Expression of Rice Small HSP Enhances Thermotolerance of Escherichia coli under Heat Stress (벼 Small HSP의 발현에 의한 대장균의 고온 stress 하에서의 내성의 증가)

  • A cDNA encoding rice chloroplast small HSP, Oshsp21, was introduced into Escherichia coli using the pET expression vector to analyze the possible function of Oshsp21 under heat stress. We compared the viability of E. coli ${\lambda}BL21$ (DE3) cells transformed with recombinant plasmid containing Oshsp21 with the control E. coli cells transformed with pET28a vector under heat stress after IPTG induction. Upon heat treatment at $50^{\circ}C$, those cells that expressed Oshsp21 showed improved viability compared with control cells. When the cell lysates from E. coli transformants were heated at $55^{\circ}C$, the amounts of proteins denatured in the control and pEhsp21-transformed cells were about 60% and 35% of total cell proteins, respectively. These results indicate that rice chloroplast small HSP function as a molecular chaperone in cells.

  • Analysis of proteome in amniotic fluid was performed by 2-D PAGE (polyacrylamide gel electrophoresis). Proteins in amniotic fluid were separated by centrifugation and solubilized in buffer solution for IEF, using an IPG strip of pH 4-7L. Both a homogeneous slab gel of 12.5% and a gradient gel of 8-18%, were used. After 2-D PAGE, spots were stained with silver nitrate and picked up for in-gel digestion. Digested peptides were analyzed by MALDI-TOF and proteins were further identifical. More protein spots were detected in the gradient gels and a protein not previously reported was identified.

Radurization of Packaged English Sole Fillets

  • English sole (Parophrvs vetulus) fillets packaged in polymylar bags were irradiated at 500 Krad, using a Cobalt-60 irradiator and the accumulation of spoilage indices substances and bacterial growth in the irradiated and unirradiated samples were measured during 36 days of storage at 0-2$^{\circ}C$. A casein agar plate technique was developed for a direct enumeration of proteolytic bacterial population, thus enabling the determination of relative proportion of proteolytic bacteria in the total microflora at each storage interval. Irradiation at 500 Krad resulted in a ten fold reduction of microflora and throughout the storage period the level of microflora lagged behind that of the unirradiated, by as much as one thousand fold. This was accompanied by a remarkable suppression of TVB and TMA accumulation in the irradiated, never reaching a spoliage level. Proteolytic bacterial population also was reduced to below one per cent of the total viable count and remained so throughout the storage period, while proteolytic bacteria in the unirradiated increased proportionately with the storage, comprising 85.5% of the total microflora by the twenty-second day. This selective removal of proteolytic bacteria must account for the reduced rate of proteolysis occurred in the irradiated during the storage.

Protein Concentrate from Ginseng Leaf and its Nutritive Value (인삼엽 단백질의 추출 및 영양가)

  • This study was investigated to evaluate the feasibility of protein concentrate for human food from ginseng leaf. The protein concentrate was prepared from ginseng green leaf by treating with cold acetone , followed by protein extraction with 0.2% NaOH containing 0.5% 2-mercaptoethanol and 0.5% sodium dodecyl sulfate. Proximate composition of the ginseng leaf protein concentrate (LPC) showed that fat and ash was less than 1%, protein was about 75%, total sugar and total saponin was 5% and 1.2%, respectively. As compared to the provisional amino acid pattern reported by FAO/WHO, ginseng LPC was found to be poor in S-containing amino acids, which were the first limiting amino acid. The amino acid score and E/T ratio of ginseng LPC were 43.1 and 3.02, respectively. Digestibility of ginseng LPC by pepsin and trypsin was lower than that of milk casein.

The Effect of Level of Dietary Protein on Kidney Development and Function in Growing Rats (식이내 단백질 수준이 성장기 흰 쥐의 신장 기능에 미치는 영향)

  • This study was performed to investigate the effect of protein intake on kidney development and function in growing rats. Fourty-two male Spraque-Dawley rats of weighing $97.5\pm1.9g$ were divided into 3 groups and given 5%, 15% or 50% casein diets for 6 weeks. Body weight gain was higher in the 50% group. The kidney weight was selectively affected more by the level of dietary protein compared to the other organs. DNA and RNA content were significantly higher in the 15% and 50% groups than in the 5% group but the differences disappeared when DNA and RNA were expressed per g of kidney weight. Protein and protein/g kidney content were increased with increasing level of protein in diet. GFR/animal and GFR/100gB. W. were significantly higher in the 50% group compared to the 5% and 15% groups. There was no differences in PAH clearence and RBF. Osmolality was not affected by dietary protein level. BUN and urinary nitrogen excretion were increased with the increasing dietary protein level. Although urinary Ca excretion was not significantly difference among 3 groups, the rats in the 5% group showed 30% less Ca excretion compared to the other groups. Above results suggest that dietary protein level has a great effect on the kidney weight and GFR in growing rats. Especially the hyperfiltration inhanced by high protein diet may accelerate the kidney senescence.

