Anti-photoaging Effects of Flavonoid glycosides from shizophragma hydrangeoides (바위수국으로부터 분리한 플라보노이드 배당체의 광노화 예방 효과)

  • 피부 노화는 피부와 피부 지지층 등의 광범위한 퇴행 과정을 말한다. 피부 노화의 원인은 흡연, 공해, 스트레스 등이 있지만, 그 중에서도 자외선(ultra violet, UV) 조사가 가장 큰 요인으로 꼽힌다. 반복적인 자외선 조사에 의해 진행되는 피부노화를 광노화라고 하며 그 가장 큰 특징으로는 콜라겐 섬유와 엘라스틴의 감소로 야기되는 주름을 들 수 있다. 본 연구에서는 제주에서 채집한 바위수국의 추출물 및 분획물의 항산화 및 자외선으로 인한 피부노화 예방(anti-photoaging) 효능을 확인하고, 활성물질을 분리하여 광노화 예방 효능과 그 메커니즘을 확인하였다. 실험에 사용된 바위수국은 범의귀과의 덩굴성 식물로 바위면이나 나무줄기 등에 붙어서 자라며, 한국(제주, 울릉도)과 일본에 분포한다. 바위수국 추출물과 분획물에서 총 페놀 함량. 총 플라보이드 함량, DPPH 및 ABTS 라디칼소거 활성의 항산화 실험 결과, 부탄올과 에틸아세테이트 분획층에서 강력한 항산화 활성이 관찰되었다. 또한 UVA를 조사한 인간 진피 섬유아세포 (human dermal fibroblast, HDF)데 대한 콜라겐 분해효소인 matrix metalloproteinase-1(MMP-1) 생성 억제 활성을 확인한 결과, 부탄올 분획층이 세포 생장 저해 없이 가장 우수한 효능이 확인되었다. 따라서 부탄올 분획층에서 주요 성분 분리 실험을 수행하여 총 4개의 화합물을 분리하였다; Chlorogenic acid (1), Quercetin-3-O-glucosyl-(1-2)-rhamnoside (2), Quercetin-3-O-xylosyl-(1-2)-rhamnoside (3), Quercitrin (4). 분리한 4개의 물질의 MMP-1 생성 억제 활성을 비교한 결과 화합물 2가 세포독성 없이 MMP-1 생성 억제 효능이 우수하였고, 이후 화합물 2의 광노화 예방 효능과 그 메커니즘을 확인하였다. 화합물 2는 MMP-1의 생성을 억제할 뿐만 아니라 procollagen type I의 생성을 증가시켰으며, MMP-1 생성에 관여하는 mitogen-activated protein kinase (MAPK)/activator protein-1 (AP-1) 신호전달경로를 하향 조절하며, 콜라겐 생성과 관련된 Transforming growth factor-β (TGF-β)/Smad 신호전달경로를 상향 조절하여 UVA에 의한 광노화 예방에 효능을 나타내었다. 이러한 결과들을 바탕으로, 바위수국은 항노화(anti-aging) 기능성 화장품 및 이너뷰티 기능성 식품 소재로 개발이 가능할 것으로 기대된다.

In Vitro Antioxidant and Anticancer Potential of n-Hexane Extract from Ginseng Marc (인삼박 n-Hexane 추출물의 in vitro 항산화 및 항암 활성)

  • A lipid-soluble extract in ginseng marc was prepared by n-hexane extraction to evaluate its antioxidant and anticancer potential. A hexane extract of ginseng marc (HEGM) possessed a 2,2-diphenyl-1-picryl-hydrazyl free radical scavenging activity which was related to the amount of total phenolics. Also, HEGM showed a potent inhibitory activity on human non-small cell lung cancer (A549, $GI_{50}=34.0{\mu}g/mL$) and colon cancer (SNU-C4, $GI_{50}=45.2{\mu}g/mL$) cells proliferation in vitro in a concentration-dependent manner as did the hexane extract of ginseng with $GI_{50}$ values of $20.0{\mu}g/mL$ in A549 and $37.0{\mu}g/mL$ in SNU-C4. These results imply that HEGM can be utilized as an antioxidant and anticancer substance.

