A Study on the Variation of the Concentrations of Carbon Species and B(a)P Associated in Atmospheric Suspended Particles in an Urban Area of Seoul (서울지역 일부 대기중 부유먼지에 함유된 탄소 성분과 Benzo(a)pyrene 성분의 농도변동 양상에 관한 연구)

  • In order to investigate the behavior of particulate carbon and benzo(a)pyrene(B(a)P) in the ambient air, suspended particle matters were collected from April, 1990 to February, 1992, and total carbon(TC), organic carbon(OC), elemental carbon(EC) and B(a)P were measured by an elemental analyzer and a HPLC. The results were as follows the average concentration of TC was 38.6 $\mug/m^3$ and its concentration was higher in winter(45.4 $\mug/m^3$) and fa11(44.3 $\mug/m^3$) than in summer(36.8 $\mug/m^3$) and spring(28.9 $\mug/m^3$). The monthly concentration trends both of EC and OC was similar, but seasonal variation of EC concentrations was larger than that of OC. The average concentration of B (a)P was 2.2 ng/m$^3$, and was higher in winter(4.1 ng/m$^3$) and fall(3.2 ng/m$^3$) than in spring(1.2 ng/m$^3$) and summer(0.6 ng/m$^3$). The seasonal behavior of carbon and B(a)P was to similar except for summer. The correlation coefficient(r) between EC and B(a)P was 0.71, and the correlation coefficient(r) between OC and B(a)P was 0.66.

Desmutagenicity of Tea Extracts from Green Tea, Oolong Tea and Black Tea (녹차, 오룡차 및 홍차 추출물의 돌연변이원성 억제작용)

  • Desmutagenicities against 2-amino-1-methyl-6-phenylimidazo[4, 5-b] pyridine(PhIP) and 2-amino-3, 8-dimethylimidazo[4, 5-f]quinoxaline(MelQx) of tea extracts (steamed green tea, roasted green tea, oolong tea and black tea) were investigated. All the fractions obtained from tea extracts showed strong desmutagenic activity against PhIP and MeIQx toward S. typhimurium TA 98 in the presence of the S-9 mix. The crude catechin fraction exhibited the strongest desmutagenic activity. Among these tea extracts, black tea especially exhibited the strongest desmutagenic activity and the activity was 70.9~91.0% against PhIP and 92.2~98.8% against MelQx at a concentration(0.5~1.0mg/plate) for drinking. The activity of authentic catechins of (-)-EGC, (-)-EGCg, (-)-ECg and (-)-EC were 79.5%, 60.2%, 46.1% and 43.5% against PhIP, and were 52.3%, 11.6%, 8.2% and 22.1% against MelQx by addition of 1.0mg/plate, respectively. The desmutagenic activity was supposedly due to the (-)-EGCg, (-)-EGC and (-)-EC in tea polyphenols, and the browning materials. The desmutagenicity was stronger when mutagens were preincubated with S-9 mix after reaciton with black tea extracts than when preincubated with them after reaction with S-9 mix. The desmutagenicity of tea extracts was rather expressed by reacting directly with mutagens than by deactivating the activated forms of mutagens.

화학독립영양미생물 Aeromonas sp. strain JS-1의 RubisCO 정제 및 특성

  • A Chemoautotroph identified as an Aeromonas sp. strain JS-1 was isolated from fresh water. Aeromonas sp. strain JS-1 used the $H_2$ and $CO_2$ as energy and carbon sources, respectively. Growth characteristics for improving the $CO_2$ fixation rate were examined in batch cultivation. Its results shown that the optimal growth appeared at culture conditions of $35^{\circ}C$, pH 7 and NaCl 0.1%(w/v). Some hydrogen-oxidizing bacteria were reported that the enzyme activity of ribulose 1,5-bisphosphate carboxylase- oxygenase (RubisCO-EC 4.1.1.39), in the key enzyme of the Calvin-Benson cycle. A RubisCO was purified from a chemoautotrophic bacterium, Aeromonas sp. strain JS-1. the enzyme was purified by ammonium sulfate precipitation, DEAE-sepharose CL-6B and gel filtration chromatography. The RubisCO showed that molecular mass was about 560KDa from gel filtration chromatography and nondenaturing PAGE, and the RubisCO was confirmed to consist of $L_8S_8$ enzyme structure by sodium dodecyl sulfate polyacrylamide gel electrophoresis. A large subunit was about 56KDa and small one was about 15kDa. The Km values of the enzyme for ribulose 1,5-bisphosphate(RUBP), $NaH^{14}CO_3$, and $Mg^{++}$ were estimated to be 0.25mM, 5.2mM, and 0.91mM, respectively. The optimum temperature for RubisCO enzymatic activity were $50^{\circ}C$, and the enzymatic activity was stable up to $45^{\circ}C$.

