The Bacteriological Survey of the Main Streams in Kyong-Ju City - on Samonellae - (경주시를 관류하는 하천수에 대한 세균학적 조사 - 살모넬라에 대하여 -)

  • The studies on the standard plate count, most probable number(MPN) and isolation of the Enterobacteriaceae, especially Salmonella species from stream waters of Kyong-Ju city were carried out from June to July, 1988. The relationship between water temperature, weather and standard plate count were observed. The locations where all of the MPN was higher than 1,600 were Mun-chon bridge, Wol-song bridge, Go-hun bridge and the area adjoining the Hyong-san river. The higher the water temperature, the more the viable counts without rain. Bacteriological survey of the Salmonella species was performed with the total 80 specimens isolated from 16 areas which are the main streams in Kyong-Ju city. The author identified S strains(2.36%) of Salmonella typhi, 5 strains(2.3%) of Salmonella paratyphi, 2 strains(1.42%) of Salmonella paratyphi B, 63 strains(29.72%) of other Salmonella groups by biochemical tests and serotyping tests with slide agglutination.

Study on microbiological safety of simple processed agricultural products (유통 단순가공 농산물의 위생지표세균 및 식중독균 오염실태 조사연구)

  • The purpose of this study was to explore methods for efficient management of the quality and safety of simple processed agricultural products in Busan. We tested 258 samples of simple processed agricultural products for distribution of aerobic bacteria and coliforms, and identified food-borne pathogens. The average aerobic bacterial and coliforms counts were 7.1 and 4.1 log CFU/g in simple processed vegetables, 6.8 and 3.5 log CFU/g in dried vegetables, and 6.2 and 2.9 log CFU/g in simple processed fruits. Additionally Staphylococcus aureus, Salmonella spp., Campylobacter jejuni/coli and Listeria monocytogenes were not detected in any samples. However, Bacillus cereus, Clostridium perfringens and E. coli were detected in 41 samples (16.3%), 2 samples (0.8%), and 4 samples (1.6%), respectively. This analysis revealed that none of C. perfringens and E. coli isolates harbored pathogenic toxic genes. However, all of B. cereus isolates carried at least 1 toxin gene.

Changes of Nitrifying Bacteria Depending on the Presence and Absence of Organic Pollutant in Nak-Dong River (낙동강에서의 유기성 오염 유무에 따른 질화세균의 변화)

  • This study was performed at 2 sites of Nak-Dong River to investigate the changes of nitrifiers depending on the presence and absence of organic pollutants (due to the effluents of domestic wastewater treatment plant, WWTP). Conventional chemical parameters such as T-N, $NH_4$-N, $NO_2$-N, $NO_3$-N were measured and the quantitative nitrifiers at the 2 sites were analyzed comparatively by fluorescent in situ hybridization (FISH) with NSO190 and NIT3, after checking the presence of gene amoA of ammonia oxidizing bacteria (AOB) and 16S rDNA signature sequence for Nitrobacter sp. that belongs to nitrite oxidizing bacteria (NOB). Also ${\alpha}{\cdot}{\beta}{\cdot}{\gamma}$-Proteobacteria were detected using FISH to get a glimpse of the general bacterial community structure of the sites. Based on the distribution structure of the ${\alpha}{\cdot}{\beta}{\cdot}{\gamma}$-Proteobacteria and the measurement of nitrogen in different phases, it could be said that the site 2 was more polluted with organics than site 1. Corresponding to the above conclusion, the average numbers of AOB and NOB detected by NSO160 and NIT3, respectively, at site 2 [AOB, $9.3{\times}10^5$; NOB, $1.6{\times}10^6$ (cells/ml)] was more than those at site 1 [AOB, $7.8{\times}10^5$; NOB, $0.8{\times}10^6$ (cells/ml)] and also their ratios to total counts were higher at site 2 (AOB, 27%; NOB, 34%) than those at site 1 (AOB, 18%; NOB, 23%). Thus, it could be concluded that the nitrification at site 2 was more active due to continuous loading of organics from the effluents of domestic WWTP, compared to site 1 located closed to raw drinking water supply and subsequently less polluted with organics.