Antioxidant Activities of Leaf, Stem and Root of Panax ginseng C. A. Meyer (인삼의 부위별 항산화 활성)

  • To develop a new functional material, leaf, stem and root of Panax gjnseng were analyzed in their antioxidant activities. Root and leaf of ginseng collected from 3 regions, exhibited inhibition activity as $45.2{\sim}54.3%\;and\;90.1{\sim}96.5%$ on peroxidation of low density lipoprotein and linoleic acid, respectively. Scavenging activities of stem, leaf and root of ginseng on superoxide anion radical were $35.6{\sim}76.1%,\;60.1{\sim}69.3%\;and\;-5.6{\sim}20.1%$, respectively. Total phenol contents of leaf, stem and root of ginseng were $147{\sim}200\;mg%,\;110{\sim}153\;mg%\;and\;61{\sim}86\;mg%$ respectively as tannic acid equivalent.

Chemical Compositions and Antioxidant Activity of Extract from a Extruded White Ginseng (압출성형 백삼추출물의 화학적 조성 및 항산화 활성)

  • Chemical components and antioxidative activities of white ginseng, red ginseng and extruded white ginseng (EWG) were evaluated. Extrusion condition was 20% moisture content, 100 and $140^{\circ}C$ barrel temperature. The results showed that total sugar and acidic polysaccharide contents of white ginseng powder were increased after extrusion treatment of which EWG at $140^{\circ}C$ barrel temperature had higher value than EWG at $100^{\circ}C$ barrel temperature. Free radical scavenging activity of EWG at $140^{\circ}C$ barrel temperature was 80.2 and 45.6% respectively. The butanol fraction of polyphenolic compound and acidic polysaccharide were $27.2{\pm}0.1\;mg/g$ and $217.6{\pm}0.7\;mg/g$, respectively. The ginsenosides were quantified by HPLC and the yield of ginsenoside-Rg3s and Rg3r were achieved by extrusion process.

Quality Characteristics of the Yanggaeng made by Chestnut Powder (밤 분말을 첨가한 양갱의 품질 특성)

  • The objective of this study was to asses the quality characteristics of yanggaeng prepared with different ratios of chestnut powder: 0%, 5%, 10%, 15%, and 20%. The moisture content of the control yanggaeng was higher than that of any other chestnut powder added groups. In terms of color, The lightness (L) and yellowness (b) decreased significantly, but redness (a) increased with increasing contents of chestnut powder. The total polyphenol content and DPPH free radical scavenging activities increased as the amount of chestnut powder. Texture profile analysis showed that the hardness, adhesiveness, cohesiveness, and chewiness were higher than the control group. The result of sensory evaluation test and the overall acceptability showed that the taste, color, flavor, chewiness, softness and overall acceptability were the highest in the 10~15% added group while the color and softness were identified no significant different group. Based on these results, it is suggested that yanggaeng with up 10~15% added chestnut powder can be developed as products.

Antioxidative Effects of Pine (Pinus denstifora) Needle Extracts. (솔잎 추출물의 항산화 효과)

  • Antioxidative activities of pine (Pinus denstifora) needle extracts were tested in vitro experimental models. The concentration of total polyphenolic compound of water extracts from pine needle was 1.61 %. In DPPH ($\alpha$, $\alpha'$-diphenyl-$\beta$-picrylhydrazyl) method, the electron donating activity of 0.1 % water extracts from pine needle was as high as BHT (0.05%, w/v). The antioxidative activity was measured by inhibition against lipid peroxidation of rat liver microsome, and this activity was shown in the following: 67.7% at 0.1% concentration >63.1% at 0.05% concentration > 28.2% at 0.01% concentration. In antioxidative activity determined by thiocyanate method against lipid peroxidation using linoleic acid, the antioxidative activities at all concentration of 0.01 %, 0.05% and 0.1 % were much higher than control during 7 days. In TBA method, the antioxidative activity was increased with increasing concentration until 6 days. These results support that water extracts from pine needle contain antioxidative compounds.

