Enhanced antimicrobial effectiveness of Omija (Schizamdra chinesis Baillon) by ClO2 (chlorine dioxide) treatment (오미자 수확 후 이산화염소수를 이용한 표면 세척에 따른 미생물 저감 효과)

  • The purpose of this study was to investigate antimicrobial effectiveness of Omija (Schizandra chinensis Baillon) treated with $ClO_2$ (chlorine dioxide) concentration (10, 15, 20, 25 and 30 ppm), washing time (30, 60 and 90 sec) and multiple proportion (x1, x2, x3 and x4). The seven groups were divided into control (Omija without washing water treatment), W-T (Omija treated by tap water ($20{\pm}1^{\circ}C$) for 30 seconds), $ClO_2$-10 (Omija treated by 10 ppm $ClO_2$), $ClO_2$-15 (Omija treated by 15 ppm $ClO_2$), $ClO_2$-20 (Omija treated by 20 ppm $ClO_2$), $ClO_2$-25 (Omija treated by 25 ppm $ClO_2$), $ClO_2$-30 (Omija treated by 30 ppm $ClO_2$), and then they were detected number of total aerobic bacteria, yeast and mold. The rate of inactivation was found, for microorganisms of total aerobic bacteria, yeast and mold, to increase with a increase of $ClO_2$ treatment concentration and multiple proportion. No total aerobic bacteria, yeast and mold in $ClO_2$-30 sample treated for 30 sec, $ClO_2$-15 treated for 60 sec and $ClO_2$-10 treated for 90 sec were detected, and in $ClO_2$-30 Omija with multiple proportion ${\times}1$ (Omija : 30 ppm $ClO_2$ solution ratio was 1:1 (w/w)), $ClO_2$-20 with ${\times}2$ (Omija : 20 ppm $ClO_2$ solution ratio was 1:2 (w/w)) and $ClO_2$-15 with ${\times}4$ (Omija : 15 ppm $ClO_2$ solution ratio was 1:4 (w/w)) respectively.

A comparison of community structure and denitrifying ratio for denitrifying bacteria dependent on agricultural methods and seasons (농법과 계절에 따른 탈질세균의 군집 구조와 탈질율 비교)

  • We studied soil composition, $N_2O$ production, a number of denitrifying bacteria, community structure and T-RFLP patterns of denitrifying bacteria dependent on agricultural methods with the change of seasons. Analyses of the soil chemical composition revealed that total carbon and total organic carbon contents were 1.57% and 1.28% in the organic farming soil, 1.52% and 1.24% in the emptiness farming soil, and 1.40% and 0.95% in traditional farming soil, respectively. So, the amount of organic carbon was relatively high in the environment friendly farming soils than traditional farming soils. In case of $N_2O$ production, the amount of $N_2O$ production was high in May and November soils, but the rate of $N_2O$ production was fast in August soil. The average number of denitrifying bacteria were $1.32{\times}10^4MPN{\cdot}g^{-1}$ in the organic farming soil, $1.17{\times}10^4MPN{\cdot}g^{-1}$ in the emptiness farming soil, and $6.29{\times}10^3MPN{\cdot}g^{-1}$ in the traditional farming soil. It was confirmed that the environment friendly farming soil have a larger number of denitrifying bacteria than the traditional farming soil. As a result of the phylogenetic analyses, it was confirmed that six clusters were included in organic farming soil among total 10 clusters. And the result of PCA profile distribution of T-RFLP pattern on agricultural methods, the range of distribution showed wide in the organic farming method, relatively narrow in the conventional farming method, and middle in the emptiness farming method. Therefore, we could concluded that the distribution and the community structure of denitrifying bacteria were changed according to the agricultural methods and seasons.

Studies on Storage Characteristics of Tofu with Herb (허브첨가 두부의 저장 특성)

  • Tofu was prepared with various herbs (green tea, rosemary, lavender and thyme) and the tofu quality was investigated during storage. In the measurement of tofu color, the L and b values increased during storage period. Especially, the b value was very high at 8 days after storage. The pH value of tofu increased until 6 days of storage, but then decreased. The turbidity gradually increased until 6 days of storage and rapidly increased at 8 days of storage. The microorganism count of herb tofu was lower an that of control tofu during storage. Especially green tea tofu showed the lowest psychrotrophic microorganism count as $1.3{\times}10^8\;CFU/g$. In addition, lavender tofu showed the lowest aerobic mesophilic microorganism count $(2.0{\times}10^7\;CFU/g)$ at 8 days of storage. In texture analysis, hardness and chewiness of herb tofu increased with increasing storage period. Springiness increased to 2 or 4 days of storage but decreased after 6 or 8 days. Therefore, herb tofu is expected to have good quality physiologically as well as microbiologically.