The Effect of Chitosan and Wood Vinegar Treatment on the Growth of Eggplant and Leaf Lettuce (키토산과 목초액 처리가 가지 및 잎상추의 생육에 미치는 영향)

  • From the experiments on the effect of chitosan and wood vinegar, which are environment-friendly materials, on the growth of eggplant and leaf lettuce, the following results were obtained. 1. The effect of chitosan and wood vinegar treatment on the growth of eggplant 1) There was no difference in soil component elements before and after treatment with chitosan and wood vinegar. The total number of microorganisms tended to increase after treatment with chitosan and wood vinegar, while the number of bacteria increased among microorganisms. However, there was no consistent tendency in the number of Acinomycetes, fungi, and trichodema between treatments. 2) The growth and the yield of eggplant increased compared with the control plot in both the chitosan-treated plot and the wood vinegar-treated plot. However, there was no significant difference between the treatments. The yield of eggplant per 10a increased in chitosan- and wood vinegar-treated plots compared with the controlled plot. There was no significant difference in treatments, however, the plot treated with 800 times-diluted solution showed a higher growth. 2. The effect of chitosan and wood vinegar treatment on the growth of leaf lettuce 1) There was no difference in soil component elements before and after treatment with chitosan and wood vinegar. The number of bacteria increased among microorganisms. However, there was no consistent tendency in the number of Acinomycetes, fungi, and trichodema between treatments. 2) The growth of leaf lettuce in both chitosan-treated plot and wood vinegar-treated plot increased compared with the control plot, however, there was no significant difference between the treatments. On the whole, the plot treated with greater concentration showed a higher growth.

Prediction of Shelf-Life of Cold Buckwheat Noodles Mixed with Vitamin D2 Enriched Siitake Mushroom and Seaweed Derived Calcium (비타민 D2 강화 표고버섯과 해조칼슘 첨가 냉면의 저장성 예측 - 연구노트 -)

  • Cold buckwheat noodles with 3% vitamin D2 enriched Siitake mushroom and seaweed derived calcium (Aquacal) added as functional ingredients were developed. For the prediction of shelf-life of these cold buckwheat noodles, classified as unsterilized cooked noodles, bacterial counts and sensory evaluations were examined during 8 days under refrigerated conditions (5oC) and 8 weeks of storage under frozen conditions $(-18^{\circ}C)$. Results of bacterial count showed that cold buckwheat noodles were microbiologically safe within 6 days at the storage temperature of $5^{\circ}C$ and the shelf-life of these noodles could be extended to 8 whole weeks when stored at $-18^{\circ}C$. Apparent qualities (off-flavor and fungi development) of cold buckwheat noodles kept in $5^{\circ}C$ were not decreased within 8 days. Sensory characteristics (gloss, mushroom flavor, bitterness, chewiness and elasticity) of noodles were not changed during 8 weeks at $-18^{\circ}C$.

새로운 Quinolone 항균제(Q-35)의 제 1 상 임상 연구

  • Q-35의 일회 및 반복투여 연구결과, 자타학 증상은 단지반복투여 피험자에서 중등도의 두통(1예)과 설사(3예)가 관찰되었으나 이들 설사증례는 장내 세균총 검사상 복감염에 의한 것이 아니었다. Q-35 투여 후 Q-EEG 및 평행기능의 장애 등은 관찰되지 않았으며 임상화학 검사상 반복투여 3예에서 SGOT, SGPT의 경미한 상승을 보였으나 이러한 변화는 정상 범위내에서의 변화이었다. Q-35의 반복투여에 따라 투약 3일에서 8일에 걸처 장내세균총은 일부 호기성 및 혐기성 세균총의 감소를 보였으나 투약종료 10일 후에는 투약전 상태로 회복되었다. Q-35는 투여량의 약 70 % 가 24시간 뇨중으로 배설되었으며 일회 및 반복투여의 결과 용량의존적인 동태양상은 관찰할 수 없었다. 50 mg에서 400 mg까지 일회 투여시 5.6-7.1 시간의 혈장반감기를 보였으며, 반복투여 시험에서는 평균 5.6 $\pm$ 0.7 시간의 반감기를 보였다. Q-35는 타액내로 신속히 이행되었으며 타액내 AUC는 혈장 AUC의 약 75 %에 해당하였고, 식사에 의해 약간의 흡수속도지연(Cmax 0.4 시간지연)과 공복시에 비해 82 %의 상대적 생체이용율을 보였다.

Phylogenetic diversity of bacterial communities in a gray solar saltern and isolation of extremely halophilic bacteria using culturomics (토판염전 결정지 내 세균군집의 계통학적 다양성 및 Culturomics법을 이용한 고도 호염균의 분리)

  • In this study, we investigated the phylogenetic diversity of the bacterial community and isolation of extremely halophilic bacteria using culturomics in a gray solar saltern. The number of bacterial living cells, enumerated in a gray solar saltern by direct fluorescence microscopy was three to four orders of magnitude greater than those enumerated by plate counts, suggesting the distribution of 'viable but non-culturable bacteria'. The biodiversity of bacterial communities in a gray solar saltern was investigated by pyrosequencing, 1,778 OTUs of bacteria were comprised of 18 phyla 46 classes 85 orders 140 families 243 genera with 6.16 diversity index. Archaea communities were composed of 3 phyla 6 classes 7 orders 7 families 38 genera with 4.95 diversity index from 643 OTUs. Totally 137 isolates were isolated by 59 different cultural methods based on culturomics considering culture media and conditions suitable for the growth of extremely halophilic bacteria. Phylogenetic analyses of extremely halophilic isolates based on 16S rRNA gene sequences, extremely halophilic isolates were composed of 4 phyla and 11 genera. Haloterrigena and Haloferax can be successfully isolated from culturomics. These culturomics were effective methods for collection of diversity of extremely halophilic bacteria.

