Anti-platelet Aggregation Effect of Cheongpyesagan-tang In Vitro (청폐사간탕(淸肺瀉肝湯)의 혈소판 응집억제 작용에 대한 in vitro 연구)

  • Objective : The study was designed to test the anti-platelet effect of the extract Cheongpyesagan-tang and compare it with aspirin in vitro. Methods : The extract from Cheongpyesagan-tang was made by the pharmacy department of Kyung Hee Oriental Medical Hospital. The extract was investigated for inhibition against the collagen induced aggregation of human platelet suspensions on aggregometry. Aspirin and aspirin-Cheongpyesagan-tang were investigated together. Results : 1. In collagen induced human platelet aggregation test, the extract from Cheongpyesagan-tang significantly inhibited in concentration 30mg/ml (p<0.05), 40mg/ml, 50mg/ml (p<0.001) and the effect depended on concentration over 20mg/ml. 2. Aspirin and aspirin-Cheongpyesagan-tang inhibited collagen induced human platelet aggregation significantly (p<0.001). Aspirin-extract of Cheongpyesagan-tang inhibition rate was higher than aspirin only (p<0.05). Conclusions : The extract of Cheongpyesagan-tang has anti-platelet aggregation and synergic effect with aspirin on human platelet in vitro.

  • Object : This study was designed to assess effect of Chungpesagan-Tang and herbs on Mouse neuroblastoma 2a cells damaged by hypoxia-reoxygenation. Method : Mouse neuroblastoma 2a (N2a) cells were measured by MTT assay and LDH assay after 48h hypoxia and 6h reoxygenation. Mouse neuroblastoma 2a (N2a) cells were treated by Chungpesagan-Tang and herbs. Result : In MTT assay of hypoxia all of herbs were almost ineffective and Hubak was a little effective. 2. In MTT assay of reoxygenation most of herbs were not effective. But Hubak was some effective. 3. In LDH assay of hypoxia all of herbs were effective. Especially Chungpesagan-Tang were equally effective on all of concentration. 4. In LDH assay of reoxygenation all of herbs were generally effective. Especially Chungpesagan-Tang and Baekji were highly effective and Kilkyung was also effective on low concentration. 5, The herbs were generally effective on LDH assay of hypoxia and reoxygenation. Conclusion : The results suggest that Chungpesagan-Tang and all of herbs may have protective effect on condition of oxidative stress and can be applied on the development of a new medicine for neurodegenerative disease like dementia.

Effects of Chungpyesagantang on the Diabetic Rats induced by Streptozotocin (청폐사간탕(淸肺瀉肝湯)이 Streptozotocin으로 유발(誘發)된 흰쥐의 실험적(實驗的) 당뇨(糖尿)에 미치는 영향(影響))

  • In order to study the effects of Chungpyesagantang(淸肺瀉肝湯) on the diabetic rats induced by streptozotocin, during 15 days rats were administered Chungpyesagantang extract after streptozotocin injection (50mg/kg) (sample group). On 2nd, 9th, and 15th day, I investigated the levels of body weight, serum glucose, serum total cholestrol, serum triglyceride. The last day, I killed rats and investigated hepatic lipid peroxidase. The results were obtained as follows: 1. Body weight of the diabetic rats induced by streptozotocin and Chungpyesagantang, as compared with the control group on the 15th day, increased effectively. (p〈0.05 respectively) 2. Glucose levels in serum of the diabetic rats induced by streptozotoc in and Chungpyesagantang, as compared with the control group on the 9th and 15th day, decreased effectively.(p〈0.05 respectively) 3. Total cholesterol levels in serum of the diabetic rats induced by streptozotocin and Chungpyesagantang, as compared with the control group on the 9th and 15th day, decreased effectively.(p〈0.05 respectively ) 4. Triglyceride levels in serum of the diabetic rats induced by streptozotocin and Chungpyesagantang, as compared with the control group on the 9th and 15th day, decreased effectively.(p〈0.05 respectively) 5. On hepatic lipid peroxidethe of the diabetic rats induced by streptozotocin and Chungpyesagantang, as compared with the control group on the 9th and 15th day, decreased effectively.(p〈0.01 respectively)

Clinical Study on the Utilization of Yeoldahanso-tang and Cheongpyesagan-tang (열다한소탕(熱多寒少湯)과 청폐사간탕(淸肺瀉肝湯)의 활용에 대한 임상적 연구)

  • Objective: The purpose of this study is to examine the characteristics and the improvement in the symptoms of the patients who showed improvement after the administration of Yeoldahanso-tang and Cheongpyesagan-tang. Methods: The patients whose Sasang Constitution were examined by an expert at six Oriental medical hospitals and three regional Oriental-medicine clinics, 89 showed a predetermined symptom improvement when Yeoldahanso-tang and Cheongpysagan-tang were administered to them from November 2007 to July 2008. The traits of the patients and the effects of their treatment were examined. Results and Conclusions: 1. There were about as many male patients as there were female patients, but 21.3% of the patients who were female and who were in their 40's showed the most improvement, which makes them the biggest category in this study. 2. The main syndromes in which the patients showed improvement were hemi-paralysis, vertigo, and headache, and the ordinary symptoms in which the patients showed improvement were diarrhea, fatigue, and dry mouth. 3. The average food intake of the patients was highest and their feces had intermediate hardness (semi-soft). 4. The amount of the patients' sweat was either high(38.2%) or appropriate(38.2%) and they showed a dislike of heat over coldness. 5. Among the diseases that the patients previously had, high blood pressure. The above results show that Yeoldahanso-tang and Cheongpyesagan-tang can be considered beneficial to females in their 40's and to stroke patients whose food intake is normal, whose sweat level is either copious or appropriate, who dislike heat and who are suffering from high blood pressure.