Physicochemical Properties and Antioxidant Activities of Steam-Dried Allium hookeri Root (증포 처리한 삼채 뿌리의 이화학적 특성 및 항산화 활성)

  • This study was carried out to investigate the effect of steam-drying method on physicochemical properties and antioxidant activities of Alliun hookeri root (AHR). Moisture, crude protein, crude fat, and crude ash contents of raw and steam-dried AHRs were 10.91~14.15%, 11.14~13.49%, 0.83~3.02%, and 7.55~8.98%, respectively. Sulfur contents of steam-dried AHRs were 2.0 and 2.2 times lower than that of raw AHR (0.51%), respectively. pH and total sugar contents of AHRs were reduced by steam-drying, whereas titrate acidity and browning intensity were increased. The L and b values of AHRs in Hunter's value were also reduced, but a value was increased by steam-drying. Among hot water extracts from raw and steam-dried AHRs, four times steam-drying showed the lowest $EC_{50}$ values (0.44, 9.01, and 0.48 mg/mL, respectively) in DPPH radical assay, ABTS radical assay, and reducing power, whereas four times steam-drying had the highest total phenolic content ($34.47{\mu}g/mg$) and browning intensity (2.05 and 0.20 at 280 and 420 nm, respectively). The antioxidant activities of hot water extracts from raw and steam-dried AHRs were closely correlated with their total phenolic contents and browning intensity, showing coefficient of determination ($R^2$) values higher than 0.87. From the results, we suggest that steam-drying method could be used as an effective process for increasing the antioxidant activity of AHR.

Impact of Physicochemical Properties of Root Substrates on Growth of Mother Plants and Occurence of Daughter Plants in 'Seolhyang' Strawberry Propagation through Bag Culture ('설향' 딸기 번식을 위한 자루재배시 상토의 물리·화학성이 모주 생육과 자묘 발생에 미치는 영향)

  • The influence of physicochemical properties of root substrates on the growth of mother plants and occurrence of daughter plants in 'Seolhyang' strawberry propagation were investigated through plastic bag cultivation. Six different formulations of root substrates were coir dust + perlite (5:5, A), coir dust + perlite (6:4, B), coir dust + perlite (7:3, C), coir dust + coconut chip (7:3, D), coir dust + coconut chip (6:4, E), and peatmoss + vermiculite (5:5, v/v; F). The total porosities (TP) and container capacities (CC) of all root substrates were higher than 85% and 55%, respectively, indicating that all substrates were in the acceptable range. But the TP and CC of F substrate were 91.5% and 60%, respectively, which were the highest among the root substrates tested. In the soil chemical properties analyzed before planting and after harvesting of 'Seolhyang' strawberry mother plants, the root substrates of A, B, C, and F had higher electrical conductivity and $NO_3$-N concentrations than those of D and F. The root substrates of A, B, C, and F had heavier runner fresh and dry weights, longer runner lengths, and more daughter plant occurrence than those of D and F. The treatment F had higher tissue N content than any other treatments at 120 days after the transplanting of 'Seolhyang' strawberry and statistical differences were not observed among remained 5 substrates. The treatment of F also had the higher tissue contents of other nutrients except N analyzed at 120 days after transplanting. These results indicated that soil chemical properties rather than physical properties severely influenced the growth of runners and occurrence of daughter plants.