Microbiological changes and quality characteristics of dried persimmon by chlorine dioxide gas fumigation treatment (이산화염소 가스 훈증 처리에 따른 곶감의 미생물학적 변화 및 품질특성)

  • This study was conducted to investigate the effect of chlorine dioxide fumigation as a substitute for sulfur fumigation which has been used as a method to prevent the quality change of persimmon during storage and distribution process. Dried persimmons were treated with chlorine dioxide gas concentration (0, 15 30, and 45 ppm) and time (0, 15, 30, and 45 min) and microbiological changes, texture properties and color of the treated samples were investigated during storage at room temperature. Total aerobic bacteria, yeast and mold numbers after chlorine dioxide gas fumigation were decreased when compared with the control group. The inhibitory effects of total aerobic bacteria, yeast, and mold were observed during storage. The texture properties and color value of dried persimmons were not affected by chlorine dioxide gas fumigation concentration and time. There was no difference in quality between chlorine dioxide gas fumigation treatment group and control group. These results suggested that chlorine dioxide gas fumigation treatment can be utilized as a processing technique to secure microbiological storage stability of dried persimmons.

The Addition Effect of Lentil and Opuntia ficus-indica on Storage Stability for Sausages (렌틸과 백년초의 첨가가 소시지의 저장 안정성에 미치는 영향)

  • This study was carried out to clarify the addition effects of lentil and Opuntia ficus-indica instead of nitrite on storage stability for sausages. The antioxidant activity, acid value, peroxide value, volatile basic nitrogen and total aerobes were determined. As the concentration of lentils and Opuntia ficus-indica increased, total polyphenol and DPPH radical scavenging effects increased significantly. The F6 batch which had the maximum concentration of lentil and Opuntia ficus-indica, had the highest polyphenol value and DPPH radical scavenging effects. In addition, lentil and Opuntia ficus-indica supplementation lowered acidity, peroxide value and volatile basic nitrogen of sausages. Therefore, addition of lentil and Opuntia ficus-indica showed inhibition effects on lipid oxidation and protein deterioration of sausages. Also, Opuntia ficus-indica dropped pH, while lentil raised the pH. Bacterial counts didn't have an effect with the concentration of lentil and Opuntia ficus-indica on initial storage day. Overtime, the bacterial growth in the supplemented sausage group was less than the growth in no supplementation sausages. These results indicate that the addition of lentil and Opuntia ficus-indica increase antioxidative activity and inhibit lipid oxidation, protein deterioration and microbial growth. Lentil and Opuntia ficus-indica are useful materials in sausage production.

Identification of bacteria from the peri-implant sulcus of orthodontic mini-implants using 16S rDNA clone library (16S rDNA 클론 library 제작 및 핵산염기서열 결정을 통한 교정용 미니임플랜트 주위 열구의 세균 동정)

  • Objective: The purpose of this study was to compare the bacterial flora at the peri-implant sulcus of the orthodontic mini-implant placed in the alveolar mucosa with the bacterial flora at the adjacent healthy gingival sulcus. Methods: Two plaque samples from 7 patients were collected by inserting paper points into the sulcus between the mini-implant and ligature wire connected to the mini-implant head and inflamed alveolar mucosa, and from the gingival sulcus of a healthy tooth adjacent to the mini-implant. Results: Using 16S rDNA clone library, the 24 kinds of bacteria including Haemophilus aphrophilus, Sphingomonas species, Capnocytophaga species, Prevotella melaninogenica, Lachnospiraceae species, Porphyromonas species, Neisseria flava were identified only from the sulcus around the mini-implant. These bacteria constituted only 9.2% of total clones, and the bacteria identified from both the sulcus around mini-implants and the gingival sulcus constituted 80.4% of total clones. Of these bacteria, clones of Prevotella species, Atopobium rimae, Veillonella species, Streptococcus intermedius/constellatus, Streptococcus salivarius were more frequently isolated from the peri-implant sulcus. Conclusion: This study suggests that a broad epidemiological study is needed to find causative bacteria which induce inflammation from the peri-implant sulcus.