Effect of Chungpaesagan-tang on cerebral Ischemic Damage Induced by MCAO in Rats (청폐사간탕(淸肺潟肝湯)이 MCAO에 의한 흰쥐의 뇌허혈 손상에 미치는 영향)

  • This study aimed to validate neuroprotective effect of Chungpaesagan-tang on the early stage of cerebral ischemic damage. Cerebral ischemic damage was induced by the middle cerebral artery occlusion (MCAO) for 2 hours in the Sprague-Dawley rats. Water extract of Chungpaesagan-tang(8.7g/kg) was administered orally twice at 1 and 4 hours after the MCAO. Neurological score was tested at 3 and 24 hours after the MCAO and Chungpaesagan-tang administration. At 24 hours after the MCAO, infarct volume and edema ratio was evaluated with the TTC staining. Apoptotic cell death in cerebral cortex and caudate putamen was observed with cresyl violet staining and TUNEL labeling. Bax expression in the MCAO rat brain was stained with immunohistochemistry. Chungpaesagan-tang improved neurological and behavioral impairment of the MCAO rats and reduced infarct area, infarct volume and brain edema formation. Chungpaesagan-tang attenuated cell death percentage in cortex penumbra and reduced TUNEL positive cells in cortex penumbra and in caudate putamen of the MCAO rats. Chungpaesagan-tang reduced Bax positive neurons in caudate putamen and reduced c-Fos positive neurons in cortex penumbra of the MCAO rats. Chungpaesagan-tang intensified neuronal HSP72 expression in cortex penumbra of the MCAO rats. In results, Chunpaesagan-tang reduces infarct volume and edema formation through anti-apoptotic effect. This result suggests that Chunapaesagan-tang has an adequate neuroprotective effect on the early stage of cerebral ischemic damage.

An Experiment of the Purgative Action of Rhei Rhizoma and Chungpesagan-tang (청폐사간탕(淸肺瀉肝湯)과 대황(大黃)의 사하효과(瀉下效果)에 관한 실험적 연구)

  • Objectives: This study was conducted to analyze scientifically the purgative action of Chungpesagan-tang, which was widely used for stroke In Oriental Medicine. Methods: The contents of sennoside A on the water-extraction of Rhei Rhizoma, the transforming activity of sennoside A to rheinanthrone by human intestinal bacteria, and the anti-costive activity of Chungpesagan-tang, Rhei Rhizoma and Yuldahanso-tang on mice were investigated. Results: Rhei Rhizoma (Rheum palmatum, from China) had dose-dependent potent purgative action. Chungpesagan-tang containing this Rhei Rhizoma had more potent purgative action than Rhei Rhizoma only. However, Korean cultivated Rhei Rhizoma (Rheum undulatum) and Chungpesagan-tang containing this Korean cultivated Rhei Rhizoma had no purgative action. Conclusions: These results suggest that Chungpesagan-tang polyprescription should be considered to enhance the purgative action of Rhei Rhizoma.

  • 1. PURPOSE : The purpose of this study is to investigate the effect of Chungpyesagantang on LPS induced Arthritis in Mice. 2. METHOD : All the BALB/C Mice used in this study were 4wks of age at the start of the experiment. The experimental model of Arthritis was induced by injectection of $300{\mu}g/kg$ LPS in mice knee joint. The experiment was compare daily CS treatment group after Arthritis elicitation with Arthritis elicitated group at day 4, 7, 14 after Arthritis elicitation. 3. RESULTS 1) The hyperplasia of synoviocytes of CS treatment group after Arthritis elicitation is soften than Arthritis elicitated group. 2) The aggregation of collagen fibers CS treatment group after Arthritis elicitation is decreased than Arthritis elicitated group. 3) The distribution of TUNEL positive cells(apoptotic cell) of CS treatment group was remarkably increased than Arthritis elicitated group. 4) The distribution of $TNF-{\alpha}$, $NF-{\kappa}B\;p50$, COX-2 positive cells of CS treatment group after Arthritis elicitation in synovial membrane was decreased than Arthritis elicitated group. 5) The distribution of $IL-2R-{\alpha}$, ICAM-1 positive cells of CS treatment group after Arthritis elicitation in apical surface of synovial membrane was decreased than Arthritis elicitated group. 6) The distribution of $NF-{\kappa}B\;p50$, $IL-2R-{\alpha}$ in common iliac lymph node of CS treatment group after Arthritis elicitation positive cells was decreased than Arthritis elicitated group. 4. CONCLUSION : As a result of these experimental results, it may be concluded that Chungpyesagantang used for treatment of LPS induced Arthritis in Mice. Inflamation activity in CS treatment group after Arthritis elicitation was decreased than Arthritis elicitated group.

