Effect of Cnidium officinale Extract on Recovery Capability of Allergic Contact-Dermatitis in Rat (흰쥐의 알레르기성 접촉피부염의 회복에 미치는 천궁추출물의 영향)

  • The epidermal recovering-effects of the Cnidium officinale extract after inducing contact-dermatitis by 1-chloro-2,4-dinitrochlorobenzene (DNCB) showed by measuring thickness of skin, identifying IgE amounts in plasma, and activities of anti-oxidant enzymes using histochemical and biochemical methods. By three-time applications of the extract, morphological changes of dermatitis in impaired region were recovered dramatically and shape of skin surface and thickness of epidermis were restored to be normal. Also, lipid content was recovered to the level of normal state. We suggested that extract treatment lowered a hypersensitive reaction by decreasing of IgE level in blood and restored activities of superoxide dismutase and catalase. In conclusion, we proposed that C. officinale extract might be used as natural resources for treating effectively allergic contact-dermatitis.

Volatile Flavor Components and Free Radical Scavenging Activity of Cnidium officinale (천궁(Cnidium officinale)의 휘발성 향기성분 및 유리기 소거활성)

  • This study was performed to develop natural spices and functional foods using Cheongung (Cnidium officinale) which is one of the Korean medicinal plants. The volatile flavor patterns of Cnidium officinale were detected by electronic nose with 6 metal oxide sensors, and the principal component analysis was carried out. The volatile flavor components of Cnidium officinale were isolated by simultaneous steam-distillation extraction with pentane and diethylether (1 : 1), and essential oils were analyzed by capillary GC and GC/MS. The free radical scavenging activity of ethanol and methanol extracts from Cnidium officinale was measured by using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and compared with ${\alpha}-tocopherol$ as reference. The principal component analysis showed the difference of principal components between fresh and drying samples. Eighty-five volatile flavor components (643.64 ppm) from fresh Cnidium officinale were identified and the major components were butyl phthalide, sabinene, neocnidilide. Sixty-four volatile flavor components (218.15 ppm) from hot air dried one were identified and the major components were butyl phthalide, sabinene, 3-N-butyl phthalide. And 73 volatile flavor components (784.15 ppm) from freeze dried one were identified and the major components were butyl phthalide, sabinene, ${\beta}-selinene$. The free radical scavenging activity of methanol cold extract (500 ppm) of freeze dried Cnidium officinale was higher than other samples. And methanol and ethanol cold extracts (above 250 ppm) of freeze dried sample were higher than ${\alpha}-tocopherol$ $25\;{\mu}M$ (22.34%).

Influence of shading and polyethylen vinyl mulching on growth and yield of Cinidium officinale Makino (차광(遮光), 비닐피복(被覆)이 궁궁이(천궁(川芎))의 생육(生育) 및 수량(收量)에 미치는 영향(影響))

  • The study was carried out to elucidate the influenced of shading and mulching with polyethylen vinyl on growth and yield of Cinidium officinale Makino, and improve the cultivation method of those medicinal plants in northern area of Gyeong-buk province, Korea. Top fresh weight of Cinidium officinale Makino was increased by both 35 % and 55% shading compared to the non-shadingand decreased by 75 % shading, but that of Ligusticum chuanxing Hart. Was decreased by the shading tested The summer wilt of Cinidium officinale Makino severely occurred in non-shading but greatly reduced by 55 % or 75 %shading, while no difference was observed in Ligusticum chuanxing Hort, between the shading and the non-shading. Dry root yield Cinidium officinale Makino was increased by 17 % and 19 % in 35 % and 55 % shading levels, respectively, compared to the dry root yield(254kg/10a) in non-shading. However, it was decreased in Ligusticum chuaxing Hort. by the shading. Emergence ratio of Cinidium officinale Makino was increased by black polyethylen vinyl mulch but decreased by transparent polyethylen vinyl mulch, compard to non-mulch. Polyethylen vinyl mulch increa­sed the top fresh weight of Cinidium officinale Makino Especialy black polythylen vinyl mulch was very effective on increasing the top fresh weight. Dry root yield of Cinidium officinale Makino was increased by 32 % and 30 %, respectively, compared to the non-shading.

EFFECT OF METHANOL EXTRACT OF CNIDII RHIZOMA ON THE FUNCTION OF RECEPTORS FOR GABA AND GLYCINE (천궁(Cnidii Rhizoma)의 메탄올 추출물이 GABA 및 Glycine 수용체에 미치는 영향)

  • Cnidii Rhizoma (CR) was subjected to extraction with 70% methanol and tested to determine whether it has anxiolytic activity in mouse by employing staircase and rotarod tests. In addition, to understand the mechanism of anxiolytic action, CR, picrotoxin, yohimbine, isoniazid and strychnine were utilized to deliniate the potential involvement of GABA and glycine receptors in the action of Cnidii Rhizoma. To gain insights into the safety of Cnidii Rhizoma extract, behavioral and MTT tests were carried out. The results were obtained as follows: 1. CR extract had little effect on climbing numbers in the stair case test. 2. CR extract had considerable anti-anxiety effects as evidenced by the reduction of rearing numbers in the stair case test. 3. CR extract had little effect on muscle relaxation. 4. Anxiolytic actions of CR extract appeared to be mediated by glycine receptor activation. 5. Cytotoxicity in the neuronal cell was not observed and no strange behaviors were found. In short, these results indicate that CR extract has the ability to exert anxiolytic activity, possibly by activating glycine receptor with little side effects in mouse.