Reduction of Pathogenic Bacteria in Organic Fertilizer using Gamma-Irradiation (감마선 조사를 이용한 유기질 퇴비의 병원성 미생물 저감화)

  • Organic compost is a useful fertilizer for organic farming. However, it poses a microbiological hazard to the farm produce because most of the compose are originated from excrementitious matters of domestic animals. Irradiation was performed to improve microbiological safety of organic compost and the effectiveness of gamma-irradiation for inactivating Salmonella typhimurium and E. coli was investigated. Total aerobic bacteria and coliform bacteria in 16 produce or commercial compose were ranged from $10^5\;to\;10^7\;CFU/mL\;and\;0\;to\;10^3\;CFU/mL$, respectively. All coliform bacteria in the composts were eliminated by irradiation at 5 kGy, while about $10^2\;CFU/mL$ of total aerobic bacteria survived up to 10 kGy of irradiation. In the inoculation test the test organisms (inoculated at $10^7\;CFU/mL$) were eliminated by irradiation at 3 kGy. $D_{10}$ values of inoculated Salmonella typhimurium and E. coli in the compost were $0.4{\pm}0.05\;and\;0.39{\pm}0.03kGy$. It was considered that $3{\sim}5kGy$ of gamma irradiation was effective for radicidation (radiation sterilization of pathogenic microbes) of organic fertilizer.

Biological Hazard Analysis of Paprikas, Strawberries and Tomatoes in the Markets (유통중인 파프리카, 딸기 및 토마토의 생물학적 위해요소 분포 조사)

  • The consumption of "ready-to-eat" agricultural products is recently increasing and the safety of these agricultural products is forefront of public concerns. The 120 samples of paprikas, strawberries and tomatoes, which are the representative exported agricultural products, were purchased at the department stores and discount stores in Daejeon. And we determined the microbiological and parasitological contamination level of these agricultural products using culture media, multiplex PCR, commercial bacterial detection kit and microscopy, and also evaluated the decontamination method. Mean counts of total aerobic bacteria from these agricultural products ranged from $1.3{\times}10^4$ CFU/g to $1.8{\times}10^5$ CFU/g, and mean counts of coliforms ranged from $1.4{\times}10^3$ CFU/g to $9.6{\times}10^3$ CFU/g. There was no significant difference in the level of bacterial contamination between the agricultural products from department stores and the ones from discount stores. Strawberry showed the highest contamination level for the bacteria and we also found the unidentified parasite eggs. Enterobacter cloacae was the most frequently isolated bacteria strain, but no food poisoning pathogenic bacteria except Staphylococcus aureus was isolated from the products by multiplex PCR. Compared to unwashed products, tab water-washed ones showed 80% decrease of the counts of total aerobic bacteria on the agricultural products, and the rates decreased more by incorporating detergent or ultrasonic wave treatment. We concluded that the biological contamination levels among paprikas, strawberries and tomatoes were the highest in strawberries, but there were not significant difference according to distribution systems.

Antibiotic Susceptibility to Isolated Bacteria and Fungi from the Indoor-air (실내 공기중에서 세균 및 진균의 분포와 분리균의 항생물질 감수성)

  • This investigation was performed to isloate and identify the total bacteria, Staphylococcus spp. and fungi from the indoor air exposed for 30 minutes on the blood agar plate at the 27 places in a hospital. Antibiotic susceptibility tests of the isolated bacteria were also studied. The mean numbers of total bacteria, Staphylococci spp. and fungi were 26, 17, and 2 in the summer and 19, 8, and 2 in the winter, respectively. Staphylococcus epidermidis was the most common isolated bacteria, and the next was Staphylococcus aureus, Aerococcus spp., Micrococcus spp., and Bacillus spp. from the indoor-air of hospital. Aspergillus spp., Cephalosporum spp., Curvularia spp., penicillium spp., and Phialophora spp. was frequently isolated from the indoor-air of hospital. The 109 strains of isolated Staphylococcus epidermidis sho-wed resistance to tetracycline(45.0%), methicillin(40.2%), erythromycin(31.2%), and kanamycin(24.8%). The 76 strains of isolated Staphylococcus aureus showed resistance to erythromycin(71.7%), methicillin(63.2%), kanamycin (44.7%), tetracycline(39.5%), and ampicillin(32.9). The 67 strains of isolated Aerococcus spp. showed resistance to erythromycin(26.9%), methicillin(25.4%), kanamycin(22%), and tetracycline(22.4%). The 48 strains of isolated Micrococus spp. showed resistance to tetracycline(27.0%), methicillin(22.9%), erythromycin(22.9%), and kanamycin(20.8%).

Detection of Denitrifying Bacteria in Groundwater by PCR (PCR을 이용한 지하수 내의 탈질화 세균의 검출)

  • Groundwater samples were collected at 6 sites in Seoul area. DNA extraction from the sample was performed by the boiling method. Samples were boiled with guanidinium thyocyanate and phenol-chloroform. One set of primer was designed for amplification of 16S rDNA. For detection of denitrifying bacteria in groundwater sample, the author used primer sets consensus regions in gene sequences encoding the two forms of nitrite reductase (NIR), a key enzyme in the denitrification pathway. Two sets of PCR primer were designed to amplify $cd_1$-and Cu-nir. We confirmed the existence of denitrifying bacteria in 3 sites using $cd_1$-nir primer and in 4 sites using Cu-nir primer.