Anti-oxidative Effect of Chungpyesagan-tang in LPS Induced RAW 264.7 Cells (LPS로 유도된 RAW 264.7 세포에서 청폐사간탕(淸肺瀉肝湯)의 항산화 효과)

  • Objectives : This study was conducted to confirm the anti-oxidative effect of Chungpyesagan-tang(CPSGT) extract. Methods : In this study, MTT assay was performed to confirm cell viability, and DPPH and ABTS were performed to confirm radical scavenging ability. The ROS scavenging ability and the protective effect against DNA damage were confirmed by 2,7-dichlorofluorescin diacetate(DCF-DA) and 4',6-diamidino-2-phenylindole(DAPI) staining and comet assay. mRNA expression of Heme oxygenase-1(HO-1) was measured by real-time PCR, and expression of HO-1 and Kelch-like ECH-associated protein 1(Keap1) proteins was measured by western blot. Results : CPSGT was not cytotoxic at 50-400㎍/㎖. The radical scavenging activity was increased, and the ROS scavenging activity and the protective effect against DNA damage were increased compared to the LPS-treated group. The mRNA expression and protein expression of HO-1 were increased in a concentration-dependent manner. The protein expression level of Keap1 was decreased in a concentration-dependent manner. Conclusion : This suggests that CPSGT has an antioxidant effect and can be used as a potential material for skin diseases.

Effects of Cheongpyesagan-tang and YKK012 on in vitro and in vivo Colon Cancer Cell Growth with and without CPT-11 (청폐사간탕(淸肺瀉肝湯)과 YKK012의 항암제 CPT-11과 병용투여 시 대장암 성장억제에 미치는 효과)

  • Objectives : The aim of this study was to evaluate the antitumor effects of Cheongpyesagan-tang(CST) and YKK012 on colon cancer. Methods : MTT assay was used to evaluate the cytotoxicity of Single herbs and combinations of CST and YKK012 on murine colon cancer cells, Colon 38. To explain effects of apoptosis in colon cancer, we performed the western blot. Effects of CST and YKK012 on antitumor activity of CPT-11 using the murine colon38 allograft tumor in BDF1 mice. Results : Single herbs and combinations of CST and YKK012 was tested in vitro, Rhei Radix (RH) and Scutellariae Radix (SC) and YKK012 showed dose-response cytotoxicity on Colon 38. This might be due to the apoptosis, as we see Bax and Caspase-3, which are apoptotic factors, was expressed in RH and SC treated cells. YKK012 also showed increased expression of Caspase-3. In mouse colorectal cancer xenograft model of colon38 cells, herbal combinations showed tendencies of tumor regression, but was not significant. Furthermore, because toxicity was observed in CST group, we reduced the dose of CST for the next experiment. The anti-tumor effects of herbal combinations were insufficient to be used as single anti-tumor agent. With simultaneous usage of CPT-11, contrary to that CST showed no synergistic effects, YKK012 which was composed by the combination of four $ER{\beta}$ selective herbs, significantly reduced the size of tumor and Bax expression was increased. Conclusions : We suggest YKK012 can be a effective cancer adjuvant therapy with CPT-11 on colon cancer.

Effects of Chungpyesagan-tang on arterial stiffness and pulse pressure in acute stroke patients (청폐사간탕(淸肺瀉肝湯)이 급성기 중풍환자의 동맥경직도 및 맥압에 미치는 영향)

  • Objectives : Arterial stiffness and pulse pressure are related to cardiovascular and cerebrovascular survival and longevity. This study is aimed at examining the effects of Chungpyesagan-tang on arterial stiffness and Pulse pressure in acute stroke Patients. Methods: The subject of this study was acute strike Patients within 1 week after ictus, with Cardio-ankle vascular index(CAVI) higher than 9.0. They were divided into two groups: A treatment group (n=44) and a control group(n=46). For two weeks, Chungpyesagan-tang was given to the former, other herbal medicines to the latter. used for stroke patients for the control group for 2 weeks. At the end of first and second week, CAVI, pulse Pressure, National Institute of Health stroke scale(NIHSS), Modified Barthel Index(MBI) were measured. Serum lipid Profile, aspartate transaminase(AST), alanine transaminase(ALT). blood urea nitrogen(BUN), creatinine were also measured at the end of the study. Results : After 2 weeks, CAVI and Pulse Pressure in Chungpyesagan-tane group were significantly tower than those in the control group(P<0.05). NIHSS and MBI were improved in both groups. But there was no significant difference between the treatment group and the control group in terms of the NIHSS and MBI. Conclusions : We suggest Chungpyesagan-tanghas desirable effects on arterial stiffness and Pulse Pressure of acute stroke patients. It can improve morbidity and mortality of patients on the basis of influencing vascular stiffness and increased pulse pressure.