The Effects of Ovoinhibitor on nyoblast Digerentiation (근세포 분화에 미치는 ovoinhibitor의 영향)

  • 단백질 분해효소와 ovoinhibitor가 계배근세포 융합에 미치는 영향을 알아보기 위하여 세포 운합지수의 측정, Western blot 분석법에 의한 ovoinhibitor의 세포 내농도 측정 및 단백질 분해효소의 활성도를 조사하였다. 세포 배양 30시간과 72시간에 처리된 ovoinhibitor는 상당히 근세포 융합 억제시켰다. 근세포 내 ovoinhibitor의 농도는 배 양 48시간에서 가장 높았고, 배양이 진행됨에 따라 계속 떨어져서 대체로 융합지수가 올라갈수록 그 농도가 낮아지는 경향을 보였다. 근세포 분화가 진행됨에 따라 총 단백질 분해효소의 환성도는 계속 증가하였는데 특히 배양 48시간에서 72시간 사이의 급격한 증가를 나타내는 시기는 세포 융합이 가장 활발히 일어나는 시기와 일치했다. 이상의 결과는 단백질 분해효소 저해제가 근세포 분화에 상당한 영향을 끼침을 보여 주며, 단백질 분해효소와 단백질 분해효소 저해제는 하나의 조절단위로써 근세포 분화에 어떤 작용을 할 것으로 사료된다.

A Study of Protein Nutritional Status and Bone Metabolism of Postmenopausal Vegetarian Women (채식을 하는 폐경 후 여성의 단백질 영양상태와 골대사)

  • Recently, interests in the influences of vegetarian diet on bone mineral density after menopause have been rapidly increased. The purpose of this study was to compare the protein nutritional status and bone mineral density of postmenopausal vegetarian women with that of the omnivores. Vegetarian (n=38, seven day adventists) were chosen from the subjects in previous study, and the subjects were matched with omnivores counterparts with respect to age and BMI. Anthropometric measurements, dietary intakes, and bone mineral density (BMD) were taken. The bone metabolism related marker including urinary deoxypyridinoline and urinary pH, and serum protein and albumin concentrations were evaluated. The average age of vegetarians and omnivores were 60.7 yrs and 60.5 yrs, respectively md, there was no significant difference. The mean daily energy intake of vegetarians and omnivores were 1518.5 ㎉ (82.7% of RDA) and 1355.5 ㎉ (72.6% of RDA), respectively. The mean calcium intake of vegetarians (492.6 mg, 70.3% of RDA) was not significantly different from that of omnivores (436.6 mg, 62.3% of RDA). There was no significant difference in BMDs of spine and femoral neck between vegetarians and omnivores. Urinary deoxypyridinoline (DPD) level was not significantly different. In the vegetarians, the intakes of total protein (p<0.05) and plant protein (p<0.05) had significant negative correlations with urinary DPD. In the omnivores, serum albumin showed significant positive correlations with urinary DPD (p<0.05). In conclusion, we can not find the beneficial roles of vegetarian diet on bone mineral metabolism. For the postmenopausal vegetarian woman, protein intake would be an important factor to promote skeletal health.

Variation of Grain Traits in Landraces of Rice Collected from Vietnam (베트남 지방수집종 벼의 종실특성변이)

  • A total of 437 landraces of rice from Vietnam were analyzed for total seed protein by SDS-PAGE and phenol reaction. The different types of glutelin $\alpha$ subunits were detected. The level of wx protein with 60kDa molecular weight was divided into 3 groups, corresponding to non-glutinous, intermediate and glutinous starch types. Based on the variation in seed storage protein and wx protein, landraces were classified into 7 groups. Frequency distribution of types A and B of glutelin $\alpha$ subunits changed with the latitude at which rice landraces were collected. Geographical cline for phenol reaction was detected.

Effects of Feed Protein Quality on the Protein Metabolism of Growing Pigs - Using a Simulation Model - (성장기 돼지의 단백질대사에 사료단백질의 질이 미치는 영향 -수치모델을 사용하여-)

  • This study was conducted to describe qualitatively the protein metabolism of pigs during growth depending on the feed protein quality and to describe quantitatively amino acids requirements, using a simulation model. The used model has a non-linear structure. In the used model, the protein utilization system of a pig, which is in the non-steady-state, is described with 15 flux equations and 11 differential equations and is composed with two compartments. Protein deposition(g/day) of pigs on the 30th, 60th, 90th, and 120th day of feeding duration with three-quality protein, beginning with body weight 20kg, were calculated according to the empirical model, PAF(the product of amino acid functions) of Menke, and was used as object function for the simulation. The mean of relative difference between the simulated protein deposition and PAF calculated values, lied in a range of 8.8%. The simulated protein deposition showed different behavior according to feed protein quality. In the high-quality protein, it showed paraboloidal form with extending growth simulation up to 150eh day. So the maximum of protein deposition was acquired on the 105th day of simulate growth time and then it decreased fast. In the low-quality protein, this form of protein deposition in the course of simulated growth did not appear until 150th day. The simulated protein mass also showed a difference in accordance with feed protein quality. The difference was small on the 30th day of simulated growth, but with duration of the simulated growth it was larger. On the 150th day the simulated protein deposition of high quality protein was 1.5 times higher as compared to the low-quality protein. The simulated protein synthesis and break-down rates(g/day) in the whole body showed a parallel behavior in the course of growth, according to feed protein quality. It was found that the improvement of feed protein quality increased protein deposition in the whole body through a increase of both protein synthesis and breakdown during growth. Also protein deposition efficiency, which was calculated from simulated protein deposition and protein synthesis, showed a difference in dependence on the protein qualify of feed protein. The protein deposition efficiency was higher in pigs fed with high quality protein, especially at the simulation time 30th day. But this phenomena disappeared with growth, so on the 150th day of growth, the protein deposition of the high feed protein quality was lowest among the three different quality of feed protein. The simulated total requirement of the 10 essential amino acids for the growth of pigs was 28.1(g/100g protein), similar to NRC. The requirement of lysine was 4.2(g/100g protein).