  • The investigation and the analysis of soil chemical properties, microbial flora and cultural practices in plastic house of 60 farms (pepper 20, cucumber 20, oriental melon 20) were conducted in Chonnam area film. An amount of fertilizer application of farms was more than that of standard fertilizer application. That was phosphate at two and a half times, potassium at twice and manure at three times the amount of standard application. It was inferred that excessive fertilizer application would lead to salts accumulation in soil. Continuous cropping injury would be incurred by excesive salts accumulation in soil. Even if inorganic elements in soil culturing peppers, cucumbers and oriental melons were greater than optimum level, base-saturation degree of soil culturing peppers, cucumbers and oriental melons was 40:18:12, 55:16:14 and 40:27:11 respectively. That was lower than the optimum levels (60:15:5). Therefore, crops would be affected by uptake inhibition by nutrition imbalance. Density of Fusarium is related to plant decease in soil i.e. pepper $26.2{\times}10^2\;cfu\;g^{-1}$, cucumber $75.8{\times}10^2\;cfu\;g^{-1}$ and oriental melon $6.5{\times}10^2\;cfu\;g^{-1}$. B/F of soil planted with pepper, cucumber and oriental melon was 91.2, 80.4 and 18.8, respectively.

Developmental characteristics of Hemiptarsenus sp.(Hymenoptera : Eulophidae), a parasitoid of Liriomyza trifolii (Burgess) and effect of the insectcides (아메리카잎굴파리 기생봉, Hemiptarsenus sp.의 발육특성 및 살충제의 영향)

  • The study was conducted to investigate the developmental periods and effect of several insecticides on Hemiptarsenus sp., ectoparasitoid of Liriomyza trifolii. The mean length and width of egg were 0.5mm and 0.1mm. The mean length of larva, pupae, abult female, and abult male were 1.9mm, 2.0mm, 2.2mm, and 1.8mm, respectively. Developmental periods of Hemiptarsenus sp. from egg to larva at 15, 20, 25, $30^{\circ}C$ were 16.9, 8.8, 5.9, and 4.5 days, and those of pupa were 20.7, 9.7, 5.6, and 3.4 days, respectively. Based on these results, developmental threshold temperatures and effective temperatures were $9.5^{\circ}C$, 91.5 degree-days in egg-larval stage, $10.9^{\circ}C$, 142.3 degree-days in pupal stage. When several insecticides were evaluated to Hemiptarsenus sp. at the recommended concentrations, B.t WP, diflubenzuron WP, and cyromazin were negligiblly effective all life stages. Fipronil SC, cartap SP, spinosad GW were less toxic to larva and pupa, but highly toxic to adults. Abamectin EC was less toxic to all life stages, but inhibited oviposition of 50% more to Hemiptarsenus sp. female.

Leaf Mineral Contents and Growth Characteristics of Strawberry Grown in Aquaponic System with Different Growing Media in a Plant Factory (식물공장형 아쿠아포닉스 시스템에서 배지 종류에 따른 딸기 잎의 무기이온 함량과 생육 특성)

  • This study was aimed to determine the effects of grow media on the mineral contents of the leaves and growth characteristics of strawberry grown under aquaponics system in a plant factory. For aquaculture, 12 fish (Cyprinus carpio) (total weight, 2.0 kg) were raised in an aquaponics tank (W 0.7 m × L 1.5 m × H 0.45 m, 472.5 L) filled with 367.5 L of water at a density of 5.44 kg·m-3 and total 34 of strawberry seedlings were transplanted in the pots filed with 200 g of orchid stone, hydroball or polyurethane sponge in the growing bed (W 0.7 m × L 1.5 m × H 0.22 m) laid out with holly acrylic sheet (140×60 mm, Ø80) on the top of the system. The pH and EC of the aquaponic solution was ranged from 7.6 to 4.9 and 0.24-0.91 dS·m-1, respectively. The concentration of NO3-N was about 28% lower than that of the hydroponic standard solution, and K, Fe and B were 10, 27 and 3.8 times lower, respectively; however, the mineral contents of strawberry leaves were in the appropriate ranges with lower contents in the leaves grown with sponge media. The organic content (OM), nitrogen (N), phosphorus (P), and potassium (K) of the sludge were 61.5, 5.72, 8.92, and 0.24%, respectively. The leaf area, leaf number, and dry and fresh weights of shoot at 81 DAT were significantly higher in the hydroball, and the average number of fruits per plant was significantly higher in both the orchid stone and hydroball. There was no significant difference in the fresh and dry weights of fruits. Integrated all the results suggest that the orchid stone and hydroball media are more effective to utilize nutrients in solid particles of aquaponic solution, compared to the polyurethane sponge.