Antioxidant Activity of Water-soluble Extracts from Kalopanacis cortex (추출조건이 해동피 열수 추출물의 항산화 효과에 미치는 영향)

  • In this study, we established the optimal conditions for obtaining water-soluble extracts with antioxidant activity from Kalopanacis cortex. The extraction conditions tested included cold treatment, extraction time (1, 5, 10, 15, and 24 h), and extraction temperature (55, 75, and $95^{\circ}C$). The highest total polyphenol compound content from water soluble extracts ($612{\mu}g/mL$) was obtained at $95^{\circ}C$ for 15 h after cold treatment. The 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenger activity was also highest (78.8%) under these conditions, which was comparable to 70.2% of ascorbic acid. The hydroxyl radical scavenging activity (HRSA) was also highest (69.0%) under these conditions, stronger than 56.6% of ascorbic acid. These results may provide critical evidence supporting the use of Kalopanacis cortex as a source of antioxidants in functional foods.

Study on the Alleviation of an Alcohol Induced Hangover and the Antioxidant Activity by Mulberry Fruit (오디 추출물의 알코올 분해능 및 항산화 효과)

  • This study investigated the effects of mulberry fruit extract on antioxidant activities and the alleviation of an alcoholinduced hangover, which was measured with alcohol dehydrogenase(ADH) enzyme activity. The antioxidative capacity of each mulberry extract was measured by total phenolic compounds, electron donating ability, superoxide dismutase-like ability, thiobarbituric acid reactive substances, and nitrite scavenging ability. The 60% methanol extract yielded the highest total phenolic compounds. The electron donating ability of the 40% ethanol and 40% methanol extracts were 68% and 67%, respectively. The superoxide dismutase-like abilities of the 40% methanol and 40% ethanol extracts were 30% and 28%, respectively, when extracts were assayed at 2.5 mg/$m\ell$. The nitrite scavenging ability of the 60% ethanol and 40% methanol extracts were 98% and 97%, respectively. The 60% ethanol extract yielded the highest ADH.

Chemical Composition and Antioxidant Activities of Prunus salicina Formosa Produced in Gimcheon (김천산 자두 후무사의 화학적 성분과 항산화 활성)

  • The present study was designed to investigate the chemical composition and antioxidant activities of Prunus salicina Formosa produced in Gimcheon. The free sugar amounts in Prunus salicina Formosa were fructose>glucose>sucrose>maltose, and malic acid contents in Prunus salicina Formosa were the highest among all the organic acids found in the present study. Each Prunus salicina Formosa extract was obtained by treating Prunus salicina Formosa with distilled water, 80% ethanol, 60% acetone, and 80% methanol at 25 and $50^{\circ}C$. The highest contents of total polyphenol and flavonoid were observed in the 60% acetone extract and 80% ethanol extract, respectively. The 60% acetone extract exhibited the greatest DPPH radical scavenging ability, reducing power, and nitrite scavenging ability. However, SOD-like activity was not considerably different for all the extracts studied. The results from the present study could be helpful for developing antioxidant products using Prunus salicina Formosa produced in Gimcheon.

Antioxidant Activity of Vinegars Commercially Available in Korean Markets (국내 시판되는 식초의 항산화 활성)

  • The aim of this study was to determine the antioxidant compounds (polyphenolics and flavonoids) and to evaluate antioxidant activities using 2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical scavenging activity and reducing power in 19 vinegar samples. In general, antioxidant activities and antixoidant compounds in drinking vinegars were higher than those in seasoning vinegars. The highest polyphenolic content was found in balsamic vinegar (372.53 mg/100 mL) and also the highest flavonoid contents was in balsamic vinegar (114.40 mg/100 mL). Organic acid content in glacial acetic acid among 19 samples was found in highest amount. There was no relationships between antioxidant compounds/activities and organic acid content in vinegar samples. Therefore, it is assumed that antioxidant activity of vinegars is due to the polyphenolic and/or flavonoid contents of the vinegar samples.
