti-inflammatory effect of Eggplant-cheongyeolsodokum composed of 8 herbs (Solanum melongena L., Lonicera japonica Thunb., Glycyrrhiza uralensis Fisch., Ligusticum chuanxiong Hort., Angelica gigas Nakai., Coptis deltoidea C. Y. Cheng et Hsiao., Gardenia jasminoides J. Ellis., Forsythia suspensa Vahl) to confirm the possibilities as useful cosmetic material. We used the modified prescription of 'cheongyeolsodokum' contained in Korean traditional medical book 'Donguibogam' as composition of Eggplant-cheongyeolsodokum and their proportions. Eggplant-cheongyeolsodokum were extracted with hot water, 70% ethanol and then powdered. To confirm anti-oxidant effect, we investigated radical scavenging ability (DPPH, $ABTS^+$, superoxide), superoxide dismutase (SOD)-like activity, total polyphenolic contents. Also to confirm anti-inflammatory effect, we investigated inhibition effect of nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated RAW264.7 macrophages, and Inhibition effect of the expression of inflammatory-related proteins (iNOS, COX-2) by western blot analysis. As a result, Eggplant-cheongyeolsodokum showed good anti-oxidant and anti-inflammation effects, we suggest that it can be used as an active ingredient for cosmetics.