Effect of Dietary Streptococcus faecium on the Performances and the Changes of Intestinal Microflora of Broiler Chicks (Streptococcus faecium의 급여가 육계의 성장과 장내 세균총 변화에 미치는 영향)

  • Effect of Streptococcus faecium(SF) and an antibiotic, Colistin(Col), supplemented to diets singly or in combination, on the performances and changes of intestinal population of microflora of broiler chicks studied. A total of 252, day-old chicks(Arbor Acre) of mixed sex(M:F=1:1) were alloted into six groups. A diet with no Col and SF was referred as a control diet. The basal diets were added with two levels of SF, 0.04 and 0.08%, singly or in combination with Col 10ppm Another diet was prepared by adding only Col 10 ppm. Numbers of the microorganism in diets added with SF 0.04% and 0.08% were 7$\times$10$^{4}$ and 1.4$\times$10$^{5}$ /g diet respectively The diets consisting of corn and soybean meal as major ingredients were fed for a period of seven weeks . During the feeding trial, fresh excreta were sampled at the end of every week in a sterilized condition to count microbial changes from each dietary group. Microbial changes of large intestine were also measured from nine birds sacrificed at the end of the 4th and 7th weeks each time per dietary group. Excreta from all the groups were also collected quantitatively at the end of 3rd and 6th weeks to measure digestibility of the diets, At the end of 7th week, nine birds from each group were also sacrificed to measure weight changes of gastrointestinal tracts . Average body weight gains of broilers fed the diets added with SF 0.08% (2.37kg) or SF 0. 08%+col 10ppm(2.34kg) were significantly larger than that of the control(2.18kg). The weight gains of the other groups were not statistically different from that of the control Feed/gain ratios of the supplemental groups were better than that of control (P<0.05) except that of birds fed the diet added only with SF 0.04%. Digestibilities of nutrients such as dry matter, crude protein, crude fat and total carbohydrates were not altered by the consumption of the diets added with SF and/or Col throughout the whole feeding period. As expected, the numbers of Streptococci in the excreta from birds fed diets added with SF increased significantly with a statistical difference between groups with SF 0.04% and SF 0.08% most of the time. However. addition of Colistin to the diets supplemented with SF did not give any effects on the number of the microorganism. Numbers of coliforms in the excreta were apparently reduced by feeding the diets added with SF and/or Col(P<0.05). There were, however, no additive effects observed between the two feed additives in this regard when supplementing Col to the SF diets. Distributions of intestinal microflora exhibited exactly the same pattern as those of the excreta. Length of small intestine of the birds fed diets added with SF 0.08% with or without Col 10 ppm became significantly longer with a range of about 10% than those of the birds fed diets without SF. However, the empty weight of the small inestine of the former group was lighter than that of control These changes resulted in a significant reduction in weight/unit length of the intestine of the birds fed diets supplemented with Col and SF singly or in combination. In overall conclusion, diet added with SF 0.08% appeared most effective in improving broiler performances. Colistin added at a level of 10ppm was not beneficial at all in itself or in combination with SF in terms of broiler performances or changes of intestinal microflora population. The efficacy of SF and Col could be attributed to the changes of wall thickness of the small intestine.

Studies on Bacterial and Fungal Contamination in the Herbal Medicines (한약재에서의 세균과 진균 오염에 관한 연구)

  • The study has been done for about two months through June 2 to July 30 of 2010. The study subjects are three herbal-pharmaceutical companies located in Seoul. Each of them purchased thirteen types of medicinal herbs, then the study did analysis for microbial contamination status of bacteria and fungi. Here, the study focuses on settling out fundamental data bases regarding the investigation standards of microbial contamination. As comparing the study results with contamination limits of bacteria and fungi which are represented by $10^7$ CFU/g and $10^4$ CFU/g in number respectively, the total percentage of fungi contamination which is 12.8% is higher than that of bacteria is only 7.7%. In the DNA homology analysis regarding 16S rRNA gene, 117 of colonization have been selected as study subjects. Including B. cereus composing of resistant spores, soil microbes account for approximately 96.6%. It indicates that it is important to establish collection and preservation systems in handling medicinal herbs. Also, it is critical to manage microbial contamination limits. In conclusion, the study proposes the needs to study on possible mingling of bacteria and fungi in manufacturing process, and microbial contamination status in medicinal herbs.

Impacts of Soil Type on Microbial Community from Paddy Soils in Gyeongnam Province (경남지역 논 토양 유형에 따른 미생물 군집 변화)

  • This study evaluated the soil microbial communities by fatty acid methyl ester (FAME) method in soils (6 sites for immatured paddy, 9 sites for normal paddy, and 5 sites for sandy paddy) in Gyeongnam Province. The soil microbial biomass carbon content in normal and sandy paddy were 1,235 and $441mg\;kg^{-1}$, respectively, showing the soil microbial biomass carbon content in normal paddy was higher than that in sandy paddy. The soil organic matter contents $33g\;kg^{-1}$ of immatured and normal paddy were higher than sandy paddy $18g\;kg^{-1}$ (p<0.05). The communities of total bacteria and Gram-negative bacteria in normal paddy were significantly higher than those in sandy paddy (p<0.05). Total bacteria communities should be considered as a potential responsible factor for the obvious microbial community differentiation.