Increased Antioxidative Activity of Fermented Ligusticum striatum Makino Ethanol Extract by Bioconversion using Lactobacillus plantarum BHN-LAB 129 (Lactobacillus plantarum BHN-LAB 129의 생물전환공정을 통한 천궁 발효 추출물의 항산화 활성 증대)

  • Phytochemical compounds of Ligusticum striatum Makino are used as traditional medicinal herbs in Asia. These compounds are reported to have pain relief and antioxidant activities in gynecological and brain diseases. In this study, we investigated the antioxidant effects of Ligusticum fermented ethanol extract from Lactobacillus plantarum BHN-LAB 129 isolated from Kimchi, a Korean traditional food. The total polyphenol and total flavonoid contents increased by about 116.2% and 281.0% respectively, in the fermented Ligusticum extract as compared with those in the nonfermented Ligusticum ethanol extract. Superoxide dismutase-like (SOD), DPPH radical scavenging, ABTS radical scavenging, and reducing power activities increased by around 139.9%, 199.6%, 301.0%, and 137.1%, respectively, in the fermented Ligusticum ethanol extract as compared with these parameters in the nonfermented Ligusticum ethanol extract, respectively. In conclusion, the fermented Ligusticum ethanol extract with L. plantarum BHN-LAB 129 was effective in increasing the antioxidant effects. The bioconversion process in this study points to the potential of using Ligusticum to produce phytochemical-enriched natural antioxidant agents with high added value. The findings may prove useful in the development of improved foods and cosmetic materials.

A Study on the Antioxidant Activity and Phenolic Compound Content of Cnidium officinale Makino Cultivated in a Temperature and Carbon Dioxide-Controlled Environment (온도 및 이산화탄소 조절 환경에서 재배한 천궁(Cnidium officinale Makino)의 항산화 활성 및 페놀 화합물 함량 연구)

  • This study aimed to investigate the growth parameters and antioxidant activity of Cnidium officinale under controlled temperature and carbon dioxide levels during the cultivation period. The plants were cultivated for four months, each group being set at the average temperature of the cultivation area +1.8℃/445ppm(SSP1), +3.6℃/872ppm(SSP3), and +4.4℃/1,142ppm(SSP5), respectively. During the cultivation period, the growth, Top/Root ratio, and leaf weight ratio(LWR) of C. officinale slightly decreased in SSP3 and SSP5 compared to SSP1, while the root weight ratio(RWR) increased. The antioxidant activity and related phenolic compound content in the aerial parts of C. officinale increased proportionally with temperature and CO2 concentration. However, an adverse effect was observed in the high-concentration SSP5 group. Conversely, in the roots, the SSP5 group exhibited the highest antioxidant activity. This study suggests that it can be utilized as fundamental data necessary for understanding the correlation between environmental conditions and the growth as well as physiological activities of medicinal plants.

Discovering the Anti-cancer Effects of Ligusticum Chuanxiong through Network-based Pharmacology Analysis and Molecular Docking: An Inquiry into Natural Products (네트워크 기반 약리학 분석 및 분자 도킹을 통한 천궁의 항암 효과 예측: 천연물에 대한 탐구)

  • In some cases of head and neck cancers (HNC), surgical interventions may result in the loss of organs and/or changes to their functions, thereby significantly affecting the patient's quality of life. As a result, the surgical treatment of HNC patients is often limited to specific cases, and alternative treatment modalities, such as chemotherapy, are considered. However, serious adverse effects caused by chemotherapy, such as severe nausea and vomiting, necessitate the need for the development of adjunctive methods to minimize patient suffering. Chuanxiong, Ligusticum chuanxiong (L. chuanxiong), is a natural herb used in Eastern medicine to treat cerebrovascular disorders and headaches. This study aimed to predict the effect and potential of L. chuanxiong as an auxiliary anticancer drug through network-based pharmacology and molecular docking analysis. The study results showed that 40 out of 41 genes of L. chuanxiong shared common targets of HNC and their proteins could be used to target HNC cells to prevent cancer progression. The results of the functional enrichment analysis confirmed that L. chuanxiong is associated with the neuroactive-ligand metabolism and neurotransmitter pathways, indicating its potential medicinal value as an adjuvant in HNC treatment. Lastly, our findings demonstrated that the active ingredient of L. chuanxiong, (Z)-Ligustilide, has the ATP binding site of heat shock protein 90, a protein known to promote the activation of cancer cells. These results suggest that L. chuanxiong is a promising candidate for developing auxiliary anticancer drugs, and further research could potentially lead to the discovery of newer and safer anti-cancer agents.

Screening for Antagonistic Natural Materials Against Alternaria alternata (Alternaria alternata에 항균력이 있는 천연물 조사)

  • 79종의 식물재료의 Alternaria alternata에 대한 항균활성을 조사한 결과, 7가지 식물재료가 항균활성이 있었고, 11가지는 확실치 않았으며, 나머지 61가지는 항균활성이 나타나지 않았다. A. alternata에 항균활성이 나타난 7가지 식물재료 중 계피가 활성이 가장 높았으며, 오미지와 천궁이 중정도의 활성을 보였으며, 나머지는 약한 활성을 나타내었다. 항균 활성 성분은 계피와 천궁의 경우는 diethyl ether에 분리되었고, 오미자의 경우는 butanol에 분리되었다. 계피의 diethyl ether 추출물은 1,000 배액 이하에서 식별 가능한 항균활성이 나타났다. 계피의 성분으로 cinnam aldehyde가 가장 높은 활성을 보여, 이 물질이 계피의 주요 항균물질로 사료된다.
